| 1 | Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual. | Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang | 2020 | Protein & Cell2020,11,4: | 44 |

| 10 | The benefits of PCSK9 inhibitors in patients with acute coronary syndrome:a systematic review and meta-analysis显示文摘Background:Proprotein convertase subtilisin/kexin 9(PSCK9)inhibitors have been beneficial for many patients with hyperlipidemia.The objective of this study was to investigate the benefit of PSCK9 inhibitors in patients with acute coronary syndrome(ACS).Methods:We systematically searched PubMed,EMBASE,and Cochrane Clinical Trials(published before January 2023;no language restriction)to compare the treatment of patients with ACS using PCSK9 inhibitors and placebo.The primary end points were major adverse cardiovascular events,nonfatal myocardial infarction,cardiogenic death,stroke,hospitalization for recurrent ACS,and coronary revascularization.Fixed-or random-effects models were used to assess the aggregated data.Results:Of the 1686 identified studies,5 were eligible and included in our analysis(of a total of 38,005 participants,18,609 cases were placed in the PCSK9 inhibitor treatment group and 19,396 cases in the placebo group).Compared with the placebo group,PCSK9 inhibitors significantly reduced the major adverse cardiovascular events(odds ratio[OR]:0.83;95%confidence interval[CI]:0.77–0.88;P<0.00001)for patients following ACS.The incidence of nonfatal myocardial infarction(relative risk:0.80;95%CI:0.74–0.87;P<0.00001),cardiovascular death(OR:0.96;95%CI:0.83–1.10;P=0.56),stroke(OR:0.74;95%CI:0.63–0.88;P=0.0007),hospitalization for recurrent ACS(OR:0.57;95%CI:0.40–0.83;P=0.003),or coronary revascularization(OR:0.82;95%CI:0.76–0.88;P<0.00001)all demonstrated a significant decrease in the comparison between the 2 groups.Conclusion:This meta-analysis demonstrated that treatment with PCSK9 inhibitors in patients with ACS reduced the probability of multiple cardiovascular events and improved patient prognosis. | Guanzhao Zhang Shuting Chang Faming Zhao Xiangfeng Guan Zifan Nie Wenhao Liu Bo Li | 2024 | Emergency and Critical Care Medicine2024,4,1: | 0 |

| 13 | Reconstruction and Dynamics of the Human Intestinal Microbiome Observed In Situ显示文摘The human gut microbiome has primarily been studied through the use of fecal samples,a practice that has generated vital knowledge on the composition and functional capacities of gastrointestinal microbial communities.However,this reliance on fecal materials limits the investigation of microbial dynamics in other locations along the gastrointestinal tract(in situ),and the infrequent availability of fecal samples prevents analysis at finer temporal scales(e.g.,hours).In our study,we utilized colonic transendoscopic enteral tubing,a technology originally developed for fecal microbiota transplantation,to sample the ileocecal microbiome twice daily;metagenomic and metatranscriptomic analyses were then conducted on these samples.A total of 43 ileocecal and 28 urine and fecal samples were collected from five healthy volunteers.The ileocecal and fecal microbiomes,as profiled in the five volunteers,were found to be similar in metagenomic profiling,yet their active genes(metatranscriptome)were found to be highly distinct.Both microbiomes were perturbed after laxative exposure;over time,they exhibited reduced dissimilarity to their pre-treatment state,thereby demonstrating resilience as an innate property of the gut microbiome,although they did not fully recover within our observation time window.Sampling of the ileocecal microbiome during the day and at night revealed the existence of diurnal rhythms in a series of bacterial species and functional pathways,particularly those related to short-chain fatty acid production,such as Propionibacterium acnes and coenzyme A biosynthesis Ⅱ.Autocorrelation analysis and fluctuations decomposition further indicated the significant periodicity of the diurnal oscillations.Metabolomic profiling in the fecal and urine samples mirrored the perturbance and recovery in the gut microbiome,indicating the crucial contribution of the gut microbiome to many key metabolites involved in host health.This study provides novel insights into the human gut microbiome and its inner resilience and diurnal rhythms,as well as the potential consequences of these to the host. | Xiaolin Liu Min Dai Yue Ma Na Zhao Ziyu Wang Ying Yu Yakun Xu Huijie Zhang Liyuan Xiang He Tian Guanghou Shui Faming Zhang Jun Wang | 2022 | Engineering2022,8,8: | 0 |
| 14 | Effects of Chemical Reactions in the Hypersonic Reacting Flow around Blunt Bodies显示文摘The effects of chemical reactions in the hypersonic reacting flow are investigated using an integrated algorithm considering simultaneously two different reaction mechanisms,i.e.,including the high temperature air nonequilibrium chemical reactions and the H_2-air combustion reactions. The program is validated by the air non-equilibrium flow at Mach number of 25.9 with the RAM C-II configuration and the shock-induced combustion flow at Mach number of 4.512 6 around a sphere,respectively. Furthermore,the mixed reacting flow with the Mach number of 10.0 with an opposing jet of hydrogen is numerically analyzed. The results show that the program is reliable,and the effects of chemical reactions engender in the decrease of peak temperature along characteristic lines,as well as on the surface.
