| 2 | First six months quality assessment of HY-2A SCAT wind products using in situ measurements显示文摘The first Chinese microwave ocean environment satellite HY-2A,carrying a Ku-band scatteromenter(SCAT),was successfully launched in August 2011.The first quality assessment of HY-2A SCAT wind products is presented through the comparison of the first 6 months operationally released SCAT products with in situ data.The in situ winds from the National Data Buoy Center(NDBC)buoys,R/V Polarstern,Aurora Australis,Roger Revelle and PY30-1 oil platform,were converted to the 10 m equivalent neutral winds.The temporal and spatial differences between the HY-2A SCAT and the in situ observations were limited to less than 5min and 12.5 km.For HY-2A SCAT wind speed products,the comparison and analysis using the NDBC buoys yield a bias of–0.49 m/s,a root mean square error(RMSE)of 1.3 m/s and an increase negative bias with increasing wind speed observation above 3 m/s.Although less accurate of HY-2A SCAT wind direction at low winds,the RMSE of 19.19 with a bias of 0.92 is found for wind speeds higher than 3 m/s.These results are found consistent with those from R/Vs and oil platform comparisons.Moreover,the NDBC buoy comparison results also suggest that the accuracy of HY-2A SCAT winds is consistent over the first half year of 2012.The encouraging assessment results over the first 6 months show that wind products from HY-2A SCAT will be useful for scientific community. | WANG He ZHU Jianhua LIN Mingsen HUANG Xiaoqi ZHAO Yili CHEN Chuntao ZHANG Youguang PENG Hailong | 2013 | Acta Oceanologica Sinica2013,32,11: | 10 |

| 5 | Biomarker evidence for changes in terrestrial organic matter input into the Yellow Sea mud area during the Holocene显示文摘We present lipid biomarker records of two cores(ZY1 and ZY3) from the central South Yellow Sea mud area to investigate the changes in sources and transport processes of the sedimentary organic matter(OM) throughout the Holocene. Based on the analysis of marine biomarker content(SPB(Phytoplankton Biomarker, total content of brassicasterol, dinosterol and C_(37)-alkenones) and crenarchaeol), and terrestrial biomarkers(Sn-alkanols and br GDGTs) as well as TMBR′ and BIT index values, the marine organic matter(MOM) and terrestrial organic matter(TOM) deposition history was reconstructed. Changes in TOM and MOM were related to variations in land vegetation density and marine productivity, as well as transport processes dominated by the oceanic circulation system. The marine biomarker contents from the South Yellow Sea have generally increased throughout the Holocene, indicating that the increased MOM contents were mainly controlled by the strengthening of the circulation system. The terrestrial biomarkers, on the other hand, were more variable, indicating more complex influence of TOM burial in the Yellow Sea. During the Early Holocene(7200–6000 cal yr BP), the moderate TOM input revealed by the terrestrial proxy records may result from abundant land source supply by strong river transport despite the lack of transport via circulation system. The Mid-Holocene(6000–3000 cal yr BP) was characterized by decreased terrestrial biomarker contents. The balance between the decrease in land source supply and increase of transportation by the current system of the TOM resulted in the lower but stable contents of TOM. | 7 | Acetylglutamate kinase is required for both gametophyte function and embryo development in Arabidopsis thaliana显示文摘The specific functions of the genes encoding arginine biosynthesis enzymes in plants are not well characterized. We report the isolation and characterization of Arabidopsis thaliana N-acetylglutamate kinase(NAGK), which catalyzes the second step of arginine biosynthesis. NAGK is a plastid-localized protein and is expressed during most developmental processes in Arabidopsis. Heterologous expression of the Arabidopsis NAGK gene in a NAGK-deficient Escherichia coli strain fully restores bacterial growth on arginine-deficient medium. nagk mutant pollen tubes grow more slowly than wild type pollen tubes and the phenotype is restored by either specifically through complementation by NAGK in pollen, or exogenous supplementation of arginine. nagk female gametophytes are defective in micropylar pollen tube guidance due to the fact that female gametophyte cell fate specification was specifically affected. Expression of NAGK