| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | Immune Responses to the Attenuated Mycoplasma hyopneumoniae 168 Strain Vaccine by Intrapulmonic Immunization in Piglets显示文摘To investigate the immune responses to the attenuated Mycoplasma hyopneumoniae 168 strain vaccine, 8-15 d old piglets were immunized with M. hyopneumoniae 168 strain vaccine by intrapulmonic route. And the specific IgG antibody in serum, lymphoproliferation, IFNγ, and specific secretory IgA (SIgA) antibody in bronchoalveolar lavage fluid were detected on 30 and 60 d post-immunization (DPI), respectively. On 60 DPI, all the pigs except for those in health control group were challenged with a field M. hyopneumoniae strain JS. Necropsy was performed on 30 d post-challenge (DPC). The results showed that IFNγ and specific SIgA were stimulated on surface of respiratory tract after immunization. And peripheral blood mononuclear cells could also be proliferated about 1.81 and 2.12 fold on 30 and 60 DPI when stimulated by M. hyopneumoniae protein in vitro. However, no serum IgG antibody against M. hyopneumoniae was detected during the whole immune phage. After challenge, vaccinated pigs were observed with only very slight histological lesion in individual lobes. None of vaccinated pigs showed any clinical signs. While the unvaccinated pigs from challenge control group showed varying degrees of clinical sign and severe macroscopical lesion of mycoplasmal pneumonia of swine (MPS). | 6 | Research on Formation Mechanisms of Hot Dry Rock Resources in China显示文摘As an important geothermal resource, hot dry rock(HDR) reserves have been studied in many countries. HDR resources in China have huge capacity and have become one of the most important resources for the potential replacement of fossil fuels. However, HDR resources are difficult to develop and utilise. Technologies for use with HDR, such as high–temperature drilling, reservoir characterisation, reservoir fracturing, microseismic monitoring and high–temperature power stations, originate from the field of oil and drilling. Addressing how to take advantage of these developed technologies is a key factor in the development of HDR reserves. Based on the thermal crustal structure in China, HDR resources can be divided into four types: high radioactive heat production, sedimentary basin, modern volcano and the inner–plate active tectonic belt. The prospective regions of HDR resources are located in South Tibet, West Yunnan, the southeast coast of China, Bohai Rim, Songliao Basin and Guanzhong Basin. The related essential technologies are relatively mature, and the prospect of HDR power generation is promising. | 7 | Genotypic Characterization of Methicillin-resistant Staphylococcus aureus Isolated from Pigs and Retail Foods in China显示文摘Objective To investigate the genotypic diversity of Methicillin-resistant Staphylococcus aureus(MRSA) isolated from pigs and retail foods from different geographical areas in China and further to study the routes and rates of transmission of this pathogen from animals to food. Methods Seventy-one MRSA isolates were obtained from pigs and retail foods and then characterized by multi-locus sequencing typing(MLST), spa typing, multiple-locus variable number of tandem repeat analysis(MLVA), pulsed-field gel electrophoresis(PFGE), and antimicrobial susceptibility testing. Results All isolated MRSA exhibited multi-drug resistance(MDR). Greater diversity was found in food-associated MRSA(7 STs, 8 spa types, and 10 MLVA patterns) compared to pig-associated MRSA(3 STs, 1 spa type, and 6 MLVA patterns). PFGE patterns were more diverse for pig-associated MRSA than those of food-associated isolates(40 vs. 11 pulse types). Among the pig-associated isolates, CC9-ST9-t899-MC2236 was the most prevalent clone(96.4%), and CC9-ST9-t437-MC621(20.0%) was the predominant clone among the food-associated isolates. The CC9-ST9 isolates showed significantly higher antimicrobial resistance than other clones. Interestingly, CC398-ST398-t034 clone was identified from both pig-and food-associated isolates. Of note, some community-and hospital-associated MRSA strains(t030, t172, t1244, and t4549) were also identified as food-associated isolates. Conclusion CC9-ST9-t899-MC2236-MDR was the most predominant clone in pigs, but significant genetic diversity