2Mesenchymal stem cells and the neuronal microenvironment in the area of spinal cord injury显示文摘Cell-based technologies are used as a therapeutic strategy in spinal cord injury(SCI). Mesenchymal stem cells(MSCs), which secrete various neurotrophic factors and cytokines, have immunomodulatory, anti-apoptotic and anti-inflammatory effects, modulate reactivity/phenotype of astrocytes and the microglia, thereby promoting neuroregeneration seem to be the most promising. The therapeutic effect of MSCs is due to a paracrine mechanism of their action, therefore the survival of MSCs and their secretory phenotype is of particular importance. Nevertheless, these data are not always reported in efficacy studies of MSC therapy in SCI. Here, we provide a review with summaries of preclinical trials data evaluating the efficacy of MSCs in animal models of SCI. 3Noninvasive investigations for non alcoholic fatty liver disease and liver fi brosis显示文摘Non-alcoholic fatty liver disease (NAFLD) includes a spectrum of diseases that have insulin resistance in common and are associated with metabolic conditions such as obesity, type 2 diabetes mellitus, and dyslipidemia. NAFLD ranges from simple liver steatosis, which follows a benign course, to nonalcoholic steatohepatitis (NASH), a more severe entity, with necroinflmmation and f ibrosis, which can progress to cryptogenic cirrhosis and end-stage liver disease. Liver biopsy remains the gold standard for evaluating the degree of hepatic necroinflammation and f ibrosis; however, several noninvasive investigations, such as serum biomarkers, have been developed to establish the diagnosis and also to evaluate treatment response. These markers are currently neither available in all centers nor validated in extensive studies. Examples include high-sensitivity C reactive protein and plasma pentraxin 3, which are associated with extensive liver f ibrosis in NASH. Interleukin-6 correlates with inflammation, and cytokeratin-18 represents a marker of hepatocyte apoptosis (prominent in NASH and absent in simple steatosis). 6Histopathology of nonalcoholic fatty liver disease显示文摘Histological analysis of liver biopsies remains a standard against which other methods of assessment for the presence and amount of hepatic injury due to nonalcoholic fatty liver disease(NAFLD) are measured.Histological evaluation remains the sole method of distinguishing steatosis from advanced forms of NAFLD,i.e.nonalcoholic steatohepatitis(NASH) and fibrosis.Included in the lesions of NAFLD are steatosis,lobular and portal inflammation,hepatocyte injury in the forms of ballooning and apoptosis,and fibrosis.However,patterns of these lesions are as distinguishing as the lesions themselves.Liver injury in adults and children due to NAFLD may have different histological patterns.In this review,the rationale for liver biopsy,as well as the histopathological lesions,the microscopically observable patterns of injury,and the differential diagnoses of NAFLD and NASH are discussed.Elizabeth M Brunt Dina G Tiniakos 2010World Journal of Gastroenterology2010,16,42:16
7New technologies to investigate the brain-gut axis显示文摘Functional gastrointestinal disorders are commonly encountered in clinical practice, and pain is their commonest presenting symptom. In addition, patients with these disorders often demonstrate a heightened sensitivity to experimental visceral stimulation, termed visceral pain hypersensitivity that is likely to be important in their pathophysiology. Knowledge of how the brain processes sensory information from visceral structures is still in its infancy. However, our understanding has been propelled by technological imaging advances such as functional Magnetic Resonance Imaging, Positron Emission To-mography, Magnetoencephalography, and Electroen-cephalography (EEG). Numerous human studies have non-invasively demonstrated