1Dexamethasone-loaded ROS-responsive poly(thioketal)nanoparticles suppress inflammation and oxidative stress of acute lung injury显示文摘Acute lung injury(ALI)is associated with excessive inflammatory response,leading to acute respiratory distress syndrome(ARDS)without timely treatment.A fewer effective drugs are available currently to treat the ALI/ARDS.Herein,a therapeutic nanoplatform with reactive oxygen species(ROS)-responsiveness was developed for the regulation of inflammation.Dexamethasone acetate(Dex)was encapsulated into poly(thioketal)polymers to form polymeric nanoparticles(NPs)(PTKNPs@Dex).The NPs were composed of poly(1,4-phenyleneacetonedimethylene thioketal)(PPADT)and polythioketal urethane(PTKU),in which the thioketal bonds could be cleaved by the high level of ROS at the ALI site.The PTKNPs@Dex could accumulate in the pulmonary inflammatory sites and release the encapsulated payloads rapidly,leading to the decreased ROS level,less generation of pro-inflammatory cytokines,and reduced lung injury and mortality of mice.RNA sequencing(RNA-seq)analysis showed that the therapeutic efficacy of the NPs was associated with the modulation of many immune and inflammation-linked pathways.These findings provide a newly developed nanoplatform for the efficient treatment of ALI/ARDS.Zihe Zhai Wei Ouyang Yuejun Yao Yuqi Zhang Haolan Zhang Feng Xu Changyou Gao 2022Bioactive Materials2022,7,8:5
2Regulation of phenylpropanoid biosynthesis by MdMYB88 and MdMYB124 contributes to pathogen and drought resistance in apple显示文摘MdMYB88 and MdMYB124 have been demonstrated to be responsible for lignin accumulation in apple under drought stress.In this study,using a metabolomic approach,we identified differentially accumulated phenylpropanoid and flavonoid metabolites in MdMYB88/124 transgenic RNAi plants under control and long-term drought stress conditions in apple roots.We confirmed the regulation of phenylalanine by MdMYB88 and MdMYB124 via UPLC-MS in apple roots under both control and drought conditions.Using Electrophoretic Mobility Shift Assay(EMSA)and ChIPquantitative PCR(qPCR)analyses,we found that MdMYB88 positively regulates the MdCM2 gene,which is responsible for phenylalanine biosynthesis,through binding to its promoter region.Under long-term drought conditions,MdMYB88/124 RNAi plants consistently accumulated increased amounts of H2O2 and MDA,while MdMYB88 and MdMYB124 overexpression plants accumulated decreased amounts of H2O2 and MDA.We also examined the accumulation of metabolites in the phenylpropanoid biosynthesis pathway in the leaves of MdMYB88 and MdMYB124 transgenic apple plants after long-term drought stress.We found that metabolites responsible for plant defense,including phenylpropanoids and flavonoids,accumulated less in the RNAi plants but more in the overexpression plants under both control and drought conditions.We further demonstrated that MdMYB88/124 RNAi plants were more sensitive to Alternaria alternata f.sp.mali and Valsa mali,two pathogens that currently severely threaten apple production.In contrast,MdMYB88 and MdMYB124 overexpression plants were more tolerant to these pathogens.The cumulative results of this study provided evidence for secondary metabolite regulation by MdMYB88 and MdMYB124,further explained the molecular roles of MdMYB88 and MdMYB124 in drought resistance,and provided information concerning molecular aspects of their roles in disease resistance.Dali Geng Xiaoxia Shen Yinpeng Xie Yusen Yang Ruiling Bian Yuqi Gao Pengmin Li Liying Sun Hao Feng Fengwang Ma Qingmei Guan 2020Horticulture Research2020,7,1:5

5Screening of amino acids in dried blood spots by stable isotope derivatization-liquid chromatography-electrospray ionization mass spectrometry显示文摘Direct infusion mass spectrometry(DIMS) is a powerful technique in clinical diagnosis for screening neonatal amino acid metabolic disorders from dried blood spots(DBS).However,DIMS sometimes generated false-positive results for analysis of amino acids.In this work,we utilized a stable isotope derivatization method,combining with liquid chromatography tandem mass spectrometry(SID-LC-MS),to improve the specificity for screening amino acids in DBS specimens.A pair of isotope reagents,p-(dimethylamino)phenyl isothiocyanate(DMAP-NCS) and 4-isothiocyanato-N,N-bis(methyl-[2H2])aniline([2H4]DMAP-NCS),was synthesized and used to label amino acids in DBS specimens.The [2H4]DMAP-NCS labelled amino acid standards were used as internal standards to compensate the matrix effect.This method was validated by measuring linearity,recovery and accuracy.The results showed that the developed SID-LC-MS method can be used for sensitive and selective determination of 12 diagnostically important amino acids in DBS specimens.Huaming Xiao Ping Liu Shujian Zheng Xian Wang Jun Ding Yuqi Feng 2020Chinese Chemical Letters2020,31,9:3

