1Cluster analysis for syndromes of real-world coronary heartdisease with angina pectoris显示文摘Syndromes of coronary heart disease with angina pectoris were analyzed to provide guidance forclinical practice and to improve accuracy of traditional Chinese medicine (TCM) diagnoses and efficacy of TCMtreatment. A total of 860 cases with coronary heart disease with angina pectoris were selected from TCM ClinicalResearch Information Sharing System for TCM clinics and research. Syndromes were automatically extractedwith the cluster method and were analyzed to provide objective evidence for clinical studies. Final syndromeclassifications were recognized and confirmed by clinical experts. Popular syndromes included Qi and blooddeficiency, blood stasis and obstruction collaterals, liver depression and spleen deficiency, and Qi stagnation andblood stasis. Syndromes Qi and blood deficiency and blood stasis and obstruction collaterals accounted for 28.61%of total syndromes, whereas liver depression and spleen deficiency and Qi stagnation and blood stasis accountedfor 26.44%. 3Comparsion analysis of data mining models applied to clinical research in Traditional Chinese Medicine显示文摘OBJECTIVE: To help researchers selecting appropriate data mining models to provide better evidence for the clinical practice of Traditional Chinese Medicine(TCM) diagnosis and therapy.METHODS: Clinical issues based on data mining models were comprehensively summarized from four significant elements of the clinical studies:symptoms, symptom patterns, herbs, and efficacy.Existing problems were further generalized to determine the relevant factors of the performance of data mining models, e.g. data type, samples, parameters, variable labels. Combining these relevant factors, the TCM clinical data features were compared with regards to statistical characters and informatics properties. Data models were compared simultaneously from the view of applied conditions and suitable scopes.RESULTS: The main application problems were the inconsistent data type and the small samples for the used data mining models, which caused the inappropriate results, even the mistake results. 4Clinical characteristics and effectiveness of antiviral agents in hospitalized children with infectious mononucleosis in China:A multicenter retrospective study显示文摘Importance:The clinical characteristics of infectious mononucleosis(IM)in Chinese children have not been evaluated in multicenter studies,and the effectiveness of antiviral treatment are controversial.Objective:To investigate the clinical characteristics of Chinese children with IM and current status of antiviral therapy for affected patients.Methods:Hospitalized patients with IM were enrolled between 2018 and 2020 in five children’s hospitals in China.The clinical characteristics were compared among four age groups:<3 years,3-<6 years,6-<10 years,and≥10 years.The clinical characteristics of IM and effectiveness of antiviral therapy were compared among patients receiving acyclovir(ACV),ganciclovir(GCV),and no antiviral therapy(i.e.,non-antiviral group).Results:In total,499 patients were analyzed;most patients were 3-<6 years of age.The most common symptoms and signs included fever(100%),lymphadenopathy(98.6%),pharyngitis(86.4%),eyelid edema(76.8%),and snoring(72.9%).There were significant differences in rash,hepatomegaly,and liver dysfunction among the four age groups.Patients aged<3 years had a lower incidence of liver dysfunction and a higher incidence of rash.Among the 499 patients,50.1%were treated with GCV,26.3%were treated with ACV,and 23.6%received no antiviral therapy.Compared with the non-antiviral group,patients in the ACV and GCV groups had longer durations of fever(P<0.001).There were no significant differences in the incidences of complications among the three treatment groups.Interpretation:The incidence of IM in Chinese children peaked at 3-<6 years of age.Clinical features of IM varied according to age.Patients receiving antiviral therapy exhibited more serious clinical manifestations than did patients without antiviral therapy.The effectiveness of antiviral therapy for IM requires further analysis.Huili Hu Huiling Deng Jing Bi Yi Xu Shuangjie Li Yue Xie Xinrong Sun Dongmeng Wang Xufang Li Wenxian Ouyang Bing Hu Yufeng Zhang He Tang Chunxiao Fang Hui Zhang Lingyun Guo Chen Wang Tianyi Wang Fengxia Yang Tao Jiang Zhengde Xie Gang Liu 2021Pediatric Investigation2021,5,3:12
