1Olmutinib(HM61713) reversed multidrug resistance by inhibiting the activity of ATPbinding cassette subfamily G member 2 in vitro and in vivo显示文摘Overexpressing of ATP-binding cassette(ABC) transporters is the essential cause of multidrug resistance(MDR), which is a significant hurdle to the success of chemotherapy in many cancers.Therefore, inhibiting the activity of ABC transporters may be a logical approach to circumvent MDR.Olmutinib is an epidermal growth factor receptor(EGFR) tyrosine kinase inhibitor(TKI), which has been approved in South Korea for advanced EGFR T790 M-positive non-small cell lung cancer(NSCLC). Here,we found that olmutinib significantly increased the sensitivity of chemotherapy drug in ABCG2-overexpressing cells. Furthermore, olmutinib could also increase the retention of doxorubicin(DOX) and rhodamine 123(Rho 123) in ABC transporter subfamily G member 2(ABCG2)-overexpressing cells. In addition, olmutinib was found to stimulate ATPase activity and inhibit photolabeling of ABCG2 with [^(125) I]-iodoarylazidoprazosin(IAAP). However, olmutinib neither altered ABCG2 expression at protein and m RNA levels nor blocked EGFR, Her-2 downstream signaling of AKT and ERK. Importantly,olmutinib enhanced the efficacy of topotecan on the inhibition of S1-MI-80 cell xenograft growth. All the results suggest that olmutinib reverses ABCG2-mediated MDR by binding to ATP bind site of ABCG2 and increasing intracellular chemotherapeutic drug accumulation. 3Genome and transcriptome of Papaver somniferum Chinese landrace CHM indicates that massive genome expansion contributes to high benzylisoquinoline alkaloid biosynthesis显示文摘Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy.Li Pei Baishi Wang Jian Ye Xiaodi Hu Lihong Fu Kui Li Zhiyu Ni Zhenlong Wang Yujie Wei Luye Shi Ying Zhang Xue Bai Mengwan Jiang Shuhui Wang Chunling Ma Shujin Li Kaihui Liu Wanshui Li Bin Cong 2021Horticulture Research2021,8,1:4
4Maneuvering target tracking algorithm based on cubature Kalman filter with observation iterated update显示文摘Reasonable selection and optimization of a filter used in model estimation for a multiple model structure is the key to improve tracking accuracy of maneuvering target.Combining with the cubature Kalman filter with iterated observation update and the interacting multiple model method,a novel interacting multiple model algorithm based on the cubature Kalman filter with observation iterated update is proposed.Firstly,aiming to the structural features of cubature Kalman filter,the cubature Kalman filter with observation iterated update is constructed by the mechanism of iterated observation update.Secondly,the improved cubature Kalman filter is used as the model filter of interacting multiple model,and the stability and reliability of model identification and state estimation are effectively promoted by the optimization of model filtering step.In the simulations,compared with classic improved interacting multiple model algorithms,the theoretical analysis and experimental results show the feasibility and validity of the proposed algorithm.胡振涛 Fu Chunling Cao Zhiwei Li Congcong 2015High Technology Letters2015,21,1:4
5A novel multi-sensor multiple model particle filter with correlated noises for maneuvering target tracking显示文摘Aiming at the effective realization of particle filter for maneuvering target tracking in multi-sensor measurements,a novel multi-sensor multiple model particle filtering algorithm with correlated noises is proposed.Combined with the kinetic evolution equation of target state,a multi-sensor multiple model particle filter is firstly constructed,which is also used as the basic framework of a new algorithm.In the new algorithm,in order to weaken the adverse influence from random measurement noises in the measuring process of particle weight,a weight optimization strategy is introduced to improve the reliability and stability of particle weight.In addition,considering the correlated noise existing in the practical engineering,a decoupling method of correlated noise is given by the rearrangement and transformation of the state transition equation and measurement equation.Since the weight optimization strategy and noise decoupling method adopt respectively the center fusion structure and the off-hne way,it improves the adverse effect effectively on computational complexity for increasing state dimension and sensor number.Finally,the theoretical analysis and experimental results show the feasibility and efficiency of the proposed algorithm.胡振涛 Fu Chunling 2014High Technology Letters2014,20,4:3