8Analytical methods for locating modifications in nucleic acids显示文摘In addition to the canonical nucleobases, a variety of chemical modifications have been identified presence in nucleic acids. These modifications have been demonstrated to involve in regulating the spatiotemporal expression of genes. Up to date, over 150 types of chemical modifications have been found existence in nucleic acids. Understanding the functional roles of modifications relies on deciphering the location information of modifications in nucleic acids. Analytical methods for studying nucleic acid modifications have greatly advanced in the last decade. To locate the modifications in nucleic acids, various mass spectrometry (MS)-based analytical strategies have been established. Recent progress in next-generation sequencing (NGS) in conjugation with immunoprecipitation, chemical reaction, enzyme-mediated mutation, or nanomaterials offer genome-wide or transcriptome-wide mapping of modifications, which greatly revolutionize the field of epigenetic modifications. Herein, we reviewed and summarized the established methods and the breakthrough of the techniques for locating modifications in nucleic acids. In addition, we discussed the principles, applications, advantages and drawbacks of these methods. 12The influence of the metal net charge of non-metallocene early transition metal catalyst on the ethylene polymerization activity显示文摘The net charges on central metals of a serial non-metallocene early transition metal catalysts (FI catalyst) with similar steric hindrance were caculated with MM-QEq (molecular mechmism-charge equilibration) method and associated with ethylene polymerization activities of these FI catalyts. It was found that the activity increased with the net charge on metal if ignoring the influence of the steric hindrance. In other words, introduction of strong and/or more electron-withdrawing groups onto the ligand of FI catalyst would enhance the activity of the catalyst. 13Characteristics of Uranium Mineralization in Red Clastic Formations in the Southwestern Margin of the Ordos Basin显示文摘In the southwestern margin of the Ordos Basin,uranium mineralization is primarily hosted by predominantly oxidative red clastic formations in the Lower Cretaceous.The main target layers for uranium exploration are the Madongshan and Liwaxia formations of the Liupanshan Group,followed by the Jingchuan Formation of the Zhidan Group.The host rocks(medium-fine feldspar quartz sandstone),which are bleached to a light grayish white color,contain a minor organic matter component and pyrite.Uranium mineralization changes from surficial infiltration or phreatic oxidation in the upper part to interlayer oxidation in the lower part.Uranium ore bodies are mostly lenticular or tabular in shape,locally shaped like crescent rolls.Individual ore bodies are typically small and shallow.Uranium predominantly manifests as pitchblende and coffinite.Coffinite is usually short and columnar or granular in habit,whereas pitchblende occurs as an irregular colloidal covering on the surface or in fissures of ferric oxide,silicate,clay or carbonate.Secondary uranium minerals are torbernite,uranophane,and uranopilite.Minerals associated with uranium are mainly pyrite,chalcopyrite and,to a minor extent,arsenopyrite and fluorite.The associated elements are Mo,V,Se,Co,Ni,and Mn,the host sandstone being high in Cu and Ba.Overall,the red clastic formations in the southwestern margin of the Ordos Basin are characterized by’five multiples but one low’which means multiple target layers,multiple stages of mineralization,multiple ore body shapes,multiple kinds of uranium minerals,multiple associated elements,but low organic matter.This implies an overall complex uranium metallogenic environment and mineralization process.It is recommended that future uranium exploration should take into consideration regional metallogenic conditions and mineralization features,with target layers in the wide-smooth synclinal slope being focused on.Most uranium deposits are small to medium in size,and the main type of uranium mineralization can vary by target layer.ZHANG Zilong HE Feng YI Longsheng FAN Honghai CAI Yuqi LIU Hongxu LIU Xinyang CHEN Hongbin JIA Cui YANG Mengjia 2020Acta Geologica Sinica(English Edition)2020,94,5:1

17Formation of an anticyclonic eddy and the mechanism involved: a case study using cruise data from the northern South China Sea显示文摘Data from satellite altimetry and in situ observations together with the Hybrid Coordinate Ocean Model (HYCOM) reanalysis data were used to investigate the mechanism and formation of an anticyclonic eddy in the northeastern South China Sea (SCS). Analysis of water mass using cruise data indicated that the water captured in the eddy diff ers from those in the SCS, the Kuroshio intrusion, and the eddy-forming region. Data from sea surface height (SSH) and sea level anomaly (SLA) indicate that the eddy formed due both to the Kuroshio intrusion and the local circulation in the SCS. The Kuroshio intrusion is present at the start of the eddy growth (March 5-9) before Kuroshio leaps the Luzon Strait. The eddy then becomes larger and stronger in the absence of the Kuroshio intrusion. From the eddy budget of the HYCOM reanalysis data, the formation of the eddy goes in three steps. By the third step, the eddy had become aff ected by variations of local SCS circulation, which is more strongly than in the fi rst step in which it is aff ected more by the Kuroshio intrusion. The variability of the temperature and salinity inside the eddy provide a support to this conclusion.
