1LSECtin on tumor-associated macrophages enhances breast cancer sternness via interaction with its receptor BTN3A3显示文摘Macrophages have bee n suggested to con tribute to con structing a cancer stem cell(CSC)niche.However,whether and how macrophages regulate the activity of CSCs through juxtacrine signaling are poorly understood.Here we report LSECtin,a transmembrane protein highly expressed on tumor-associated macrophages(TAMs),enhances sternness of breast cancer cells(BCCs).We identified BTN3A3,a B7 family member with previously unknown functions as the receptor for LSECtin on BCCs responsible for stemness-promoting effect of LSECtin.In mice bearing human tumor xenografts,either macrophage-specific ablation of LSECtin or silencing of BTN3A3 in BCCs decreased CSC frequency and tumor growth.Admixture of LSECtin-positive macrophages increased the tumorigenic activity of BCCs dependent on BTN3A3.Disruption of the LSECtin-BTN3A3 axis with BTN3A3-Fc or anti-BTN3A3 mAb has a therapeutic effect on breast cancer.These findings define a juxtacrine signaling mechanism by which TAMs promote cancer stemness.Targeting this axis in the CSC niche may provide potential therapies to breast cancer.Di Liu Qian Lu Xing Wang Jun Wang Ning Lu Zefei Jiang Xiaopeng Hao Jianbin Li Jing Liu Pengbo Cao Guilin Peng Yuandong Tao Dianyuan Zhao Fuchu He Li Tang 2019Cell Research2019,29,5:8
2Connectivity in finite ad-hoc networks显示文摘Research on ad-hoc network connectivity has mainly focused on asymptotic results in the number of nodes in the network. For a one-dimensional ad-hoc network G1, assuming all the nodes are independently uniform distributed in a closed interval [0,Z](z ∈ R+), we derive a generic formula for the probability that the network is connected. The finite connected ad-hoc networks is analyzed. And we separately suggest necessary conditions to make the ad-hoc network to be connected in one and two dimensional cases, facing possible failed nodes (f-nodes). Based on the necessary condition and unit-disk assumption for the node transmission, we prove that the nodes of the connected two-dimensional ad-hoc networks (G2) can be divided into at most five different groups. 3Multivariate similarity clustering analysis:a new method regarding biogeography and its application in global insects显示文摘A new method,multivariate similarity clustering analysis(MSCA)method,was established for biogeographical distribution analyzing.General similarity formula(GSF),the core of MSCA method,can be used to calculate the similarity coefficients between 2 and among any≥3 geographical units.Taking the global insects as example,we introduced the steps to use of GSF and consequent clustering processes of this method in details.Firstly,geographical distributions of certain taxa(e.g.Insecta)were categorized into basic geographical units(BGUs);Secondly,similarity coefficients between 2 and among n BGUs were calculated using GSF.Thirdly,hierarchical clustering was conducted according to values of similarity coefficients(from high to low);then a clustering diagram was generated.Finally,a framework of biogeographical division map was established for the target taxa(e.g.Insecta).We concluded that the MSCA method was efficiently applied in analyzing the biogeographical distribution of given biological taxa;the geographical regions regarding global insects were categorized into 7 Realms with 20 sub-Realms based on the results of MSCA method.Xiaocheng SHEN Shujie ZHANG Qi SHEN Guilin HU Jiqi LU 2021Integrative Zoology2021,16,3:2
4Virus-inspired surface-nanoengineered antimicrobial liposome:A potential system to simultaneously achieve high activity and selectivity显示文摘Enveloped viruses such as SARS-CoV-2 frequently have a highly infectious nature and are considered effective natural delivery systems exhibiting high efficiency and specificity.Since simultaneously enhancing the activity and selectivity of lipopeptides is a seemingly unsolvable problem for conventional chemistry and pharmaceutical approaches,we present a biomimetic strategy to construct lipopeptide-based mimics of viral architectures and infections to enhance their antimicrobial efficacy while avoiding side effects.Herein,a surface-nanoengineered antimicrobial liposome(SNAL)is developed with the morphological features of enveloped viruses,including a moderate size range,lipid-based membrane structure,and highly lipopeptide-enriched bilayer surface.The SNAL possesses virus-like infection to bacterial cells,which can mediate high-efficiency and high-selectivity bacteria binding,rapidly attack and invade bacteria via plasma membrane fusion pathway,and induce a local“burst”release of lipopeptide to produce irreversible damage of cell membrane.Remarkably,viral mimics are effective against multiple pathogens with low minimum inhibitory concentrations(1.6-6.3μg mL1),high bactericidal efficiency of>99%within 2 h,>10-fold enhanced selectivity over free lipopeptide,99.8%reduction in skin MRSA load after a single treatment,and negligible toxicity.This bioinspired design has significant potential to enhance the therapeutic efficacy of lipopeptides and may create new opportunities for designing next-generation antimicrobials.Yin Shi Xiaoqian Feng Liming Lin Jing Wang Jiaying Chi Biyuan Wu Guilin Zhou Feiyuan Yu Qian Xu Daojun Liu Guilan Quan Chao Lu Xin Pan Jianfeng Cai Chuanbin Wu 2021Bioactive Materials2021,6,10:1

