| 1 | Sinomenine ester derivative inhibits glioblastoma by inducing mitochondria-dependent apoptosis and autophagy by PI3K/AKT/mTOR and AMPK/mTOR pathway显示文摘Glioblastoma multiforme(GBM)in the central nervous system is the most lethal advanced glioma and currently there is no effective treatment for it.Studies of sinomenine,an alkaloid from the Chinese medicinal plant,Sinomenium acutum,showed that it had inhibitory effects on several kinds of cancer.Here,we synthesized a sinomenine derivative,sino-wcj-33(SW33),tested it for antitumor activity on GBM and explored the underlying mechanism.SW33 significantly inhibited proliferation and colony formation of GBM and reduced migration and invasion of U87 and U251 cells.It also arrested the cell cycle at G2/M phase and induced mitochondria-dependent apoptosis.Differential gene enrichment analysis and pathway validation showed that SW33 exerted anti-GBM effects by regulating PI3 K/AKT and AMPK signaling pathways and significantly suppressed tumorigenicity with no obvious adverse effects on the body.SW33 also induced autophagy through the PI3 K/AKT/mTOR and AMPK/mTOR pathways.Thus,SW33 appears to be a promising drug for treating GBM effectively and safely. | Xiangjin Zheng Wan Li Huanli Xu Jinyi Liu Liwen Ren Yihui Yang Sha Li Jinhua Wang Tengfei Ji Guanhua Du | 2021 | Acta Pharmaceutica Sinica B2021,11,11: | 15 |
| 2 | Genistein suppresses the mitochondrial apoptotic pathway in hippocampal neurons in rats with Alzheimer's disease显示文摘Genistein is effective against amyloid-β toxicity,but the underlying mechanisms are unclear.We hypothesized that genistein may protect neurons by inhibiting the mitochondrial apoptotic pathway,and thereby play a role in the prevention of Alzheimer's disease.A rat model of Alzheimer's disease was established by intraperitoneal injection of D-galactose and intracerebral injection of amyloid-β peptide(25–35).In the genistein treatment groups,a 7-day pretreatment with genistein(10,30,90 mg/kg) was given prior to establishing Alzheimer's disease model,for 49 consecutive days.Terminal deoxyribonucleotidyl transferase-mediated d UTP nick end labeling assay demonstrated a reduction in apoptosis in the hippocampus of rats treated with genistein.Western blot analysis showed that expression levels of capase-3,Bax and cytochrome c were decreased compared with the model group.Furthermore,immunohistochemical staining revealed reductions in cytochrome c and Bax immunoreactivity in these rats.Morris water maze revealed a substantial shortening of escape latency by genistein in Alzheimer's disease rats.These findings suggest that genistein decreases neuronal loss in the hippocampus,and improves learning and memory ability.The neuroprotective effects of genistein are associated with the inhibition of the mitochondrial apoptotic pathway,as shown by its ability to reduce levels of caspase-3,Bax and cytochrome c. | Yan Wang Biao Cai Jing Sha Ting-ting Wang Run-ze Cai Chang-ju Ma Tao Han Jun Du | 2016 | Neural Regeneration Research2016,11,7: | 12 |
| 3 | Evapotranspiration and Its Energy Exchange in Alpine Meadow Ecosystem on the Qinghai-Tibetan Plateau显示文摘To understand the water and energy exchange on the Qinghai-Tibetan Plateau, we explored the characteristics of evapotranspiration (ET) and energy fluxes from 2002 to 2005 over a Kobresia meadow ecosystem using the eddy covariance method. The ratio of annual ET to precipitation (P) of meadow ecosystem was about 60%, but varied greatly with the change of season from summer to winter. The annual ET/P in meadow was lower than that in shrub, steppe and wetland ecosystems of this plateau. The incident solar radiation (R s ) received by the meadow was obviously higher than that of lowland in the same latitude; however the ratio of net radiation (R n ) to R s with average annual value of 0.44 was significantly lower than that in the same latitude. The average annual ET was about 390 mm for 2002-2005, of which more than 80% occurred in growing season from May to September. The energy consumed on the ET was about 44% of net radiation in growing season, which was lower than that of shrub, steppe and wetland on this plateau. | 6 | Circulating MicroRNAs as Novel Diagnostic Biomarkers for Very Early-onset(≤40 years) Coronary Artery Disease显示文摘Objective Very early-onset