2Hepatitis B virus-induced oncogenesis显示文摘Hepatocellular carcinoma (HCC) is one of the most com- mon cancers in the world with an annual incidence of more than 500 000 in the year 2000. Its incidence is rising in many countries. Recently, it has been estimated that about 53% of HCC cases in the world are related to hepatitis B virus (HBV). The epidemiological association of HBV with HCC is well established. In recent studies, it was revealed that HBsAg carriers have a 25-37 times increased risk of developing HCC as compared to non-infected people. At present, HBV-associated carcinogenesis can be seen as a multi-factorial process that includes both direct and indirect mechanisms that might act synergistically. The integration of HBV DNA into the host genome occurs at early steps of clonal tumor expansion. The integration has been shown in a number of cases to affect a variety of cancer- related genes and to exert insertional mutagenesis. The permanent liver inflammation, induced by the immune response, resulting in a degeneration and regeneration process confers to the accumulation of critical mutations in the host genome. In addition to this, the regulatory proteins HBx and the PreS2 activators that can be encoded by the integrate exert a tumor promoter-like function resulting in positive selection of cells producing a functional regulatory protein. 5盐胁迫下胡杨和毛白杨叶细胞中的离子区隔化显示文摘该文以抗盐的胡杨和盐敏感的毛白杨组培再生苗为实验材料,利用X--射线微区分析技术研究盐胁迫下(为期20d的盐处理)叶片各种类型细胞内主要元素Na+、Cl-、K+的分布情况,测定的细胞类型包括:①叶脉细胞(叶脉维管束导管、叶脉维管束内部和外部的薄壁细胞);②叶肉细胞(栅栏组织、海绵组织);③表皮细胞(上、下表皮细胞).结果表明,胡杨和毛白杨叶细胞对盐离子的区隔化有明显差异:与毛白杨相比,盐处理后的胡杨叶细胞不但具有较高的拒Na+能力,即将Na+贮存在质外体中,而且液泡区隔Na+的能力也强于毛白杨.在Cl-的区隔化方面,对照处理胡杨各种类型的细胞(包括叶脉、叶肉和表皮细胞)Cl-浓度都高于毛白杨,但在盐胁迫下,胡杨叶片的细胞壁、液泡甚至叶绿体中Cl-的含量都低于毛白杨,这是胡杨根细胞限制Cl-离子根冠运输的结果.盐处理使胡杨叶片各种类型细胞中的K+浓度降低;与胡杨不同,毛白杨叶片细胞中质外体的K+浓度下降,而液泡中的K+水平却相应提高,这是毛白杨细胞内K+重新分配的结果:由于盐处理后毛白杨液泡内积累了过多的Cl-,K+从细胞壁进入液泡,用以平衡液泡内Cl-积累所带来的负电荷.戴松香 陈少良 Eberhard Fritz Andrea Olbrich Christine Kettner Andrea Polle Aloys Hüttermann 2006北京林业大学学报2006,28,S2:14

7Implication of the intestinal microbiome in complications of cirrhosis显示文摘The intestinal microbiome(IM) is altered in patients with cirrhosis,and emerging literature suggests that this impacts on the development of complications.The Pub Med database was searched from January 2000 to May 2015 for studies and review articles on the composition,pathophysiologic effects and therapeutic modulation of the IM in cirrhosis.The following combination of relevant text words and MeS H terms were used,namely intestinal microbiome,microbiota,or dysbiosis,and cirrhosis,encephalopathy,spontaneous bacterial peritonitis,hepatorenal syndrome,variceal bleeding,hepatopulmonary syndrome,portopulmonary hypertension and hepatocellular carcinoma.The search results were evaluated for pertinence to the subject of IM and cirrhosis,as well as for quality of study design.The IM in cirrhosis is characterized by a decreased proportion of Bacteroides and Lactobacilli,and an increased proportion of Enterobacteriaceae compared to healthy controls.Except for alcoholic cirrhosis,the composition of the IM in cirrhosis is not affected by the etiology of the liver disease.The percentage of Enterobacteriaceae increases with worsening liver disease severity and decompensation and is associated with bacteremia,spontaneous bacterial peritonitis and hepatic encephalopathy.Lactulose,rifaximin and Lactobacillus-containing probiotics have been shown to partial y reverse the cirrhosis associated enteric dysbiosis,in conjunction with improvement in encephalopathy.The IM is altered in cirrhosis,and this may contribute to the development of complications associated with end-stage liver disease.Therapies such as lactulose,rifaximin and probiotics may,at least partially,reverse the cirrhosisassociated changes in the IM.This,in turn,may prevent or alleviate the severity of complications.Mamatha Bhat Bianca M Arendt Venkat Bhat Eberhard L Renner Atul Humar Johane P Allard 2016World Journal of Hepatology2016,8,27:13
8Liver transplantation and non-alcoholic fatty liver disease显示文摘Non-alcoholic fatty liver disease(NAFLD) is an important health problem worldwide. NAFLD encompasses a histological spectrum ranging from bland liver steatosis to severe steatohepatitis(nonalcoholic steatohepatitis, NASH) with the potential of progressing to cirrhosis and its associated morbidity and mortality. NAFLD is thought to be the hepatic manifestation of insulin resistance(or the metabolic syndrome); its prevalence is increasing worldwide in parallel with the obesity epidemic. In many developed countries, NAFLD is the most common cause of liver disease and NASH related cirrhosis is currently the third most common indication for liver