5Regulatory effects of antitumor agent matrine on FOXO and PI3K-AKT pathway in castration-resistant prostate cancer cells显示文摘We previously demonstrated that matrine could inhibit the proliferating, migrating, as well as invading processes of both PC-3 and DU145 cells. However, the underlying molecular mechanisms have not yet been clearly defined. In this study, using various techniques such as high throughput sequencing technology, bioinformatics, quantitative real-time PCR, and immunoblot analysis,we aimed to understand whether matrine serves as a novel regulator of FOXO and PI3K-AKT signaling pathway. DU145 and PC-3 cell lines were cultured for 24 h in vitro. Cells were treated with either matrine or control serum for 48 h, followed by extraction of total RNA. The RNA was sequenced using HiSeq 2500 high-throughput sequencing platform (Illumina). A gene library was established and quality analysis of read data carried out. Integrated database from the website DAVID was used to analyze Gene Ontology (GO), and Kyoto encyclopedia of genes and genomes (KEGG) pathway of differential genes was used for pathway analysis, screening for fold differences of more than two times. The FOXO and PI3K-AKT signaling pathways were screened, and expression levels of mRNA and core protein detected by real-time PCR and immunoblotting, respectively. High throughput sequencing and GO analysis revealed that differentially expressed genes before and after treatment played an important role in cell metabolic process, growth process, anatomical structure formation, cellular component organization, and biological regulation. KEGG signal pathway analysis revealed that FOXO and PI3K-AKT signal pathways had a significant difference between before and after matrine-treated androgen-independent prostate cancer cells PC-3 and DU145. Real-time PCR showed that matrine treatment led to a significant increase in the expression levels of FOXO1A, FOXO3A, FOXO4, and FOXO6 in DU145 and PC-3 cells (P<0.01 or P<0.05), whereas the PI3K expression levels decreased (P<0.01). Similarly, immunoblotting revealed a significant increase (P<0.05) in the expression levels of FOXO1A FOXO3A, FOXO4, and FOXO6 in both PC-3 and DU145 cells, whereas PI3K expression levels decreased (P<0.05). Matrine had a broad regulating effect on the mRNA expression profiles of both PC-3 and DU145 cells. Matrine may inhibit cell proliferation, migration, as well as invasion, and induce apoptosis in both PC-3 and DU145 cells through FOXO and PI3K-AKT signaling pathways. 7CRISPR-Cas13a mediated nanosystem for attomolar detection of canine parvovirus type 2显示文摘Canine parvovirus type 2(CPV-2) infection is the most lethal disease of dogs with higher mortality in puppies worldwide.In today’s world,dogs are an integral part of our communities as well as dogs breeding and rearing has become a lucrative business.Therefore,a fast,accurate,portable,and costeffective CPV-2 detection method with the ability for on-site detection is highly desired.In this study,we for the first time proposed a nanosystem for CPV-2 DNA detection with RNA-guided RNA endonuclease Cas13 a,which upon activation results in collateral RNA degradation.We expressed LwCasl3 a in prokaryotic expression system and purified it through nickel column.Activity of Cas13 a was verified by RNA-bound fluorescent group while using a quenched fluorescent probe as signals.Further Cas13 a was combined with Recombinase polymerase amplification(RPA) and T7 transcription to establish molecular detection system termed specific high-sensitivity enzymatic reporter un-locking(SHERLOCK) for sensitive detection of CPV-2 DNA.This nanosystem can detect 100 amol/L CPV-2 DNA within 30 min.The proposed nanosystem exhibited high specificity when tested for CPV-2 and other dog viruses.This CRISPR-Cas13 a mediated sensitive detection approach can be of formidable advantage during CPV-2 outbreaks because it is time-efficient,less laborious and does not involve the use of sophisticated instruments.Haroon Khan Adeel Khan Yufeng Liu Su Wang Sumaira Bibi Hongpan Xu Yuan Liu Samran Durrani Lian Jin Nongyue He Tao Xiong 2019Chinese Chemical Letters2019,30,12:8