coronary artery disease(CAD) is a great challenge in cardiovascular medicine throughout the world,especially regarding its early diagnosis.This study explored whether circulating micro RNAs(mi RNAs) could be used as potential biomarkers for patients with very early-onset CAD.Methods We performed an initial screening of mi RNA expression using RNA isolated from 20 patients with angiographically documented very early-onset CAD and 20 age-and sex-matched normal controls.For further confirmation,we prospectively examined the mi RNAs selected from 40 patients with very early-onset CAD and 40 angiography-normal controls.Results A total of 22 overexpressed mi RNAs and 22 underexpressed mi RNAs were detected in the initial screening.RT-q PCR analysis of the mi RNAs obtained from the initial screening revealed that four mi RNAs including mi R-196-5p,mi R-3163-3p,mi R-145-3p,and mi R-190a-5p exhibited significantly decreased expression in patients compared with that in controls(P<0.05).The areas under the receiver operating characteristic curve for these mi RNAs were 0.824(95% CI,0.731-0.917;P<0.001),0.758(95% CI,0.651-0.864;P<0.001),0.753(95% CI,0.643-0.863;P<0.001),and 0.782(95% CI,0.680-0.884;P<0.001),respectively,in the validation set.Conclusion To our knowledge,this is an advanced study to report about four serum mi RNAs(mi R-196-5p,mi R-3163-3p,mi R-145-3p,and mi R-190a-5p) that could be used as novel biomarkers for the diagnosis of very early-onset CAD. | DU Ying YANG Sheng Hua LI Sha CUI Chuan Jue ZHANG Yan ZHU Cheng Gang GUO Yuan Lin WU Na Qiong GAO Ying SUN Jing DONG Qian LIU Geng LI Jian Jun | 2016 | Biomedical and Environmental Sciences2016,29,8: | 5 |

| 8 | Systematic analyses of glutamine and glutamate metabolisms across different cancer types显示文摘Background: Glutamine and glutamate are known to play important roles in cancer biology. However, no detailed information is available in terms of their levels of involvement in various biological processes across different cancer types, whereas such knowledge could be critical for understanding the distinct characteristics of different cancer types. Our computational study aimed to examine the functional roles of glutamine and glutamate across different cancer types.Methods: We conducted a comparative analysis of gene expression data of cancer tissues versus normal control tissues of 11 cancer types to understand glutamine and glutamate metabolisms in cancer. Specifically, we developed a linear regression model to assess differential contributions by glutamine and/or glutamate to each of seven biological processes in cancer versus control tissues.Results: While our computational predictions were consistent with some of the previous observations, multiple novel predictions were made:(1) glutamine is generally not involved in purine synthesis in cancer except for breast cancer, and is similarly not involved in pyridine synthesis except for kidney cancer;(2) glutamine is generally not involved in ATP production in cancer;(3) glutamine's contribution to nucleotide synthesis is minimal if any in cancer;(4) glutamine is not involved in asparagine synthesis in cancer except for bladder and lung cancers; and(5) glutamate does not contribute to serine synthesis except for bladder cancer.Conclusions: We comprehensively predicted the roles of glutamine and glutamate metabolisms in selected metabolic pathways in cancer tissues versus control tissues, which may lead to novel approaches to therapeutic development targeted at glutamine and/or glutamate metabolism. | 10 | Efficacy and economic analysis of Ex- PRESS implantation versus trabeculectomy in uncontrolled glaucoma: a systematic review and Meta-analysis显示文摘AIM:To systematically review the current evidence based on the efficacy and cost of Ex-PRESS implantation and trabeculectomy(Trab)for uncontrolled glaucoma.·METHODS:Clinical trials were identified by electronic databases(Pub Med,EMBASE,ISI Web of science and Cochrane library),and data,such as intraocular pressure(IOP),the complete and qualified success rate,the postoperative complications and the cost,were exacted from these relevant studies.Weighted mean difference(WMD),odds ratio(OR)and 95%confidence intervals(CIs)were calculated and were pooled using a randomeffects model.