| 1 | Wnt/b-catenin signaling plays an ever-expanding role in stem cell self-renewal,tumorigenesis and cancer chemoresistance显示文摘Wnt signaling transduces evolutionarily conserved pathways which play important roles in initiating and regulating a diverse range of cellular activities,including cell proliferation,calcium homeostasis,and cell polarity.The role of Wnt signaling in controlling cell proliferation and stem cell self-renewal is primarily carried out through the canonical pathway,which is the best-characterized the multiple Wnt signaling branches.The past 10 years has seen a rapid expansion in our understanding of the complexity of this pathway,as many new components of Wnt signaling have been identified and linked to signaling regulation,stem cell functions,and adult tissue homeostasis.Additionally,a substantial body of evidence links Wnt signaling to tumorigenesis of cancer types and implicates it in the development of cancer drug resistance.Thus,a better understanding of the mechanisms by which dysregulation of Wnt signaling precedes the development and progression of human cancer may hasten the development of pathway inhibitors to augment current therapy.This review summarizes and synthesizes our current knowledge of the canonical Wnt pathway in development and disease.We begin with an overview of the components of the canonical Wnt signaling pathway and delve into the role this pathway has been shown to play in stemness,tumorigenesis,and cancer drug resistance.Ultimately,we hope to present an organized collection of evidence implicating Wnt signaling in tumorigenesis and chemoresistance to facilitate the pursuit of Wnt pathway modulators that may improve outcomes of cancers in which Wnt signaling contributes to aggressive disease and/or treatment resistance. | Maryam K.Mohammed Connie Shao Jing Wang Qiang Wei Xin Wang Zachary Collier Shengli Tang Hao Liu Fugui Zhang Jiayi Huang Dan Guo Minpeng Lu Feng Liu Jianxiang Liu Chao Ma Lewis L.Shi Aravind Athiviraham Tong-Chuan He Michael J.Lee | 2016 | Genes & Diseases2016,3,1: | 70 |
| 2 | The Chromosome-Level Genome Sequence of the Autotetraploid Alfalfa and Resequencing of Core Germplasms Provide Genomic Resources for Alfalfa Research显示文摘Alfalfa(Medicago sativa)is one of the most important forage crops in the world;however,its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome.Here,we report a de novo assembled 816-Mb high-quality,chromosome-level haploid genome sequence for‘Zhongmu No.1’alfalfa,a heterozygous autotetraploid.The contig N50 is 3.92 Mb,and 49165 genes are annotated in the genome.The alfalfa genome is estimated to have diverged from M.truncatula approximately 8 million years ago.Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity,weak population structure,and extensive gene flow from wild to cultivated alfalfa.Genome-wide association studies identified many candidate genes associated with important agronomic traits.Furthermore,we showed that MsFTa2,a Flowering Locus T homolog,whose expression is upregulated in salt-resistant germplasms,may be associated with fall dormancy and salt resistance.Taken together,these genomic resources will facilitate alfalfa genetic research and agronomic improvement. | Chen Shen Huilong Du Zhuo Chen Hongwei Lu Fugui Zhu Hong Chen Xiangzhao Meng Qianwen Liu Peng Liu Lihua Zheng Xiuxiu Li Jiangli Dong Chengzhi Liang Tao Wang | 2020 | Molecular Plant2020,13,9: | 21 |
| 3 | Type Synthesis of 4-DOF Parallel Kinematic Mechanisms Based on Grassmann Line Geometry and Atlas Method显示文摘Many methods are proposed to deal with the type synthesis of parallel kinematic mechanisms(PKMs),but most of them are less intuitive to some extent.Thus,to propose a concise and intuitive type synthesis method for engineering application is a very challenging issue,which should be further studied in the field.Grassmann line geometry,which can investigate the dimensions of spatial line-clusters in a concise way,is taken as the mathematic foundation.Atlas method is introduced to visually describe the degrees of freedom(DOFs)and constraints of a mechanism,and the dual rule is brought in to realize the mutual conversion of the freedom-space and constraint-space.Consequently,a systematic method based on Grassmann line geometry and Atlas method is generated and the entire type synthesis process is presented.Three type 4-DOF PKMs,i.e.,1T3R,2T2R and 3T1R(T:translational DOF;R:rotational DOF),are classified according to the different combinations of the translational DOFs and rotational DOFs.The type synthesis of4-DOF PKMs is carried out and the possible configurations are thoroughly investigated.Some new PKMs with useful functions are generated during this procedure.The type synthesis method based on Grassmann line geometry and Atlas method is intuitive and concise,and can reduce the complexity of the PKMs’type synthesis.Moreover,this method can provide theoretical guidance for other PKMs’type synthesis and engineering application.A novel type synthesis method is proposed,which solves the existing methods’problems in terms of complicated,not intuitive and unsuitable for practical application. | XIE Fugui LI Tiemin LIU Xinjun | 2013 | Chinese Journal of Mechanical Engineering2013,26,6: | 21 |
