| 1 | 三氯生对离体鹅尾臀腺和人乳腺癌细胞脂肪酸合成酶的抑制作用显示文摘背景与目的:研究表明,人乳腺癌早期和病程中癌细胞脂肪酸合成酶(fattyacidsynthase,FAS)呈高效表达。抑制FAS可引起癌细胞凋亡。本研究采用酶促反应动力学实验观察三氯生对离体鹅尾臀腺FAS的影响,建立实验方法,研究三氯生对人乳腺癌SKBr3细胞FAS的抑制作用。方法:运用超速离心和SuperdexPG200层析技术分离纯化鹅尾臀腺FAS,用超速离心技术部分纯化人乳腺癌SKBr3细胞FAS。不同浓度三氯生与FAS相互作用不同时间后,加入底物,用分光光度法观察三氯生对FAS的抑制作用。结果:在鹅尾臀腺组,FAS被纯化为SDS-PAGE电泳单条区带(分子量250kDa),三种浓度三氯生(12.5、25.0、100.0μmol/L)与FAS在催化作用前相互作用0、5和10min,对FAS的抑制率分别为26.40%、28.30%和43.93%(12.5μmol/L);46.22%、50.28%和97.05%(25μmol/L);98.11%、97.75%和97.37%(100μmol/L)。在人乳腺癌SKBr3细胞组FAS被部分纯化,25、50、100和200μmol/L三氯生分别与FAS在催化前相互作用5min,对FAS活性的抑制率分别为20.00%、26.67%、60.00%和100.00%。结论:三氯生对离体鹅尾臀腺FAS和人乳腺癌SKBr3细胞FAS均具有抑制作用;作用强度既依赖于抑制剂浓度,又依赖于抑制剂与酶相互作用时间。 | 王映强 赖炳森 Vernon E.Anderson | 2003 | 癌症2003,22,3: | 5 |

| 5 | Genetic Evidence of Middle East Respiratory Syndrome Coronavirus(MERS-Cov) and Widespread Seroprevalence among Camels in Kenya显示文摘We describe the first genome isolation of Middle East respiratory syndrome coronavirus(MERS-CoV) in Kenya. This fatal zoonotic pathogen was first described in the Kingdom of Saudi Arabia in 2012. Epidemiological and molecular evidence revealed zoonotic transmission from camels to humans and between humans. Currently, MERS-CoV is classified by the WHO as having high pandemic potential requiring greater surveillance. Previous studies of MERS-CoV in Kenya mainly focused on site-specific and archived camel and human serum samples for antibodies. We conducted active nationwide cross-sectional surveillance of camels and humans in Kenya, targeting both nasal swabs and plasma samples from 1,163 camels and 486 humans collected from January 2016 to June 2018. A total of 792 camel plasma samples were positive by ELISA. Seroprevalence increased with age, and the highest prevalence was observed in adult camels(82.37%, 95%confidence interval(CI) 79.50–84.91). More female camels were significantly seropositive(74.28%, 95% CI 71.14–77.19)than male camels(P \ 0.001)(53.74%, 95% CI 48.48–58.90). Only 11 camel nasal swabs were positive for MERS-CoV by reverse transcription-quantitative PCR. Phylogenetic analysis of whole genome sequences showed that Kenyan MERSCoV clustered within sub-clade C2, which is associated with the African clade, but did not contain signature deletions of orf4 b in African viruses. None of the human plasma screened contained neutralizing antibodies against MERS-CoV. This study confirms the geographically widespread occurrence of MERS-CoV in Kenyan camels. | 11 | On the development of optical peripheral nerve interfaces显示文摘Limb loss and spinal cord injury are two debilitating conditions that continue to grow in prevalence. Prosthetic limbs and limb reanimation present two ways of providing affected individuals with means to interact in the world. These techniques are both dependent on a robust interface with the peripheral nerve. Current methods for interfacing with the peripheral nerve tend to suffer from low specificity, high latency and insufficient robustness for a chronic implant. An optical peripheral nerve interface may solve some of these problems by decreasing invasiveness and providing single axon specificity. In order to implement such an interface three elements are required:(1) a transducer capable of translating light into a neural stimulus or translating neural activity into changes in fluorescence,(2) a means for delivering said transducer and(3) a microscope for providing the stimulus light and detecting the fluorescence change. There are continued improvements in both genetically encoded calcium and voltage indicators as well as new optogenetic actuators for stimulation. Similarly, improvements in specificity of viral vectors continue to improve expression in the axons of the peripheral nerve. Our work has recently shown that it is possible to virally transduce axons of the peripheral nerve for recording from small fibers.
