1Effects of cytokines on carbon tetrachloride-induced hepatic fibrogenesis in rats显示文摘AIM: To observe the possible effects of transforming growth factor (TGF) β1, interleukin (IL)-6, tumor-necrosis factor (TNF) α and IL-10 on experimental rat hepatic fibrosis. METHODS: One hundred SD rats were divided randomly into the three groups. Control group received intraperitoneal injection of saline (2 ml-kg^-1), twice a week. Fibrogenesis group was injected intraperitoneally with 50% carbon tetrachloride (CCI4) (2 ml-kg^-1) twice a week. Fibrosisintervention group was given IL-10 at a dose of 4 μg-kg^-1 20 minutes before CCI4 administration from the third week.At the fifth, seventh, and ninth weeks, 7 to 10 rats in each group were sacrificed to collect serum. Levels of TGF-β1,TNF-α, IL-6 and IL-10 were determined by enzyme-linkedimmunosorbent assay (ELISA). The liver tissues were taken for routine histological examination. RESULTS: Hepatic fibrosis was developed with the injection of CCI4. Values of the circulating TGFβ, TNFα, IL-6 and IL-10 in the control group were 25.495.56 ng.L^-1, 15.18±3.83ng.L^-1, 63.64±13.03 ng.L-^1 and 132.90±12.13 ng.L^-1,respectively. Their levels in the CCI4-intoxication group were 31.13±6.41 ng.L^-1, 18.91±5.31 ng.L^-1, 89.08±25.39 ng.L^-1 and 57.63±18.88 ng.L^-1, respectively, and those in the IL-10-intervention group were 26.11±5.32 ng.L^-1,13.99±1.86 ng.L^-1,74.71±21.15 ng.L^-1 and 88.19±20.81 ng.L^-1, respectively. A gradual increase was observed in the levels of TGFβ1, TNFα and IL-6 during hepatic fibrogenesis. These changes were pardally reversed by simultaneous administration of IL-10. The histological parameters, characterized by CCl4-intoxificatJon,also seemed to be improved with IL-10 treatment, the collagen production was reduced at the ninth week and the histological activity index was decreased from 7.9±1.2 to 4.7±0.9. 2Overexpression of HBxAg in hepatocellular carcinoma and its relationship with Fas/FasL system显示文摘AIM: To study the expression and serum level of HBxAg,Fas and FasL in tissues of HCC patients, and to assess the relationship between HBxAg and Fas/FasL system.METHODS: Tissues from 50 patients with HCC were tested for the expression of HBxAg, Fas and FasL by S-P immunohistochemistry. Serum levels of sFas/sFasL and HBsAg/HBeAg were measured by ELISA assay. HBV X gene was detected by PCR in serum and confirmed by automatic sequencing. Fifty cases of liver cirrhosis and 30 normal controls were involved in serum analysis.RESULTS: The expression of HBxAg, Fas and FasL in carcinoma tissues was 96 %, 84 % and 98 %, respectively.Staining of HBxAg, Fas and FasL was observed predominately in cytoplasms, no significant difference was found in intensity between HBxAg, Fas and FasL (P>0.05). HBxAg, Fas and FasL might express in the same area of carcinoma tissues and this co-expression could be found in most patients with HCC. The mean levels of sFas in serum from HCC, cirrhosis and normal controls were 762.29±391.56 μg@ L-1 835.36±407.33 μg@L-1 and 238.27±135.29 μg@L-1. The mean levels of sFasL in serum from HCC, cirrhosis and normal controls were 156.36±9.61iμg@ L-1, 173.63±18.74 μg@L-1 and 121.96±7.83 μg@ L-1.Statistical analysis showed that both sFas and sFasL in HCC and cirrhosis patients were significantly higher than those in normal controls (P<0.01). Serum HBV X gene was found in 32 % of HCC patients and ,46 % of cirrhotic patients.There was no significant relationship between serum level of sFas/sFasL and serum X gene detection (P>0.05). Eight percent of HCC patients with negative HBsAg and HBeAg in serum might have X gene in serum and HBxAg expression in carcinoma tissues.CONCLUSION: Our data suggest that HBxAg and Fas/FasL system plays an important role in the development of human HCC. 3Molecular cloning and functional analysis of the FLOWERING LOCUS T(FT) homolog Gh FT1 from Gossypium hirsutum显示文摘FLOWERING LOCUS T(FT) encodes a member of the phosphatidylethanolamine-binding protein(PEBP) family that functions as the mobile floral signal, playing an important role in regulating the floral transition in angiosperms. We isolated an FT-homolog(Gh FT1) from Gossypium hirsutum L. cultivar,Xinluzao 33 Gh FT1 was predominantly expressed in stamens and sepals, and had a relatively higher expression level during the initiation stage of fiber development. Gh FT1 m RNA displayed diurnal oscillations in both long-day and short-day condition,suggesting that the expression of this gene may be under the control of the circadian clock. Subcellular analysis revealed that Gh FT1 protein located in the cytoplasm and nucleus. Ectopic expression of Gh FT1 in transgenic arabidopsis plants resulted in early flowering compared with wild-type plants. In addition,ectopic expression of Gh FT1 in arabidopsis ft-10 mutants partially rescued the extremely late flowering phenotype. Finally, several Researchflowering related genes functioning downstream of At FT were highly upregulated in the 35S::Gh FT1 transgenic arabidopsis plants. 