1Detection of hepatitis B virus DNA by real-time PCR using TaqMan-MGB probe technology显示文摘AIM: To develop a real-time PCR for detecting hepatitis B virus-(HBV) DNA based on TaqMan technology using a new MGB probe.METHODS: Plasmid containing the sequence of X gene (1414-1744 nt) was constructed as HBV-DNA standard for quantitative analysis. A TaqMan-MGB probe between primers for amplification was designed to detect PCR products. The interested sequence contained in the plasmid and in clinical specimens was quantitatively measured.RESULTS: The detection limit of the assay for HBV DNA was 1 genome equivalent per reaction. A linear standard curve was obtained between 100 and 109 DNA copies/reaction (r>0.990). None of the negative control samples showed false-positive reactions in duplicate. HBV DNA was detected in 100% (50/50) of HBV patients with HbeAg, and in 72.0% (36/50) with HBsAg, HBeAb and HBcAb. 2Grouting techniques for the unfavorable geological conditions of Xiang'an subsea tunnel in China显示文摘One of the major challenges during subsea tunnel construction is to seal the potential water inflow.The paper presents a case study of Xiang'an subsea tunnel in Xiamen,the first subsea tunnel in China.During its construction,different grades of weathered geomaterials were encountered,which was the challenging issue for this project.To deal with these unfavorable geological conditions,grouting was adopted as an important measure for ground treatment.The grouting mechanism is first illustrated by introducing a typical grouting process.Then the site-specific grouting techniques employed in the Xiang'an subsea tunnel are elaborated.By using this ground reinforcement technique,the tunneling safety of the Xiang'an subsea tunnel was guaranteed.Dingli Zhang Qian Fang Haicheng Lou 2014Journal of Rock Mechanics and Geotechnical Engineering2014,6,5:12
3Evolution of particle number size distribution in an urban atmosphere during episodes of heavy pollution and new particle formation显示文摘This study discusses the evolution of particle number size distribution during episodes of heavy pollution and new particle formation in the urban atmosphere of Beijing to quantify the effects of dynamic processes (coagulation and condensation) on the particle number size distribution.During a heavy-pollution event,an extremely low number concentration of 3-10 nm particles (on average 46 cm-3) was observed.This is because nucleation-mode particles were easily removed by strong coagulational scavenging of larger particles under this condition.In addition,a large condensation sink (on average 0.13 s-1) restrained nucleation,which is one of the major sources of nucleation-mode particles.Conversely,during a new-particle formation event,the small condensation sink (0.01 s-1) of precursor facilitated nucleation.At the same time,preexisting particles had little ability to scavenge newly formed particles (around 1 nm) and allowed them to grow to a detectable size (larger than 3 nm currently).We suggest that the effects of dynamic processes (coagulation and condensation) on particle size distribution should be stressed under some extreme conditions of the relatively polluted urban atmosphere in addition to traffic and meteorological factors.WU ZhiJun, HU Min YUE DingLi Birgit WEHNER Alfred WIEDENSOHLER 2011Science China Earth Sciences2011,54,11:8
4Analytical study on pretensioned bolt-cable combined support of large cross-section tunnel显示文摘To study the mechanical responses of large cross-section tunnel reinforced by pretensioned rock bolts and anchor cables, an analytical model is proposed. Considering the interaction between rock mass and bolt-cable support, the strain softening characteristic of rock mass, the elastic-plastic characteristic of bolt-cable support, and the delay effect of installation are considered in the model. To solve the different mechanical cases of tunneling reinforced by bolt-cable support, an analytical approach has been put forward to get the solutions of stress and displacement associated with tunneling. The proposed analytical model is verified by numerical simulation. Moreover, parametric analysis is performed to study the effects of pretension force,cross-section area, length, and supporting density of bolt-cable support on tunnel reinforcement, which can provide references for determining these parameters in tunnel design. Based on the analytical model, a new Ground Response Curve(GRC)considering the reinforcement of bolt-cable support is obtained, which shows the pretension forces and the timely installation are important in bolt-cable support. 