| 4 | Error Modeling and Sensitivity Analysis of a Parallel Robot with SCARA(Selective Compliance Assembly Robot Arm) Motions显示文摘Parallel robots with SCARA(selective compliance assembly robot arm) motions are utilized widely in the field of high speed pick-and-place manipulation. Error modeling for these robots generally simplifies the parallelogram structures included by the robots as a link. As the established error model fails to reflect the error feature of the parallelogram structures, the effect of accuracy design and kinematic calibration based on the error model come to be undermined. An error modeling methodology is proposed to establish an error model of parallel robots with parallelogram structures. The error model can embody the geometric errors of all joints, including the joints of parallelogram structures. Thus it can contain more exhaustively the factors that reduce the accuracy of the robot. Based on the error model and some sensitivity indices defined in the sense of statistics, sensitivity analysis is carried out. Accordingly, some atlases are depicted to express each geometric error's influence on the moving platform's pose errors. From these atlases, the geometric errors that have greater impact on the accuracy of the moving platform are identified, and some sensitive areas where the pose errors of the moving platform are extremely sensitive to the geometric errors are also figured out. | 9 | Design and experimental study of the SPKM165, a five-axis serial-parallel kinematic milling machine显示文摘A five-axis serial-parallel kinematic milling machine, the SPKM 165, is introduced. This machine consists of a three-degree of-freedom parallel module and a two-degree-of-freedom serial table. The SPKM 165 is capable of five-face machining. A discussion of the inverse kinematics of the five-axis control is provided. A dimensional synthesis procedure is presented in terms of motion/force transmissibility. Finite-element analysis was used to evaluate the stiffness of a CAD model before the machine was manufactured. Kinematic calibration was implemented to improve the accuracy of the end effector. The results of a calibration experiment are presented. The stiffness of the developed machine was then measured. | 13 | Beclin-1/LC3-II dependent macroautophagy was uninfluenced in ischemia-challenged vascular endothelial cells显示文摘Autophagy has been extensively studied and occurs in many biological settings.However,a question remains as to whether ischemia enhances Beclin-1/LC3-II-dependent macroautophagy in vascular endothelial cells,as has been previously thought.Furthermore,the effect of the level of autophagy on cell or skin flap survival still requires elucidation.We created a lethal ischemia model in human umbilical vascular endothelial cells(HUVECs),performed quantitative proteomics and bioinformatics analyses,and verified the autophagic status and effect both in vitro and in vivo.The significantly upregulated proteins encoded by autophagy-related genes(ATGs)included ATG2A,ATG3,ATG4B,ATG5,ATG7,ATG9A,ATG12,ATG16,and ATG101.The significantly enhanced lysosomal proteins were cathepsin B,cathepsin D,lysosome-associated membrane protein 1(LAMP1),and LAMP2.However,the differentially expressed proteins excluded Beclin-1,microtubule-associated protein light chain 3(LC3)-I,and LC3-II.Western blot analyses verified that the protein expression levels of Beclin-1,LC3-I,and LC3-II were neither upregulated nor downregulated in ischemia-challenged HUVECs.The autophagic status was not enhanced by rapamycin in ischemic HUVECs but appeared to be inhibited by chloroquine.Our in vivo study on rats showed that a downregulation in autophagic status jeopardized skin flap survival.In conclusion,Ischemia neither enhanced nor inhibited Beclin-1/LC3-II-dependent canonical macroautophagy both in vitro and in vivo,in contradiction to previous studies.An appropriate autophagic homeostasis can minimize cell or skin flap damage. | Yaping Ma Chaofan Li Yan He Tiwei Fu Li Song Qingsong Ye Fugui Zhang | 2022 | Genes & Diseases2022,9,2: | 3 |