in synergid cells rescues the defect of nagk female gametophytes. Lossof-function of NAGK results in Arabidopsis embryos not developing beyond the four-celled embryo stage. The embryo-defective phenotype in nagk/NAGK plants cannot be rescued by watering nagk/NAGK plants with arginine or ornithine supplementation. | 9 | Selection of culture conditions for callus induction and proliferation by somatic embryogenesis of Pinus koraiensis显示文摘The induction and proliferation of embryogenic callus are key steps for large-scale propagation of somatic embryogenesis pathway and long-term preservation of coniferous germplasm.Callus can be induced from immature embryos of Korean pine(Pinus koraiensis Sieb.et Zucc.;Pinaceae)as explants,but there are problems,such as low proliferation efficiency,loss of embryogenicity,poor vigor;thus,best conditions for proliferation and culture of immature embryos of Korean pine are not yet clear.To solve the problems with somatic embryogenesis of Korean pine and determine the best culture conditions for callus induction and proliferation,we varied hormone concentration,subculture cycle of proliferation and other plant growth regulators combinations in media to induce callus formation by megagametophytes of three Korean pine families at different developmental stages,then analyzed the effects on embryogenic callus retention and cell proliferation using a quadratic regression orthogonal rotation design.The results showed that the family origin and collection date of explants significantly affected callus induction(induction rate reached 1.67%).Embryogenic maintenance and callus proliferation were best on DCR medium supplemented with 0.25 mg L^(−1)6-benzyl adenine,1 mg L^(−1)naphthaleneacetic acid,30 g L^(−1)sucrose,500 mg L^(−1),L-glutamine,500 mg L^(−1)casein hydrolysis and 6.5 g L^(−1)agar.In addition,the combination of 2,4-dichlorophenoxyacetic acid+6-benzyl adenine also had a better proliferative effect on callus.The effects of different combinations of growth regulators on callus proliferation efficiency were significantly different.Transfer to new medium every 13–15 days not only maintained robust callus vigor,but also yielded a larger proliferation coefficient.The techniques and conditions for embryogenic callus induction and proliferation of Korean determined here will serve as a foundation for establishing a large-scale system for somatic embryogenesis and propagation of Korean pine. | Fang Gao Chunxue Peng Hao Wang Hailong Shen Ling Yang | 2021 | Journal of Forestry Research2021,32,2: | 3 |
| 10 | OGT Mediated Histone H2B S112 Glc NAcylation Regulates DNA Damage Response显示文摘O-GlcNAcylation is an important post-translational modification and has been implicated in many fundamental cellular processes. Recent studies showed that O-linked N-acetylglucosamine(GlcNAc) transferase(OGT) mediated O-Glc NAcylation of histone H2B Ser 112(H2B S112 Glc NAcylation) plays an important role in gene transcription. However, the role of this histone modification in DNA damage response has not been studied yet. In this study, we found that OGT and OGT mediated H2B S112 Glc NAcylation are involved in DNA damage response for maintaining genomic stability and are required for resistance to many DNA-damaging and replication stressinducing agents. OGT mediated H2B S112 GlcNAcylation increased locally upon the induction of double-strand breaks(DSBs), and depletion of OGT or overexpression of H2B S112A mutant impaired homologous recombination(HR) and nonhomologous end-joining(NHEJ). Mechanistically, H2B S112 GlcNAcylation could bind Nijmegen breakage syndrome 1(NBS1) and regulate NBS1 foci formation. But accurate calibration is important for all applications, especially for climate studies. HY-2a altimeter has been operational since April 2012 and its products are available to the scientific community. In this work, SWH data from HY-2A altimeters are calibrated against in situ buoy data from the National Data Buoy Center(NDBC), Distinguished from previous calibration studies which generally regarded buoy data as 'truth', the work of calibration for HY-2A altimeter wave data against in situ buoys was applied a more sophisticated statistical technique—the total least squares(TLS) method which can take into account errors in both variables. We present calibration results for HY-2A radar altimeter measurement of wave height against NDBC buoys.
