| 3 | Genomic insights into the fast growth of paulownias and the formation of Paulownia witches' broom显示文摘Paulownias are among the fastest growing trees in the world,but they often suffer tremendous loss of wood production due to infection by Paulownia witches'broom(PaWB)phytoplasmas.In this study,we have sequenced and assembled a high-quality nuclear genome of Paulownia fortunei,a commonly cultivated paulownia species.The assembled genome of P.fortunei is 511.6 Mb in size,with 93.2%of its sequences anchored to 20 pseudo-chromosomes,and it contains 31985 protein-coding genes.Phylogenomic analyses show that the family Paulowniaceae is sister to a clade composed of Phrymaceae and Orobanchaceae.Higher photosynthetic efficiency is achieved by integrating C3 photosynthesis and the crassulacean acid metabolism pathway,which may contribute to the extremely fast growth habit of paulownia trees.Comparative transcriptome analyses reveal modules related to cambial growth and development,photosynthesis,and defense responses.Additional genome sequencing of PaWB phytoplasma,combined with functional analyses,indicates that the effector PaWB-SAP54 interacts directly with Paulownia PfSPLa,which in turn causes the degradation of PfSPLa by the ubiquitin-mediated pathway and leads to the formation of witches'broom.Taken together,these results provide significant insights into the biology of paulownias and the regulatory mechanism for the formation of PaWB. | Yabing Cao Guiling Sun Xiaoqiao Zhai Pingluo Xu Liming Ma Minjie Deng Zhenli Zhao Haibo Yang Yanpeng Dong Zhonghai Shang Yujie Lv Lijun Yan Haifang Liu Xibing Cao Bingbing Li Zhe Wang Xiaogai Zhao Haiyan Yu Fan Wang Wen Ma Jinling Huang Guoqiang Fan | 2021 | Molecular Plant2021,14,10: | 7 |
| 4 | Water softening by induced crystallization in fluidized bed显示文摘Fluidized bed and induced crystallization technology were combined to design a new type of induced crystallization fluidized bed reactor. The added particulate matter served as crystal nucleus to induce crystallization so that the insoluble material, which was in a saturated state, could precipitate on its surface. In this study, by filling the fluidized bed with quartz sand and by adjusting water p H, precipitation of calcium carbonate was induced on the surface of quartz sand, and the removal of water hardness was achieved.With a reactor influent flow of 60 L/hr, a fixed-bed height of 0.5 m, pH value of 9.5, quartz sand nuclear diameter of 0.2–0.4 mm, and a reflux ratio of 60%, the effluent concentration of calcium hardness was reduced to 60 mg/L and 86.6% removal efficiency was achieved. The resulting effluent reached the quality standard set for circulating cooling water. Majority of the material on the surface of quartz sand was calculated to be calcium carbonate based on energy spectrum analysis and moisture content was around 15.994%. | 7 | Reducing energy loss via tuning energy levels of polymer acceptors for efficient all-polymer solar cells显示文摘The open-circuit voltage(Voc) of all-polymer solar cells(all-PSCs) is typically lower than 0.9 V even for the most efficient ones.Large energy loss is the main reason for limiting Voc and efficiency of all-PSCs. Herein, through materials design using electron deficient building blocks based on bithiophene imides, the lowest unoccupied molecular orbital(LUMO) energy levels of polymer acceptors can be effectively tuned, which resulted in a reduced energy loss induced by charge generation and recombination loss due to the suppressed charge-transfer(CT) state absorption. Despite a negligible driving force, all-PSC based on the polymer donor and acceptor combination with well-aligned energy levels exhibited efficient charge transfer and achieved an external quantum efficiency over 70% while maintaining a large Voc of 1.02 V, leading to a 9.21% efficiency. | 8 | YTHDF2 is essential for spermatogenesis and fertility by mediating a wave of transcriptional transition in spermatogenic cells显示文摘The dynamic and reversible regulation roles of m^(6)A modification and the characterization of m^(6)A readers have provided new insights into spermatogenesis at the post-transcriptional level.YTHDF2,as an m^(6)A reader,has been reported to mediate the m^(6)A-containing transcript decay during the mouse oocyte maturation,embryonic stem cell differentiation,neural development,and zebrafish maternal-to-zygotic transition.However,the roles of YTHDF2 in mammalian spermatogenesis are uncertain.Here,we generated germ cell-specific