transplantation. NASH related cirrhosis is anticipated to become the leading indication for liver transplantation within the next one or two decades. In this review, we discuss how liver transplantation is affected by NAFLD, specifically the following:(1) the increasing need for liver transplantation due to NASH;(2) the impact of the increasing prevalence of NAFLD in the general population on the quality of deceased and live donor livers available for transplantation;(3) the long term graft and patient outcomes after liver transplantation forNASH,and finally;and(4)the de novo occurrence of NAFLD/NASH after liver transplantation and its impact on graft and patient outcomes.Petros Zezos Eberhard L Renner 2014World Journal of Gastroenterology2014,20,42:12
9Identification and characterization of a novel bipartite nuclear localization signal in the hepatitis B virus polymerase显示文摘AIM:To characterize the nuclear import of hepatitis B virus(HBV)polymerase(P)and its relevance for the viral life cycle.METHODS:Sequence analysis was performed to predict functional motives within P.Phosphorylation of P was analyzed by in vitro phosphorylation.Phosphorylation site and nuclear localization signal(NLS)were destroyed by site directed mutagenesis.Functionality of the identified NLS was analyzed by confocal fluorescence microscopy and characterizing the karyopherin binding.Relevance of the structural motives for viral life cycle was studied by infection of primary Tupaia hepatocytes with HBV.RESULTS:We identified by sequence alignment and functional experiments a conserved bipartite NLS containing a casein kinaseⅡ(CKⅡ)phosphorylation site located within the terminal protein domain(TP)of the HBV polymerase.Inhibition of CKⅡimpairs the functionality of this NLS and thereby prevents the nuclear import of the polymerase.Binding of the import factor karyopherin-α2 to the polymerase depends on its CKⅡ-mediated phosphorylation of the bipartite NLS.In HBV-infected primary Tupaia hepatocytes CKⅡinhibition in the early phase(post entry phase)of the infection process prevents the establishment of the infection.CONCLUSION:Based on these data it is suggested that during HBV infection the final import of the genome complex into the nucleus is mediated by a novel bipartite NLS localized in the TP domain of HBV polymerase.Joachim Lupberger Stephanie Schaedler Alexander Peiran Eberhard Hildt 2013World Journal of Gastroenterology2013,19,44:8
10Adaptive nonlinear model predictive control design of a flexible-link manipulator with uncertain parameters显示文摘This paper presents a novel adaptive nonlinear model predictive control design for trajectory tracking of flexible-link manipulators consisting of feedback linearization, linear model predictive control, and unscented Kalman filtering. Reducing the nonlinear system to a linear system by feedback linearization simplifies the optimization problem of the model predictive controller significantly, which,however, is no longer linear in the presence of parameter uncertainties and can potentially lead to an undesired dynamical behaviour. An unscented Kalman filter is used to approximate the dynamics of the prediction model by an online parameter estimation, which leads to an adaptation of the optimization problem in each time step and thus to a better prediction and an improved input action. Finally,a detailed fuzzy-arithmetic analysis is performed in order to quantify the effect of the uncertainties on the control structure and to derive robustness assessments. 13Thermal Reversion of Plant Phytochromes显示文摘Phytochromes are red/far-red reversible photoreceptors essential for plant growth and development.Phytochrome signaling is mediated by the physiologically active far-red-absorbing Pfr form that can be inactivated to the red-absorbing Pr ground state by light-dependent photoconversion or by light-independent thermal reversion,also termed dark reversion.Although the term“dark reversion”is justified by historical reasons and frequently used in the literature,“thermal reversion”more appropriately describes the process of light-independent but temperature-regulated Pfr relaxation that not only occurs in darkness but also in light and is used throughout the review.Thermal reversion is a critical parameter for the light sensitivity of phytochrome-mediated responses and has been studied for decades,often resulting in contradictory findings.Thermal reversion is an intrinsic property of the phytochrome molecules but can be modulated by intra-and intermolecular interactions,as well as biochemical modifications,such as phosphorylation.In this review,we outline the research history of phytochrome thermal reversion,highlighting important predictions that have been made before knowing the molecular basis.We further summarize and discuss recent findings about the molecular mechanisms regulating phytochrome thermal reversion and its functional roles in light and temperature sensing in plants.Cornelia Klose Ferenc Nagy Eberhard Schafer 2020Molecular Plant2020,13,3:4