4Highly sensitive wearable sensor based on a flexible multi-layer graphene film antenna显示文摘The use of advanced carbon nanomaterials for flexible antenna sensors has attracted great attention due to their outstanding electromechanical properties. However, carbon nanomaterial based composites have yet to overcome drawbacks, such as low conductivity and toughness. In this work, a flexible multi-layer graphene film(FGF) with a high conductivity of 10~6 S/m for antenna based wearable sensors is investigated. A 1.63 GHz FGF antenna sensor exhibits significantly high strain sensitivity of 9.8 for compressive bending and 9.36 for tensile bending, which is super than the copper antenna sensor(5.39 for compressive bending and 4.05 for tensile bending). 5Influence of expressed TRAIL on biophysical properties of the human leukemic cell line Jurkat显示文摘The cDNA fragment of human TRAIL (TNF-related apoptosis inducing ligand) was cloned into RevTet-On, a Tetregulated and high-level gene expression system. The gene expression system was constructed in a human leukemic cell line: Jurkat. By using RevTet-On TRAIL gene expression system in Jurkat as a cell model, we studied the influence of TRAIL gene on the changes of cellular apoptosis before and after the TRAIL gene expression, which was induced by adding tetracycline derivative doxycycline (Dox). The results indicated that the cellular apoptosis ratio was largely dependent on the TRAIL gene expression level. Moreover, it was found that the apoptosis-inducing TRAIL could cause significant changes in the biophysical properties of Jurkat cells. The cell surface charge density decreased, the membrane fluidity declined, the elastic coefficients K1 increased, and the proportion of o-helix in membrane protein secondary structure decreased. 6Travel Behaviours of Sharing Bicycles in the Central Urban Area Based on Geographically Weighted Regression: The Case of Guangzhou, China显示文摘Mobile information and communication technologies(MICTs) have fully penetrated everyday life in smart societies;this has greatly compressed time, space, and distance, and consequently, reshaped residents’ travel behaviour patterns. As a new mode of shared mobility, the sharing bicycle offers a variety of options for the daily travel of urban residents. Extant studies have mainly examined the travel characteristics and influencing factors of public bicycles with piles, while the travel patterns for sharing bicycles and their driving mechanisms have been largely ignored. Using one week’s travel data for Mobike, this study investigated the spatial and temporal distribution patterns of sharing bicycle travel behaviours in the central urban area of Guangzhou, China;furthermore, it identified the influences of built environment density factors on sharing bicycle travel behaviours based on the geographically weighted regression method. Obvious morning and evening peaks were observed in the sharing bicycle travel patterns for both weekdays and weekends. The old urban area, which had a high degree of mixed function, dense road networks, and cycling-friendly built environments, was the main travel area that attracted sharing bicycles on both weekdays and weekends. Furthermore, factors including the point of interest(POI) for the density of public transport stations, the functional mixing degree, and the density of residential POIs significantly affected residents’ travel behaviours. 7Performance of bioferric-submerged membrane bioreactor for dyeing wastewater treatment显示文摘Adding iron salt or iron hydroxide to sludge-mixed liquor in an aeration tank of a conventional activated sludge processes(bioferric process)can simultaneously improve the sludge’s filterability and enhance the system’s treatment capacity.In view of this,Fe(OH)3 was added to a submerged membrane bioreactor(SMBR)to enhance the removal efficiency and to mitigate membrane fouling.Bio-ferric process and SMBR were combined to create a novel process called Bioferric-SMBR.A side-by-side comparison study of Bioferric-SMBR and common SMBR dealing with dyeing wastewater was carried out.Bioferric-SMBR showed potential superiority,which could enhance removal efficien-cy,reduce membrane fouling and improve sludge character-istic.When volumetric loading rate was 25% higher than that of common SMBR,the removal efficiencies of Bioferric-SMBR on COD,dye,and NH_(4)^(+)-N were 1.0%,9.5%,and 5.2%higher than that of common SMBR,respectively.The trans-membrane pressure of Bioferric-SMBR was only 36%of that in common SMBR while its membrane flux was 25% higher than that of common SMBR.The stable running period in Bioferric-SMBR was 2.5 times of that in common SMBR when there was no surplus sludge discharged.The mixed liquor suspended solids concentration of Bioferric-SMBR was higher than that of common SMBR with more diversified kinds of microorganisms such as protozoans and metazoans.The mean particle diameter and specific oxygen uptake rate of Bioferric-SMBR were 3.10 and 1.23 times the common SMBR,respectively.ZOU Haiyan XI Danli 2007Frontiers of Environmental Science & Engineering2007,1,3:5

