1Autologous CD34^+ and CD133^+ stem cells transplantation in patients with end stage liver disease显示文摘AIM:To assess the utility of an autologous CD34 + and CD133 + stem cells infusion as a possible therapeutic modality in patients with end-stage liver diseases.METHODS:One hundred and forty patients with endstage liver diseases were randomized into two groups.Group 1,comprising 90 patients,received granulocyte colony stimulating factor for five days followed by autologous CD34 + and CD133 + stem cell infusion in the portal vein.Group 2,comprising 50 patients,received regular liver treatment only and served as a control group.RESULTS:Near normalization of liver enzymes and improvement in synthetic function were observed in 54.5% of the group 1 patients;13.6% of the patients showed stable states in the infused group.None of the patients in the control group showed improvement.No adverse effects were noted.CONCLUSION:Our data showed that a CD34 + and CD133 + stem cells infusion can be used as supportive treatment for end-stage liver disease with satisfactory tolerability.Hosny Salama Abdel-Rahman N Zekri Abeer A Bahnassy Eman Medhat Hanan A Halim Ola S Ahmed Ghada Mohamed Sheren A Al Alim Ghada M Sherif 2010World Journal of Gastroenterology2010,16,42:16
2Pivotal role of long non-coding ribonucleic acid-X-inactive specific transcript in regulating immune checkpoint programmed death ligand 1 through a shared pathway between miR-194-5p and miR-155-5p in hepatocellular carcinoma显示文摘BACKGROUND Anti-programmed death therapy has thrust immunotherapy into the spotlight.However,such therapy has a modest response in hepatocellular carcinoma(HCC).Epigenetic immunomodulation is a suggestive combinatorial therapy with immune checkpoint blockade.Non-coding ribonucleic acid(ncRNA)driven regulation is a major mechanism of epigenetic modulation.Given the wide range of ncRNAs that co-opt in programmed cell-death protein 1(PD-1)/programmed death ligand 1(PD-L1)regulation,and based on the literature,we hypothesized that miR-155-5p,miR-194-5p and long non-coding RNAs(lncRNAs)X-inactive specific transcript(XIST)and MALAT-1 are involved in a regulatory upstream pathway for PD-1/PD-L1.Recently,nutraceutical therapeutics in cancers have received increasing attention.Thus,it is interesting to study the impact of oleuropein on the respective study key players.AIM To explore potential upstream regulatory ncRNAs for the immune checkpoint PD-1/PD-L1.METHODS Bioinformatics tools including microrna.org and lnCeDB software were adopted to detect targeting of miR-155-5p,miR-194-5p and lncRNAs XIST and MALAT-1 to PD-L1 mRNA,respectively.In addition,Diana tool was used to predict targeting of both aforementioned miRNAs to lncRNAs XIST and MALAT-1.HCC and normal tissue samples were collected for scanning of PD-L1,XIST and MALAT-1 expression.To study the interaction among miR-155-5p,miR-194-5p,lncRNAs XIST and MALAT-1,as well as PD-L1 mRNA,a series of transfections of the Huh-7 cell line was carried out.RESULTS Bioinformatics software predicted that miR-155-5p and miR-194-5p can target PDL1,MALAT-1 and XIST.MALAT-1 and XIST were predicted to target PD-L1 mRNA.PD-L1 and XIST were significantly upregulated in 23 HCC biopsies compared to healthy controls;however,MALAT-1 was barely detected.MiR-194 induced expression elevated the expression of PD-L1,XIST and MALAT-1.However,overexpression of miR-155-5p induced the upregulation of PD-L1 and XIST,while it had a negative impact on MALAT-1 expression.Knockdown of XIST did have an impact on PD-L1 expression;however,following knockdown of the negative regulator of X-inactive specific transcript(TSIX),PD-L1 expression was elevated,and abolished MALAT-1 activity.Upon co-transfection of miR-194-5p with siMALAT-1,PD-L1 expression was elevated.Co-transfection of miR-194-5p with siXIST did not have an impact on PD-L1 expression.Upon co-transfection of miR-194 with siTSIX,PD-L1 expression was upregulated.Interestingly,the same PD-L1 expression pattern was observed following miR-155-5p cotransfections.Oleuropein treatment of Huh-7 cells reduced the expression profile of PD-L1,XIST,and miR-155-5p,upregulated the expression of miR-194-5p and had no significant impact on the MALAT-1 expression profile.CONCLUSION This study reported a novel finding revealing that opposing acting miRNAs in HCC,have the same impact on PD-1/PD-L1 immune checkpoint by sharing a common signaling pathway.Sara M Atwa Heba Handoussa Karim M Hosny Margarete Odenthal Hend M El Tayebi 2020World Journal of Hepatology2020,12,12:9
