| 1 | Comprehensive analysis of NAC transcription factors and their expression during fruit spine development in cucumber (Cucumis sativus L.)显示文摘The cucumber(Cucumis sativus L.)is an important vegetable crop worldwide,and fruit trichomes or spines are an important trait for external fruit quality.The mechanisms underlying spine formation are not well understood,but the plant-specific NAC family of transcription factors may play important roles in fruit spine initiation and development.In this study,we conducted a genome-wide survey and identified 91 NAC gene homologs in the cucumber genome.Clustering analysis classified these genes into six subfamilies;each contained a varying number of NAC family members with a similar intron–exon structure and conserved motifs.Quantitative real-time PCR analysis revealed tissue-specific expression patterns of these genes,including 10 and 12 that exhibited preferential expression in the stem and fruit,respectively.Thirteen of the 91 NAC genes showed higher expression in the wild-type plant than in its near-isogenic trichome mutant,suggesting their important roles in fruit spine development.Exogenous application of four plant hormones promoted spine formation and increased spine density on the cucumber fruits;several NAC genes showed differential expression over time in response to phytohormone treatments on cucumber fruit,implying their essential roles in fruit-trichome development.Among the NAC genes identified,12 were found to be targets of 13 known cucumber micro-RNAs.Collectively,these findings provide a useful resource for further analysis of the interactions between NAC genes and genes underlying trichome organogenesis and development during fruit spine development in cucumber. | Xingwang Liu Ting wang Ezra Bartholomew Kezia Black Mingming Dong Yaqi Zhang Sen Yang Yanling Cai Shudan Xue Yiqun Weng Huazhong Ren | 2018 | Horticulture Research2018，5，1： | 10 |
| 2 | Apoptotic pathways as a therapeutic target for colorectal cancer treatment显示文摘Colorectal cancer is the second leading cause of death from cancer among adults. The disease begins as a benign adenomatous polyp, which develops into an advanced adenoma with high-grade dysplasia and then progresses to an invasive cancer. Appropriate apoptotic signaling is fundamentally important to preserve a healthy balance between cell death and cell survival and in maintaining genome integrity. Evasion of apoptotic pathway has been established as a prominent hallmark of several cancers. During colorectal cancer development, the balance between the rates of cell growth and apoptosis that maintains intestinal epithelial cell homeostasis gets progressively disturbed. Evidences are increasingly available to support the hypothesis that failure of apoptosis may be an important factor in the evolution of colorectal cancer and its poor response to chemotherapy and radiation. The other reason for targeting apoptotic pathway in the treatment of cancer is based on the observation that this process is deregulated in cancer cells but not in normal cells. As a result, colorectal cancer therapies designed to stimulate apoptosis in target cells would play a critical role in controlling its development and progression. A better understanding of the apoptotic signaling pathways, and the mechanisms by which cancer cells evade apoptotic death might lead to effective therapeutic strategies to inhibit cancer cell proliferation with minimal toxicity and high responses to chemotherapy. | 5 | Antioxidant therapy for chronic hepatitis C after failure of interferon:Results of phase Ⅱ randomized,double-blind placebo controlled clinical trial显示文摘AIM:TO assess the safety and efficacy of antioxidant therapy for patients with chronic hepatitis C virus(HCV) infection. METHODS:One hundred chronic HCV infection patients failed in interferon treatment were enrolled and randomly assigned to receive combined intravenous and oral antioxidants or placebo,or oral treatment alone. Primary end points were liver enzymes,HCV-RNA levels and histology. RESULTS:Combined oral and intravenous antioxidant therapy was associated with a significant decline in ALT levels in 52% of patients who received antioxidant therapy vs 20% of patients who received placebo(P= 0.05).Histology activity index(HAI)score at the end of treatment was reduced in 48% of patients who received antioxidant therapy vs 26% of patients who received placebo(P=0.21).HCV-RNA levels decreased by 1-log or more in 28% of patients who received antioxidant therapy vs 12% who received placebo(P=NS).In partⅡof the trial,oral administration of antioxidants was not associated with significant alterations in any of the end points. CONCLUSION:Antioxidant therapy has a mild beneficial effect on the inflammatory response of chronic HCV infection patients who are non-responders to interferon.
