1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

3Hydroisomerization performance of platinum supported on ZSM-22/ZSM-23 intergrowth zeolite catalyst显示文摘Hydroisomerization catalysts Pt/ZSM-22,Pt/ZSM-23,and Pt/ZSM-22/ZSM-23 were prepared by supporting Pt on ZSM-22,ZSM-23,and intergrowth zeolite ZSM-22/ZSM-23,respectively.The typical physicochemical properties of these catalysts were characterized by X-Ray Diffraction(XRD),N2 absorption-desorption,Pyridine-Fourier Transform Infrared(Py-FTIR),Transmission Electron Microscopy(TEM),X-Ray Fluorescence(XRF),Scanning Electron Microscopy(SEM) and NH3-Temperature Programmed Desorption(NH3-TPD),and the performance of these catalysts in n-dodecane hydroisomerization was evaluated in a continuous down-flow fixed bed with a stainless steel tubular reactor.The characterization results indicated that the intergrowth zeolite ZSM-22/ZSM-23 possessed the dual structure of ZSM-22 and ZSM-23,and the catalyst Pt/ZSM-22/ZSM-23 had similar pores and weak acidity to Pt/ZSM-22 and Pt/ZSM-23 catalysts.Moreover,Pt/ZSM-22/ZSM-23 catalyst showed a high selectivity in hydroisomerization of long chain n-alkanes to mono-branched isomers.The evaluation results for n-dodecane hydroisomerization indicated that the activity of Pt/ZSM-22/ZSM-23 was the lowest,while the hydroisomerization selectivity was the highest among the three catalysts.The maximum yield of i-dodecane product was 68.3% over Pt/ZSM-22/ZSM-23 at 320 oC.Chi Kebin Zhao Zhen Tian Zhijian Hu Sheng Yan Lijun Li Tianshu Wang Bingchun Meng Xiangbin Gao Shanbin Tan Mingwei Liu Yanfeng 2013Petroleum Science2013,10,2:8
4Neural Mechanisms of Mental Fatigue Revisited: New Insights from the Brain Connectome显示文摘Maintaining sustained attention during a prolonged cognitive task often comes at a cost: high levels of mental fatigue. Heuristically, mental fatigue refers to a feeling of tiredness or exhaustion, and a disengagement from the task at hand;it manifests as impaired cognitive and behavioral performance. In order to effectively reduce the undesirable yet preventable consequences of mental fatigue in many real-world workspaces, a better understanding of the underlying neural mechanisms is needed, and continuous efforts have been devoted to this topic. In comparison with conventional univariate approaches, which are widely utilized in fatigue studies, convergent evidence has shown that multivariate functional connectivity analysis may lead to richer information about mental fatigue. In fact, mental fatigue is increasingly thought to be related to the deviated reorganization of functional connectivity among brain regions in recent studies. In addition, graph theoretical analysis has shed new light on quantitatively assessing the reorganization of the brain functional networks that are modulated by mental fatigue. This review article begins with a brief introduction to neuroimaging studies on mental fatigue and the brain connectome, followed by a thorough overview of connectome studies on mental fatigue. 5Mining geographic episode association patterns of abnormal events in global earth science data显示文摘Abnormal events in earth science have great influence on both the natural envi-ronment and the human society. Finding association patterns among these events has great significance. Because data in earth science has characteristics of mass,high dimension,spatial autocorrelation and time delay,existing mining technolo-gies cannot be directly used on it. We propose a RSNN (range-based searching nearest neighbors) spatial clustering algorithm to reduce the data size and auto-correlation. Based on the clustered data,we propose a GEAM (geographic episode association pattern mining) algorithm which can deal with events time lags and find interesting patterns with specific constraints,to mine the association patterns. We carried out experiments on global climate datasets and found many interesting association patterns. 7Enhancing corrosion resistance of ZK60 magnesium alloys via Ca microalloying: The impact of nanoscale precipitates显示文摘Enhancing corrosion resistance of Mg-Zn alloys with high strength and low cost was critical for broadening their large-scale practical applications. Here we prepared solutionized, peak-and over-aged ZK60 alloys with and without microalloying Ca(0.26 wt.%) to explore the effects of nanoscale precipitates on their corrosion behavior in detail via experimental analyses and theoretical calculations. The results suggested the peak-aged ZK60 alloy with Ca addition showed improved corrosion resistance in comparison with the alloys without Ca,owing to the contribution of Ca on the refinement of precipitates and increase in their number density. Although the precipitates and Mg matrix formed micro-galvanic couples leading to dissolution, the fine and dense precipitates could generate “in-situ pinning” effect on the corrosion products, forming a spider-web-like structure and improving the corrosion inhibition ability accordingly. The pinning effect was closely related to the size and number density of precipitates.