5Ambient volatile organic compounds at a receptor site in the Pearl River Delta region:Variations,source apportionment and effects on ozone formation显示文摘We present the continuously measurements of volatile organic compounds(VOCs)at a receptor site(Wan Qing Sha,WQS)in the Pearl River Delta(PRD)region from September to November of 2017.The average mixing ratios of total VOCs(TVOCs)was 36.3±27.9 ppbv with the dominant contribution from alkanes(55.5%),followed by aromatics(33.3%).The diurnal variation of TVOCs showed a strong photochemical consumption during daytime,resulting in the formation of ozone(O_(3)).Five VOC sources were resolved by the positive matrix factorization(PMF)model,including solvent usage(28.6%),liquid petroleum gas(LPG)usage(24.4%),vehicle exhaust(21.0%),industrial emissions(13.2%)and gasoline evaporation(12.9%).The regional transport air masses from the upwind cities of south China can result in the elevated concentrations of TVOCs.Low ratios of TVOCs/NO_(x)(1.53±0.88)suggested that the O_(3) formation regime at WQS site was VOC-limited,which also confirmed by a photochemical box model with the master chemical mechanism(PBM-MCM).Furthermore,the observation on high-O_(3) episode days revealed that frequent O_(3) outbreaks at WQS were mainly caused by the regional transport of anthropogenic VOCs especially for aromatics and the subsequent photochemical reactions.This study provides valuable information for policymakers to propose the effective control strategies on photochemical pollution in a regional perspective.Yao Meng Junwei Song Lewei Zeng Yingyi Zhang Yan Zhao Xufei Liu Hai Guo Liuju Zhong Yubo Ou Yan Zhou Tao Zhang Dingli Yue Senchao Lai 2022Journal of Environmental Sciences2022,34,1:5
6A microarray-based gastric carcinoma prewarning system显示文摘AIM: To develop a microarray-based prewarning system consisting of gastric cancer chip, prewarning data and analysissoftware for early detection of gastric cancer and pre-cancerous lesions.METHODS: Two high-density chips with 8 464 human cDNA sites were used to primarily identify potential genes specific for normal gastric mucosa, pre-cancerous lesion and gastric cancer. The low-density chips, composed of selected genes associated with normal gastric mucosa,precancerous lesion and gastric cancer, were fabricated and used to screen 150 specimens including 60 specimens of gastric cancer, 60 of pre-cancerous tissues and 30 of normal gastric mucosa. CAD software was used to screen out the relevant genes and their critical threshold values of expression levels distinguishing normal mucosa from pre-cancerous lesion and cancer. All data were stored in a computer database to establish a prewarning data library for gastric cancer. Two potential markers brcaa1 and ndr1were identified by Western blot and immunohistochemistry.RESULTS: A total of 412 genes associated with three stages of gastric cancer development were identified.There were 216 genes displaying higher expression in gastric cancer, 85 genes displaying higher expression in pre-cancerous lesion and 88 genes displaying higher expression in normal gastric mucosa. Also 15 genes associated with metastasis of gastric cancer and 8 genes associated with risk factors were screened out for target genes of diagnosis chip of early gastric cancer. The threshold values of 412 selected genes to distinguish gastric cancer, pre-cancerous lesion from normal gastric mucosa were defined as 6.01±2.40, 4.86±1.94 and 5.42±2.17, respectively. These selected 412 genes and critical threshold values were compiled into an analysis software, which can automatically provide reports by analyzing the results of 412 genes obtained by