Variations of pore water sulfate gradients in sediments as indicator for underlying gas hydrate in Shenhu Area, the South China Sea显示文摘Shenhu Area is one of the most promising areas for gas hydrate exploration in the northern South China Sea (SCS). Pore water sulfate gradient, sulfate-methane interface (SMI) depth, and sulfate flux were analyzed at 53 sites in this area. SO42 gradient ranges between 0.33 and 4.43 mmol L-1 m-1 . SMI depths are from 7.7 to 87.9mbsf. Sulfate flux varies between 2.0 and 26.9 mmol m-2 yr-1 , with a mean of 11.7 mmol m-2 yr-1 . Correlation coefficient between SMI depth and methane flux for the 53 sites is 0.80, implying that methane flux regulates the rate of anaerobic methane oxidation (AMO), SMI depth, and sulfate flux. Twelve anomalous fields with high methane flux and steep sulfate gradients were recognized. Bottom simulating reflector (BSR) is distributed mainly in areas where SMI depth is less than 50 mbsf or places with sulfate flux larger than 3.5 mmol m-2 yr-1 . It is suggested that the Baiyun Sag and the Southern Uplift are potential areas for gas hydrate exploration. | WU LuShan YANG ShengXiong LIANG JinQiang SU Xin FU ShaoYing SHA ZhiBin YANG Tao | 2013 | Science China Earth Sciences2013,56,4: | 24
Gas Sources of Natural Gas Hydrates in the Shenhu Drilling Area, South China Sea: Geochemical Evidence and Geological Analysis显示文摘The Shenhu gas hydrate drilling area is located in the central Baiyun sag, Zhu Ⅱ depression, Pearl River Mouth basin, northern South China Sea. The gas compositions contained in the hydrate-bearing zones is dominated by methane with content up to 99.89% and 99.91%. The carbon isotope of the methane (δ13C1 ) are 56.7‰ and 60.9‰, and its hydrogen isotope (δD) are 199‰ and 180‰, respectively, indicating the methane from the microbial reduction of CO2 . Based on the data of measured seafloor temperature and geothermal gradient, the gas formed hydrate reservoirs are from depths 24-1699 m below the seafloor, and main gas-generation zone is present at the depth interval of 416-1165 m. Gas-bearing zones include the Hanjiang Formation, Yuehai Formation, Wanshan Formation and Quaternary sediments. We infer that the microbial gas migrated laterally or vertically along faults (especially interlayer faults), slump structures, small-scale diapiric structures, regional sand beds and sedimentary boundaries to the hydrate stability zone, and formed natural gas hydrates in the upper Yuehai Formation and lower Wanshan Formation, probably with contribution of a little thermogenic gas from the deep sedments during this process. | ZHU Youhai HUANG Xia FU Shaoying SU Pibo | 2013 | Acta Geologica Sinica(English Edition)2013,87,3: | 13
Elemental sulfur in northern South China Sea sediments and its significance显示文摘Elemental sulfur(ES) is one of the intermediates in the inorganic sulfur cycle and thus plays a key role in the fractionation of stable sulfur isotopes in different reservoirs and the marine environment. In this study, solid ES is discovered in sediments near the Jiulong Methane Reef in the northern South China Sea by scanning electron microscopy and Raman spectroscopy. Combining the morphology and distribution of ES, pyrite concentrations, and sulfur isotopes, we conclude that:(1) solid ES coexists with pyrite microcrystals and sulfide(oxyhydr)oxides as well as clay minerals, and they are mainly distributed on the surface of mineral aggregates;(2) ES mainly occurs within and near the sulfate-methane transition zone(SMTZ) despite little morphological diversity;(3) ES formation might be related to hydrogen sulfide oxidation and is therefore linked with fluctuations in the SMTZ. Within the SMTZ, hydrogen sulfide is produced and pyrite precipitates because of enhanced anaerobic oxidation of methane coupled with dissimilatory sulfate reduction. This enhances the efficiency of the inorganic sulfur cycle and provides favorable conditions for ES formation. The discovery of solid ES in sediments near the Jiulong Methane Reef suggests an important relationship with SMTZ fluctuations that could have implications for the evolution of methane hydrate in the South China Sea. | LIN Qi WANG JiaSheng FU ShaoYing LU HongFeng BU QingTao LIN RongXiao SUN Fei | 2015 | Science China Earth Sciences2015,58,12: | 6
