3Discovery of spirooxindole-ferrocene hybrids as novel MDM2 inhibitors显示文摘A series of spirooxindole-ferrocene hybrids bearing five or four contiguous chiral centers were designed and synthesized via organocatalysis.In vitro protein binding and cellular proliferation assays suggested that compound 5 d was the most potent mouse double minute 2 homolog(MDM2)inhibitor.In addition,mechanistic studies indicated that compound 5 d suppressed MDM2-mediated p53 degradation,induced apoptosis and promoted oxidative damage.Molecular docking studies have suggested that 5 d binds to MDM2 by mimicking the Trp23 and Leu26 residues of p53.This work can provide a basis for the development of novel multifunctional MDM2 inhibitors.The further exploration of more derivatives from this library and additional investigation of organocatalysis application in the development of new molecules may generate new potential lead compounds for cancer-targeted therapy.Jun Mu Xin Xie Shanshan Xiong Yuehua Zhang Yuting Wang Qian Zhao Hongping Zhu Wei Huang Gu He 2021Chinese Chemical Letters2021,32,6:1
4Abnormal gonad development in Kit^(W-2Bao) mice caused by a Kit gene missense mutation显示文摘KitW-2Bao mice are single-gene autosomal dominant mutation mice with a B6 background that were bred in our laboratory. Heterozygotes had morphological characteristics including albinism of the abdomen, extremities, and tail, whereas the homozygotes had albinism of the body, black eyes, and infertility. The homozygous mutants showed small, structurally abnormal gonads, and lacked germ cells. Heterozygous male mice lacked germ cells in some contorted seminiferous tubules. This mutation has been mapped at 43.8 cM from the centromere in chromosome 5 by linkage analysis and Kit has been identified as the candidate gene. After Kit full-length mRNA amplification, it was found that a G to T conversion at position 1228 in the ORF changed the 410th amino acid from V to F. This amino acid change could affect the protein’s secondary structure. Heterozygous mutant mice were intercrossed and homozygous mutant mice were bred and genotyped. We found that no primordial germ cells (PGCs) appeared in the urogenital ridge area at fetus day 11.5 in the homozygotes. The number of PGCs also significantly decreased in heterozygotes. At fetus day 15.5, the differentiation of the testis tubule structure was unclear; as well, they contained no spermatogonia. Female homozygotes contained no primordial follicles in the ovary. The numbers of PGCs and primordial follicles were significantly decreased in heterozygous mice. 5Effects of residual chlorine on the mortality, grazing and respiration of Labidocera euchaeta (Copepoda)显示文摘In this study,the authors investigated the effects of residual chlorine on mortality,grazing and respiration of Labidocera euchaeta in laboratory.The grazing rate was evaluated by subtraction method of food concentration and respiration rate was measured using oxygen electrode.It was found that the lethal effect of residual chlorine on L.euchaeta increased with enhanced concentration and prolonged duration.The medium lethal concentration (LC 50 ) of chlorine for L.euchaeta in 24 h was about 0.58 mg/L and the safe concentration was about 0.21 mg/L.However,the grazing and respiration of L.euchaeta decreased by 32.6% and 18.9% when exposed to 0.2 mg/L residual chlorine for 4 h.It indicated that the physiological activities of zooplankton could be suppressed by the residual chlorine less than the safety concentration.Therefore,both survival and physiological activities of the organisms living in the thermal (nuclear) power plant discharging waters should be considered when carrying out the ecological risks assessment.MA Zengling LIN Hongping GU Xiaolian XU Zhaoli 2011Acta Oceanologica Sinica2011,30,2:0