the complexity involved in functional pain processing, and highlighted a number of subcortical and cortical regions involved. This review will focus on the neurophysiological pathways (primary afferents, spinal and supraspinal transmission), brain- imaging techniques and the influence of endogenous and psychological processes in healthy controls and patients suffering from functional gastrointestinal dis- orders. Special attention will be paid to the newer EEG source analysis techniques. Understanding the pheno- typic differences that determine an individual's response to injurious stimuli could be the key to understanding why some patients develop pain and hyperalgesia in response to inflammation/injury while others do not. 9Increased resistance of drought by Trichoderma harzianum fungal treatment correlates with increased secondary metabolites and proline content显示文摘Plant secondary metabolites play vital role in plant stress response. In this study we investigated whether root colonization of tomato(Solanum lycopersicum) infected by Trichoderma harzianum leads to alterations in the biosynthesis of secondary plant metabolites including phytohormones and osmolyte proline under drought stress. Exposure of tomato to drought caused a drastic decline in plant growth and physiological parameters. Tomato inoculated with T. harzianum showed increased root and shoot growth and chlorophyll pigments as compared to uninoculated controls as well as drought stressed plants. Proline and total soluble protein content was increased in plants inoculated with T. harzianum under both normal as well as drought conditions. An obvious increase in phenol and flavonoid content was observed due to T. harzianum. In addition, T. harzianum inoculated plants maintained higher levels of growth regulators indole acetic acid, indole butyric acid, and gibberellic acid under drought stress. 10圆二色光谱、红外光谱法解析羊乳和牛乳β-酪蛋白结构及性质差异显示文摘羊乳β-酪蛋白比牛乳β-酪蛋白更容易被婴幼儿消化吸收,主要原因是二者结构的不同。目前对牛乳β-酪蛋白结构的研究较多,但对羊乳β-酪蛋白的结构以及羊乳和牛乳β-酪蛋白结构差异的研究还鲜有报道。蛋白质二级结构的信息可由光谱获得,其中圆二色光谱是利用蛋白质分子中具有光学活性的生色基团对左、右平面圆偏振光吸收不同,对蛋白质结构进行表征的方法,可以测定溶液状态下的蛋白质样品,使蛋白质构象更接近其生理状态,而且具有快速简便,对构象变化灵敏等优点;红外光谱则是利用蛋白质分子在振动过程中不同化学键或官能团对红外光吸收频率不同,对蛋白质结构进行表征的方法,可以测定固体状态下的蛋白质样品,具有扫描速度快、分辨率高、可测波长范围广、不易受蛋白质样品的分子大小和外界条件影响等优点。圆二色光谱和红外光谱已被广泛应用于蛋白质构象的研究中,但是结合使用这两种方法分析β-酪蛋白结构的研究还鲜有报道。因此,该研究采用圆二色光谱和红外光谱比较羊乳和牛乳β-酪蛋白的结构特点,并利用分光光度法对二者的巯基含量及溶解性进行了分析,从功能性质方面的不同对两种蛋白结构的差异进行更好的说明。圆二色光谱测得羊乳和牛乳β-酪蛋白二级结构中主要以无规卷曲为主,但羊乳β-酪蛋白的无规卷曲含量(50.2%±0.16%)显著高于牛乳β-酪蛋白(43.8%±0.14%),其有序结构中α-螺旋含量(2.7%±0.21%)、β-折叠含量(15.3%±0.08%)显著低于牛乳β-酪蛋白(4.3%±0.13%, 19.5%±0.12%),β-转角含量分别为31.8%±0.11%和32.4%±0.09%,差异不显著;红外光谱测得羊乳β-酪蛋白二级结构中α-螺旋、β-折叠、β-转角含量分别比牛乳β-酪蛋白低18%~20%, 9%~10%, 0.6%~1%,无规卷曲含量比牛乳β-酪蛋白高17%~19%。对两种蛋白功能性质的研究表明,羊乳β-酪蛋白与牛乳β-酪蛋白表面巯基含量基本一致19~20μmol·g^-1,但羊乳β-酪蛋白总巯基含量[(28.35±0.13)μmol·g^-1]显著低于牛乳β-酪蛋白[(46.72±0.21)μmol·g^-1];羊乳β-酪蛋白与牛乳β-酪蛋白的等电点较为接近(pH为4~5),且在等电点附近前者的溶解性低于后者,而远离等电点时前者溶解性则高于后者。研究结果说明与牛乳β-酪蛋白相比,羊乳β-酪蛋白分子的无序性和柔韧性更高,胶束内部结构更加的柔软疏松。李萌 王娟 魏子凯 康佳欣 张玲 Tabys Dina 刘宁 张爽 2020光谱学与光谱分析2020,40,3:12
11Pathophysiology of chronic pancreatitis显示文摘Chronic pancreatitis(CP)is an inflammatory disease of the pancreas characterized by progressive fibrotic destruction of the pancreatic secretory parenchyma.Despite the heterogeneity in pathogenesis and involved risk factors,processes such as necrosis/apoptosis,inflammation or duct obstruction are involved.This fibrosing process ultimately leads to progressive loss of the lobular morphology and structure of the pancreas,deformation of the large ducts and severe changes in the arrangement and composition of the islets.These conditions lead to irreversible morphological and structural changes resulting in impairment of both exocrine and endocrine functions.The prevalence of the disease is largely dependent on culture and geography.The etiological risk-factors associated with CP are multiple and involve both genetic and