| 1 | Use of genotype-environment interactions to elucidate the pattern of maize root plasticity to nitrogen deficiency显示文摘Maize(Zea mays L.) root morphology exhibits a high degree of phenotypic plasticity to nitrogen(N) de ficiency,but the underlying genetic architecture remains to be investigated Using an advanced BC_4F_3 population,we investigated the root growth plasticity under two contrasted N levels and identi fied the quantitative trait loci(QTLs) with QTL-environment(Q×E)interaction effects. Principal components analysis(PCA) on changes of root traits to N de ficiency(D LN-HN) showed that root length and biomass contributed for 45.8% in the same magnitude and direction on the first PC,while root traits scattered highly on PC_2 and PC_3. Hierarchical cluster analysis on traits for D LN-HN further assigned the BC_4F_3 lines into six groups,in which the special phenotypic responses to N de ficiency was presented These results revealed the complicated root plasticity of maize in response to N de ficiency that can be caused by genotype environment(G×E) interactions. Furthermore,QTL mapping using a multi-environment analysis identi fied 35 QTLs for root traits. Nine of these QTLs exhibited signi ficant Q×E interaction effects. | 2 | 姜黄素诱导髓系白血病细胞凋亡的机制研究(英文)显示文摘Objective: To investigate the mechanism of apoptosis of myelocytic leukemia lines NB4, K562 and THP-1 cells induced by curcumin. Methods: After the cells were treated with curcumin at different concentration (25, 12.5, 6.25, 3.125, 0 μmol/L) for various times (0, 12, 24, 48 h), flow cytometry (FCM) was used to determine the rate of apoptosis of cells. After the cells were treated with curcumin at 25 μmol/L for 24 h, flow cytometry was used to determine the expression level of Fas, and Western blot was performed to determine the expression of Caspase-8 and Caspase-9. Results: (1) Curcumin could induced the apoptosis of NB4, K562 and THP-1 cells in a time- and dose-dependent manner. After the cells were treated with curcumin at 25 μmol/L for 48 h, the rate of apoptosis of cells was over fifty percent. (2) Fas level showed remarkable increase (P < 0.01) after above cells were treated with 25 μmol/L curcumin for 24 h. (3) The apoptosis proteins of Caspase-8 and Cas- pase-9 were also increased obviously (P < 0.01) after the cells were treated with 25 μmol/L curcumin for 24 h. | 4 | 睾丸的原发性NK/T细胞淋巴瘤一例报告及文献复习(英文)显示文摘Objective: To study the clinical and pathological features of primary NK/T cell lymphoma of testis and to investi- gate the effective diagnosis and treatment of this disease. Methods: The surgical specimens of a patient with primary NK/T cell lymphoma of the testis were observed by light microscopy, immunohistochemistry and examined by the polymerase chain reaction (PCR) for Epstein-Barr virus (EBV) DNA and T-cell receptor (TCR) gene rearrangement, and the literature were reviewed. Results: The patient presented with left-sided painless testicular enlargement and the lymphoma had a propensity to spread to the contralateral testis, spleen, central nervous system, and so on. The neoplastic cells were positive for CD56, CD45R0 and CD3ε, while the expressions of CD20, CD79α, CD5, Bcl-2 and PLAP were negative. In addition, the EBV DNA was detected in the lymphoma by PCR. And the results of gene rearrangement studies for the γ chain of the T-cell receptor were negative. The pathological diagnosis was NK/T cell lymphoma of the left testis. Conclusion: Primary NK/T cell lym- phoma of the testis is a rare entity and progressed rapidly. The histopathological, immunohistochemical, EBV examination and TCR gene rearrangement studies should be carried out as soon as possible in order to get the defined diagnosis. | 5 | Lineage-specific positive selection on ACE2 contributes to the genetic susceptibility of COVID-19显示文摘The Angiotensin-Converting Enzyme-2(ACE2)gene,located on Xp22.2,attracts a great deal of attention because the protein it encodes is believed to be the functional cellular receptor for the new coronavirus(SARS-CoV-2).However,recent studies are controversial,especially concerning the intrinsic link between ACE2 diversity and COVID-19 susceptibility.Here,we conduct a population genetic study on ACE2 in6354 individuals representing 210 present-day populations and 5329 individuals of ancient or archaic groups.We dissected the genetic architecture ofACE2 and identified two major haplogroups(hg)in East Asians,i.e.ACE2-hgl(43%)and ACE2-hg2(53%),while other populations harbor more diverse A CE2-hgs.Accordingly,there was a significant loss of ACE2 common variations in East Asians in contrast to the X-chromosome-wide and genome-wide patterns.Notably,association analysis between ACE2-hgs and COVID-19 severity in 1229 Han Chinese individuals with various levels of COVID-19 severity showed a higher risk ofACE2-hgl(odds ratio=1.56,P<0.01)and a lower risk of ACE2-hg2(odds ratio=0.65,P<0.01).Interestingly,ACE2-hgl is in strong linkage disequilibrium with rs1849863-C,which is an assumed risk factor of elevated plasma ACE2 level and is related to a higher risk of COVID-19 severity,hospitalization and infection.Strikingly,remarkable signatures of positive selection were detected,especially on ACE2-hg2,and were traced back to 100000 years ago(but rose to a strong level during the Bronze Age,5000~3000 years ago,in East Asians).The selection pressures could have stemmed from multiple sources,but pre-COVID-19 viral epidemics and pandemics might have been potential driving forces,which consequently contributed to the genetic susceptibility to COVID-19 within and between populations. | Yuwen Pan Panhong Liu Fang Wang Peng Wu Fanjun Cheng Xin Jin Shuhua Xu | 2022 | National Science Review2022,9,9: | 0 |
