| 4 | An electron microscopic-cytochemical localization of plasma membrane Ca^(2+)-ATPase activity in poplar apical bud cells during the induction of dormancy by short-day photoperiods显示文摘Plasma membrane (PM) Ca2+-ATPase activity in poplar apical bud meristematic cells during short-day (SD)-induced dormancy development was examined by a cerium precipitation EM-cytochemical method. Ca2+-ATPase activity, indicated by the status of cerium phosphate precipitated grains, was localized mainly on the interior face (cytoplasmic side) of the PM when plants were grown under long days and reached a deep dormancy. A few reaction products were also observed on the nuclear envelope.When plant buds were developing dormancy after 28 to 42 d of SD exposure, almost no reaction products were present on the interior face of the PM. In contrast, a large number of cerium phosphate precipitated grains were distributed on the exterior face of the PM. After 70 d of SD exposure, when buds had developed a deep dormancy, the reaction products of Ca2+-ATPase activity again appeared on the interior face of the PM. The results seemed suggesting that two kinds of Ca2+-ATPases may be present on the PM during the SD-induced dormancy in poplar.One is the Ca2+-punlping ATPase, which is located on the interior face of the PM, for maintaining and restoring the Ca2+homeostasis. A timed complexation colorimetric analysis method of A1 Fe ferron in view of the total concentration of {A1+Fe} was then established to determine the species distribution of polymeric Al Fe. The testing wavelength was recommended at 362 nm and the testing pH value was 5. With a comparison of the ratios of n Al / n Fe , the standard adsorption curves of the polymeric Al Fe solutions were derived from the experimental results. | 10 | Risks of rapid decline renal function in patients with type 2 diabetes显示文摘Progressive rising population of diabetes and related nephropathy, namely, diabetic kidney disease and associated end stage renal disease has become a major global public health issue. Results of observational studies indicate that most diabetic kidney disease progresses over decades; however, certain diabetes patients display a rapid decline in renal function, which may lead to renal failure within months. Although the definition of rapid renal function decline remained speculative, in general,it is defined by the decrease of estimated glomerular filtration rate(e GFR) in absolute rate of loss or percent change. Based on the Kidney Disease: Improving Global Outcomes 2012 clinical practice guidelines, a rapid decline in renal function is defined as a sustained declinein e GFR of > 5 m L/min per 1.73 m2 per year. It has been reported that potential factors contributing to a rapid decline in renal function include ethnic/genetic and demographic causes, smoking habits, increased glycated hemoglobin levels, obesity, albuminuria, anemia, low serum magnesium levels, high serum phosphate levels, vitamin D deficiency, elevated systolic blood pressure, pulse pressure, brachial-ankle pulse wave velocity values, retinopathy, and cardiac autonomic neuropathy. | 11 | Calcitriol analog ZK191784 ameliorates acute and chronic dextran sodium sulfate-induced colitis by modulation of intestinal dendritic cell numbers and phenotype显示文摘AIM: To investigate the effects of ZK1916784, a low calcemic analog of calcitriol on intestinal inflammation. METHODS: Acute and chronic colitis was induced by dextran sodium sulfate (DSS) according to standard procedures. Mice were treated intraperitoneally with ZK1916784 or placebo and colonic inflammation was evaluated. Cytokine production by mesenterial lymph node (MLN) cells was measured by ELISA. Immunohistochemistry was performed to detect intestinal dendritic cells (DCs) within the colonic tissue, and the effect of the calcitriol analog on DCs was investigated. RESULTS: Treatment with ZK191784 resulted in significant amelioration of disease with a reduced histological score in acute and chronic intestinal inflammation. In animals with acute DSS colitis, down- regulation of colonic inflammation was associated with a dramatic reduction in the secretion of the proinflammatory cytokine interferon (IFN)-γ and a significant increase in intereleukin (IL)-10 by MLN cells. Similarly, in chronic colitis, IL-10 expression in colonic tissue increased 1.4-fold when mice were treated with ZK191784, whereas expression of the Th1-specific transcription factor T-beta decreased by 81.6%. Lower numbers of infiltrating activated CD11c+ DCs were found in the colon in ZK191784-treated mice with acute DSScolitis, and secretion of proinflammatory cytokines by primary mucosal DCs was inhibited in the presence of the calcitriol analog.