9Low Complexity Multiuser Detection with Recursively Successive Zero-Forcing and SIC Based on Nullspace for Multiuser MIMO-OFDM System显示文摘To improve the spectrum efficiency, this paper considers the multiuser detection with the MU-MIMO technology for multiuser MIMO-OFDM system uplink with the same subcarrier shared by multiple users. A low complexity multiuser detection algorithm with recursively successive zero-forcing and successive interference cancellation(RSZF-SIC) based on nullspace is proposed. The RSZF process based on the block diagonalization(BD) technique eliminates the co-channel interference(CCI) by a recursive method based on the nullspace orthogonal theorem. The SIC process detects the user signals respectively with the reasonable user detection sequence based on the results of the RSZF process. The computational complexity of the proposed algorithm is effectively reduced by reducing the total number of singular value decomposition(SVD) operations and the dimension of the SVD matrix in the recursive procedure. 11Cytochrome C oxidase III interacts with hepatitis B virus X protein in vivo by yeast two-hybrid system显示文摘AIM: To screen and identify the proteins which interact with hepatitis B virus (HBV) X protein in hepatocytes by yeast two-hybrid system and to explore the effects of X protein in the development of hepatocellular carcinoma (HCC).METHODS: With HBV X gene amplified by polymerase chainreaction (PCR), HBV X bait plasmid, named pAS2-1-X, was constructed by yeast-two hybridization system3 and verified by auto-sequencing assay, pAS2-1-X was transformed into the yeast AH109, and X-BD fusion protein expressed in the yeast cells was detected by Western blotting. The yeast cells cotransformed with pAS2-1-X and normal human liver cDNA library were grown in selective SC/-trp-leu-his-ade medium. The second screen was performed with β-gal activity detection, and false positive clones were eliminated by segregation analysis, true positive clones were amplified,sequenced and analyzed with bioinformatics. Mating experiment was peformed to confirm the binding of putative proteins to X protein in the yeast cells. RESULTS: Bait plasmid pAS2-1-X was successfully constructed and pAS2-1-X correctly expressed BD-X fusion protein in yeast AH109. One hundred and three clones grew in the selective SC/-trp-leu-his-ade medium, and only one clone passed through 13-gal activity detection and segregation analysis. The inserted cDNA fragment showed high homology with Homo sapiens cytochrome C oxidase III (COXIII). 13Efficacy of Sanqi(Radix Notoginseng)in treating cerebral hemorrhage in rats with traumatic brain injury显示文摘OBJECTIVE:To evaluate the protective efficacy of Sanqi(Radix Notoginseng)on cerebral hemorrhage in a rat model of traumatic brain injury(TBI)by investigating plasminogen activator inhibitor-1(PAI-1),tissue-type plasminogen activator(t-PA),nuclear factor-κB(NF-κB,p-p65),nitric oxide(NO),endothelin(ET),cluster differentiation(CD61CD62),and coagulation.METHODS:The free-fall method was used to create a rat model of TBI.Forty-eight rats were randomly divided into six groups:the blank group,sham group,model group,low-dose Sanqi(Radix Notoginseng)group,middle-dose Sanqi(Radix Notoginseng)group,and high-dose Sanqi(Radix Notoginseng)group.At 24 h after the model was created,we investigated brain MRI,brain tissue morphology using HE staining,flow cytometry,and immunohistochemical changes.RESULTS:Cerebral hemorrhage was aggravated in TBI rats(observed in brain specimens,brain MRI,and brain tissue HE).Cerebral immunohistochemistry results demonstrated that the expression of t-PA,PAI-1 and p-p65 increased significantly in TBI rats,while t-PA/PAI-1 had a significant decrease.In addition,CD61CD62,D2D,and ET were significantly increased in TBI rats,and PT and APTT were significantly prolonged;in contrast,NO was significantly decreased.Sanqi(Radix Notoginseng)decreased cerebral hemorrhage in TBI rats(observed in brain MRI and brain tissue HE),and increased t-PA/PAI-1,CD61CD62 significantly.It also significantly decreased the expression of t-PA,PAI-1,and p-p65 in brain immunohistochemistry and significantly decreased PT,APTT,D2D,and ET.However,there were no differences in NO between the model group and the Sanqi(Radix Notoginseng)group.CONCLUSION:Sanqi(Radix Notoginseng)can decrease the expression of p-p65,increase t-PA/PAI-1,and stem traumatic intracranial hemorrhage in a TBI rat model.JIANG Tao ZHOU Xiaqing JIANG Huifang YING Rongbiao ZHANG Zhirong CAI Danli WU Yu FANG Haijun WANG Lingcong 2021Journal of Traditional Chinese Medicine2021,41,2:2
