1Pyogenic liver abscesses associated with nonmetastatic colorectal cancers: An increasing problem in Eastern Asia显示文摘AIM: To elaborate the clinicopathologic features of colorectal cancer-related pyogenic liver abscess (PLA). METHODS: Reported cases of colorectal cancer-related PLAs were collected from the literature published up to October 2011 and evaluated for their clinicopathologic features. Data of collected cases included demographics, clinical presentation, microbial findings and treatment. Categorical variables were compared by 2 analysis and continuous variables were evaluated using Student's t test. RESULTS: A total 96 cases of colorectal cancer-related PLA were collected from the previous literature. Most patients (60%) were male and 40% cases occurred in the age group of 61-70 years. Apart from some special types of PLA, there were significant differences in the microbiological spectrum between Eastern Asia and non-Eastern Asian countries, which implied different risk factors and courses of the disease. Gram negative bacteria especially Klebsiella pneumoniae (K. pneu- moniae ) PLA was predominant in Eastern Asia (80.0%) in contrast to non-Eastern Asian countries (P < 0.01). Meanwhile, most of the Eastern Asian patients exhibited smaller size of liver abscess and atypical presentation. Sigmoid colon and rectum (72.73%) were the main sites of tumor in Eastern Asian patients, whereas tumor sites were uneven among most of the non-Easter Asian PLA patients. 2Beneficial Effect of Moderately Increasing Hypothermic Machine Perfusion Pressure on Donor after Cardiac Death Renal Transplantation显示文摘Background:Vascular resistance and flow rate during hypotherrnic machine perfusion (HMP)of kidneys is correlated with graft function. We aimed to determine the effects of increasing HMP pressure versus maintaining the initial pressure on kidney transplantation outcomes. Methods:We retrospectively reviewed the data of 76 primary transplantation patients who received HMP-preserved kidneys from 48 donors after cardiac death between September 1,2013,and August 31,2015.HMP pressure was increased from 30 to 40mmHg (1mmHg =0.133kPa)in kidneys with poor flow and/or vascular resistance (increased pressure [IP]group;36 patients);otherwise,the initial pressure was maintained (constant pressure group;40 patients).Finally,the clinical characteristics and transplantation outcomes in both groups were assessed. Results:Delayed graft function (DGF)incidence,1-year allograft,patient survival,kidney function recovery time,and serum creatinine level on day 30 were similar in both groups,with improved flow and resistance in the IP group.Among patients with DGF,kidney function recovery time and DGF duration were ameliorated in the IP group.Multivariate logistic regression analysis revealed that donor hypertension (odds ratio [OR]:1.43,95%confidence interval [CI]:1.02-2.06,P =0.035),donor terminal serum creatinine (OR:1.27,95%C7:1.06-1.62,P =0.023),warm ischemic time (OR:3.45,95%CI:1.97-6.37,P =0.002),and terminal resistance (OR:3.12,95%CI:1.76-6.09,P =0.012)were independent predictors of DGF.Cox proportional hazards analysis showed that terminal resistance (hazard ratio:2.06,95%C1:1.32-5.16,P =0.032)significantly affected graft survival. 3BAFF, involved in B cell activation through the NF-κB pathway, is related to disease activity and bone destruction in rheumatoid arthritis显示文摘B cell activating factor of TNF family(BAFF)is a member of TNF ligand superfamily and plays a key role in B cell homeostasis,proliferation,maturation,and survival.In this study,we detected BAFF level,the expressions of BAFF receptors and important molecules in NF-κB pathway in rheumatoid arthritis(RA)patients and analyzed the correlation between BAFF level and clinical variables,laboratory parameters or X-ray scores in order to elucidate the roles of BAFF in RA.A total of 50 RA patients and 50 healthy controls(HCs)were enrolled.We showed that the serum BAFF level in RA patients was significantly higher than that of HCs,and the percentages of B cell subsets(CD19^(+)B cells,CD19^(+)CD27^(+)B cells,CD19^(+)CD20^(+)CD27^(+)B cells,and CD19^(+)CD20−CD27^(+)B cells)in the serum of RA patients were significantly increased compared with those of HCs.The percentages of CD19^(+)BAFFR^(+)B cells,CD19^(+)BCMA^(+)B cells,and CD19^(+)TACI^(+)B cells in RA patients were significantly increased compared with those in HCs.The expression of important molecules in the NF-κB pathway(MKK3,MKK6,p-P38,p-P65,TRAF2,and p52)was significantly higher in RA patients than in HCs,but p100 level in RA patients was lower than that in HCs.The serum BAFF level was positively correlated with C-reactive protein,rheumatoid factor,disease activity score(in 28 joints),swollen joint counts,tender joint counts,and X-ray scores.When normal B cells were treated with BAFF in vitro,the percentages of the B cell subset and the expression of BAFF receptors were significantly upregulated.BAFF also promoted the expression of MKK3,MKK6,p-P38,p-P65,TRAF2,and p52.In conclusion,this study demonstrates that BAFF level is correlated with the disease activity and bone destruction of RA.BAFF is involved in the differentiation,proliferation,and activation of B cells in RA through NF-κB signaling pathway,suggesting that BAFF might be an ideal therapeutic target for RA.Ling-ling Zhang Hui Xiao Feng Zhang Yu-jing Wu Jin-ling Shu Ying Li Yu Tai Sheng-qian Xu Jian-hua Xu Wei Wei 2021Acta Pharmacologica Sinica2021,42,10:9
