| 1 | 姜黄素诱导髓系白血病细胞凋亡的机制研究(英文)显示文摘Objective: To investigate the mechanism of apoptosis of myelocytic leukemia lines NB4, K562 and THP-1 cells induced by curcumin. Methods: After the cells were treated with curcumin at different concentration (25, 12.5, 6.25, 3.125, 0 μmol/L) for various times (0, 12, 24, 48 h), flow cytometry (FCM) was used to determine the rate of apoptosis of cells. After the cells were treated with curcumin at 25 μmol/L for 24 h, flow cytometry was used to determine the expression level of Fas, and Western blot was performed to determine the expression of Caspase-8 and Caspase-9. Results: (1) Curcumin could induced the apoptosis of NB4, K562 and THP-1 cells in a time- and dose-dependent manner. After the cells were treated with curcumin at 25 μmol/L for 48 h, the rate of apoptosis of cells was over fifty percent. (2) Fas level showed remarkable increase (P < 0.01) after above cells were treated with 25 μmol/L curcumin for 24 h. (3) The apoptosis proteins of Caspase-8 and Cas- pase-9 were also increased obviously (P < 0.01) after the cells were treated with 25 μmol/L curcumin for 24 h. | 2 | A homologous and molecular dual-targeted biomimetic nanocarrier for EGFR-related non-small cell lung cancer therapy显示文摘The abnormal activation of epidermal growth factor receptor(EGFR)drives the development of non-small cell lung cancer(NSCLC).The EGFR-targeting tyrosine kinase inhibitor osimertinib is frequently used to clinically treat NSCLC and exhibits marked efficacy in patients with NSCLC who have an EGFR mutation.However,free osimertinib administration exhibits an inadequate response in vivo,with only~3%patients demonstrating a complete clinical response.Consequently,we designed a biomimetic nanoparticle(CMNP^(@Osi))comprising a polymeric nanoparticle core and tumor cell-derived membrane-coated shell that combines membrane-mediated homologous and molecular targeting for targeted drug delivery,thereby supporting a dual-target strategy for enhancing osimertinib efficacy.After intravenous injection,CMNP^(@Osi)accumulates at tumor sites and displays enhanced uptake into cancer cells based on homologous targeting.Osimertinib is subsequently released into the cytoplasm,where it suppresses the phosphorylation of upstream EGFR and the downstream AKT signaling pathway and inhibits the proliferation of NSCLC cells.Thus,this dual-targeting strategy using a biomimetic nanocarrier can enhance molecular-targeted drug delivery and improve clinical efficacy. | Bin Xu Fanjun Zeng Jialong Deng Lintong Yao Shengbo Liu Hengliang Hou Yucheng Huang Hongyuan Zhu Shaowei Wu Qiaxuan Li Weijie Zhan Hongrui Qiu Huili Wang Yundong Li Xianzhu Yang Ziyang Cao Yu Zhang Haiyu Zhou | 2023 | Bioactive Materials2023,,9: | 1 |
| 3 | 睾丸的原发性NK/T细胞淋巴瘤一例报告及文献复习(英文)显示文摘Objective: To study the clinical and pathological features of primary NK/T cell lymphoma of testis and to investi- gate the effective diagnosis and treatment of this disease. Methods: The surgical specimens of a patient with primary NK/T cell lymphoma of the testis were observed by light microscopy, immunohistochemistry and examined by the polymerase chain reaction (PCR) for Epstein-Barr virus (EBV) DNA and T-cell receptor (TCR) gene rearrangement, and the literature were reviewed. Results: The patient presented with left-sided painless testicular enlargement and the lymphoma had a propensity to spread to the contralateral testis, spleen, central nervous system, and so on. The neoplastic cells were positive for CD56, CD45R0 and CD3ε, while the expressions of CD20, CD79α, CD5, Bcl-2 and PLAP were negative. In addition, the EBV DNA was detected in the lymphoma by PCR. And the results of gene rearrangement studies for the γ chain of the T-cell receptor were negative. The pathological diagnosis was NK/T cell lymphoma of the left testis. Conclusion: Primary NK/T cell lym- phoma of the testis is a rare entity and progressed rapidly. The histopathological, immunohistochemical, EBV examination and TCR gene rearrangement studies should be carried out as soon as possible in order to get the defined diagnosis. | 5 | Successful kidney transplantation in highly sensitized patients显示文摘Highly sensitized patients experience an increased number of rejection episodes and have poorer graft survival rates;hence,sensitization is a significant barrier to both access to and the success of organ transplantation.This study reports our experience in kidney transplantation in highly sensitized patients.Fourteen patients with sensitization or high levels of panelreactive antibodies(PRA)were studied.All patients were desensitized with pre-transplant intravenous immunoglobulin(IVIG)/plasmapheresis(PP)with or without rituximab and thymoglobulin induction therapy,combined with a Prograf/MMF/Pred immunosuppressive regimen.Of 14 patients,10 showed good graft functions without acute rejection(AR)episodes.Acute cellular rejection in two patients was reversed by methylprednisolone.Two patients underwent antibody-mediated rejection;one was treated with PP/IVIG successfully,whereas the other lost graft functions due to the de novo production of donor-specific antibodies(DSA).Graft functions were stable,and there were no AR episodes in other patients.Conclusively,desensitization using PP/IVIG with or without rituximab increases the likelihood of successful live-donor kidney transplantation in sensitized recipients. | Weijie ZHANG Dong CHEN Zhishui CHEN Fanjun ZENG Changsheng MING Zhengbin LIN Ping ZHOU Gang CHEN Xiaoping CHEN | 2011 | Frontiers of Medicine2011,5,1: | 0 |
