| 1 | Induction of xenogeneic islet transplantation tolerance by simultaneously blocking CD28-B7 and OX40-OX40L co-stimulatory pathways显示文摘It has been demonstrated that prolonged graft survival can be achieved through inhibiting the activation of T cells, and addition of soluble CTLA4Ig and OX40Ig proteins to mixed lymphocyte reactions can effectively inhibit T cell proliferation. To explore the potential of this type of treatment in xenotransplantation, we infected streptozotocin-induced diabetic BalB/c mice (H-2d) (200 mg/kg, IV) with 5×108 pfu AdCTLA4Ig-IRES-OX40Ig on day 1 before islets trans-plantation through the tail vein. The results showed that this treatment prolonged the islet xeno-grafts survival significantly. The reaction to exogenous glucose stimulation was normal and the cytokine secretion of the type Th1 cells was inhibited. The AdCTLA4Ig-IRES-OX40Ig-mediated treatment effectively induced the T cells into anergy and the Th1/Th2 cells into deviation. | 2 | SHANK2 is a frequently amplified oncogene with evolutionarily conserved roles in regulating Hippo signaling显示文摘Dysfunction of the Hippo pathway enables cells to evade contact inhibition and provides advantages for cancerous overgrowth.However,for a significant portion of human cancer,how Hippo signaling is perturbed remains unknown.To answer this question,we performed a genome-wide screening for genes that affect the Hippo pathway in Drosophila and cross-referenced the hit genes with human cancer genome.In our screen,Prosap was identified as a novel regulator of the Hippo pathway that potently affects tissue growth.Interestingly,a mammalian homolog of Prosap,SHANK2,is the most frequently amplified gene on 11 q13,a major tumor amplicon in human cancer.Gene amplification profile in this 11q13 amplicon clearly indicates selective pressure for SHANK2 amplification.More importantly,across the human cancer genome,SHANK2 is the most frequently amplified gene that is not located within the Myc amplicon.Further studies in multiple human cell lines confirmed that SHANK2 overexpression causes deregulation of Hippo signaling through competitive binding for a LATS1 activator,and as a potential oncogene,SHANK2 promotes cellular transformation and tumor formation in vivo.In cancer cell lines with deregulated Hippo pathway,depletion of SHANK2 restores Hippo signaling and ceases cellular proliferation.Taken together,these results suggest that SHANK2 is an evolutionarily conserved Hippo pathway regulator,commonly amplified in human cancer and potently promotes cancer.Our study for the first time illustrated oncogenic function of SHANK2,one of the most frequently amplified gene in human cancer.Furthermore,given that in normal adult tissues,SHANK2 s expression is largely restricted to the nervous system,SHANK2 may represent an interesting target for anticancer therapy. | Liang Xu Peixue Li Xue Hao Yi Lu Mingxian Liu Wenqian Song Lin Shan Jiao Yu Hongyu Ding Shishuang Chen Ailing Yang Yi Arial Zeng Lei Zhang Hai Jiang | 2021 | Protein & Cell2021,12,3: | 1 |

| 11 | Comparative study of the promotion of porcine fetal fibroblast proliferation by overexpression of two transcriptional variants of SIRT6显示文摘Mammalian Sirtuin 6(SIRT6) plays an important role in the transcriptional silencing of silent mating-type loci,telomeres,and rDNA.However,the roles of porcine SIRT6 in cell proliferation are poorly understood,and a better knowledge of these will help improve our understanding of the biological mechanisms of cell growth and development.In this study,a novel variant of porcine SIRT6(SIRT6 V2) identified by reverse transcription-polymerase chain reaction and BLAST analysis showed a 124-bp deletion compared to wild-type SIRT6 mRNA(SIRT6 V1).Two recombinant plasmids overexpressing SIRT6 V1 and SIRT6 V2 were produced and their roles in the proliferation of porcine fetal fibroblasts(PFFs) were compared.Cells transfected with SIRT6 V1 proliferated significantly faster than those transfected with SIRT6 V2(P<0.01),though both proliferated significantly faster than cells expressing an empty vector(P<0.01).These results might be caused by altered proportions of α helices and β sheets in the SIRT6 V2 structure relative to SIRT6 V1.These results indicate that overexpression of SIRT6 V1/V2 was involved in promoting PFF proliferation.Deletion of a 124-bp sequence attenuated the effects of SIRT6 on cell proliferation,possibly as a result of changes in the proportions of α helices and β sheets in the protein secondary structure. | XIE XiaoXian HE Kan ZHANG AiLing ZHANG Hao WANG QiShan LI JiaQi MA YuFang | 2013 | Chinese Science Bulletin2013,58,10: | 0 |
| 12 | Combination of CTLA4-FasL gene transfer and allogeneic bone marrow transplantation led to durable macrochimerism and donor-specific tolerance in mouse model显示文摘Mixed hemopoietic chimerism is capable of inducing donor specific tolerance, thus eliminating the chronic immunosuppressive therapy following organ trans- plantation. As yet no safe and effective tolerance protocol is available for clinical implementation. Here we describe an alternative nonmyeloablative based strategy of using a single injection of recombination adenovirus vector encoding CTLA4-FasL fusing gene and donor bone marrow cells to promote durable mixed macrochimerism (>20% on 140 d). Chimeras exhibited robust donor-specific tolerance, as evi- denced by acceptance of fully allogeneic skin grafts (the mean survival time (MST)>200 d) and rejection of third- party skin grafts in a normal manner (MST<10 d).