3Role of relevant immune-modulators and cytokines in hepatocellular carcinoma and premalignant hepatic lesions显示文摘AIM To assess the levels of different immune modulators in patients with hepatocellular carcinoma(HCC),in relation to other hepatic diseases.METHODS Eighty-eight patients were included in the current study and represented patients with HCC(20),liver cirrhosis(28) and chronic hepatitis(CH;25),and normal controls(NC;15).Peripheral blood was isolated for immunophenotyping of active myeloid dendritic cells(m DCs;CD1 c and CD40),mature inactive myeloid cells(CD1 c and HLA),active plasmacytoid cells(p DCs;CD303 and CD40),mature inactive p DCs(CD30 and HLA),active natural killer(NK) cells(CD56 and CD161),active NK cells(CD56 and CD314) and inactive NK cells(CD56 and CD158) was done by flow cytometry.Serum levels of interleukin(IL)-2,IL-10,IL-12,IL-1β,interferon(IFN)-α,IFN-γ and tumor necrosis factor(TNF)-αR2 were assessed by ELISA.RESULTS Active m DCs(CD1 C+/CD40+) and inactive m DCs(CD1 c+/HLA+) were significantly decreased in HCC patients in relation to NC(P < 0.001).CD40+ expression on active p DCs was decreased in HCC patients(P < 0.001),and its level was not significantly changed among other groups.Inactive p DCs(CD303+/HLA+),inactive NKs(CD56+/CD158+) and active NKs(CD56+/CD161+) were not statistically changed among the four groups studied;however,the latter was increased in CH(P < 0.05).NKG2 D was statistically decreased in HCC,CH and cirrhosis(P < 0.001),and it was not expressed in 63%(12/20) of HCC patients.There was significant decrease of IL-2,IFN-α and IFN-γ(P < 0.001),and a significant increase in IL-10,IL-1β,and TNF-αR2(P <0.01,P < 0.001 and P < 0.001;respectively) in HCC patients.There was inverted correlation between IL-12 and IL-1β in HCC(r =-0.565,P < 0.01),with a strong correlation between p DCs(CD303+/CD40+) and NKs(CD56+/CD161+;r = 0.512,P < 0.05) as well as inactive m DCs(CD1 c+/HLA+) and inactive NK cells(CD56+/CD158+;r = 0.945,P < 0.001).CONCLUSION NKG2 D,CD40,IL-2 and IL-10 are important modulators in the development and progression of HCC.Abdel-Rahman N Zekri Somaya El Deeb Abeer A Bahnassy Abeer M Badr Mona S Abdellateif Gamal Esmat Hosny Salama Marwa Mohanad Ahmed Esam El-dien Shimaa Rabah Assmaa Abd Elkader 2018World Journal of Gastroenterology2018,24,11:6
4Biochemical changes in grape rootstocks resulted from humic acid treatments in relation to nematode infection显示文摘Objective:To investigate the effect of humic acid on nematode infected,resistant and susceptible grapes in relation to lipid peroxidation and antioxidant mechanisms on selected biochemical parameters known as proactive substances.Methods:The grape rootstocks,superior,superior/ freedom and freedom were reacted differently to Meloidogyne incognita and Rotylenchulus reniformis according to rootstock progenitor.Two weeks after inoculation,two commercial products of humic acid were applied at the rate of(2,4 mL or grams/plant) as soil drench.After 4 months,nematode soil populations were extracted and counted.A subsample of roots from each plant was stained and gall numbers,embedded stages per root were calculated,final population, nematode build up(Pf/Pi),average of eggs/eggmass were estimated.Subsamples of fresh root of each treatment were chemically analyzed.Results:Freedom reduced significantly the nematode criteria and build up.Humic acid granules appeared to be more suppressive to nematode build up on superior and the higher dose on superior/freedom than liquid treatments.On freedom,all treatments reduced significantly the nematode build up regardless to the material nature.The higher dose was more effective than the lower one.As a result of humic acid applications,the malondialdehyde(MDA) and H_2O_2 contents were significandy reduced after humic acid treatments while the antioxidant compounds glutathione(GSH),ascorbic acid(ASA) and total phenol contents were significandy increased when compared with check.Antioxidant defense enzymes ascorbate peroxidase(AFX),superoxide dismutase(SOD),catalase(CAT) and polyphenol oxidase(PPO) showed significant increase in their specific activities in treated plants compared with nematode treated check.Conclnsions:Humic acid treatments improve we yield of grape by increasing the contents of antioxidant compounds and the specific activities of antioxidant enzymes.Hosny H Kesba Hossam S El-Beltagi 2012Asian Pacific Journal of Tropical Biomedicine2012,2,4:4
