| 1 | Adipose-derived mesenchymal stem cell transplantation promotes adult neurogenesis in the brains of Alzheimer's disease mice显示文摘In the present study,we transplanted adipose-derived mesenchymal stem cells into the hippocampi of APP/PS1 transgenic Alzheimer’s disease model mice.Immunofluorescence staining revealed that the number of newly generated(BrdU+)cells in the subgranular zone of the dentate gyrus in the hippocampus was significantly higher in Alzheimer’s disease mice after adipose-derived mesenchymal stem cell transplantation,and there was also a significant increase in the number of BrdU+/DCX+neuroblasts in these animals.Adipose-derived mesenchymal stem cell transplantation enhanced neurogenic activity in the subventricular zone as well.Furthermore,adipose-derived mesenchymal stem cell transplantation reduced oxidative stress and alleviated cognitive impairment in the mice.Based on these findings,we propose that adipose-derived mesenchymal stem cell transplantation enhances endogenous neurogenesis in both the subgranular and subventricular zones in APP/PS1 transgenic Alzheimer’s disease mice,thereby facilitating functional recovery. | Yufang Yan Tuo Ma Kai Gong Qiang Ao Xiufang Zhang Yao Gong | 2014 | Neural Regeneration Research2014,9,8: | 16 |

| 3 | Serum uric acid and risk of incident diabetes in middle-aged and elderly Chinese adults: prospective cohort study显示文摘The association between serum uric acid and the risk of incident diabetes in Chinese adults remains unknown.This study aimed to investigate this association in a community-dwelling population aged≥40 years in Shanghai,China.Oral glucose tole3rance test was conducted during baseline and follow-up visits.Relative risk regression was utilized to examine the associations between baseline gender-specific serum uric acid levels and incident diabetes risk.A total of 613(10.3%)incident diabetes cases were identified during the follow-up visit after 4.5 years.Fasting plasma glucose,postload glucose,and glycated hemoglobin A1c during the follow-up visit progressively increased across the sex-specific quartiles of serum uric acid(all Ps<0.05).The incidence rate of diabetes increased across the quartiles of serum uric acid(7.43%,8.77%,11.47%,and 13.43%).Multivariate adjusted regression analysis revealed that individuals in the highest quartile had 1.36-fold increased risk of diabetes compared with those in the lowest quartile of serum uric acid(odds ratio(95%confidence interval)=1.36(1.06−1.73)).Stratified analysis indicated that the association was only observed in women.Accordingly,serum uric acid was associated with the increased risk of incident diabetes among middle-aged and elderly Chinese women. | Di Cheng Chunyan Hu Rui Du Hongyan Qi Lin Lin Xueyan Wu Lina Ma Kui Peng Mian Li Min Xu Yu Xu Yufang Bi Weiqing Wang Yuhong Chen Jieli Lu | 2020 | Frontiers of Medicine2020,14,6: | 3 |

| 5 | Late Quaternary Activity of the Huashan Piedmont Fault and Associated Hazards in the Southeastern Weihe Graben,Central China显示文摘The Weihe Graben is not only an important Cenozoic fault basin in China but also a significant active seismic zone. The Huashan piedmont fault is an important active fault on the southeast side of the Weihe Graben and has been highly active since the Cenozoic. The well–known Great Huaxian County Earthquake of 1556 occurred on the Huashan piedmont fault. This earthquake, which claimed the lives of approximately 830000 people, is one of the few large earthquakes known to have occurred on a high–angle normal fault. The Huashan piedmont fault is a typical active normal fault that can be used to study tectonic activity and the associated hazards. In this study, the types and characteristics of late Quaternary deformation along this fault are discussed from geological investigations, historical research and comprehensive analysis. On the basis of its characteristics and activity, the fault can be divided into three sections, namely eastern, central and western. The eastern and western sections display normal slip. Intense deformation has occurred along the two sections during the Quaternary; however, no deformation has occurred during the Holocene. The central section has experienced significant high–angle normal fault activity during the Quaternary, including the Holocene. Holocene alluvial fans and loess cut by the fault have been identified at the mouths of many stream valleys of the Huashan Mountains along the central section of the Huashan piedmont fault zone. Of the three sections of the Huashan piedmont fault, the central section is the most active and was very active during the late Quaternary. The rate of normal dip–slip was 1.67–2.71±0.11 mm/a in the Holocene and 0.61±0.15 mm/a during the Mid–Late Pleistocene. As is typical of normal faults, the late Quaternary activity of the Huashan piedmont fault has produced a set of disasters, which include frequent earthquakes, collapses, landslides, mudslides and ground fissures. | 8 | Production of human lysozyme-transgenic cloned porcine embryos by somatic nuclear transfer显示文摘Due to their physiology and organ size, pigs have significant potential as human disease models and as organ transplantation donors. Genetic modification of pigs could provide benefits for both agriculture and human medicine. In this study, five fetal pig fibroblast cell lines from two species (Wuzhishan and Landrace pigs) were transfected using double-marked human lysozyme (HLY) plasmids (pBC1-HLY-GFP-NEO) by a liposome-mediated method. The ratio of green fluorescent protein (GFP)-expressing cells was >95% in sw7, sw8, slw3 and slw6 cell lines, but only 49.3% in slw9 cells. Cells from the four highly transgenic lines were used as nuclear donors to construct embryos, which were then cultured after fusion and activation by electric stimulation. The