1A novel negative regulatory mechanism of Smurf2 in BMP/Smad signaling in bone显示文摘Transforming growth factor-β(TGF-β)and bone morphogenetic protein(BMP)play important roles in bone metabolism.Smad ubiquitination regulatory factors(Smurfs)regulate TGF-β/BMP signaling via ubiquitination,resulting in degradation of signaling molecules to prevent excessive activation of TGF-β/BMP signaling.Though Smurf2 has been shown to negatively regulate TGF-β/Smad signaling,its involvement in BMP/Smad signaling in bone metabolism has not been thoroughly investigated.In the present study,we sought to evaluate the role of Smurf2 in BMP/Smad signaling in bone metabolism.Absorbable collagen sponges containing 3μg of recombinant human BMP2(rhBMP2)were implanted in the dorsal muscle pouches of wild type(WT)and Smurf2−/−mice.The rhBMP2-induced ectopic bone in Smurf2−/−mice showed greater bone mass,higher mineral apposition and bone formation rates,and greater osteoblast numbers than the ectopic bone in WT mice.In WT mice,the ectopic bone consisted of a thin discontinuous outer cortical shell and scant inner trabecular bone.In contrast,in Smurf2−/−mice,the induced bone consisted of a thick,continuous outer cortical shell and abundant inner trabecular bone.Additionally,rhBMP2-stimulated bone marrow stromal cells(BMSCs)from Smurf2−/−mice showed increased osteogenic differentiation.Smurf2 induced the ubiquitination of Smad1/5.BMP/Smad signaling was enhanced in Smurf2−/−BMSCs stimulated with rhBMP2,and the inhibition of BMP/Smad signaling suppressed osteogenic differentiation of these BMSCs.These findings demonstrate that Smurf2 negatively regulates BMP/Smad signaling,thereby identifying a new regulatory mechanism in bone metabolism.Junichi Kushioka Takashi Kaito Rintaro Okada Hiroyuki Ishiguro Zeynep Bal Joe Kodama Ryota Chijimatsu Melanie Pye Masahiro Narimatsu Jeffrey LWrana Yasumichi Inoue Hiroko Ninomiya Shin Yamamoto Takashi Saitou Hideki Yoshikawa Takeshi Imamura 2020Bone Research2020,8,4:6

3Pediatric living donor liver transplantation for congenital hepatic fibrosis using a mother's graft with von Meyenburg complex: A case report显示文摘This is the first report of living donor liver transplantation(LDLT) for congenital hepatic fibrosis(CHF) using a mother's graft with von Meyenburg complex. A 6-year-old girl with CHF, who suffered from recurrent gastrointestinal bleeding, was referred to our hospital for liver transplantation. Her 38-year-old mother was investigated as a living donor and multiple biliary hamartoma were seen on her computed tomography and magnetic resonance imaging scan. The mother's liver function tests were normal and she did not have any organ abnormality, including polycystic kidney disease. LDLT using the left lateral segment(LLS) graft from the donor was performed. The donor LLS graft weighed 250 g; the graft recipient weight ratio was 1.19%. The operation and post-operative course of the donor were uneventful and she was discharged on post-operative day(POD) 8. The graft liver function was good, and the recipient was discharged on POD 31. LDLT using a graft with von Meyenburg complex is safe and useful. 9Initial 3-month dynamics of dehydroepiandrosterone sulfate can predict responsiveness to primary androgen deprivation therapy in patients with metastatic prostate cancer显示文摘Dear Editor,Intensive treatments such as primary androgen deprivation therapy(pADT)plus docetaxel or abiraterone acetate are reportedly beneficial in metastatic prostate cancer(mPCa)patients with high-volume disease or high-risk features,respectively.1,2 High-volume or high-risk features are determined exclusively via pretreatment parameters.Predicting patient responsiveness to pADT,which is one of the major determinants of survival in these patients,would be advantageous with regard to making primary treatment decisions.If an initial 3-month period is possible,short-term serum parameter dynamics can be used to predict pADT responsiveness.Prostate-specific antigen(PSA)and alkaline phosphatase(ALP)dynamics have been indicated as potential predictors.3,4 Although pADT can reportedly cause a 20%-30%decrease in adrenal androgen levels,5 it is not known whether the dynamics of adrenal androgens shortly after the initiation of pADT can predict pADT responsiveness.The aim of the current study was to identify predictors of pADT responsiveness among pretreatment parameters and 3-month dynamics(D3m)of serum parameters including the adrenal androgen,dehydroepiandrosterone sulfate(DHEAS).Akihiro Yano Makoto Kagawa Hideki Takeshita Yohei Okada Makoto Morozumi Satoru Kawakami 2019Asian Journal of Andrology2019,21,6:1

14Efficacy and safety of endoscopic submucosal dissection under general anesthesia显示文摘AIM: To evaluate the efficacy and safety of endoscopic submucosal dissection(ESD) under general anesthesia.METHODS: From January 2011 to July 2014, 206 consecutive patients had undergone ESD under general anesthesia for neoplasms of the stomach, esophagus, and colorectum were enrolled in this retrospective study. The efficacy and safety of ESD under general anesthesia were assessed.RESULTS: The en bloc resection rate of esophageal, gastric, and colorectal lesions was 100.0%, 98.3%, and 96.1%, respectively. The complication rate of perforationand bleeding were 0.0% and 0.0% in esophageal ESD, 1.7% and 1.7% in gastric ESD, and 3.9% and 2.0% in colorectal ESD, respectively. No cases of aspiration pneumonia were observed.