14Anthropogenic footprints are invading global habitats of Indo-Pacific humpback dolphins显示文摘As ecologically fragile areas,coastal zones are affected by both anthropogenic activities and climate change.However,the impacts of these factors on large nearshore mammals,such as Indo-Pacific humpback dolphins(IPHDs,Sousa chinensis),are poorly understood.Here,modeling revealed that the suitable habitats of IPHDs are affected mainly by the sea surface temperature(SST),and the habitat suitability decreases as the distance to the nearest coastline increases.In addition,anthropogenic activities involving demersal fishing,contamination and shipping have narrowed IPHD habitats and reduced the habitat suitability.We found that climate change will further narrow suitable habitats located farther than 7 km from coastlines and trigger habitat losses in the eastern Taiwan Strait by 2090-2100 under the Representative Concentration Pathway(RCP)8.5 scenario.The projected decreases in habitat suitability and area emphasize the urgency of establishing connected marine protected areas(MPAs)while considering climate change,intergovernmental cooperation,and public involvement.Yongquan Lu Guilin Liu William W.L.Cheung Yuyang Xian Weijia Chen Dandan Yu 2023Geography and Sustainability2023,4,1:0
15Effects of Different Drying Methods on Content of Paederosidic Acid in Paederia scandens显示文摘[Objectives]To establish a method to simultaneously determine the content of paederosidic acid in Paederia scandens dried by four different methods.[Methods]Reversed-phase high performance liquid chromatography(RP-HPLC)was adopted.The chromatographic column:Thermo SCIENTIFIC Hypersil GOLD Dim.(mm);mobile phase:acetonitrile-0.1%phosphoric acid aqueous solution;gradient elution;column temperature:30℃;flow rate:1 mL/min;detection wavelength:236 nm;injection volume:10μL.[Results]The linear range of the detection injection volume of paederosidic acid was 0.64-9.60μg(R=0.9992);the limit of quantity(LOQ)was 5.10 ng,and the limit of detection(LOD)was 1.36 ng;the RSD of the precision,stability and reproducibility test was all less than 3%;the sample recovery rate was 98.87%,RSD<3.00,n=6.The results show that the content of paederosidic acid in the shade-dried P.scandens is the highest,and the order of the content is P.scandens(dry in shade)>P.scandens(dried at 50℃)>P.scandens(dried at 60℃)>P.scandens(dried at 70℃).[Conclusions]This method is sensitive,reliable and reproducible,and can be used to simultaneously determine the content of paederosidic acid in P.scandens dried by four different methods.Jingrong LU Haisheng ZENG Silu HE Xiumei MA Dongmei HUANG Guilin YANG Chengtong LIU Qiji ZHOU 2020Medicinal Plant2020,11,6:0

18Determination of Polydatin in Rat Serum and Its Changes by UPLC Method显示文摘[Objectives]To establish a UPLC-UV method for the determination of polydatin in the serum of Wistar rats.[Methods]Acquity UPLC BEH shield RP18 column(1.7μm,2.1 mm×100 mm,Waters Corporation,USA)was used as the analytical column,acetonitrile-water(55∶45)was used as the mobile phase,the flow rate was 0.5 mL/min,and the column temperature was 30℃and the detection wavelength was 306 nm.[Results]The linear range of the established serum sample analysis method was 1.0-20.0μg/mL,and the correlation coefficient was r=0.9994;the intraday and interday RSD of Wistar rat serum was less than 3.0%,and the accuracy was higher than 90%.[Conclusions]This method is sensitive,accurate,and rapid.It is suitable for monitoring the concentration of polydatin in serum after intragastric administration,and can also be used for pharmacokinetics and bioavailability studies.Haisheng ZENG Zhiqiang WANG Meiyuan LU Chao ZENG Mengxia YANG Guilin YANG 2021Medicinal Plant2021,12,2:0
19Quantitative Evaluation of the Larval Density of Sea Potato Acaudina spp.with Mitochondrial Molecular Marker显示文摘The sea potato Acaudina spp.is a species of sea cucumber,belonging to the class Holothuroidea under Phylum Echino-dermata.It becomes blooming recently in the East Sea of China,causing serious ecological problems.However,until now there are no molecular data for its larval identification and population genetic analysis.In this study,we firstly screened a mtDNA fragment and demonstrated that it was the species-specific molecular marker for the identification of Acaudina spp.We then developed a quantitative polymerase chain reaction(PCR)method to evaluate the larval density of Acaudina spp.based on this molecular probe.Utilizing this method,we examined 116 plankton samples collected in four seasons from 13 stations along the coastal region in Fu-jian province,China.The results showed that the high larval density was presented at stations 1,2,and 3,which were near a quay in the coast.The larval density increased from April and reached the highest value in June and July,suggesting temperature might be the main environmental factor on the effects of its population distribution and density.Our work provides an important molecular tool for species identification and risk evaluation of a potentially invasive species.LI Xiang WANG Guilin LI Yunfan LIU Wei LU Chao GUO Yuchen HUANG Wen WANG Jinhui DONG Bo 2021Journal of Ocean University of China2021,20,3:0
20Strength of undisturbed and reconstituted frozen soil at temperatures close to 0 ℃显示文摘The strength of warm frozen soils in permafrost is fundamentally significant to estimate and predict the ground settlements from construction activities. A study was therefore initiated to assess the strength and its behaviors of undisturbed and reconstituted frozen soils at temperatures close to 0 ℃. A series of triaxial compression tests(TCT) were performed by using a developed testing apparatus and a matching specimen-preparation method. The confinement was applied from air pressure, the temperature in the specimen was maintained using two-end refrigeration, and multi-stage loading on a single specimen was adopted to determine the strength. The test results showed that the strength, both for the undisturbed and reconstituted frozen-soil specimens, was significantly dependent on the temperatures and independent of the applied confining pressures. Additionally, the strength of undisturbed frozen soils was about 1.6 times more than that for reconstituted frozen soils.
