| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | Effect of Astragalus membranaceus injection on the activity of the intestinal mucosal mast cells after hemorrhagic shock-reperfusion in rats显示文摘Background The mechanism of mucosal damage induced by ischemia-reperfusion (IR) after hemorrhagic shock is complex; mast cells (MC) degranulation is associated with the mucosal damage. Astragalus membranaceus can protect intestinal mucosa against intestinal oxidative damage after hemorrhagic shock, and some antioxidant agents could prevent MC against degranulation. This study aimed to observe the effects of astragalus membranaceus injection on the activity of intestinal mucosal mast cells (IMMC) after hemorrhage shock-reperfusion in rats Methods Thirty-two Wistar rats were randomly divided into the normal group, model group, low dosage group, (treated with Astragalus membranacaus injection, 10 g crude medication/kg) and high dosage group (treated with Astragalus membranacaus injection, 20 g crude medication/kg). The rat model of hemorrhagic shock-reperfusion was induced by hemorrhage for 60 minutes followed by 90 minutes of reperfusion. The animals were administrated with 3 ml of the test drug solution before reperfusion. At the end of study, intestinal pathology, ultrastructure of IMMC, and expression of tryptase were assayed. The levels of malondisldehyde (MDA), TNF-α, histamine, and superoxide dismutase (SOD) activity in intestine were detected, and the number of IMMC was counted. Results The Chiu’s score of the rats in the model group was higher than in other groups (P<0.01). The Chiu’s score in the high dosage group was higher than that in the low dosage group (P<0.05). Hemorrhage-reperfusion induced IMMC degranulation: Astragalus membranaceus injection attenuated this degranulation. Expression of tryptase and the number of IMMC in the model group increased compared with the other groups (P<0.01) and was significantly reduced by the treatments of Astragalus membranaceus injection at both doses. There was no significant difference between the two treatment groups (P>0.05). MDA content and concentration of TNF-α in the model group were higher than that in the other three groups (P<0.05), and the concentration of TNF-α in the low dosage group was higher than that in the high dosage group (P<0.05). SOD activity and the concentration of histamine in the model group were lower than the other three groups (P<0.05). There was a negative correlation betweenthe Chiu’s score and the concentration of histamine and a positive correlation between the Chiu’s score and the concentration of TNF-α and between the SOD activity and the concentration of histamine in the four groups (P<0.05). | 4 | Commissioning progress of the FAST显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) was completed with its main structure installed on September 25, 2016, after which it entered the commissioning phase. This paper aims to introduce the commissioning progress of the FAST over the past two years. To improve its operational reliability and ensure effective observation time, FAST has been equipped with a real-time information system for the active reflector system and hierarchical commissioning scheme for the feed support system, which ultimately achieves safe operation of the two systems. For meeting the high-performance indices, a highprecision measurement system was set up based on the effective control methods that were implemented for the active reflector system and feed support system. Since the commissioning of the FAST, a low-frequency ultra-wideband receiver and 19-beam1.05-1.45 GHz receiver have been mainly used. Telescope efficiency, pointing accuracy, and system noise temperature were completely tested and ultimately achieved the acceptance indices of the telescope. The FAST has been in the process of national acceptance preparations and has begun to search for pulsars. | 5 | A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. | 8 | Sedimentary Characteristics and Model of Gravity Flow Depositional System for the First Member of Upper Miocene Huangliu Formation in Dongfang Area, Yinggehai Basin, Northwestern South China Sea显示文摘The gravity flow deposit were mainly developed in the lowstand systems tract(LST) of the first member of Upper Miocene Huangliu Formation(Ehl1) in Dongfang area, Yinggehai Basin, has become a valuable target for gas exploration and production. The gravity flow sedimentary characteristics of lithofacies associations, sedimentary texture, seismic facies and logging facies were described in detail on the basis of integrated analysis of cores, logging and seismic data. The sedimentary microfacies types composed of neritic sandbar, continental shelf mud, main channel, bifurcated or cross-cutting distributary channel, overspill, and natural levee are revealed under the constraint of high resolution sequence stratigraphic framework in the Ehl1. The gravity flow deposit system in the LST is divided into three evolution stages corresponding to periods of three parasequence sets. The gravity flow deposit was induced in the early LST, expanded rapidly in the middle LST and decreased slightly in the late LST. But its developing scale decreased sharply in the transgression systems tract(TST) and finally vanished in the highstand systems tract(HST). This spatial evolution rule is constrained by the integrated function of sediments supply of the Vietnam Blue River in the LST, the development of local gradient change in sea floor(micro-topography, i.e., flexure slope break), and the fall in relative sea level. | 10 | Protective effects of human umbilical