examining gastric tissues. All data were compiled into a prewarning database for gastric cancer by CGO software. Northern blot and immunohistochemistry analysis confirmed that gene and protein of brcaa1 displayed lower expression in normal gastric mucosa and higher expression in gastric cancer tissues, conversely, ndr1 displayed lower expression in gastric cancer and higher expression in normal gastric mucosa.CONCLUSION: The microarray-based prewarning system for gastric cancer was developed. This system consisted of gastric cancer-associated gene chip, prewarning data and analysis software, which has a high potential for applications in the early detection of gastric cancer. 7Determination of Mirtazapine in Human Plasma by HPLC-MS and Bioavailability of Newly Developed Mirtazapine Tablets in Healthy Volunteers显示文摘Aim To estabhsh a sensitive and specific liquid chromatography-mass spectrometry method for determination of mirtazapine in human plasma and evaluation of its relative bioavailabihty. Methods After being alkalized by 25% ammonia, mirtazapine in the plasma was extracted with n-hexane. Desloratadine was used as internal standard (IS). Solutes were separated on a C18 column with a mobile phase of methanol-ammonium acetate buffer (pH3.5) (75:25). The flow rate of the mobile phase was 1 mL·min^-1. Detection was performed on an electrospray ionization (ESI) mass spectrometer and operated in selected ion monitoring (SIM) and positive-ionization mode using target ionsat m/z 266.2 for mirtazapine and m/z 311.2 for the IS. The fragmentor voltage was 90 V. A randomized cross-over study was performed in 20 healthy volunteers. In the two study periods, twenty healthy Chinese male subjects received a single oral dose of mirtazapine 30 mg. Results The calibration curve was linear over the range of 0.3-200 ng·mL^-1. The limit of quantitation was 0.1 ng·mL^-1. The paramneters for mirtazapine test tablet and reference tablet were as follows.. T1/2 ( 24.7±4.1 ) and (23.6±4.3) h, Tmax (1.6±0.8) and (1.5±0.8) h, Cmax (95.9±29.8) and (91.9±26.7) ng·mL^-1, respectively. Conclusion The estabhshed HPLC-MS method is rapid, sensitive and specific for the detemtination of mirtazapine in human plasma. 8Scientific problems and research proposals for Sichuan–Tibet railway tunnel construction显示文摘The Sichuan–Tibet railway construction has received significant attention from both China and abroad.The new section from Ya’an to Nyingchi is about 1018.6 km long,including 69 tunnels with a total length of 841.7 km.The longest tunnel(Yigong tunnel)is 42.4 km long.The successful construction of the Sichuan–Tibet railway depends largely on tunnel construction.Due to the complex conditions,tunnel construction for the Sichuan–Tibet railway poses great challenges to the research community.This paper aims to illustrate the necessity of interdisciplinary and joint research to solve problems associated with tunnel construction that will be encountered in the immediate future and facilitate the communication and exchange of ideas between disciplines.To this end,based on a systematic analysis of the available data related to Sichuan–Tibet railway tunnels,six major aspects of the geology,environment,and engineering conditions are identified.Next,based on the engineering responses from the Sichuan–Tibet railway tunnels,potential technical problems and risk characteristics are predicted and evaluated.Further,six key scientific issues are identified and discussed based on a coupled multi-layer analysis of essential tunnel engineering issues.Finally,research directions,technical ideas,and research methods that should be carried out for this project are proposed.Dingli Zhang Zhenyu Sun Qian Fang 2022Underground Space2022,7,3:3