·RESULTS:Eleven relevant publications and two abstracts met the inclusion criteria.The efficacy of ExPRESS was similar to that of Trab in the percentage of IOP reduction(IOPR%)at 1,2y(WMD:-2.01;95%CI:-7.92-3.90;P=0.50 and WMD:2.89;95%CI:-8.05-13.83;P=0.60,respectively).Ex-PRESS possessed a significant higher complete and qualified success rate(OR:1.59;95%CI:1.07-2.35;P=0.02 and OR:1.74;95%CI:1.06-2.86;P=0.03,respectively).Moreover,Ex-PRESS exerted a significantly lower frequency of hypotony and hyphema than Trab(OR:0.39;95%CI:0.21-0.72;P=0.003 and OR:0.27;95%CI:0.10-0.69;P=0.003,respectively).However,there was no consistent result on the cost between the two groups according to the previous three studies.·CONCLUSION:Both Trab and Ex-PRESS have equivalent efficacy in lowering IOP,yet Ex-PRESS had a lower risk of hypotony and hyphema than Trab.Nevertheless,whether the cost of Ex-PRESS was less than that of Trab should be further investigated to ensure evidence-based conclusion in the long run. | Ling Wang Fang Sha Da-Dong Guo Hong-Sheng Bi Jun-Kang Si Yu-Xiang Du Kai Tang | 2016 | International Journal of Ophthalmology(English edition)2016,9,1: | 5 |

Web service discovery has drawn much attention with the development of Web service applications and big data analysis. Under this circumstance, traditional Web service discovery strategies cannot adequately meet high user requirements due to the efficiency and precision of service discovery is low. In order to improve the accuracy and efficiency of service discovery, a user requirement oriented Web service discovery approach based on Petri nets is proposed in this study. A data preprocessing strategy of Web service is first designed. Then, a service clustering method is proposed based on Petri nets, which can conduct service cluster head generation, service cluster composition, and service discovery. The proposed method utilizes a superior data preprocessing method. Using simulation experiments,the efficiency and precision of Web service discovery are illustrated. | 12 | Effectiveness and safety of Ginkgo biloba extract(GBE50)in the treatment of dizziness caused by cerebral arteriosclerosis:a multi-center,double-blind,randomized controlled trial显示文摘OBJECTIVE:To evaluate the effectiveness and safety of Ginkgo biloba extract(GBE50)in the treatment of dizziness caused by cerebral arteriosclerosis.METHODS:This was a multi-center,double-blind,double-dummy,positive-controlled,parallel randomized controlled clinical trial with 1∶1 allocation.We recruited 404 patients with dizziness caused by cerebral arteriosclerosis(blood stasis symptom pattern)in 10 hospitals in China.GBE50 group received GBE50 and Naoxinqing tablet(NXQ)of mimetic agent,control group received NXQ and GBE50 of mimetic agent.The main outcome was Traditional Chinese Medicine(TCM)symptom pattern score of blood stasis after 6 weeks.The secondary outcomes were changes in the dizziness handicap inventory(DHI)score,vertigo visual analogue scale(VAS)score,the university of California vertigo questionnaire(UCLA-DQ)score and single-item symptom score of TCM from baseline to 2,4 and 6 weeks.Safety indicators included the incidence of adverse events,severe adverse events and laboratory examination including blood routine,liver function,renal function,and so forth.RESULTS:The total effective rate of TCM symptom pattern score in the GBE50 group after 6 weeks of treatment was higher than that in the control group,the difference in rate was statistically significant(92.67%vs 83.07%,P=0.004).Compared with the control group,there was no difference in the incidence of adverse reactions(9.95%vs 14.85%,P=0.136).CONCLUSION:The treatment of dizziness caused by cerebral arteriosclerosis with GBE50 is effective,safe and reliable. | SHA Rina TANG Lu DU Yawei WU Shengxian SHI Huawei ZOU Hongxin ZHANG Xuran DONG Xinglu ZHOU Li | 2022 | Journal of Traditional Chinese Medicine2022,42,1: | 5 |