8Cross-cultural adaption and psychometric properties of the Chinese version of the Diabetes Behavior Rating Scale: a pilot study显示文摘Self-care behavior plays a major role in diabetic management. However, in China, a satisfactory instrument has not yet been developed to evaluate the compliance of self-care behavior for young patients with type 1 diabetes mellitus(T1 DM). The Diabetes Behavior Rating Scale(DBRS) has a potential to be the first mature instrument. The purpose of this study is to cross-culturally adapt the DBRS, and preliminarily evaluate its psychometric properties. The instrument translation included translation, back translation and culture adaptation. Psychometric properties were assessed in a sample of 116 young patients with T1 DM adapting insulin injection therapy. The Chinese version of the DBRS was divided to four subscales. Cronbach's α for the total scale was 0.92. The mean inter-item and item-total correlations were 0.35 and 0.54 respectively. Test-retest reliability showed good temporal stability(r=0.81, P=0.001). Negative correlations were found between DBRS scores with the Diabetes Distress Scale scores(r=.0.32, P=0.003) and hemoglobin A1 c(HbA1 c) levels(r=.0.36, P=0.002). Higher DBRS scores correlated with better glycemic control. The Chinese insulin injection therapy version of the DBRS is well translated and culturally adapted. 9Analysis of DNA Methylation Level by Methylation-Sensitive Amplification Polymorphism in Half Smooth Tongue Sole(Cynoglossus semilaevis)Subjected to Salinity Stress显示文摘Increasingly arisen environmental constraints may contribute to heritable phenotypic variation including methylation changes,which can help the animals with development,growth and survival.In this study,we assessed the DNA methylation levels in three tissues(gonad,kidney and gill) of half smooth tongue sole under the salinity stress.The methylation-sensitive amplification polymorphism(MSAP) technique was applied to illustrate the regulation of epigenetic mechanism in environmental stimuli.Fish were subjected to 15 salinity treatment for 7 and 60 days,respectively.A total of 11259 fragments were amplified with 8 pairs of selective primers.The levels of methylated DNA in different tissues of females and males without salinity stress were analyzed,which were 32.76% and 47.32% in gonad;38.13% and 37.69% in kidney;37.58% and 34.96% in gill,respectively.In addition,the significant difference was observed in gonad between females and males,indicating that discrepant regulation in gonadal development and differentiation may involve sex-related genes.Further analysis showed that total and hemi-methylation were significantly decreased under 15 salinity for 7 days,probably resulting in up-regulating salt-tolerance genes expression to adjust salt changing.With the adjustment for 60 days,total and hemi-methylation prominently went back to its normal levels to obtain equilibrium.Particularly,full methylation levels were steady along with salinity stress to maintain the stability of gene expression.Additionally,the data showed that gonads in females and gills in males were superior in adaptability.As a result,DNA methylation regulates tissue-specific epiloci,and may respond to salinity stress by regulating gene expression to maintain animal survival and activity.LI Siping HE Feng WEN Haishen LI Jifang SI Yufeng LIU Mingyuan HE Huiwen HUANG Zhengju 2017Journal of Ocean University of China2017,16,2:4
10Metabolic and Phylogenetic Profile of Bacterial Community in Guishan Coastal Water(Pearl River Estuary), South China Sea显示文摘Characteristics of a microbial community are important as they indicate the status of aquatic ecosystems. In the present study, the metabolic and phylogenetic profile of the bacterioplankton community in Guishan coastal water(Pearl River Estuary), South China Sea, at 12 sites(S1–S12) were explored by community-level physiological profiling(CLPP) with BIOLOG Eco-plate and denaturing gradient gel electrophoresis(DGGE). Our results showed that the core mariculture area(S6, S7 and S8) and the sites associating with human activity and sewage discharge(S11 and S12) had higher microbial metabolic capability and bacterial community diversity than others(S1–5, S9–10). Especially, the diversity index of S11 and S12 calculated from both CLPP and DGGE data(H >3.2) was higher than that of others as sewage discharge may increase water nitrogen and phosphorus nutrient. The bacterial community structure of S6, S8, S11 and S12 was greatly influenced by total phosphorous, salinity and total nitrogen. Based on DGGE fingerprinting, proteobacteria, especially γ- and α-proteobacteria, were found dominant at all sites. In conclusion, the aquaculture area and wharf had high microbial metabolic capability. 