11LINC00891 regulated by miR-128-3p/GATA2 axis impedes lung cancer cell proliferation,invasion and EMT by inhibiting RhoA pathway显示文摘Long non-coding RNA(lncRNA)LINC00891 knockdown is associated with poor prognosis of lung adenocarcinoma,but the underlying mechanism remains to be further explored.Here,we found that LINC00891 expression is downregulated in lung cancer tissues and cell lines compared with that in adjacent normal tissues and normal lung epithelial cells.LINC00891 overexpression impedes cell proliferation,invasion,migration and epithelial-to-me-senchymal transition(EMT)process in lung cancer cells.Mechanistic research showed that GATA2 directly binds to LINC00891 promoter and transcriptionally regulates LINC00891 expression.Meanwhile,GATA2 was identified as a target of miR-128-3p,and it is negatively regulated by miR-128-3p.Moreover,overexpression of GATA2 suppresses lung cancer cell proliferation,invasion,migration,and EMT process.Furthermore,LINC00891 restrains the RhoA pathway activity,and treatment with CCG-1423(a specific RhoA pathway inhibitor)antagonizes the promoting effect of LINC00891 knockdown on cell malignant behaviors.Additionally,silencing of LINC00891 promotes xenograft tumor growth,which can be reversed by administration with CCG-1423.In summary,LINC00891 regulated by the miR-128-3p/GATA2 axis restrains lung cancer cell malignant progression and hinders xenograft tumor growth by suppressing the RhoA pathway.Yunfeng Zhang Dingli Song Ziyang Peng Rui Wang Kai Li Hong Ren Xin Sun Ning Du Shou-Ching Tang 2022Acta Biochimica et Biophysica Sinica2022,54,3:2
12Differential Responses to Simulated Precipitation Exhibited by a Typical Shrub and a Herb Coexisted in Hunshandak Sandy Land显示文摘To assess the ecophysiological responses and adaptive strategies to rainfall exhibited by different plant functional types, we compared gas exchange, leaf water potential (ψleaf) and PS photochemical efficiency (Fv/Fm) between Hedysarum fruticosum Pall. (shrub) and Salsola collina Y. L. Chang(herb) under a series of simulated rainfalls. The experiment was conducted in Hunshandak Sandy Land, North China. The values of net photosynthetic rate (Pn), transpiration rate (E), stomatal conductance (gs), Fv/Fm and ψleaf always elevated with the increasing amount of simulated rainfall. Whereas water useefficiency (WUE) maintained constant in H. fruticosum while always decreased in S.collina. Both speciesfully recovered their Pn and Fv/Fm 3d after rainfall, while gs and ψleaf recovered only within 2 d. However, the response patterns were obviously different. H.fruticosu promote its physiological traits within 5-15 mm rainfall but no further positive enhancement was noted when rainfall was larger than 15 mm. In S.collina, however, the enhancement continued with more water applied. We inferred that H.fruticosum and S.collina responded similarly to rainfall in time courses but differential to simulated precipitation change. 13Analysis of Cardiotoxicity from rh-Endostatin Therapy Combined with Chemotherapy显示文摘OBJECTIVE To evaluate the cardotoxicity from recombinant human endostatin(rh-endostatin)combined with chemotherapy. METHODS A total of 12 cancer patients treated with rh- endostatin combined with chemotherapy were selected,and their clinical data collected.Their symptoms,including cardiopalmus, chest distress,dyspnea and changes in their electrocardiogram (ECG),myocardium enzymogram and left ventricular ejection fraction(LVEF),were observed during the drug treatment.These indicators were used for early diagnosis of cardiotoxicity. RESULTS Compared with a pre-therapeutic value,there was a significant increase in the CK-MB value at one week after starting the treatment as well as at the end of treatment(P<0.05).There was a significant change in the ECG at the end of treatment, compared to a pre-therapeutic condition(P<0.05),but there was no significant difference when comparing the pre-and post- therapeutic LVEF values. CONCLUSION It was recognized that mild cardiac adverse reactions exist in the regimen of recombinant human endostatin combined with chemotherapy.This therapy caused definite injury to the cardiac muscle,but cardiac functions were not obviously changed.CK-MB and ECG may be used as indicators for early monitoring cardiac toxicity.Vigilance against cardiac adverse reactions should be heightened during a course of rh-endostatin combined with chemotherapy.Jing Qin Penghai Zhang Xinyu Qian Aimin Li Rongcheng Luo Dingli Xu 2008Chinese Journal of Clinical Oncology2008,5,4:2
