| 2 | Inhibiting collagen Ⅰ production and tumor cell colonization in the lung via miR-29a-3p loading of exosome-/liposome-based nanovesicles显示文摘The lung is one of the most common sites for cancer metastasis.Collagens in the lung provide a permissive microenvironment that supports the colonization and outgrowth of disseminated tumor cells.Therefore,down-regulating the production of collagens may contribute to the inhibition of lung metastasis.It has been suggested that mi R-29 exhibits effective anti-fibrotic activity by negatively regulating the expression of collagens.Indeed,our clinical lung tumor data shows that mi R-29 a-3 p expression negatively correlates with collagen I expression in lung tumors and positively correlates with patients’outcomes.However,suitable carriers need to be selected to deliver this therapeutic mi RNA to the lungs.In this study,we found that the chemotherapy drug cisplatin facilitated mi R-29 a-3 p accumulation in the exosomes of lung tumor cells,and this type of exosomes exhibited a specific lung-targeting effect and promising collagen down-regulation.To scale up the preparation and simplify the delivery system,we designed a lung-targeting liposomal nanovesicle(by adjusting the molar ratio of DOTAP/cholesterol-mi RNAs to 4:1)to carry mi R-29 a-3 p and mimic the exosomes.This liposomal nanovesicle delivery system significantly down-regulated collagen I secretion by lung fibroblasts in vivo,thus alleviating the establishment of a pro-metastatic environment for circulating lung tumor cells. | Yan Yan Cancan Du Xixi Duan Xiaohan Yao Jiajia Wan Ziming Jiang Zhongyu Qin Wenqing Li Longze Pan Zhuoyu Gu Fazhan Wang Ming Wang Zhihai Qin | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 7 |
| 3 | Optimizing ultrasensitive single electron magnetometer based on nitrogen-vacancy center in diamond显示文摘The measurement of the weak magnetic field in nanoscale resolution and at room temperature is always a significant topic in biological, physical, and material science. Such detection can be used to decide the characterization of the samples, such as cells, materials, and so on. Nitrogen-vacancy (NV) center in diamond has been proved to be able to detect a magnetic field with nano Tesla sensitivity and nanometer resolution at room temperature. Here we experimentally demonstrate an optimized NV center based single electron magnetometer in a commercial diamond and under a home-built optically detected magnetic resonance (ODMR) microscope. With current technology, we change the optically detected time window to get a better signal to noise ratio, and use dynamical decoupling to increase the slope of magnetic field amplitude versus fluorescence signal. By employing the 8-pulse XY-4 dynamical decoupling sequence we achieve a sensitivity of 18.9 nT (Hz)(1/2) , which is 1.7 times better than spin echo. We also propose a NV center based scanning diamond microscope for electron and nuclear spins detection as well as nanoscale magnetic resonance imaging. | 4 | ZmSRL5 is involved in drought tolerance by maintaining cuticular wax structure in maize显示文摘Cuticular wax is a natural barrier on terrestrial plant organs,which protects plants from damages caused by a variety of stresses.Here,we report the identification and functional characterization of a cuticular-wax-related gene,Zea mays L.SEMI-ROLLED LEAF 5(ZmSRL5).The loss-of-function mutant srl5,which was created by a 3,745 bp in-sertion in the first intron that led to the premature tran-script,exhibited abnormal wax crystal morphology and distribution,which,in turn,caused the pleiotropic pheno-types including increased chlorophyll leaching and water loss rate,decreased leaf temperature,sensitivity to drought,as well as semi-rolled mature leaves.However,total wax amounts showed no significant difference between wild type and semi-rolled leaf5(srl5)mutant.The phenotype of srl5 was confirmed through the generation of two allelic mutants using CRISPR/Cas9.ZmSRL5 encodes a CASPARIAN-STRIP-MEMBRANE-DOMAIN-LIKE(CASPL)protein located in plasma membrane,and highly expressed in developing leaves.Further analysis showed that the expressions of the most wax related genes were not affected or slightly al-tered in srl5.This study,thus,primarily uncovers that ZmSRL5 is required for the structure formation of the cu-ticular wax and could increase the drought tolerance by maintaining the proper cuticular wax structure in maize. | Zhenyuan Pan Min Liu Hailiang Zhao Zengdong Tan Kun Liang Qin Sun Dianming Gong Haijun He Wenqi Zhou Fazhan Qiu | 2020 | Journal of Integrative Plant Biology2020,62,12: | 5 |