15Stability analysis of a thin-walled cylinder in turning operation using the semi-discretization method显示文摘In this work, the stability of a flexible thin cylindrical workpiece in turning is analyzed. A process model is derived based on a finite element representation of the workpiece flexibility and a nonlinear cutting force law. Repeated cutting of the same surface due to overlapping cuts is modeled with the help of a time delay. The stability of the so obtained system of periodic delay differential equations is then determined using an approximation as a time-discrete system and Floquet theory. The time-discrete system is obtained using the semi-discretization method. The method is implemented to analyze the stability of two different workpiece models of different thicknesses for different tool positions with respect to the jaw end. 18Recurrence of autoimmune liver diseases after liver transplantation显示文摘Liver transplantation(LT) is the most effective treatment modality for end stage liver disease caused by many etiologies including autoimmune processes. That said, the need for transplantation for autoimmune hepatitis(AIH) and primary biliary cirrhosis(PBC), but not for primary sclerosing cholangitis(PSC), has decreased over the years due to the availability of effective medical treatment. Autoimmune liver diseases have superior transplant outcomes than those of other etiologies. While AIH and PBC can recur after LT, recurrence is of limited clinical significance in most, but not all cases. Recurrent PSC, however, often progresses over years to a stage requiring re-transplantation. The exact incidence and the predisposing factors of disease recurrence remain debated. Better understanding of the pathogenesis and the risk factors of recurrent autoimmune liver diseases is required to develop preventive measures. 19Single port laparoscopy in gastroenterology and hepatology: A fine step forward显示文摘Single incision laparoscopy(SIL) has become an emerging technology aiming at a further reduction of abdominal wall trauma in minimally invasive surgery. Available data is encouraging for the safe application of standardized SIL in a wide range of procedures in gastroenterology and hepatology. Compared to technically simple SIL procedures, the merit of SIL in advanced surgeries, such as liver or colorectal interventions, compared to conventional laparsocopy is self-evident without any doubt. SIL has already passed the learning curve and is routinely utilized in expert centers. This minimized approach has allowed to enter a new era of surgical management that can not be acceded without a fruitful combination of prudent training, consistent day-to-day work and enthusiastic motivation for technical innovations. Both, basic and novel technical specifics as well as particular procedures are described herein. 20Effect of NaCl on growth and ion relations in two salt-tolerant strains of Paxillus involutus显示文摘The effect of NaCl on growth,biomass and ion relations of two salt-tolerant isolates of Paxillus involutus,MAJ and NAU were investigated.The two Paxillus strains were exposed to the following concentrations of NaCl:0,100,200 and 500 mmol·L-1.Growth of MAJ and NAU was enhanced by 100 mmol·L-1 NaCl but severely inhibited at the concentration of 500 mmol·L-1.NAU exhibited a greater capacity to exclude Na+ and Clunder all salinity levels,whereas the salt-includer MAJ had a higher capacity in nutrient uptake under salt stress.The ratios Na+/K+,Na+/Ca2+ and Na+/Mg2+ in NaCl-treated MAJ and NAU did not increase at levels of 100 and 200 mmol·L-1 NaCl but markedly increased at 500 mmol·L-1.This suggests that the two strains,especially MAJ,enhanced nutrient uptake corresponding to the increased Na+ influx at moderate salinity.We conclude that both MAJ and NAU are able to tolerate 500 mmol·L-1 NaCl but there are species-specific differences in retaining ionic homeostasis in the two Paxillus strains.NAU is a salt-excluder,MAJ is a salt-includer but retains a high capacity in nutrient selectivity under saline conditions.Their definite mechanisms to enhance salt tolerance of mycorrhizal hosts need further study.ZHANG Hua LI Jing CHEN Shao-liang LU Cun-fu WANG Rui-gang DAI Song-xiang ZHU Hui-juan ZHANG Yun-xia SHI Yong WANG Mei-juan LU Yan-jun LI Li-si Rosemarie LANGENFELD-HEYSER Fritz EBERHARD Andrea POLLE 2008Forestry Studies in China2008,10,2:3