rate of cleavage was 76.7%, 48 h after activation. After 7 days, 18.5% of cleaved eggs had developed to the blastocyst stage and 93.3% of blastocysts were GFP-positive. These results indicate that transgenic fetal pig flbroblast cell lines could be obtained by a liposome-mediated method, though the transfection efficiency varied between cell lines. | 9 | A Disintegrin and Metalloprotease 10 in neuronal maturation and gliogenesis during cortex development显示文摘The multiple-layer structure of the cerebral cortex is important for its functions. Such a structure is generated based on the proliferation and differentiation of neural stem/progenitor cells. Notch functions as a molecular switch for neural stem/progenitor cell fate during cortex development but the mechanism remains unclear. Biochemical and cellular studies showed that Notch receptor activation induces several proteases to release the Notch intracellular domain (NICD). A Disintegrin and Metalloprotease 10 (ADAM10) might be a physiological rate-limiting S2 enzyme for Notch activation. Nestin-driven conditional ADAM10 knockout in mouse cortex showed that ADAM10 is critical for maintenance of the neural stem cell population during early embryonic cortex development. However, the expression pattern and function of ADAM10 during later cerebral cortex development remains poorly understood. We performed in situ hybridization for ADAM10 mRNA and immunofluorescent analysis to determine the expression of ADAM10 and NICD in mouse cortex from embryonic day 9 (E14.5) to postnatal day 1 (P1). ADAM10 and NICD were highly co-localized in the cortex of E16.5 to P1 mice. Comparisons of expression patterns of ADAM10 with Nestin (neural stem cell marker), Tuj1 (mature neuron marker), and S100β (glia marker) showed that ADAM10 expression highly matched that of S100β and partially matched that of Tuj1 at later embryonic to early postnatal cortex developmental stages. | 15 | Seasonal influence on the development of parthenotes and cloned embryos in pigs显示文摘Seasonal influence on the developmental ability of porcine embryos was studied by comparisons of the number of cumulus-oocytes-complexes (COCs) per ovary pair and meiotic maturation ability of oocytes in different seasons; and by the observations on developmental competence of the parthenogenetic oocytes and cloned embryos collected from different seasons. We found that the number of COCs per ovary pairs was significantly higher in spring compared to the other seasons (P<0.05). However, no significant difference was found in the rate of oocytes progressing to MⅡ in four seasons (P<0.05). In addition, the rate of blastocyst formation of parthenogenetic embryos in summer was obviously declined compared to the other seasons (P<0.05), and an increased blastocyst rate of cloned porcine embryos was found in spring compared to autumn and winter (P<0.05). By combining the front anti-collision beam, crash box and NPR structure, a novel NPR bumper system for improving the crashworthiness is first proposed in the work. The performances of the NPR bumper system are detailed studied by comparing to traditional bumper system and aluminum foam filled bumper system. To achieve the rapid design while considering perturbation induced by parameter uncertainties, a multi-objective robust design optimization method of the NPR bumper system is also proposed. The parametric model of the bumper system is constructed by combining the full parametric model of the traditional bumper system and the parametric model of the NPR structure. Optimal Latin hypercube sampling technique and dual response surface method are combined to construct the surrogate models. The multi-objective robust optimization results of the NPR bumper system are then obtained by applying the multi-objective particle swarm optimization algorithm and six sigma criteria. | 20 | Increasing Fusarium verticillioides resistance in maize by genomicsassisted breeding:Methods,progress,and prospects显示文摘Maize(Zea mays L.)is an indispensable crop worldwide for food,feed,and bioenergy production.Fusarium verticillioides(F.verticillioides)is a widely distributed phytopathogen and incites multiple destructive diseases in maize:seedling blight,stalk rot,ear rot,and seed rot.As a soil-,seed-,and airborne pathogen,F.verticillioides can survive in soil or plant residue and systemically infect maize via roots,contaminated seed,silks,or external wounds,posing a severe threat to maize production and quality.Infection triggers complex immune responses:induction of defense-response genes,changes in reactive oxygen species,plant hormone levels and oxylipins,and alterations in secondary metabolites such as flavonoids,phenylpropanoids,phenolic compounds,and benzoxazinoid defense compounds.Breeding resistant maize cultivars is the preferred approach to reducing F.verticillioides infection and mycotoxin contamination.Reliable phenotyping systems are prerequisites for elucidating the genetic structure and molecular mechanism of maize resistance to F.verticillioides.Although many F.verticillioides resistance genes have been identified by genome-wide association study,linkage analysis,bulkedsegregant analysis,and various omics technologies,few have been functionally validated and applied in molecular breeding.This review summarizes research progress on the infection cycle of F.verticillioides in maize,phenotyping evaluation systems for F.verticillioides resistance,quantitative trait loci and genes associated with F.verticillioides resistance,and molecular mechanisms underlying maize defense against F.verticillioides,and discusses potential avenues for molecular design breeding to improve maize resistance to F.verticillioides. | Yufang Xu Zhirui Zhang Ping Lu Ruiqi Li Peipei Ma Jianyu Wu Tao Li Huiyong Zhang | 2023 | The Crop Journal2023,11,6: | 0 |