cord mesenchymal stem cell vein transplantation against spinal cord ischemia/reperfusion injury in rats显示文摘BACKGROUND: The majority of studies addressing spinal cord ischemia/reperfusion injury (SCIRI) have focused on drugs, proteins, cytokines, and various surgical techniques. A recent study reports that human umbilical cord mesenchymal stem cell (hUCMSC) transplantation achieves good therapeutic effects, but the mechanisms underlying nerve protection remain poorly understood. OBJECTIVE: To observe survival of transplanted hUCMSCs in SCIRI rat models and the influence on motor function in the hind limbs, to determine interleukin-8 expression and cellular apoptosis in spinal cord tissues, and to verify the hypothesis that hUCMSC transplantation exhibits protective effects on SCIRI. DESIGN, TIME AND SETTING: A randomized, controlled, animal experiment was performed at the Laboratory of the Department of Orthopedics in the First Affiliated Hospital of Soochow University, China between January 2007 and December 2008. MATERIALS: hUCMSCs were harvested from umbilical cord blood of healthy pregnant women after parturition in the Obstetrical Department of the First Affiliated Hospital of Soochow University, China. Rabbit anti-human BrdU monoclonal antibody was provided by DAKO, USA. Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) Kit and enzyme-linked immunosorbent assay (ELISA) Kit were purchased by Wuhan Boster, China. METHODS: A total of 72 healthy, Wistar, adult rats were randomly assigned to three groups: sham-surgery, model, and transplantation, with 24 rats in each group. SCIRI was induced in the model and transplantation groups via the abdominal aorta block method. The infrarenal abdominal aorta was not blocked in the sham-surgery group. Prior to abdominal aorta occlusion, 0.2-0.3 mL bromodeoxyuridine (BrdU)-labeled hUCMSCs suspension (cell concentration 5 × 103/μL) was injected through the great saphenous vein of the hind limb, and an equal volume of physiological saline was administered to the model and sham-surgery groups. MAIN OUTCOME MEASURES: Pathological observation of rat spinal cord tissues was performed by hematoxylin-eosin staining at 6, 24, and 48 hours post-surgery. Immunohistochemistry was applied to determine hUCMSCs survival in the spinal cord. The amount of cellular apoptosis and interleukin-8 expression in spinal cord tissues was assayed utilizing the TUNEL and ELISA methods, respectively. Motor function in the hind limbs was evaluated according to Jacob's score. RESULTS: Numerous BrdU-positive cells were observed in spinal cord tissues from the transplantation group. The number of apoptotic cells and interleukin-8 levels significantly decreased in the transplantation group (P < 0.05), pathological injury was significantly ameliorated, and motor function scores significantly increased (P < 0.05) compared with the model group. CONCLUSION: Via vein transplantation, hUCMSCs were shown to reach and survive in the injury area. Results suggested that the transplanted hUCMSCs contributed to significantly improved pathological changes in the injured spinal cord, as well as motor function, following SCIRI. | 11 | Effect of a transverse plasma jet on a shock wave induced by a ramp显示文摘We conducted experiments in a wind tunnel with Mach number 2 to explore the evolution of a transverse plasma jet and its modification effect on a shock wave induced by a ramp with an angle of 24°. The transverse plasma jet was created by arc discharge in a small cylindrical cavity with a 2 mm diameter orifice. Three group tests with different actuator arrangements in the spanwise or streamwise direction upstream from the ramp were respectively studied to compare their disturbances to the shock wave. As shown by a time-resolved schlieren system, an unsteady motion of the shock wave by actuation was found: the shock wave was significantly modified by the plasma jet with an upstream motion and a reduced angle. Compared to spanwise actuation, a more intensive impact was obtained with two or three streamwise actuators working together. From shock wave structures, the control effect of the plasma jet on the shock motion based on a thermal effect, a potential cause of shock modification, was discussed. Furthermore, we performed a numerical simulation by using the Improved Delayed Detached Eddy Simulation(IDDES) method to simulate the evolution of the transverse plasma jet plume produced by two streamwise actuators. The results show that flow structures are similar to those identified in schlieren images. | 12 | Characterizing the antibiotic resistance genes in a river catchment: Influence of anthropogenic activities显示文摘Previous studies on environmental antibiotics resistance genes(ARGs) have focused on the pollution sources such as wastewater treatment plants, aquaculture and livestock farms,etc. Few of them had addressed this issue in a regional scale such as river catchment. Hence,the occurrence and abundances of 23 ARGs were investigated in surface water samples collected from 38 sites which located from the river source to estuary of the Beijiang River.Among them, 11 ARGs were frequently detected in this region and 5 ARGs(sul I, sul II, tet B,tet C, and tet W) were selected for their distribution pattern analysis. The abundances of the selected ARGs were higher in the upstream(8.70 × 10~6 copies/ng DNA) and downstream areas(3.17 × 10~6 copies/ng DNA) than those in the midstream areas(1.23 × 10~6 copies/ng DNA), which was positively correlated to the population density and number of pollution sources. Pollution sources of ARGs along the Beijiang River not