6Establishment and characterization of arsenic trioxide resistant KB/ATO cells显示文摘Arsenic trioxide(ATO) is used as a chemotherapeutic agent for the treatment of acute promyelocytic leukemia. However, increasing drug resistance is reducing its efficacy. Therefore, a better understanding of ATO resistance mechanism is required. In this study, we established an ATO-resistant human epidermoid carcinoma cell line, KB/ATO, from its parental KB-3-1 cells. In addition to ATO, KB/ATO cells also exhibited cross-resistance to other anticancer drugs such as cisplatin, antimony potassium tartrate, and 6-mercaptopurine. The arsenic accumulation in KB/ATO cells was significantly lower than that in KB-3-1 cells. Further analysis indicated that neither application of P-glycoprotein inhibitor, breast cancer resistant protein(BCRP) inhibitor, or multidrug resistance protein 1(MRP1) inhibitor could eliminate ATO resistance. We found that the expression level of ABCB6 was increased in KB/ATO cells.In conclusion, ABCB6 could be an important factor for ATO resistance in KB/ATO cells. 15Maneuvering target tracking algorithm based on CDKF in observation bootstrapping strategy显示文摘The selection and optimization of model filters affect the precision of motion pattern identification and state estimation in maneuvering target tracking directly.Aiming at improving performance of model filters,a novel maneuvering target tracking algorithm based on central difference Kalman filter in observation bootstrapping strategy is proposed.The framework of interactive multiple model(IMM) is used to realize identification of motion pattern,and a central difference Kalman filter(CDKF) is selected as the model filter of IMM.Considering the advantage of multi-sensor fusion method in improving the stability and reliability of observation information,the hardware cost of the observation system for multiple sensors is adopted,meanwhile,according to the data assimilation technique in Ensemble Kalman filter(En KF),a bootstrapping observation set is constructed by integrating the latest observation and the prior information of observation noise.On that basis,these bootstrapping observations are reasonably used to optimize the filtering performance of CDKF by means of weight fusion way.The object of new algorithm is to improve the tracking precision of observed target by the multi-sensor fusion method without increasing the number of physical sensors.The theoretical analysis and experimental results show the feasibility and efficiency of the proposed algorithm.胡振涛 Zhang Jin Fu Chunling Li Xian 2017High Technology Letters2017,23,2:1

19Simulation of SLD Impingement on Wind Turbine Blade Airfoil显示文摘Accurate prediction of droplet impingement on wind turbine blade is one of the most important premises of anti-icing and de-icing system design.In a super-cooled large droplets(SLD)conditions,droplet no longer maintains a sphere shape,and it may deform,break up,and splash when moving or impinging on the surface.Semi-empirical models of droplet dynamic behaviours are embedded into the Eulerian droplet model to improve the accuracy of the numerical simulation of droplet impingement limits and local collection efficiency.Eulerian droplet model(Model 1)for small droplets and improved Eulerian droplet model(Model 2)for large droplet are both validated by comparing to the wind tunnel experiment results.Using the proposed methods,droplet impingement limitation and local collection efficiency on the S809 airfoil are calculated in various conditions.A detailed derivation of Model 1and Model 2is presented along with a comparison of numerical trajectories,drag coefficient and collection efficiency distributions.The results show that droplet dynamic behaviours,including splashing,break-up and deforming,must be considered to accurately simulate the impingement behaviour in the SLD conditions.And with the increase of the droplet diameters,the effects of the droplet dynamic behaviors on the impingement characteristics are more obvious.Zhu Chengxiang Zhu Chunling Fu Bin ZhaoHuanyu 2016Transactions of Nanjing University of Aeronautics and Astronautics2016,33,1:0
20Application of Systematic Clustering Method in the Classification and Changes of Atmospheric Ozone in Hunan Province显示文摘The cluster analysis method needs continuous improvement and perfection in the research and application of the spatial differentiation and change of pollutants.In this paper,the date of monthly highest concentration of ozone(O_(3))and the concentration value of that day were selected as the similarity coefficient between classes.Single-factor cluster analysis was performed on O_(3)during 2016-2019 and the COVID-19 outbreak of 2020 in Hunan Province using the Ward method.The clustering results showed that the spatial distribution of atmospheric O_(3)in the 14 regions of Hunan Province was most suitable to be classified according to class III clustering areas.That is,the Changsha-Zhuzhou-Xiangtan urban agglomeration was the center,and the high-value area was in northern Hunan.The transition area was in central and southern Hunan,while the low-value area was centered in western Hunan.The partition results were in good agreement with the homogeneous subset of one-way ANOVA and the distribution of monitoring values during the same period.The comparison showed that the inter-class plates in the two periods corresponded well,and the intra-class area showed a continuous geographical distribution,and there were dynamic changes in the spatial differentiation of the O_(3)plates in different periods.In 2020,the center of the O_(3)high-value area plate in Hunan Province moved eastward and extended southward,focusing on the middle and lower reaches of the Xiangjiang River basin,and extending to the upstream area;the regional plate in the transition area expanded significantly;the low-value area plate shrank to the two cities in western Hunan.The abnormal emissions and abnormal climate during the COVID-19 epidemic had an impact on the spatial differentiation of O_(3)in Hunan Province.Wenhui YAO Liushu FU Qian GUO Chunling XU Zhe YANG Wei ZHOU 2022Meteorological and Environmental Research2022,13,4:0
