1Collagen scaffolds modified with collagen-binding bFGF promotes the neural regeneration in a rat hemisected spinal cord injury model显示文摘Nerve conduit is one of strategies for spine cord injury(SCI)treatment.Recently,studies showed that biomaterials could guide the neurite growth and promote axon regeneration at the injury site.However,the scaffold by itself was difficult to meet the need of SCI functional recovery.The basic fibroblast growth factor(bFGF)administration significantly promotes functional recovery after organ injuries.Here,using a rat model of T9 hemisected SCI,we aimed at assessing the repair capacity of implantation of collagen scaffold(CS)modified by collagen binding bFGF(CBD-bFGF).The results showed that CS combined with CBD-bFGF treatment improved survival rates after the lateral hemisection SCI.The CS/CBD-bFGF group showed more significant improvements in motor than the simply CS-implanted and untreated control group,when evaluated by the 21-point Basso-Beattie-Bresnahan(BBB)score and footprint analysis.Both hematoxylin and eosin(H&E)and immunohistochemical staining of neurofilament(NF)and glial fibrillary acidic protein(GFAP)demonstrated that fibers were guided to grow through the implants.These findings indicated that administration of CS modified with CBD-bFGF could promote spinal cord regeneration and functional recovery.SHI Qin GAO Wei HAN XingLong ZHU XueSong SUN Jie XIE Fang HOU XiangLin YANG HuiLin DAI JianWu CHEN Liang 2014Science China(Life Sciences)2014,57,2:23

3The novel OCT4 spliced variant OCT4B1 can generate three protein isoforms by alternative splicing into OCT4B显示文摘OCT4 is one of the key transcription factors in maintaining the pluripotency and self-renewal of embryonic stem (ES) cells.Human OCT4 can generate two isoforms OCT4A and OCT4B by alternative splicing.OCT4B1 is a recently discovered novel OCT4 spliced variant,which has been considered as a putative marker of stemness.Compared with the OCT4B mRNA,OCT4B1 mRNA is generated by retaining intron 2 as a cryptic exon which contains a TGA stop codon in it.As a result,the protein product of OCT4B1 mRNA could be truncated.Interestingly,we present here that OCT4B1 can indirectly produce the same protein products as OCT4B.We have demonstrated that OCT4B1 mRNA can be spliced into OCT4B mRNA,and encode three protein isoforms.The splicing of OCT4B1 mRNA into OCT4B mRNA can be remarkably inhibited by the mutation of the classical splicing site.Our result suggests that OCT4B mRNA may originate from OCT4B1 mRNA by alternative splicing.Yuan Gao Xia Wang Jin Han Zhifeng Xiao Bing Chen Guannan Su Jianwu Dai 2010Journal of Genetics and Genomics2010,37,7:6
4Seasonal and Interannual Variations of Carbon Exchange over a Rice–Wheat Rotation System on the North China Plain显示文摘Rice–wheat(R–W) rotation systems are ubiquitous in South and East Asia,and play an important role in modulating the carbon cycle and climate. Long-term,continuous flux measurements help in better understanding the seasonal and interannual variation of the carbon budget over R–W rotation systems. In this study,measurements of CO2 fluxes and meteorological variables over an R–W rotation system on the North China Plain from 2007 to 2010 were analyzed. To analyze the abiotic factors regulating Net Ecosystem Exchange(NEE),NEE was partitioned into gross primary production(GPP) and ecosystem respiration. Nighttime NEE or ecosystem respiration was controlled primarily by soil temperature,while daytime NEE was mainly determined by photosythetically active radiation(PAR). The responses of nighttime NEE to soil temperature and daytime NEE to light were closely associated with crop development and photosynthetic activity,respectively. Moreover,the interannual variation in GPP and NEE mainly depended on precipitation and PAR. Overall,NEE was negative on the annual scale and the rotation system behaved as a carbon sink of 982 g C m-2per year over the three years. The winter wheat field took up more CO2 than the rice paddy during the longer growing season,while the daily NEE for wheat and rice were-2.35and-3.96 g C m-2,respectively. 7FEEDBACK LINEARIZATION CONTROL FOR ELECTRONICALLY CONTROLLABLE CLUTCH OF VEHICLE显示文摘An investigation is made to the friction chutch engagement control of automotive AMT systems based on a nonlinear dynamic model with double inputs. According to friction torque transmission characteristics during clutch engagement, an equivalent, fully controllable and linearized model and the feedback linearization control are derived from the original system with nonlinearities via homomorphic transforms. By the resulting mathematical modeling, computer simulations are made both for the original nonlinear and feedback linearized systems with incorporation of ordinary PID controllers to follow ideal vehicle dynamic responses. 