1Roles of Fas signaling pathway in vitamin E succinateinduced apoptosis in human gastric cancer SGC-7901 cells显示文摘AIM: To investigate the roles of Fas signaling pathway in vitamin E succinate-induced apoptosis in human gastric cancer SGC-7901 cells.METHODS: Human gastric cancer SGC-7901 cells were treated with VES at 5, 10, 20 mg@L-1, succinic acid and vitamin E as vehicle control and condition media only as untreated (UT) control. Apoptotic morphology was observed by DAPI staining. Western blot analysis was applied to measure the expression of Fas, FADD and caspase-8 proteins. After the cells were transiently transfected with Fas and FADD antisense oligonucleotides, respectively, caspase-8 activity was determined by flurometric method.RESULTS: The morphologically apoptotic changes were observed after VES treatment by DAPI staining. 23.7 % and 89.6 % apoptosis occurred after 24 h and 48 h of 20 mg@L-1 VES treatment, respectively. The protein levels of Fas, FADD and caspase-8 were evidently increased in a dose-dependent manner after 24 h of VES treatment. The blockage of Fas by transfection with Fas antisense oligonucleotides obviously inhibited the expression of FADD protein. 2Effect of cis-9,trans-11-conjugated linoleic acid on cell cycle of gastric adenocarcinoma cell line(SGC-7901)显示文摘AIM: To determine the effect of cis-9, trans-1l-conjugated linoleic acid ( c9, tl 1-CLA) on the cell cycle of gastric cancer cells( SGC-7901 ) and its possible mechanism in inhibition cancer growth.METHODS: Using cell culture and immunocytochemicaltechniques, we examined the cell growth, DNA synthesis,expression of PCNA, cyclin A, B1, D1, pl6nink4a and p21clp/wafl ofSGC-7901 cells which were heated with various c9, tll-CLAconcentrations (25,50,100 and 200μnol@L-1)of c9, tll-CLA for 24and 48 h, with a negative control (0.1% ethane).RESULTS: The cell growth and DNA synthesis of SGC-7901cells were inhibited by c9, tll-CLA. SGC-7901 cells. 3Inhibitory effects of RRR-α-tocopheryl succinate on benzo (a) pyrene (B (a) P)-induced forestomach carcinogenesis in female mice显示文摘AIM To study the inhibitory effects of VES( RRR-α-tocopheryl Succinate, VES ), aderivative of natural Vitamin E, on benzo (a)pyrene (B (a) P)-induced forestomach tumor infemale mice.METHODS The model of B (a)P-inducedforestomach tumor was established according tothe methods of Wattenberg with slightmodifications. One hundred and eighty femalemice (6 weeks old) were divided into six groupsequally; negative control (Succinic acid),vehicle control ( Succinate + B (a) P), positivecontrol(B(a) P), high VES(2.5g/kg. b. w + B(a)P), Iow VES(1 .25 g/kg. b. w + B(a) P) ig as wellas VES by ip (20 mg/kg, b. w + B(a) P). Exceptthe negative control group, the mice wereadministrated with B(a)P ig. and correspondingtreatments for 4 weeks to study the anti-carcinogenetic effect of VES during the initiationperiod. The experiment lasted 29 weeks, inwhich the inhibitory effects of VES both ontumor incidence and tumor size were tested.RESULTS The models of B (a)P-inducedforestomach tumor in female mice wereestablished successfully. Some werecauliflower-like, others looked like papilla, evena few were formed into the ulcer cavities. VES at1.25 g/kg. b. w, 2.5 g/kg. b.w. by ig and 20 mg/kg. b. w. via ip could decrease the number oftumors per mouse (1.7 ± 0. 41, 1.6 ± 0.34 and 1.1±0.43), being lower than that of B(a)P group(5.4 ± 0.32, P<0.05). The tumor incidence wasinhibited by 18.2%, 23.1% and 50.0%. 5New Collection of Crude Drugs in Chinese Pharmacopoeia 2010 Ⅰ.Callicarpa Linn. and Related Items显示文摘Callicarpa Linn. (beautyberry) is one of the major genera in Verbenaceae, about 20 of which are medicinal plants. Beautyberry, called Zizhu in China, is a generic name of those species and largely used as hemostatic medicine. The Chinese Pharmacopoeia 2010 has admitted three new crude drugs from the genus of Callicarpa Linn. including Callicarpae Macrophyllae Folium, Callicarpae Caulis et Folium, and Callicarpae Formosanae Folium for the first time since the 1977 version of the Chinese Pharmacopoeia. In order to better understand these new crude drugs, we systematically described their bibliography, admission reasons, botanical identification, chemistry, and pharmacology. 6Role of soil rhizobacteria in phytoremediation of heavy metal contaminated soils显示文摘Heavy metal pollution of soil is a significant environmental problem and has its negative impact on human health and agriculture. Rhizosphere, as an important interface of soil and plant, plays a significant role in phytoremediation of contaminated soil by heavy metals, in which, microbial populations are known to affect heavy metal mobility and availability to the plant through release of chelating agents, acidification, phosphate solubilization and redox changes, and therefore, have potential to enhance phytoremediation processes. Phytoremediation strategies with appropriate heavy metal-adapted rhizobacteria have re-ceived more and more attention. This article paper reviews some recent advances in effect and significance of rhizobacteria in phytoremediation of heavy metal contaminated soils. 