| 3 | Analysis of gene expression patterns with cDNA microarray during late stage of spermatogenesis in mice显示文摘The differentiation process of round spermat-ids to spermatozoa during the late stage of spermatogenesis is called spermiogenesis. To explore spermiogenesis-related genes, cDNA microarray was used to study expression patterns of 1176 genes in pachytene spermatocytes, round spermatids and elongating spermatids of Balb/c mice. The results showed that 208 genes were detected in all the three cell types. Most of them were down-regulated from pachytene spermatocytes to round spermatids and elongating spermatids. However, up-regulation of 7 genes expression in round spermatids and 3 genes in elongating spermatids were found. Expression of 7 differentially expressed genes in cDNA arrays was further confirmed by semi-quantitative RT-PCR study. The RT-PCR results indicated that the expression of 6 genes was consistent with that in cDNA arrays, only one gene did not show differential expression by RT-PCR. | 6 | Abnormal expression of cen-trosome protein (centrin) in spermatozoa of male human infertility显示文摘To study the relations between male infertility and centrosome protein (centrin) and the functions of centrin in spermatogenesis, the matured spermatozoa of 10 normal male people and 18 male infertility patients were stained by immunofluorescence labeling antibody against centrin. The results showed that two fluorescence signal dots appeared in the normal male spermatozoa and were located at the base of flagellum. They are proximal centriole and distal centriole. However, in some spermatozoa of the male infertility, centrin protein was located abnormally at the base of flagellum and its staining signals were spread, the normal proximal and distal centrioles were confused and could not be recognized separately. These results suggest that abnormality of centrosome protein may be related to male infertility. | 12 | Mechanisms of testicular immune privilege显示文摘The testis exhibits a distinctive form of immune privilege to protect the germ cells from the host immuneattack. The property of testicular immune privilege was originally attributed to the blood-testis barrier in theseminiferous epithelium, which sequesters antigens. Recent studies have uncovered several levels of immune controlbesides the blood-testis barrier involved in the privilege of the testis, including the mechanisms of immune tolerance,reduced immune activation, localized active immunosuppression and antigen-specific immunoregulation. The somaticcells of the testis, especially Sertoli cells, play a key role in regulating the testicular immune privileged status. Theconstitutive expression of anti-inflammatory factors in the testis by somatic cells is essential for local immunosuppression.Growing evidence shows that androgens orchestrate the inhibition of proinflammatory factors and shift cytokinebalance toward a tolerogenic environment. Disruption of these protective mechanisms, which may be caused by trauma,infection and genetic factors, can lead to orchitis and infertility.