4MiRNA-206 inhibits hepatocellular carcinoma cell proliferation and migration but promotes apoptosis by modulating cMET expression显示文摘A close relationship between cancer progression and microRNAs(miRNAs)regulation has been demonstrated.Abnormal microRNA-206(miR-206)expression has been shown to be related to the development of malignancies.However,the role of miR-206 in hepatocellular carcinoma(HCC)remains unclear.Here,we evaluated the function of miR-206 in HCC.Results showed that miR-206 expression was decreased in 27 human HCC tissues compared with that of adjacent normal tissues.Conversely,cMET was up-regulated in human HCC cancer tissues,and cMET levels were shown to be negatively correlated with miR-206 expression.Abnormally increased miR-206 expression in three HCC cell lines(SMMC-7721,HepG2,and Huh7)attenuated cell viability,migration,and invasion.Increased apoptosis was also observed in these miR-206 expressing cells.Furthermore,we identified that miR-206 targets the 3′-UTR of the cMET gene for silencing,and restoration of cMET expression reversed the inhibitory effect of miR-206 on HCC.Tumor cells expressing miR-206 also showed delayed growth in the in vivo experiments compared with the controls.Altogether,our findings provide new insights into the molecular mechanisms of HCC oncogenesis.Yuanxi Wang Qinwen Tai Jinhui Zhang Junsheng Kang Feng Gao Feng Zhong Liquan Cai Fa Fang Yi Gao 2019Acta Biochimica et Biophysica Sinica2019,51,3:7
5Paeoniflorin-6′-0-benzene sulfonate alleviates collageninduced arthritis in mice by downregulating BAFF-TRAF2-NF-kB signaling: comparison with biological agents显示文摘Paeoniflorin-6′-0-benzene sulfonate (CP-25) is a new ester derivative of paeoniflorin with improved lipid solubility and oral bioavailability, as well as better anti-inflammatory activity than its parent compound. In this study we explored whether CP-25 exerted therapeutic effects in collagen-induced arthritis (CIA) mice through regulating B-cell activating factor (BAFF)-BAFF receptors-mediated signaling pathways. CIA mice were given CP-25 or injected with biological agents rituximab or etanercept for 40 days. In CIA mice, we found that T cells and B cells exhibited abnormal proliferation;the percentages of CD19^+ total B cells, CD19^+CD27^+-activated B cells, CD19^+BAFFR^+ and CD19^+TACI^+cells were significantly increased in PBMCs and spleen lymphocytes. CP-25 suppressed the indicators of arthritis, alleviated histopathology, accompanied by reduced BAFF and BAFF receptors expressions, inhibited serum immunoglobulin levels, decreased the Bell subsets percentages, and prevented the expressions of key molecules in NF-kB signaling. Furthermore, we showed that treatment with CP-25 reduced CD19+TRAF2^+ cell expressions stimulated by BAFF and decreased TRAF2 overexpression in HEK293 cells in vitro. Thus, CP-25 restored the abnormal T cells proliferation and B-cell percentages to the normal levels, and normalized the elevated levels of IgA, lgG2a and key proteins in NF-kB signaling. In comparison, rituximab and etanercept displayed stronger antiinflammatory activities than CP-25;they suppressed the elevated inflammatory indexes to below the normal levels in CIA mice. In summary, our results provide evidence that CP-25 alleviates CIA and regulates the functions of B cells through BAFF-TRAF2-NF-kB signaling. 6Effects of rare earth La on the microstructure and melting property of(Ag-Cu_(28))-30Sn alloys prepared by mechanical alloying显示文摘To obtain novel intermediate temperature alloy solders with a melting temperature of 400-600°C,nominal(Ag-Cu28)-30Sn alloys without or with a trace addition(0.5 or 1.0 wt.%) of rare earth(RE) element La were prepared by mechanical alloying.The aim of this research is to investigate the effects of the addition of La on the microstructures,alloying process and melting properties of(Ag-Cu28)-30Sn alloys.The results show that the addition of La produces no new phase.A trace amount of La addition can effectively refine the grain size,but the excessive addition of 1.0 wt.% La inhibits the alloying process.The influence of La on the melting temperatures of solder alloys is negligible.However,the trace addition of 0.5 wt.% La can distinctly reduce the fusion zone and improve the melting property of(Ag-Cu28)-30Sn alloys.LI Liangfeng QIU Tai YANG Jian FENG Yongbao 2011Rare Metals2011,30,1:6
7Complete disassociation of adult pancreas into viable single cells through cold trypsin-EDTA digestion显示文摘The in vitro isolation and analysis of pancreatic stem/progenitor cells are necessary for understanding their properties and function; however, the preparation of high-quality single-cell suspensions from adult pancreas is prerequisite. In this study, we applied a cold trypsin-ethylenediaminetetraacetic acid (EDTA) digestion method to disassociate adult mouse pancreata into single cells. The yield of single cells and the viability of the harvested cells were much higher than those obtained via the two commonly used warm digestion methods. Flow cytometric analysis showed that the ratio of ductal or BCRP1-positive cells in cell suspensions prepared through cold digestion was consistent with that found in vivo. Cell culture tests showed that pancreatic epithelial cells prepared by cold digestion maintained proliferative capacity comparable to those derived from warm collagenase digestion. 