8Studies on Human Dietary Requirements and Safe Range of Dietary Intakes of Selenium in China and Their Application in the Prevention of Related Endemic Diseases显示文摘The human dietary selenium requirement in China has been estimated by various methods. The minimum dietary selenium requircment for the prevention of Keshan disease (KD) was found to be around 17 μg/d. On the other hand, an intake of 40 μg/d is required to maintain the plasma glutathione peroxidase (GPx) activity at plateau. Hence 40 μg/d is considered as the adequate dietary selenium requirement. Studies conducted in a chronic selenosis area indicate thai the toxic dietary selenlum inlake (adverse effect level), which would maintain the characteristic fingernail changes, was approximately 1600 μg/d. The mean value of dietary selenium intakes, which enabled the five patients to recover from fingernail lesions, was found to be 819 ±126μg/d. At a 95% confidence limit, the lower limit is around 600μg/d. 9Reduction of Chilling Injury and Ultrastructural Damage in Cherry Tomato Fruits After Hot Water Treatment显示文摘The effects of hot water treatment in alleviating chilling injury and reducing ultrastructural damage of mature-green cherry tomatoes (Lycopersicun esculentum cv. cerasiform Alef) were investigated. Mature-green cherry tomato fruits were treated in water at 40°C or 45°C for 5 min or 15 min, and then stored at 5°C for 19 days followed by ripening at 20°C. Water treatment at 40°C for 15 min increased tolerance of cherry tomato fruits to chilling stress, indicating as low outbreak of skin lesion, high color a* value, and low electrolyte leakage. Treated fruits showed typical climacteric respiration and developed normal red color with chlorophyll degradation and lycopene accumulation during ripening, while fruits without treatment failed to develop red color and suffered skin lesion. After 19 days of chilling, heated fruits showed the conversion of chloroplast to chromoplast with the disappearance of thylakoids. Mitochondria and other cell organelles were not adversely affected in treated fruits. However, ultrastructures in pericarp cells in control fruits severely damaged with extensive disorganization of cytoplasm, swelled chloroplasts, distorted and unstacked thylakoids. Chloroplast was the first and most severely impacted organelle by chilling stress. Hot water treatment (40°C for 15 min) before storage alleviated chilling injury in cherry tomato fruits. 10Methyl Jasmonate Reduces Chilling Injury and Maintains Postharvest Quality in Peaches显示文摘Zhaohui peaches (Prunus persica Batsch) were treated with 0 (CK), 1, 10 or 100 μmol L-1methyl jasmonate (MeJA) vapor at 20℃ for 24 h before stored at 0℃ for 35 d. The untreated fruits showed chilling injury (CI) symptoms after 4 wk of storage, as indicated by increased fruit firmness and reduced extractable juice, which is referred as leatheriness. Treatment with 1 and 10 μmol L-1 MeJA promoted normal ripening and softening, maintained higher levels of extractable juice, titratable acidity, pectinesterase (PE)and polygalacturonase (PG) activities, inhibited increases in fruit fresh weight loss, decay incidence, electrolyte leakage and MDA content, and improved color development, thereby preventing chilling injury symptoms development and maintaining edible quality. MeJA treatment also delayed the climacteric rise in respiratory rate, but promoted ethylene production during the later period of cold storage, suggesting that ethylene may involve in CI development of peaches. 11Primary structure and configuration of tea polysaccharide显示文摘The monosaccharide composition of a tea polysaccharide (TGC) was determined by GC-MS method. Furthermore, the primary structure of tea polysaccharide and its configura-tion in the aqueous solution were investigated utilizing a combination of classical chemical methods and modern instrumental techniques including GC-MS, Proton NMR, UV and CD. The results indicate that TGC consists of 6 monosaccharides: Rha, Ara, Xyl, Glu, Man and Gal. The configuration of TGC in water solution is proposed to be an ordered helix. The possible primary structure of TGC was outlined as below: the basic structure of the main chain consists of Rha, Glu and Gal units. All three monosaccharides can potentially be connected to branch chains consisting of mainly Ara, and the linkages could be in b1→2, b 1→3, b 2→3 forms. 13The Effect of Chinese Tea on Occurrence of Esophageal Tumor Induced by N-Nitrosomethylbenzylamine in Rats显示文摘Five groups of rats (40 rats per group) were given five different varieties of Chinese tea (1 g/50 ml) and another two groups of rats (a positive control and a negative control) were given tap water as drinking water. Except for the negative control group, all the animals were intubated with A’-nitrosomethyibenzylamine (NMBzA, 5 mg/kg body wt/week). Half of the animals in each group were sacrificed after 6 weeks and the remaining animals at the end of 12 weeks. The incidences of esophageal mucosa lesions (16-59%) at the end of6 weeks were significantly lower in the tea-treated rats than in the positive control group (100%). The incidences of esophageal tumor at the end of 12 weeks (42-67%) were also significantly lower in the tea-treated groups than in the positive control group (90%). The same phenomenon was found with respect to the size and number of tumors in each tumor-bearing animal. The relative anticarcinogenic effects of the five varieties of Chinese tea were different. The Fujian oolong tea and jasmine tea exhibited the strongest effects. 