| 1 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 2 | NADPH is superior to NADH or edaravone in ameliorating metabolic disturbance and brain injury in ischemic stroke显示文摘Our previous studies confirm that exogenous reduced nicotinamide adenine dinucleotide phosphate(NADPH)exerts a neuroprotective effect in animal models of ischemic stroke,and its primary mechanism is related to anti-oxidative stress and improved energy metabolism.However,it is unknown whether nicotinamide adenine dinucleotide(NADH)also plays a neuroprotective role and whether NADPH is superior to NADH against ischemic stroke?In this study we compared the efficacy of NADH,NADPH,and edaravone in ameliorating brain injury and metabolic stress in ischemic stroke.Transient middle cerebral artery occlusion/reperfusion(t-MCAO/R)mouse model and in vitro oxygen glucose deprivation/reoxygenation(OGD/R)model were established.The mice were intravenously administered the optimal dose of NADPH(7.5 mg/kg),NADH(22.5 mg/kg),or edaravone(3 mg/kg)immediately after reperfusion.We showed that the overall efficacy of NADPH in ameliorating ischemic injury was superior to NADH and edaravone.NADPH had a longer therapeutic time window(within 5 h)after reperfusion than NADH and edaravone(within 2 h)for ischemic stroke.In addition,NADPH and edaravone were better in alleviating the brain atrophy,while NADH and NADPH were better in increasing the long-term survival rate.NADPH showed stronger antioxidant effects than NADH and edaravone;but NADH was the best in terms of maintaining energy metabolism.Taken together,this study demonstrates that NADPH exerts better neuroprotective effects against ischemic stroke than NADH and edaravone. | Xin-xin Wang Fan Wang Guang-hui Mao Jun-chao Wu Mei Li Rong Han Jing She Rong Zhang Rui Sheng Zhong Chen Zheng-hong Qin | 2022 | Acta Pharmacologica Sinica2022,43,3: | 4 |
| 3 | CPI-17-mediated contraction of vascular smooth muscle is essential for the development of hypertension in obese mice显示文摘Several factors have been implicated in obesity-related hypertension, but the genesis of the hypertension is largely unknown. In this study, we found a significantly upregulated expression of CPI-17(C-kinasepotentiated protein phosphatase 1 inhibitor of 17 kDa) and protein kinase C(PKC) isoforms in the vascular smooth muscles of high-fat diet(HFD)-fed obese mice. The obese wild-type mice showed a significant elevation of blood pressure and enhanced calcium-sensitized contraction of vascular smooth muscles. However, the obese CPI-17-deficient mice showed a normotensive blood pressure, and the calcium-sensitized contraction was consistently reduced. In addition, the mutant muscle displayed an abolished responsive force to a PKC activator and a 30%-50% reduction in both the initial peak force and sustained force in response to various G protein-coupled receptor(GPCR) agonists. Our observations showed that CPI-17-mediated calcium sensitization is mediated through a GPCR/PKC/CPI-17/MLCP/RLC signaling pathway. | 6 | Identifying the Risk of SARS-CoV-2 Infection and Environmental Monitoring in Airborne Infectious Isolation Rooms(AIIRs)显示文摘Healthcare workers(HCWs)are at high risk of occupational exposure to the new pandemic human coronavirus,SARSCoV-2,and are a source of nosocomial transmission in airborne infectious isolation rooms(AIIRs).Here,we performed comprehensive environmental contamination surveillance to evaluate the risk of viral transmission in AIIRs with 115 rooms in three buildings at the Shanghai Public Health Clinical Center,Shanghai,during the treatment of 334 patients infected with SARS-CoV-2.The results showed that the risk of airborne transmission of SARS-CoV-2 in AIIRs was low(1.62%,25/1544)due to the directional airflow and strong environmental hygiene procedures.However,we detected viral RNA on the surface of foot-operated openers and bathroom sinks in AIIRs(viral load:55.00–3154.50 copies/mL).This might be a source of contamination to connecting corridors and object surfaces through the footwear and gloves used by HCWs.The risk of infection was eliminated by the use of disposable footwear covers and the application of more effective environmental and personal hygiene measures.With the help of effective infection control procedures,none of 290 HCWs was infected when working in the AIIRs at this hospital.This study has provided information pertinent for infection control in AIIRs during the treatment of COVID-19 patients. | Zhi-Gang Song Yan-Mei Chen Fan Wu Lin Xu Bang-Fang Wang Lei Shi Xiao Chen Fa-Hui Dai Jia-Lei She Jian-Min Chen Edward CHolmes Tong-Yu Zhu Yong-Zhen Zhang | 2020 | Virologica Sinica2020,35,6: | 3 |

| 8 | Interfacial Characteristic of as-Deformed SiCp-Reinforced Magnesium Matrix Composite显示文摘The(submicron+micron) Si Cp-reinforced magnesium matrix composite was fabricated by stir casting. After the application of forging and extrusion, the interface between Si Cp and Mg in the composite was investigated by transmission electron microscopy. Results show that the interfacial characterization was different at the interfaces of micron-Si Cp/Mg and submicron-Si Cp/Mg. While most interfaces between micron-Si Cp and Mg were clean, the precipitated Mg17Al12 phase as well as dispersedly distributed nano-Mg O particles was observed at some interfaces. Unlike the interface between micron-Si Cp and Mg, no interfacial reaction product was found at the interface between submicron-Si Cp and Mg in the present study. Besides, the specific orientation relationships were found at the interfaces between submicronSi Cp and Mg, which was thought to have developed during hot deformation process. | 9 | Photocrosslinking silver nanoparticles–aloe vera–silk fibroin composite hydrogel for treatment of full-thickness cutaneous wounds显示文摘Damage to the skin causes physiological and functional issues.The most effective treatment approach is the use of wound dressings.Silk fibroin(SF)is a promising candidate biomaterial for regulating wound healing;however,its antibacterial properties and biological activity must be further improved.In this study,a photocrosslinking hydrogel was developed to treat full-thickness cutaneous wounds.The composite hydrogel(Ag-AV-SF hydrogel)was prepared by introducing the silver nanoparticles(AgNPs)and aloe vera(AV)as the modifiers.In vitro study exhibited great antibacterial ability,biocompatibility and cell-proliferation and-migration-promoting capacities.It also showed the pH-response releasing properties which release more AgNPs in a simulated chronic infection environment.The healing effect evaluation in vivo showed the healing-promoting ability of the Ag-AV-SF hydrogel was stronger than the single-modifiers groups,and the healing rate of it reached 97.02%on Day 21,higher than the commercial wound dressing,silver sulfadiazine(SS)cream on sale.Additionally,the histological and protein expression results showed that the Ag-AV-SF hydrogel has a greater effect on the pro-healing regenerative phenotype with M2 macrophages at the early stage,reconstructing the blood vessels networks and inhibiting the formation of scars.In summary,the Ag-AV-SF hydrogel developed in this study had good physical properties,overwhelming antibacterial properties,satisfactory biocompatibility and significantly promoting effect on cell proliferation,migration and wound healing.Overall,our results suggest that the Ag-AV-SF hydrogel we developed has great potential for improving the wound healing in clinical treatment. | Yangkun Liu JinChuan Fan MingQi Lv Kepeng She Jiale Sun Qingqing Lu Changhao Han SongTao Ding Shuang Zhao GuiXue Wang YuChan Zhang GuangChao Zang | 2021 | Regenerative Biomaterials2021,8,6: | 2 |