only had a great impact on the abundances and diversity, but also on the distribution of specific ARGs in the water samples. Both sul I and sul II were likely originated from aquaculture farms and animal farms,tet W gene was possibly associated with the mining/metal melting industry and the electric waste disposal and tet C gene was commonly found in the area with multiple pollution sources.However, the abundance of tet B was not particularly related to anthropogenic impacts. | 14 | The Interplay between Artificial Intelligence and Fog Radio Access Networks显示文摘The interplay between artificial intelligence(AI) and fog radio access networks(F-RANs) is investigated in this work from two perspectives: how F-RANs enable hierarchical AI to be deployed in wireless networks and how AI makes F-RANs smarter to better serve mobile devices. Due to the heterogeneity of processing capability, the cloud, fog, and device layers in F-RANs provide hierarchical intelligence via centralized, distributed, and federated learning. In addition, cross-layer learning is also introduced to further reduce the demand for the memory size of the mobile devices. On the other hand, AI provides F-RANs with technologies and methods to deal with massive data and make smarter decisions. Specifically, machine learning tools such as deep neural networks are introduced for data processing, while reinforcement learning(RL) algorithms are adopted for network optimization and decisions. | 15 | Does Postoperative Rehabilitation for Radical Cystectomy Call for Enhanced Recovery after Surgery?A Systematic Review and Metaanalysis显示文摘The aim of this review was to systematically compare the outcomes of enhanced recovery after surgery(ERAS)with standard care(SC)after radical cystectomy.We performed a systematic search of PubMed,Ovid?Web of Science,and the Cochrane Library to identify studies published until September 2017 which involved a comparison of ERAS and SC.A meta-analysis was performed to assess the outcomes of ERAS versus SC.Sixteen studies including 8 prospective and 8 retrospective trials met the eligibility criteria.A total of 2100 participants were assigned to ERAS(1258 cases)or SC(842 cases).The time to first flatus passage[WMD=-0.95 days,95%Cl(-1.50,-0.41),P=0.0006],time until return to a regular diet[WMD=-2.15 days,95%Cl(-2.86,—1.45),P<0.00001]and the length of hospital stay[WMD=-3.75 days,95%Cl(-5.13,-2.36),P<0.00001]were significantly shorter,and the incidence of postoperative complications[OR=0.60,95%Cl(0.44,0.83),P=0.002],especially postoperative paralytic ileus[OR=0.43,95%Cl(0.30,0.62),P<0.00001]and cardiovascular complications[OR=0.28,95%Cl(0.09,0.90),P=0.03]was significantly lower in the ERAS group than those in the SC group.This meta-analysis demonstrated that ERAS was associated with a shorter time to first flatus passage,return of bowel fimction,and the length of hospital stay than SC in patients undergoing radical cystectomy,as well as a lower rate of postoperative complications,especially paralytic ileus and cardiovascular complications. | Jun XIAO Meng WANG Wei HE Jing WANG Fan YANG Xue-you MA Yu ZANG Chun-guang YANG Gan YU Zhi-hua Wang Zhang-qun YE | 2019 | Current Medical Science2019,39,1: | 5 |
| 16 | Garcinia xanthochymus extract protects PC12 cells from H2O2-induced apoptosis through modulation of PI3K/AKT and NRF2/HO-1 pathways显示文摘The aim of the present study was to investigate the protective effects and underlying mechanisms of Garcinia xanthochymus, a perennial medicinal plant native to Yunnan, China, against H2 O2-induced oxidative damage in rat pheochromacytoma PC12 cells. Preincubation of PC12 cells with fruit Et OAc fraction(fruit-EFr., 12.5–50 μmol·L^(-1)) of G. xanthochymus for 24 h prior to H_2O_2 exposure markedly improved cell viability and increased the activities of antioxidant enzymes(superoxide dismutase, catalase, and heme oxygenase-1 [HO-1]), prevented lactate dehydrogenase release and lipid peroxidation malondialdehyde production, attenuated the decrease of matrix metalloproteinases(MMP), and scavenged reactive oxygen species(ROS). Fruit-EFr. also reduced BAX and cytochrome C expression and improved BCL-2 expression, thereby decreasing the ratio of BAX to BCL-2. Fruit-EFr. activated the nuclear translocation of NRF2 to increase HO-1 and induced the phosphorylation of AKT. Its cytoprotective effect was abolished by LY294002, a specific inhibitor of PI3 K. | 19 | The fundamental performance of FAST with 19-beam receiver at L band显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) has passed national acceptance and finished one pilot cycle of ‘Shared-Risk’ observations. It will start formal operation soon. In this context, this paper describes testing results of key fundamental parameters for FAST, aiming to provide basic support for observation and data reduction of FAST for scientific researchers. The 19-beam receiver covering 1.05–1.45 GHz was utilized for most of these observations. The fluctuation in electronic gain of the system is better than 1% over 3.5 hours, enabling enough stability for observations. Pointing accuracy,aperture efficiency and system temperature are three key parameters for FAST. The measured standard deviation of pointing accuracy is 7.9′′, which satisfies the initial design of FAST. When zenith angle is less than 26.4°, the aperture efficiency and system temperature around 1.4 GHz are ~0.63 and less than 24 K for central beam, respectively. The sensitivity and stability of the 19-beam backend are confirmed to satisfy expectation by spectral HI observations toward NGC 672 and polarization observations toward 3 C 286.