5Interplay between micro RNA-17-5p, insulin-like growth factor-Ⅱ through binding protein-3 in hepatocellular carcinoma显示文摘AIM: To investigate the effect of microR NA on insulinlike growth factor binding protein-3(IGFBP-3) and hence on insulin-like growth factor-Ⅱ(IGF-Ⅱ) bioavailability in hepatocellular carcinoma(HCC).METHODS: Bioinformatic analysis was performed using microrna.org, DIANA lab and Segal lab softwares. Total RNA was extracted from 23 HCC and 10 healthy liver tissues using mir Vana mi RNA Isolation Kit. microR NA-17-5p(miR-17-5p) expression was mimicked and antagonized in Hu H-7 cell lines using Hi Per Fect Transfection Reagent, then total RNA was extracted using Biozol reagent then reverse transcribed into cD NA followed by quantification of mi R-17-5p and IGFBP-3 expression using Taq Man real-time quantitative PCR. Luciferase reporter assay was performed to validate the binding of miR-17-5p to the 3'UTR of IGFBP-3. Free IGF-Ⅱ protein was measured in transfected Hu H-7 cells using IGF-Ⅱ ELISA kit. RESULTS: Bioinformatic analysis revealed IGFBP-3 as a potential target for miR-17-5p. Screening of miR-17-5p and IGFBP-3 revealed a moderate negative correlation in HCC patients, where mi R-17-5p was extensively underexpressed in HCC tissues(P = 0.0012), while IGFBP-3 showed significant upregulation in the same set of patients(P = 0.0041) compared to healthy donors. Forcing mi R-17-5p expression in Hu H-7 cell lines showed a significant downregulation of IGFBP-3 mR NA expression(P = 0.0267) and a significant increase in free IGF-Ⅱ protein(P = 0.0339) compared to mock untransfected cells using unpaired t-test. Luciferase assay validated IGFBP-3 as a direct target of mi R-17-5p; luciferase activity was inhibited by 27.5% in cells co-transfected with miR-17-5p mimics and the construct harboring the wild-type binding region 2 of IGFBP-3 compared to cells transfected with this construct alone(P = 0.0474).CONCLUSION: These data suggest that regulating IGF-Ⅱ bioavailability and hence HCC progression can be achieved through targeting IGFBP-3 via manipulating the expression of miR NAs.Danira Ashraf Habashy Hend Mohamed El Tayebi Injie Omar Fawzy Karim Adel Hosny Gamal Esmat Ahmed Ihab Abdelaziz 2016World Journal of Hepatology2016,8,23:3
6Treatment of perforated giant gastric ulcer in an emergency setting显示文摘AIM:To study and assess clinical outcomes of various modes of treatment for perforated giant gastric ulcer in an emergency setting.METHODS:From May 2010 to February 2013,20cases of perforated giant gastric ulcer(>2 cm)were operated on in an emergency setting.All the patients presented with features of peritonitis and were resuscitated aggressively before taking for surgery.In the first4 cases,primary closure was done after taking a biopsy and among these,the 3rdcase also underwent partial distal gastrectomy and gastrojejunostomy and the 4th case underwent a radical subtotal gastrectomy with D2 lymphadenectomy and gastrojejunostomy for malignancy.All the remaining 16 cases underwent partial distal gastrectomy and gastrojejunostomy.RESULTS:Among the first 4 cases,2 had an uneventful recovery and were discharged on the 6thpostoperative day.The 3rdand 4thpatients developed gastricfistula,leading to prolonged hospitalization.For the 3rd patient,conservative management was tried for 1 wk,followed by partial distal gastrectomy and gastrojejunostomy,and he was discharged on the 20thday after admission,while the 4thpatient underwent a radical subtotal gastrectomy with D2 lymphadenectomy and gastrojejunostomy.Postoperatively,he developed adult respiratory distress syndrome,multiorgan dysfunction syndrome and expired on the 3rdpostoperative day of the second surgery.All the remaining 16 patients underwent partial distal gastrectomy and gastrojejunostomy and recovered well.Among these,4 of them were malignant and the remaining were benign ulcers.All had an uneventful recovery.The percentage of malignancy in our series was 30%(6 out of 20 cases).In our study,86%had an uneventful recovery,complications were seen in about 10%,and mortality was about 5%.CONCLUSION:In giant gastric ulcer,the chances of malignancy and leak after primary closure are high.So,we feel that partial distal gastrectomy and gastrojejunostomy is better.Pradeep Kumar Hosni Mubarak Khan Safarulla Hasanrabba 2014World Journal of Gastrointestinal Surgery2014,6,1:3