14Free Radical Generation by Selenium Compounds and Their Prooxidant Toxicity显示文摘Selemum (Se) and many of its compounds are among the most toxic of nutrients. Selenium toxicity was first described in range animals in the western United States in the 1930’s which consumed' selenium accumulator' plants of the genus Astragalus, Xylorrhiza,Oonopsis, and Stanleya. Selenites and selenates from the soil accumulate in these plants primarily as methylated selenium compounds and plants evolve dimethyldiselenide and dimethyselenide.Dietary selenium, primarily as selenomethionine and selenocysteine for humans fulfill the dietary requirement for selenoenzymes and proteins. In humans and animals excessive dietary selenium may be toxic. In vitro, selenium compounds such as selenite, selenium dioxide and diselenides react with thiols, such as glutathione, producing superoxide and other reactive oxygen species. This catalytic reaction of selenium compounds with thiols likely accounts for selenium toxicity to cells ex vivo and in vivo where the major glutathione producing organ,the liver, is also the major target organ of selenium toxicity.Selenium enzymes and selenoethers that do not readily form a selenide (RSe-) anion and compounds such as Ebselen where selenium is sequestered, are not toxic. Methylation of selenium by both plants and animals serves to detoxify selenium by generating methylselenides. Alternatively, full reduction of Se to elemental selenium (Se0) as done by some bacteria and the formation of heavy metal selenides such as Ag2Se or Hg2Se, results in a non-catalytic nontoxic form of selenium.This catalytic prooxidant attribute of some selenium compounds appears to account for its toxicity when such activity exceeds plant and animal methylation reactions and antioxidant defenses. 15Study on Molecular Chain Morphology of Konjac Glucomannan显示文摘The long-range structure of konjac glucomannan(KGM) is studied by using laser light scatter(LLS), gel permeation chromatography (GPC) and method of viscostdity. The weight-average molecularweight (Mw), root-mean-square ratio of gyration[ (S2) 1/2 ], second viral coefficient (A2) and multi-dispersioncoefficient (Mw/Mn) are 1.04× 106, 105.0±-0. 9 nm, (- 1.59±0. 28) × 10-3 mol ml g-2 and 1. 015±0. 003,respectively. Mark-Houwink equation is established as [η] = 5. 96 × 10-2Mw0.73 and the molecular chain pa-rameters are as follows: ML=982.82 nm-1 , Lp=27.93 nm, d=0.74 nm, h=0.26 nm, L=1 054.11 nm.Further more molecular chain morphology of KGM is studied by using atom force microscope (AFM) andtransmission electronic microscope (TEM), and the result shows that the KGM molecular is an extendingsemi-flexible linear chain without branch. 16Mechanical and thermal properties of sodium silicate treated moso bamboo particles reinforced PVC composites显示文摘The main objective of this research was to study the potential of sodium silicate modification on moso bamboo particles as reinforcements for thermoplastic. Moso bamboo particles were modified with sodium silicate aqueous solutions (of 0.5%, 1%, 2%, 5% and 10% concentrations). The mechanical properties of sodium silicate treated moso bamboo particles reinforced PVC composites (BPPC) were calculated and compared with raw bamboo particles filled samples. The thermal characteristics of the BPPC were studied to investigate the feasibility of sodium silicate treatment on moso bamboo particles. The particle morphology and BPPC microstructure were investigated by scanning electron microscopy. Results showed that the tensile strength and modulus of elasticity of the BPPC increased before the concentration of sodium silicate solution reached 5% and got their maximum values of 15.72 MPa and 2956.80 MPa, respectively at 5% concentration. The modulus of rupture obtained the maximum value of 27.73 MPa at 2% concentration. The mechanical curve decreased as the concentration of solution went higher. Differential scanning calorimetric analysis illustrated that the sodium silicate solution treated BPPC possesses a better compatibility. More uniform dispersion of moso bamboo particles in PVC matrix was obtained after the sodium silicate treatment. 19Study on Gel Formation Mechanism of Konjac Glucomannan显示文摘The transformation of molecular conformation after gelatinization of konjac glucomannan ischaracterized by means of GC, GPC, FT-IR, DSC, XRD and TEM. The results showed that the molecule ofKGM had no branch, the monose composition and connecting type of the molecular chain had no change,the transformation of molecule weight was little, and its drying powder sample formed an evidently newcrystalline region. The hollow double helix structure of KGM in hydrosol changed from stretch chain into in-tercross and entwisted after gelatinization.