Ythdf2 mutants(Ythdf2-vKO)at a C57BL/6J background and demonstrated that YTHDF2 is essential for mouse spermatogenesis and fertility.Ythdf2-vKO provides oligoasthenoteratozoospermia phenotype with increased apoptosis in germ cells.High-throughput RNA-seq analysis showed that a group of mRNAs is upregulated in Ythdf2-vKO mouse testis;further analysis and MeRIP-qPCR data showed that most of the upregulated genes in Ythdf2-vKO mouse testis are modified with m^(6)A and are YTHDF2 candidate binding genes.Interestingly,RNA-seq analysis combined with our previous single-cell transcriptomics data of mouse spermatogenesis pointed out the failure of a wave of transcript transition during the spermatogenesis of Ythdf2-vKO mice,which was confirmed by gene expression analysis using qPCR of diplotene spermatocytes and round spermatids obtained through fluorescence-activated cell sorting.Our study demonstrates the fundamental role of YTHDF2 during mouse spermatogenesis and provides a potential candidate for the diagnosis of male infertility with the oligoasthenoteratozoospermia syndrome. | Xinxi Zhao Zhen Lin Yong Fan Wenzhi Li Yujie Zhang Fei Li Tong Hong Hua Feng Minghan Tong Ningling Wang Yanping Kuang Qifeng Lyu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,12: | 2 |

| 10 | A large-scale outdoor atmospheric simulation smog chamber for studying atmospheric photochemical processes:Characterization and preliminary application显示文摘Understanding the formation mechanisms of secondary air pollution is very important for the formulation of air pollution control countermeasures in China.Thus,a large-scale outdoor atmospheric simulation smog chamber was constructed at Chinese Research Academy of Environmental Sciences(the CRAES Chamber),which was designed for simulating the atmospheric photochemical processes under the conditions close to the real atmospheric environment.The chamber consisted of a 56-m^(3) fluorinated ethylene propylene(FEP) Teflon film reactor,an electrically-driven stainless steel alloy shield,an auxiliary system,and multiple detection instrumentations.By performing a series of characterization experiments,we obtained basic parameters of the CRAES chamber,such as the mixing ability,the background reactivity,and the wall loss rates of gaseous compounds(propene,NO,NO_(2),ozone) and aerosols(ammonium sulfate).Oxidation experiments were also performed to study the formation of ozone and secondary organic aerosol(SOA),including α-pinene ozonolysis,propene and 1,3,5-trimethylbenzene photooxidation.Temperature and seed effects on the vapor wall loss and SOA yields were obtained in this work:higher temperature and the presence of seed could reduce the vapor wall loss;SOA yield was found to depend inversely on temperature,and the presence of seed could increase SOA yield.The seed was suggested to be used in the chamber to reduce the interaction between the gas phase and chamber walls.The results above showed that the CRAES chamber was reliable and could meet the demands for investigating tropospheric chemistry. | Junling Li Hong Li Xuezhong Wang Weigang Wang Maofa Ge Hao Zhang Xin Zhang Kun Li Yan Chen Zhenhai Wu Fahe Chai Fan Meng Yujing Mu Abdelwahid Mellouki Fang Bi Yujie Zhang Lingyan Wu Yongchun Liu | 2021 | Journal of Environmental Sciences2021,33,4: | 2 |

| 14 | Mixed secondary chromatin structure revealed by modeling radiation-induced DNA fragment length distribution显示文摘Spatial chromatin structure plays fundamental roles in many vital biological processes including DNA replication, transcription,damage and repair. However, the current understanding of the secondary structure of chromatin formed by local nucleosomenucleosome interactions remains controversial, especially for the existence and conformation of 30 nm structure. Since chromatin structure influences the fragment length distribution(FLD) of ionizing radiation-induced DNA strand breaks, a 3D chromatin model fitting FLD patterns can help to distinguish different models of chromatin structure. Here, we developed a novel '30-C' model combining 30 nm chromatin structure models with Hi-C data, which measured the spatial contact frequency between different loci in the genome. We first reconstructed the 3D coordinates of the 25 kb bins from Hi-C heatmaps. Within the25 kb bins, lower level chromatin structures supported by recent studies were filled. Simulated FLD patterns based on the 30-C model were compared to published FLD patterns induced by heavy ion radiation to validate the models.