| 5 | Detection of radio-frequency field with a single spin in diamond显示文摘Detection of a.c. magnetic field is consequential for many developments in physical and biological sciences,and various designs of magnetometer have been proposed recently. However, the large size of sensor and the extreme measurement conditions required strongly limit their application. It remains a challenge to reconstruct the vector of a.c. field with nanoscale spatial resolution using a single spin under ambient conditions. In this work, we choose the radio-frequency(RF) field as a typical case and realize the measurement of RF field based on a nitrogen-vacancy(NV)center in diamond. We build a solid sensor through measuring the effect of RF field on NV electron spin energy levels and the transition between them. | 6 | Core-shell lipid-polymer nanoparticles as a promising ocular drug delivery system to treat glaucoma显示文摘Nowadays,tremendous researches have been focused on the core-shell lipid-polymer nanoparticles(LPNs) due to the advantages of both liposomes and polymer nanoparticles.In this work,LPNs were applied to encapsulate brinzolamide(Brz-LPNs) for achieving sustained drug release,improving drug corneal permeation and enhancing drug topical therapeutic effect.The structure of Brz-LPNs was composed of poly(lactic-co-glycolic) acid(PLGA) nanocore which encapsulated Brz(Brz-NPs) and lipid shell around the core.Brz-LPNs were prepared by a modified thin-film dispersion method.With the parameters optimization of Brz-LPNs,optimal Brz-LPNs showed an average particle size of151.23±1.64 nm with a high encapsulation efficiency(EE) of 86.7%±2.28%.The core-shell structure of Brz-LPNs were confirmed by transmission electronic microscopy(TEM).Fourier transformed infrared spectra(FTIR) analysis proved that Brz was successfully entrapped into Brz-LPNs.Brz-LPNs exhibited obvious sustained release of Brz,compared with AZOPT^■ and Brz-LPs.Furthermore,the corneal accumulative permeability of Brz-LPNs significantly increased compared to the commercial available formulation(AZOPT^■) in vitro.Moreover,Brz-LPNs(1 mg/mL Brz) showed a more sustained and effective intraocular pressure(IOP) reduction than Brz-LPs(1 mg/mL) and AZOPT^■(10 mg/mL Brz) in vivo.In conclusion,Brz-LPNs,as promising ocular drug delivery systems,are well worth developing in the future for glaucoma treatment. | Yang Zhou Aiping Fang Fazhan Wang Huili Li Quansheng Jin Lingjing Huang Chunmei Fu Jun Zeng Zhaohui Jin Xiangrong Song | 2020 | Chinese Chemical Letters2020,31,2: | 3 |
| 7 | Semi-elastic core-shell nanoparticles enhanced the oral bioavailability of peptide drugs显示文摘The rigidity of nanoparticles was newly reported to influence their oral delivery.Semi-elastic nanoparticles can enhance the penetration in mucus and uptake by epithelial cells.However,it is still challenging and unclear that the semi-elastic core-shell nanoparticles can enhance the oral bioavailability of peptide drugs.This study was for the first time to validate the semi-elastic coreshell poly(lactic-co-glycolic acid)(PLGA)-lipid nanoparticles(LNPs)as the carrier of the oral peptide drug.The antihypertensive peptide Val-Leu-Pro-Val-Pro(VP5)loaded LNPs(VP5-LNPs)were prepared by a modified thin-film ultrasonic dispersion method.Uptake experiment was performed in Caco-2 and HT-29 cells and monito red by high content screening(HCS)and flow cyto metric(FCM).Pharmacokinetics of VP5-LNPs was carried out in Sprague-Dawley(SD)rats and analyzed by DAS 2.0.The optimal VP5-LNPs had an average particle size of 247.3±3.8 nm,zeta potential of-6.57±0.45 mV and excellent entrapment efficiency(EE)of 89.88%±1.23%.Transmission electron microscope(TEM)and Differential scanning calorimeter(DSC)further confirmed the core-shell structure.VP5-LNPs could increase the cellular uptake in vitro and have a 2.55-fold increase in AUC0-72 h,indicating a great promotion of the o ral bioavailability.The semi-elastic LNPs remarkably improved the oral availability of peptide and could be a promising oral peptide delivery system for peptide drugs in the future. | Shengnan Zhao Jinhua Li Fazhan Wang Ting Yu Yang Zhou Lili He Yi Zhang jian Yang | 2020 | Chinese Chemical Letters2020,31,5: | 2 |

| 16 | Mechanism of enhanced removal of quinonic intermediates during electrochemical oxidation of Orange Ⅱ under ultraviolet irradiation显示文摘The effect of ultraviolet irradiation on generation of radicals and formation of intermediates was investigated in electrochemical oxidation of the azo-dye Orange II using a TiO2-modified-PbO2 electrode. It was found that a characteristic absorbance of quinonic compounds at 255 nm, which is responsible for the rate-determining step during aromatics degradation, was formed only in electrocatalytic oxidation. The dye can be oxidized by either HO radicals or direct electron transfer. Quinonic compounds were produced concurrently. The removal of TOC by photo-assisted electrocatalytic oxidation was 1.56 times that of the sum of the other two processes, indicating a significant synergetic effect. In addition, once the ultraviolet irradiation was introduced into the process of electrocatalytic oxidation, the degradation rate of quinonic compounds was enhanced by as much as a factor of two.