8Direct Effects of Activin A on the Activation of Mouse Macrophage RAW264.7 Cells显示文摘Macrophages play critical roles in innate immune and acquired immune via secreting pro-inflammatory mediators, phagocytosing microorganisms and presenting antigens. Activin A, a member of transforming growth factor β (TGF-β) superfamily, is produced by macrophages and microglia cells. In this study, we reported a direct effect of activin A as a pro-inflammatory factor on mouse macrophage cell line RAW264.7 cells. Our data revealed that activin A could not only increase IL-1β and IL-6 production from RAW264.7 cells, but also promote pinocytic and phagocytic activities of RAW264.7 cells. In addition, activin A obviously up-regulated MHC II expression on the surface of RAW264.7 cells, whereas did not influence MHC I expression. Activin A also enhanced CD80 expression, which is a marker of activated macrophages, but did not influence RAW264.7 cell proliferation. 9Monitoring Value of Multimodal Magnetic Resonance Imaging in Disease Progression of Amyotrophic Lateral Sclerosis: A Prospective Observational Study显示文摘Background: Ongoing efforts have been made to identify new neuroimaging markers to track amyotrophic lateral sclerosis (ALS) progression. This study aimed to explore the monitoring value of multimodal magnetic resonance imaging (MRI) in the disease progression of ALS. Methods: From September 2015 to March 2017, ten patients diagnosed with ALS in Peking Union Medical College Hospital completed head MRI scans at baseline and during follow-up. Multimodal MRI analyses, including gray matter (GM) volume measured by voxel-based morphometry;cerebral blood flow (CBF) evaluated by arterial spin labeling;functional connectivity, including low-frequency fluctuation (fALFF) and regional homogeneity (ReHo), measured by resting-state functional MRI;and integrity of white-matter (WM) fiber tracts evaluated by diffusion tensor imaging, were performed in these patients. Comparisons of imaging metrics were made between baseline and follow-up using paired t-test. Results: In the longitudinal comparisons, the brain structure (GM volume of the right precentral gyri, left postcentral gyri, and right thalami) and perfusion (CBF of the bilateral temporal poles, left precentral gyri, postcentral gyri, and right middle temporal gyri) in both motor and extramotor areas at follow-up were impaired to different extents when compared with those at baseline (all P < 0.05, false discovery rate adjusted). Functional connectivity was increased in the motor areas (fALFF of the right precentral gyri and superior frontal gyri, and ReHo of right precentral gyri) and decreased in the extramotor areas (fALFF of the bilateral middle frontal gyri and ReHo of the right precuneus and cingulate gyri) (all P < 0.001, unadjusted). No significant changes were detected in terms of brain WM measures. 11Effects of nano-sized Ag reinforcing particulates on the microstructure of Sn-0.7Cu solder joints显示文摘Composite solders were prepared by mechanically dispersing different volumes of nano-sized Ag particles into the Sn-0.7Cu eutectic solder. The effects of Ag particle addition on the microstructure of Sn-0.7Cu solder joints were investigated. Besides, the effects of isothermal aging on the microstructural evolution in the interfacial intermetallic compound (IMC) layer of the Sn-0.7Cu solder and the composite solder reinforced with 1vol% Ag particles were analyzed, respectively. Experimental results indicate that the growth rate of the interfacial IMC layer in the Ag particles reinforced composite solder joint is much lower than that in the Sn-0.7Cu solder joint during isothermal aging. 15Theoretical Study of Similar Experimental Method for Durability of Concrete under Artifi cial Climate Environment显示文摘Based on the similarity theory,a new experimentalmethod named Similar ExperimentalMethod for Durability of Concrete(SEMDC)was established.The existing experimentalmethods for durability of concrete were summarized,and the merits and demerits of these experimentalmethods were analyzed.Major factors affecting the durability of concrete were found through literature review.These factors were analyzed and the similarity criteria were established according to the similarity theory,and then the SEMDC was established according to the rules of these criteria.The various influentialfactors of the experimentalmethod were analyzed and the merits and demerits of this new experimentalmethod were discussed.According to SEMDC,changing the geometry shrinkage ratio was the only way to accelerate the test in order to keep the experiment similar to the reality.There were few other parameters which need to be changed in SEMDC,making the test easy to be achieved.According to SEMDC,time shrinkage ratio was the square of geometric shrinkage ratio,so an appropriate increase of the geometric shrinkage ratio could accelerate the test.Finally,an example of experimentaldesign for durability of concrete was devised theoretically base on SEMDC theory.耿欧 FENG Tai LI Debao LI Qingtao 2016Journal of Wuhan University of Technology(Materials Science)2016,31,2:2