8Puerarin accelerates neural regeneration after sciatic nerve injury显示文摘Puerarin is a natural isoflavone isolated from plants of the genus Pueraria and functions as a protector against cerebral ischemia. We hypothesized that puerarin can be involved in the repair of peripheral nerve injuries. To test this hypothesis, doses of 10, 5, or 2.5 mg/kg per day puerarin(8-(β-D-Glucopyranosyl-7-hydroxy-3-(4-hydroxyphenyl)-4H-1-benzopyran-4-one) were injected intraperitoneally into mouse models of sciatic nerve injury. Puerarin at the middle and high doses significantly up-regulated the expression of growth-associated protein 43 in the L4–6 segments of the spinal cord from mice at 1, 2, and 4 weeks after modeling, and reduced the atrophy of the triceps surae on the affected side and promoted the regeneration of nerve fibers of the damaged spinal cord at 8 weeks after injury. 9Seasonal Responses of Net Primary Productivity of Vegetation to Phenological Dynamics in the Loess Plateau, China显示文摘With global warming, the great changes in the patterns of plant growth have occurred. The conditions in early spring and late autumn have changed the process of vegetation photosynthesis, which are expected to have a significant impact on net primary productivity(NPP) and affect the global carbon cycle. Currently, the seasonal response characteristics of NPP to phenological changes in dryland ecosystems are still not well defined. This article calibrated and analyzed the normalized difference vegetation index(NDVI)time series of Advanced Very-High-Resolution Radiometer(AVHRR) data from 1982 to 2015 in the Loess Plateau, China. The spatial and temporal distributions of vegetation phenology and NPP in the Loess Plateau under semihumid and semiarid conditions were investigated. The seasonal variation in the NPP response to vegetation phenology under the climate change was also analyzed. 10Microvascular endothelial cells derived from spinal cord promote spinal cord injury repair显示文摘Neural regeneration after spinal cord injury (SCI) closely relates to the microvascular endothelial cell (MEC)- mediated neurovascular unit formation. However, the effects of central nerve system-derived MECs on neovascularization and neurogenesis, and potential signaling involved therein, are unclear. Here, we established a primary spinal cord-derived MECs (SCMECs) isolation with high cell yield and purity to describe the differences with brain-derived MECs (BMECs) and their therapeutic effects on SCI. Transcriptomics and proteomics revealed differentially expressed genes and proteins in SCMECs were involved in angiogenesis, immunity, metabolism, and cell adhesion molecular signaling was the only signaling pathway enriched of top 10 in differentially expressed genes and proteins KEGG analysis. SCMECs and BMECs could be induced angiogenesis by different stiffness stimulation of PEG hydrogels with elastic modulus 50-1650 Pa for SCMECs and 50-300 Pa for BMECs, respectively. Moreover, SCMECs and BMECs promoted spinal cord or brain-derived NSC (SNSC/BNSC) proliferation, migration, and differentiation at different levels. At certain dose, SCMECs in combination with the NeuroRegen scaffold, showed higher effectiveness in the promotion of vascular reconstruction. The potential underlying mechanism of this phenomenon may through VEGF/AKT/eNOS- signaling pathway, and consequently accelerated neuronal regeneration and functional recovery of SCI rats compared to BMECs. 18Differential effects of recombinant fusion proteins TAT-OCT4 and TAT-NANOG on adult human fibroblasts显示文摘OCT4 and NANOG are two important tran-scription factors for maintaining the pluripotency and self-renewal abilities of embryonic stem(ES)cells.Meanwhile they play key roles in the induced pluripotent stem(iPS)cells.In this study,recombinant transcript factors TAT-NANOG and TAT-OCT4,which contained a fused powerful protein transduction domain(PTD)TAT from human immunodeficiency virus(HIV),were produced.Each fusion protein could be transported into human adultfibroblasts(HAF)successfully and activated the endogen-ous transcription of both nanog and oct4.Our study revealed the inter-regulation and autoregulation abilities of solo oct4 or nanog in the process of iPS cell reprogram-ming.Meanwhile the transduction of TAT-NANOG could accelerate the growth rate of HAF cells,and the key cell cycle regulator cdc25a was up-regulated.Thus cdc25a may be involved in the regulation of cell growth by NANOG.In addition,the TAT fusion protein technology provided a novel way to improve cell growth that is more controllable and safer.Jiani CAO Zhifeng XIAO Bing CHEN Yuan GAO Chunying SHI Jinhuan WANG Jianwu DAI 2010Frontiers in Biology2010,5,5:0