9Expression levels of pro-inflammatory interleukin-8 and certain antimicrobial peptides in concurrent with bacterial conjunctivitis显示文摘AIM:To detect the quantitative expression levels of the pro-inflammatory interleukin-8(IL8),antimicrobial peptides human beta defense-2(HBD2),and human beta defense-3(HBD3)genes in bacterial conjunctivitis.METHODS:The human conjunctival epithelial cells were obtained using the impression cytology technique from healthy controls and patients.The genes expression levels were determined utilizing a reverse transcription quantitative polymerase chain reaction(RT-q PCR).The contribution of causative agent type,the number of isolates and severity of clinical features,in the increase of genes expression was also determined.RESULTS:The RT-q PCR showed that IL8,HBD2,and HBD3 expression increased in bacterial conjunctivitis as compared to healthy control(P<0.001).In gram-negative bacterial conjunctivitis,HBD2 was highly up-regulated(P<0.001)compared to other types of bacterial conjunctivitis.In mixed bacterial conjunctivitis,a direct correlation between HBD2 up-regulation and HBD3 up-regulation was observed(P<0.05).The severity of clinical features was related to the up-regulation of IL8 and HBD2(P<0.05).CONCLUSION:IL8,HBD2,and HBD3 are immuneeffectors in infectious conjunctivitis.HBD2 is active during different bacterial conjunctivitis but is more released with gram-negative bacteria compared to gram-positive bacteria.HBD3 is an obvious defender in different bacterial conjunctivitis.Alaa El-Dien Shawky Hosny Zeinab El-Demerdash El-Bazza Mohammed Abdelhalim Ramadan Maha Ahmed Shafik Mahmoud Ahmed Shafeek Rania Abdelmonem Khattab 2021International Journal of Ophthalmology(English edition)2021,14,5:2

12Synthesis, spectral, optical and anti-inflammatory activity of complexes derived from 2-aminobenzohydrazide with some rare earths显示文摘Three new metal complexes derived from Er(III), Dy(III) and Zr(IV) with 2-aminobenzohydrazide(ABH) were synthesized and characterized by elemental analyses, IR, 1H-NMR, ES-MS and transmission electron microscopy(TEM). The morphology and the particle size were determined by TEM. The results showed that the ligand acted as neutral bi-dentate coordinating to the metal ions through the carbonyl oxygen and amidic amino nitrogen. The aromatic amine group remained inert towards coordination. The optical band gap was measured and found to be 3.3, 3.5 and 4.3 e V for Er(III), Dy(III) and Zr(IV), respectively. The optical band gap values indicated a semi-conducting nature of the investigated complexes. 13Deep Learning Prediction Model for Heart Disease for Elderly Patients显示文摘The detection of heart disease is a problematic task in medical research.This diagnosis utilizes a thorough analysis of the clinical tests from the patient’s medical history.The massive advances in deep learning models pursue the devel-opment of intelligent computerized systems that aid medical professionals to detect the disease type with the internet of things support.Therefore,in this paper,we propose a deep learning model for elderly patients to aid and enhance the diag-nosis of heart disease.The proposed model utilizes a deeper neural architecture with multiple perceptron layers with regularization learning techniques.The mod-el performance is verified with a full and minimum set of features.Fewer features enhance the processing time of the classification process while the accuracy is compromised.The performance of classifiers with less features has been analyzed with experimental results.The proposed system is built on the Internet of Things Platform for medical data for the classification process which aids medical profes-sionals to detect heart diseases through cloud platforms.The results accuracy is matched to classical learning models such as Convolutional Neural Network(CNN),Deep CNN,and neural ensemble models.The analysis of the proposed diagnostic system can determine the heart disease risks efficiently.Experimental results demonstrate thatflexible modeling and tuning of the hyperparameters can attain an accuracy of up to 97.11%.Abeer Abdulaziz AlArfaj Hanan Ahmed Hosni Mahmoud 2023Intelligent Automation & Soft Computing2023,,2:2