Coupled carbon and sulfur isotope behaviors and other geochemical perspectives into marine methane seepage显示文摘Methane seepage is the signal of the deep hydrocarbon reservoir. The determination of seepage is significant to the exploration of petroleum, gas and gas hydrate. The seepage habits microbial and macrofaunal life which is fueled by the hydrocarbons, the metabolic byproducts facilitate the precipitation of authigenic minerals. The study of methane seepage is also important to understand the oceanographic condition and local ecosystem. The seepage could be active or quiescent at different times. The geophysical surveys and the geochemical determinations reveal the existence of seepage. Among these methods, only geochemical determination could expose message of the dormant seepages. The active seepage demonstrates high porewater methane concentration with rapid SO_4^(2–) depleted, low H_2S and dissolved inorganic carbon(DIC), higher rates of sulfate reduction(SR) and anaerobic oxidation of methane(AOM). The quiescent seepage typically develops authigenic carbonates with specific biomarkers, with extremely depleted ~13C in gas, DIC and carbonates and with enriched ~34S sulfate and depleted ~34S pyrite. The origin of methane, minerals precipitation, the scenario of seepage and the possible method of immigration could be determined by the integration of solutes concentration, mineral composition and isotopic fractionation of carbon, sulfur. Numerical models with the integrated results provide useful insight into the nature and intensity of methane seepage occurring in the sediment and paleooceanographic conditions. Unfortunately, the intensive investigation of a specific area with dormant seep is still limit. Most seepage and modeling studies are site-specific and little attempt has been made to extrapolate the results to larger scales. Further research is thus needed to foster our understanding of the methane seepage. | LIU Lihua FU Shaoying ZHANG Mei GUAN Hongxiang WU Nengyou | 2017 | Acta Oceanologica Sinica2017,36,6: | 2
SLC1A1-mediated cellular and mitochondrial influx of R-2-hydroxyglutarate in vascular endothelial cells promotes tumor angiogenesis in IDH1-mutant solid tumors显示文摘Muta nt isocitrate dehydrog en ase 1(mlDH1)drives tumorigenesis via produci ng on cometabolite R-2-hydroxyglutarate(R-2-HG)across various tumor types. However,mlDHl in hibitors appear only effective in hematological tumors. The therapeutic ben efit in solid tumors remains elusive, likely due to the complex tumor microenvironment. In this study,we discover that R-2-HG produced by IDH1-mutant tumor cells is preferentially imported into vascular endothelial cells and remodels mitochondrial respiration to promote tumor angiogenesis, conferring a therapeutic vulnerability in IDH1-mutant solid tumors.Mechanistically,SLC1 Alz a Na'-depe ndent glutamate tran sporter that is prefere ntially expressed in en dothelial cells, facilitates the in flux of R-2-HG from the tumor microenvironment into the endothelial cells as well as the intracellular trafficking of R-2-HG from cytoplasm to mitochondria.R-2-HG hijacks SLC1A1 to promote mitochondrial Na^(+)/Ca^(2+)exchange,which activates the mitochondrial respiratory chain and fuels vascular en dothelial cell migratio n in tumor an giogenesis.SLC1A1 deficiency in mice abolishes mlDHl-promoted tumor an giogenesis as well as the therapeutic benefit of mlDHl in hibitor in solid tumors. Moreover,we report that HH2301,a newly discovered mlDHl inhibitor, shows promising efficacy in treating IDH1-mutant cholangiocarcinoma in preclinical models.Together,we identify a new role of SLC1A1 as a gatekeeper of R-2-HG-mediated crosstalk between IDH1-mutant tumor cells and vascular en dothelial cells, and dem on strate the therapeutic potential of mlDHl in hibitors in treating IDH1-muta nt solid tumors via disrupting R-2-HG-promoted tumor angiogenesis. | Xiaomin Wang Ziqi Chen Jun Xu Shuai Tang Nan An Lei Jiang Yixiang Zhang Shaoying Zhang Qingli Zhang Yanyan Shen Shijie Chen Xiaojing Lan Ting Wang Linhui Zhai Siyuwei Cao Siqi Guo Yingluo Liu Aiwei Bi Yuehong Chen Xiameng Gai Yichen Duan Ying Zheng Yixian Fu Yize Li Liang Yuan Linjiang Tong Kun Mo Mingcheng Wang Shu-Hai Lin Minjia Tan Cheng Luo Yi Chen Jia Liu Qiansen Zhang Leping Li Min Huang | 2022 | Cell Research2022,32,7: | 2

A comparative study of alteration in retinal layer segmentation alteration by SD-OCT in neuromyelitis optica spectrum disorders: A systematic review and meta-analysis显示文摘Background:To evaluate the feature of different retinal layer segmentation in neuromyelitis optica spectrum disorders(NMOSD)with spectral-domain optical coherence tomography(SD-OCT)and to compare it with that in multiple sclerosis(MS),healthy controls(HC),and idiopathic optic neuritis(ION).Methods:We retrieved four electronic databases, including Pubmed,Embase,Cochrane Library, and Web of Science from inception to September 1st,2021.A meta-analysis was performed to compare different retinal layer segmentation thicknesses between patients with or without a history of optic neuritis(ON)in NMOSD and the control group, including