Due to the combined influence of human activities and natural factors in the past half a century, water quality in the lower reaches of the Shiyang River decreased and ecological environment deteriorated, which influenced sustainable development of this area and surrounding area seriously. This paper analyzed current situation and reasons of the deteriorated ecological environment in the lower reaches of the Shiyang River, and proposed that water environment regulation was the primary task in the eco-environment control of Minqin area, specifically,(a) By using existing policies, accelerating the construction of water-saving agriculture in the middle reaches of the Shiyang River, water quantity in the lower reaches would be guanranted, water shortage for agricultural production and ecological use in Minqin oasis would be relieved;(b) Unreasonable human activities such as overexploitation of underground water should be controlled strictly;(c) Water conservancy project should be developed vigorously to ensure rate of flow. | 17 | Short-term effect of supplemental yeast extract without or with feed enzymes on growth performance, immune status and gut structure of weaned pigs challenged with Escherichia coli lipopolysaccharide显示文摘Background: This study investigated the response of piglets receiving a yeast extract without or with a multi-enzyme mixture compared with an antimicrobial growth promoter(AGP) on performance, immune status and gut structure after an E. coli lipopolysaccharide(LPS) challenge. Thirty-six pigs were allotted to six treatments including: a non-challenged control(NCC); LPS-challenged control(CC); CC + AGP; CC + yeast extract; CC + enzymes; and CC + enzymes + yeast extract. On d 7, pigs were bled and thereafter injected with LPS or sterile saline. Blood samples were collected at 6, 48, and 96 h post-challenge. After 96 h post-challenge, pigs were euthanized to obtain duodenal, jejunal and ileal samples.Results: Overall(d 1 to 11), compared with CC pigs, AGP attenuated the LPS-induced reduction in ADG(P = 0.004),ADFI(P = 0.03) and gain/feed ratio(P = 0.01). At 6 h post-challenge, AGP pigs had lower plasma urea N(PUN; P = 0.02)and serum TNF-α concentration(P = 0.07), and higher platelet count(P = 0.04) and serum IL-10 concentration(P = 0.02) than CC pigs. At 48 h post-challenge, AGP pigs had lower PUN(P = 0.02) than CC pigs, whereas enzymes + yeast extract interacted non-additively(P = 0.001) to reduce PUN. At 96 h post-challenge, AGP pigs had lower PUN(P = 0.02)and higher duodenal(P = 0.03), jejunal(P = 0.01) and ileal(P = 0.07) villus height than CC pigs. In addition, enzymes +yeast extract interacted additively and non-additively to reduce ileal IFN-γ(P < 0.0001) and IL-10(P = 0.012) expression,respectively. | 19 | Transcriptome analysis reveals genes expression pattern of seed response to heat stress in Brassica napus L.显示文摘Seeds might deteriorate,and lose the ability to germinate when stored under high temperature and high humidity.Brassica napus is one of the most important oil crops in China.However,B.napus seeds were generally stored through summer season with high ambient temperature,which lead to seed viability loss.In order to understand the mechanism of seed response to heat stress and improve seed heat tolerance,B.napus seeds were treated with high temperature(40℃)and ultra-high temperature(60℃)for 4 h.The germination of heatstressed seeds were obviously slower,the germination index and vigor index decreased with temperature increase from 40 to 60℃,and the 40℃ pretreatment could improve the seed tolerance to 60℃ heat stress.Transcriptomics results showed that 442 differentially expressed genes(DEGs)were identified in seeds after heat stress.Gene ontology and KEGG pathway enrichment analysis revealed that some of the genes were involved in posttranslational modification,protein turnover,chaperones and carbohydrate transport,metabolic pathways and secondary metabolites biosynthesis pathway.Among these DEGs,sHSP and transcription factors genes were involved in heat stress tolerance.Thirty-two overlapping genes under different high temperature stress(40℃ and 60℃)were enriched in biological processes of response to oxidative stress and abiotic stimulus.The expression trends of 12 genes randomly selected from the RNA-seq data were almost consistent with the results of qRT-PCR.Our results revealed several potential candidate genes and pathways related to heat responsive by high temperature,which is beneficial for further improving the heat tolerance in B.napus seeds. | Guizhen Gao Jihong Hu Xiaojun Zhang Fugui Zhang Mei Li Xiaoming Wu | 2021 | Oil Crop Science2021,6,2: | 1 |