was observed in food-associated MRSA. | 10 | A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. | 11 | Chinese Herbal Medicine Combined with Entecavir for HBeAg Positive Chronic Hepatitis B:Study Protocol for a Multi-Center,Double-Blind Randomized-Controlled Trial显示文摘Background: The domestic prevalence of chronic hepatitis B(CHB) in China is 7.18% in 2006, imposing great societal healthcare burdens. Nucleot(s)ide analogues(NUCs) anti-hepatitis B virus(HBV) therapies are widely applied despite the relatively low rate of seroconversion and high risk of drug-resistant mutation. More effective treatments for CHB deserve further explorations. Combined therapy of NUCs plus Chinese herbal medicine(CHM) is widely accepted in China, which is recognized as a prospective alternative approach. The study was primarily designed to confirm the hypothesis that Tiaogan-Yipi Granule(调肝益脾颗粒, TGYP) or Tiaogan-Jianpi-Jiedu Granule(调肝健脾解毒颗粒, TGJPJD) plus entecavir tablet(ETV) was superior over ETV monotherapy in enhancing HBeA g loss rate. Methods: The study was a nationwide, large-scale, multi-center, double-blind, randomized, placebo-control ed trial with a designed duration of 108 weeks. A total of 16 hospitals and 596 eligible Chinese HBeA g positive CHB patients were enrolled from November 2012 to September 2013 and randomly allocated into 2 groups in 1:1 ratio via central randomization system: experimental group(EG) and control group(CG). Subjects in EG received CM formulae(TGYP or TGJPJD, 50 g per dose, twice daily) plus ETV tablet(or ETV placebo) 0.5 mg per day in the first 24 weeks(stage 1), and CHM granule plus ETV tablet(0.5 mg per day) from week 25 to 108(stage 2). Subjects in CG received CHM Granule placebo plus ETV tablet(0.5 mg per day) for 108 weeks throughout the trial. The assessments of primary outcomes(HBV serum markers and HBV-DNA) were conducted by a third-party College of American Pathologists(CAP) qualified laboratory. Adverse effects were observed in the hospitals of recruitment. Discussion: The study was designed to compare the curative effect of CM plus ETV and ETV monotherapy in respect of HBe Ag loss, which is recognized by the European Association for the Study of the Liver as 'a valuable endpoint'. We believe this trial could provide a reliable status for patients' 'journey' towards durable responses after treatment discontinuation. | 13 | How does the motor relearning program improve neurological function of brain ischemia monkeys?显示文摘The motor relearning program can significantly improve various functional disturbance induced by ischemic cerebrovascular diseases. However, its mechanism of action remains poorly understood. In injured brain tissues, glial fibrillary acidic protein and neurofilament protein changes can reflect the condition of injured neurons and astrocytes, while vascular endothelial growth factor and basic fibroblast growth factor changes can indicate angiogenesis. In the present study, we induced ischemic brain injury in the rhesus macaque by electrocoagulation of the M1 segment of the right middle cerebral artery. The motor relearning program was conducted for 60 days from the third day after model establishment. Immunohistochemistry and single-photon emission CT showed that the numbers of glial fibrillary acidic protein-, neurofilament protein-, vascular endothelial growth factor- and basic fibroblast growth factor-positive cells were significantly increased in the infarcted side compared with the contralateral hemisphere following the motor relearning program. Moreover, cerebral blood flow in the infarcted side was significantly improved. The clinical rating scale for stroke was used to assess neurological function changes in the rhesus macaque following the motor relearning program. Results showed that motor function was improved, and problems with consciousness, self-care ability and balance function were significantly ameliorated. | 14 | Elevated serum prostaglandin E2 predicts the risk of infection in hepatitis B virus-related acute-on-chronic liver failure patients显示文摘Objective: To evaluate the serum Prostaglandin E2(PGE2) level in Acute-on-chronic liver failure(ACLF) and determine its predicative value for infection.Methods: From April 2014 to April 2015, ninety-one patients with hepatitis B virus and ACLF but without