patients with MS,HC,and ION.Results:Forty-two studies were included and the interval between the last ON onset and examination was greater than 3 months.Compared with that in HC eyes, the loss of retinal nerve fiber layer(RNFL)and macular ganglion cell and inner plexiform layer(GC-IPL)was serious in NMOSD eye especially after ON.Moreover, compared with that in ION eyes or MS-related-ON eyes, the injury to the peripapillary retinal nerve fiber layer(pRNFL)was severe in NMOSD-related-ON eyes.In addition, the correlation coefficient between pRNFL and prognostic visual acuity was 0.43.However,the one-arm study revealed the inner nuclear layer(INL)was thickened in NMOSDrelated-ON eyes compared with HC eyes. Conclusions:Inclusion of the RNFL and macular GC-IPL is recommended for monitoring disease progression and attention should be paid to changes in the INL. | Junxia Fu Shaoying Tan Chunxia Peng Huanfen Zhou Shihui Wei | 2021 | Advances in Ophthalmology Practice and Research2021,1,1: | 1
The clinical application of critical flicker fusion frequency in demyelinating optic neuritis显示文摘Purpose:To investigate the application of critical flicker fusion frequency(CFF)in demyelinating optic neuritis(DON).Methods:A cross-sectional study.A total of 127 eyes in 69 DON patients and 63 eyes in 33 healthy control(HC)groups were included between January 2021 to September 2021 from Department of Ophthalmology,PLA General Hospital.Patients underwent best-corrected visual acuity(BCVA),visual field, optical coherence tomography(OCT),flash visual evoked potential(F-VEP),and CFF examinations. The affected eyes were divided into aquaporins 4(AQP4-),myelin oligodendrocyte glycoprotein(MOG-),and double negative DON according to serum antibody;mild, moderate, severe degree visual impairment according to BCVA0.5,0.1-0.5,<0.1;and 4 groups:<1,1~<3,3~<6 and>6 months according to time interval from onset to CFF examination.One-way ANOVA was used to perform above subgroup analysis. The correlations between CFF and F-VEP peak time, peak value,BCVA and mean visual filed defect(MD)were analyzed in order via Pearson correlation analysis. Results:he trichromatic values of red, green, and yellow in DON affected eyes were 21.83±9.03,23.66±10.21,24.09±10.77 Hz,respectively, which was significantly reduced compared with the HC group(t=-14.82,-14.22,-14.00;P<0.001).The subgroup analysis showed no significant difference between different antibody subtypes(P=0.914<0.848<0.604),whereas,a significant decrease of CFF trichromatic value was found in severe visual acuity impairment group(P<0.001).There was a significant difference in CFF-trichromatic values at different time points(P<0.001),to be specific,CFF fluctuated under 20Hz within 3 months after onset and tended to be stable around 24-28Hz.Correlation analysis showed that the peak time of F-VEP(r=-0.486,-0.515,-0.526;P<0.001),BVCA(r=-0.640,-0.659,-0.642;P<0.001),were negatively correlated with CFF trichromatic values,MD and CFF were positively correlated(r=0.486,0.453,0.476;P=0.003,0.006,0.004).Conclusions:A significant decrease of CFF value was found in DON-affected eyes, and it has a good correlation with BCVA,MD and latency of F-VEP,and can better reflect the impairment of visual function. | Junxia Fu Yongping Wang Shaoying Tan Guangcan Xu Huanfen Zhou Quangang Xu Shihui Wei | 2021 | Advances in Ophthalmology Practice and Research2021,1,2: | 0
‘C-type’ closed state and gating mechanisms of K2P channels revealed by conformational changes of the TREK-1 channel显示文摘Two-pore domain potassium (K2P) channels gate primarily within the selectivity filter, termed ‘C-type’ gating. Due to the lack of structural insights into the nonconductive (closed) state, ‘C-type’ gating mechanisms remain elusive. Here, molecular dynamics (MD) simulations on TREK-1, a K2P channel, revealed that M4 helix movements induce filter closing in a novel ‘deeper-down’ structure that represents a ‘C-type’ closed state. The ‘down’ structure does not represent the closed state as previously proposed and instead acts as an intermediate state in gating. The study identified the allosteric ‘seesaw’ mechanism of M4 helix movements in modulating filter closing. Finally, guided by this recognition of K2P gating mechanisms, MD simulations revealed that gain-of-function mutations and small-molecule activators activate TREK-1 by perturbing state transitions from open to closed states. Together, we reveal a ‘C-type’ closed state and provide mechanical insights into gating procedures and allosteric regulations for K2P channels. | Qiansen Zhang Jie Fu Shaoying Zhang Peipei Guo Shijie Liu Juwen Shen Jiangtao Guo Huaiyu Yang | 2022 | Journal of Molecular Cell Biology2022,14,1: | 0