11Prediction Research of Deformation Modulus of Weak Rock Zone under In-situ Conditions显示文摘Weak rock zone (soft interlayer, fault zone and soft rock) is the highlight of large-scale geological engineering research. It is an important boundary for analysis of rock mass stability. Weak rock zone has been formed in a long geological period, and in this period, various rocks have undergone long-term consolidation of geostatic stress and tectonic stress; therefore, under in-situ conditions, their density and modulus of deformation are relatively high. Due to its fragmentary nature, once being exposed to the earth's surface, the structure of weak rock zone will soon be loosened, its density will be reduced, and its modulus of deformation will also be reduced significantly. Generally, weak rock zone can be found in large construction projects, especially in the dam foundation rocks of hydropower stations. These rocks cannot be eliminated completely by excavation. Furthermore, all tests nowadays are carried out after the exposure of weak rock zone, modulus of deformation under in-situ conditions cannot be revealed. In this paper, a test method explored by the authors has been introduced. This method is a whole multilayered medium deformation method. It is unnecessary to eliminate the relatively complete rocks covering on weak rock zone. A theoretical formula to obtain the modulus of deformation in various mediums has also been introduced. On-site comparative trials and indoor deformation modulus tests under equivalent density conditions have been carried out. We adopted several methods for the prediction researches of the deformation modulus of weak rock zone under in-situ conditions, and revealed a fact that under in-situ conditions, the deformation modulus of weak rock zone are several times higher than the test results obtained after the exposure. 13Evolution of micro-pores in a single crystal nickel-based superalloy during 980℃ creep显示文摘The evolution of micro-pores in a single crystal nickel-based superalloy during creep at 980 ℃/220 MPa was investigated by X-ray computed tomography. Time-dependent ex-situ 3D information including the number, volume fraction, distribution and morphology of micro-pores was analyzed. The results reveal that the signifi cant formation and growth of micro-pores occur at the end of secondary/beginning of tertiary creep stage. The irregular large pores as well as high density pores located at strain concentration region are the major detrimental factors facilitating the creep damage. 20Preparation and antioxidant activity of albumin binding Salen Schiff-base metal complexes显示文摘A kind of novel biopolymer antioxidant (BSA/HOSalenM, M=Co, Mn, Zn) is prepared with conjugation, which increases the antioxidant activity of the bovine serum albumin (BSA). The conjugations have been characterized by IR spectra, UV-Vis spectra, Fluorescence spectra, Circular dichroism (CD) spectra and Native-PAGE. The BSA is used as a biopolymer scaffold, and the insoluble Salen Schiff-base metal complexes HOSalenM make axial coordination with the amino acid residues of the BSA. The structure of the BSA is unchanged when the binding rate of HOSalenCo is less than 10. The HOSalenCo conjugations show an excellent hydroxyl radical (·OH) scavenging activity, and the activity (EC50 ) of BSA/HOSalenCo(10) (BSA : HOSalenCo=1 : 10) is improved by two orders of magnitude compared with the BSA, while the activity of the BSA/HOSalenMn is weak and the BSA/HOSalenZn shows no scavenging activity.LI Gang ZHANG HuiFang WANG RongMin HE YuFeng XIONG YuBing 2013Chinese Science Bulletin2013,58,24:1