infection were enrolled into this prospective study that was carried out at our Hospital. Twenty patients with stable chronic hepatitis B were enrolled from the outpatient department and twenty healthy control subjects without any disease were enrolled from hospital staff. Serum PGE2 levels were determined using ELISA at enrollment. Clinical and laboratory parameters were collected. Receiver operating characteristic(ROC) curves were used to determine optimal cut-off values to predict infection.Results: Significantly higher PGE2 levels were found in patients with ACLF in comparison with healthy controls and patients with stable CHB(P < 0.000 1). In ACLF patients, PGE2 levels were significantly higher in patients that eventually developed infection than those without this complication(P < 0.000 1). ROC analysis showed that serum PGE2(area under the ROC curve, 0.83) could predict infection in patients with ACLF with sensitivity of 78.4% and specificity of 81.5% using a threshold of 141 pg/m L.Conclusions: Serum PGE2 is associated with the susceptibility to secondary infections for patients with ACLF. The concentrations of total mercury (Hg) and stable isotope compositions of mercury in the peat core were measured using a LUMEX 915 instrument and multi-collector inductively coupled plasma mass spectrometer, respectively. Total mercury (Hg) concentrations in the peat core had a clearly increasing trend from the bottom to top of the core while δ 202Hg values (relative to NIST 3133) of peat had a decreasing trend. The total mercury (Hg) concentration varied from 16.7 to 101.3 ng/g and the δ 202Hg values ranged from –0.44‰± 0.14‰ to –1.45‰± 0.22‰. Utilizing a self-injection locking framework and a booster optical amplifier, the noise sidebands with relative amplitudes as high as 20 dB are completely suppressed.The RIN is remarkably reduced by more than 64 dB at the relaxation oscillation peak to retain below-150 dB∕Hz in a frequency range from 75 kHz to 50 MHz, while the quantum noise limit is -152.9 d B∕Hz.Furthermore, a laser linewidth narrower than 600 Hz, a polarization-extinction ratio of more than 23 dB, and an optical signal-to-noise ratio of more than 73 dB are acquired simultaneously. | 17 | Risk factors of interstitial lung diseases in clinically amyopathic dermatomyositis显示文摘Background:Clinically amyopathic dermatomyositis(CADM)is a unique sub-type of idiopathic inflammatory myopathies with a high prevalence of interstitial lung disease(ILD).Poor prognosis of the patients was strongly associated with rapid progressive ILD.The aim of this study was to identify risk factors for prediction of different types of ILD in CADM.Methods:In this study,data of 108 inpatients with CADM were collected,including 87 with ILD.The baseline clinical data and laboratory parameters,including myositis-specific and associated antibodies and tumor-associated antigens were analyzed to identify risk factors for acute or subacute interstitial pneumonitis(A/SIP)and chronic interstitial pneumonitis(CIP).Results:In 87 patients with CADM-ILD,39(36.1%)were A/SIP,and 48(44.4%)were CIP.There were 22(20.4%)patients with asymptomatic ILD who were detected by routine high resolution computed tomography.Cytokeratin-19 fragment(CYFRA21-1)was significantly higher in CADM-ILD than that in CADM patients without ILD;carcinoembryonic antigen and neuron-specific enolase were significantly elevated in A/SIP than that in CIP.Patients with A/SIP had a higher positive rate of anti-melanoma differentiation-associated gene 5(MDA5),while patients with CIP had a higher positive rate of anti PL-12 and anti-Ro-52.Logistic regression analysis indicated that elevation of CYFRA21-1 was a risk factor for ILD,higher titer of anti-MDA5 indicated increased likelihood for A/SIP,and higher titer of anti-Ro-52 was also clearly associated with CIP.Conclusions:This study indicated that the prevalence of ILD was high in CADM.Asymptomatic ILD has been previously underestimated.Anti-MDA5 was a risk factor for the presence of A/SIP,and CYFRA21-1 was a risk factor for ILD. | Yu-Zhou Gan Li-Hua Zhang Lin Ma Feng Sun Yu-Hui Li Yuan An Zhan-Guo Li Hua Ye | 2020 | Chinese Medical Journal2020,,6: | 7 |