| 15 | Tumorigenic bacteria in colorectal cancer:mechanisms and treatments显示文摘Colorectal cancer(CRC)is the third most common and the second most fatal cancer.In recent years,more attention has been directed toward the role of gut microbiota in the initiation and development of CRC.Some bacterial species,such as Fusobacterium nucleatum,Escherichia coli,Bacteroides fragilis,Enterococcus faecalis,and Salmonella sp.have been associated with CRC,based upon sequencing studies in CRC patients and functional studies in cell culture and animal models.These bacteria can cause host DNA damage by genotoxic substances,including colibactin secreted by pks+Escherichia coli,B.fragilis toxin(BFT)produced by Bacteroides fragilis,and typhoid toxin(TT)from Salmonella.These bacteria can also indirectly promote CRC by influencing host-signaling pathways,such as E-cadherin/β-catenin,TLR4/MYD88/NF-κB,and SMO/RAS/p38 MAPK.Moreover,some of these bacteria can contribute to CRC progression by helping tumor cells to evade the immune response by suppressing immune cell function,creating a proinflammatory environment,or influencing the autophagy process.Treatments with the classical antibacterial drugs,metronidazole or erythromycin,the antibacterial active ingredients,M13@Ag(electrostatically assembled from inorganic silver nanoparticles and the protein capsid of bacteriophage M13),berberine,and zerumbone,were found to inhibit tumorigenic bacteria to different degrees.In this review,we described progress in elucidating the tumorigenic mechanisms of several CRC-associated bacteria,as well as progress in developing effective antibacterial therapies.Specific bacteria have been shown to be active in the oncogenesis and progression of CRC,and some antibacterial compounds have shown therapeutic potential in bacteria-induced CRC.These bacteria may be useful as biomarkers or therapeutic targets for CRC. | Sha Li Jinyi Liu Xiangjin Zheng Liwen Ren Yihui Yang Wan Li Weiqi Fu Jinhua Wang Guanhua Du | 2022 | Cancer Biology & Medicine2022,19,2: | 3 |

| 17 | Benzimidazoles induce concurrent apoptosis and pyroptosis of human glioblastoma cells via arresting cell cycle显示文摘Glioblastoma multiforme(GBM)is the most malignant and lethal primary brain tumor in adults accounting for about 50%of all gliomas.The only treatment available for GBM is the drug temozolomide,which unfortunately has frequent drug resistance issue.By analyzing the hub genes of GBM via weighted gene co-expression network analysis(WGCNA)of the cancer genome atlas(TCGA)dataset,and using the connectivity map(CMAP)platform for drug repurposing,we found that multiple azole compounds had potential anti-GBM activity.When their anti-GBM activity was examined,however,only three benzimidazole compounds,i.e.flubendazole,mebendazole and fenbendazole,potently and dose-dependently inhibited proliferation of U87 and U251 cells with IC50 values below 0.26μM.Benzimidazoles(0.125−0.5μM)dose-dependently suppressed DNA synthesis,cell migration and invasion,and regulated the expression of key epithelial-mesenchymal transition(EMT)markers in U87 and U251 cells.Benzimidazoles treatment also dose-dependently induced the GBM cell cycle arrest at the G2/M phase via the P53/P21/cyclin B1 pathway.Furthermore,the drugs triggered pyroptosis of GBM cells through the NF-κB/NLRP3/GSDMD pathway,and might also concurrently induced mitochondria-dependent apoptosis.In a nude mouse U87 cell xenograft model,administration of flubendazole(12.5,25,and 50 mg·kg^(−1)·d^(−1),i.p,for 3 weeks)dose-dependently suppressed the tumor growth without obvious adverse effects.Taken together,our results demonstrated that benzimidazoles might be promising candidates for the treatment of GBM. | Li-wen Ren Wan Li Xiang-jin Zheng Jin-yi Liu Yi-hui Yang Sha Li Sen Zhang Wei-qi Fu Bin Xiao Jin-hua Wang Guan-hua Du | 2022 | Acta Pharmacologica Sinica2022,43,1: | 3 |
| 18 | Protection of 3'-methoxy-puerarin against focal brain ischemia and its association with c-fos expression显示文摘The present study established a rat model of focal brain ischemia by occlusion of the middle cerebral artery covered with FeCl3, and investigated the protective effect of 3'-methoxy-puerarin. Hippocampal and cortical c-fos gene expression was determined using in situ hybridization. Results showed that 3'-methoxy-puerarin reduced neurological deficit scores, cerebral infarcted zone and water content of brain tissues, dramatically increased the activity of catalase and glutathione peroxidase in the ischemia zone of the hippocampus, increased the activity of catalase in the cortex, decreased lipid peroxide and lactic acid contents in the hippocampus and cerebral cortex, and down-regulated c-fos gene expression in brain ischemic rats.