| 18 | Modeling and simulation of urea-water-solution droplet evaporation and thermolysis processes for SCR systems显示文摘A reliable mathematical model of urea-water-solution(UWS) droplet evaporation and thermolysis is developed.The well known Abramzon–Sirignano evaporation model is corrected by introducing an adjustment coefficient considering the different evaporation behaviors of UWS droplet at different ambient temperatures. A semidetailed kinetic scheme of urea thermolysis is developed based on Ebrahimian's work. Sequentially, the evaporation characteristics, decomposition efficiency of a single UWS droplet and deposit formation are simulated. As a result, the relation of evaporation time, relative velocity, exhaust temperature and droplet initial diameter is presented. Synchronously, it indicates that temperature is the decisive factor for urea thermolysis. Different temperatures result in different deposit components, and deposit yield is significantly influenced by temperature and decomposition time. | 19 | The Lyman-alpha Solar Telescope(LST) for the ASO-S mission——Ⅲ. data and potential diagnostics显示文摘The Lyman-alpha Solar Telescope(LST)is one of the three payloads onboard the Advanced Space-based Solar Observatory(ASO-S)mission.It aims at imaging the Sun from the disk center up to 2.5 R⊙targeting solar eruptions,particularly coronal mass ejections(CMEs),solar flares,prominences/filaments and related phenomena,as well as the fast and slow solar wind.The most prominent speciality of LST is the simultaneous observation of the solar atmosphere in both Lyαand white light(WL)with high temporospatial resolution both on the solar disk and the inner corona.New observations in the Lyαline together with traditional WL observations will provide us with many new insights into solar eruptions and solar wind.LST consists of a Solar Corona Imager(SCI)with a field of view(FOV)of 1.1–2.5 R⊙,a Solar Disk Imager(SDI)and a full-disk White-light Solar Telescope(WST)with an identical FOV up to 1.2 R⊙.SCI has a dual waveband in Lyα(121.6±10 nm)and in WL(700±40 nm),while SDI works in the Lyαwaveband of 121.6±7.5 nm and WST works in the violet narrow-band continuum of 360±2.0 nm.To produce high quality science data,careful ground and in-flight calibrations are required.We present our methods for different calibrations including dark field correction,flat field correction,radiometry,instrumental polarization and optical geometry.Based on the data calibration,definitions of the data levels and processing procedures for the defined levels from raw data are described.Plasma physical diagnostics offer key ingredients to understand ejecta and plasma flows in the inner corona,as well as different features on the solar disk including flares,filaments,etc.Therefore,we are making efforts to develop various tools to detect the different features observed by LST,and then to derive their physical parameters,for example,the electron density and temperature of CMEs,the outflow velocity of the solar wind,and the hydrogen density and mass flows of prominences.Coordinated observations and data analyses with the coronagraphs onboard Solar Orbiter,PROBA-3,and Aditya are also briefly discussed. | Li Feng Hui Li Bo Chen Ying Li Roberto Susino Yu Huang Lei Lu Bei-Li Ying Jing-Wei Li Jian-Chao Xue Yu-Tong Yang Jie Hong Jian-Ping Li Jie Zhao Wei-Qun Gan Yan Zhang | 2019 | Research in Astronomy and Astrophysics2019,19,11: | 7 |
| 20 | Costunolide alleviates HKSA-induced acute lung injury via inhibition of macrophage activation显示文摘Staphylococcus aureus (S. aureus) infection leads to a severe inflammatory response and causes acute lung injury (ALI), eventually threatening human life. Therefore, it is of importance to find an agent to inhibit infiammation and reduce ALI. Here, we found that costunolide, a sesquiterpene lactone, displays anti-infiammatory effects and ameliorates heat-killed S. aureus (HKSA)-induced pneumonia. Costunolide treatment attenuated HKSA-induced murine ALI in which pulmonary neutrophil infltration was inhibited, lung edema was decreased, and the production of pro-inflammatory cytokines was significantly reduced. In addition, costunolide dose-dependently inhibited the generation of IL-6, TNF-α, IL-1β, and keratinocyte-derived cytokine (KC), as well as the expression of iNOS, in HKSA-induced macrophages. Furthermore, costunolide attenuated the phosphorylation of p38 MAPK and cAMP response element-binding protein (CREB).