environmental factors.Throughout this review the M-ANNHEIM classification system will be used,comprising a detailed description of risk factors such as:alcohol-consumption,nicotineconsumption,nutritional factors,hereditary factors,efferent duct factors,immunological factors and miscellaneous and rare metabolic factors.Increased knowledge of the different etiological factors may encourage the use of further advanced diagnostic tools,which potentially will help clinicians to diagnose CP at an earlier stage.However,in view of the multi factorial disease and the complex clinical picture,it is not surprising that treatment of patients with CP is challenging and often unsuccessful.Christina Brock Lecia Mφller Nielsen Dina Lelic Asbjφrn Mohr Drewes 2013World Journal of Gastroenterology2013,19,42:11
12E2F transcription factors and digestive system malignancies: How much do we know?显示文摘E2F family of transcription factors regulates various cellular functions related to cell cycle and apoptosis. Its individual members have traditionally been classified into activators and repressors, based on in vitro studies. However their contribution in human cancer is more complicated and difficult to predict. We review current knowledge on the expression of E2Fs in digestive system malignancies and its clinical implications for patient prognosis and treatment. E2F1, the most extensively studied member and the only one with prognostic value, exhibits a tumor-suppressing activity in esophageal, gastric and colorectal adenocarcinoma, and in hepatocellular carcinoma (HCC), whereas in pancreatic ductal adenocarcinoma and esophageal squamous cell carcinoma may function as a tumorpromoter. In the latter malignancies, E2F1 immunohistochemical expression has been correlated with higher tumor grade and worse patient survival, whereas in esophageal, gastric and colorectal adenocarcinomas is a marker of increased patient survival. E2F2 has only been studied in colorectal cancer, where its role is not considered significant. E2F4's role in colorectal, gastric and hepatic carcinogenesis is tumor-promoting. E2F8 is strongly upregulated in human HCC, thus possibly contributing to hepatocarcinogenesis. Adenoviral transfer of E2F as gene therapy to sensitize pancreatic cancer cells for chemotherapeutic agents has been used in experimental studies. Other therapeutic strategies are yet to be developed, but it appears that targeted approaches using E2F-agonists or antagonists should take into account the tissue-dependent function of each E2F member. 13Vascular calcification:When should we interfere in chronic kidney disease patients and how?显示文摘Chronic kidney disease(CKD) patients are endangered with the highest mortality rate compared to other chronic diseases. Cardiovascular events account for up to 60% of the fatalities. Cardiovascular calcifications affect most of the CKD patients. Most of this calcification is related to disturbed renal phosphate handling. Fibroblast growth factor 23 and klotho deficiency were incriminated in the pathogenesis of vascular calcification through different mechanisms including their effects on endothelium and arterial wall smooth muscle cells. In addition, deficient klotho gene expression, a constant feature of CKD, promotes vascular pathology and shares in progression of the CKD. The role of gut in the etio-pathogenesis of systemic inflammation and vascular calcification is a newly discovered mechanism. 19Liver histology in ICU patients dying from sepsis:A clinico-pathological study显示文摘AIM:To determine end-stage pathologic changes in the liver of septic patients dying in the intensive care unit. METHODS: Needle liver biopsies obtained immediately after death from 15 consecutive patients with sepsis and no underlying liver disease were subjected to routine histological examination. Liver function tests and clinical monitoring measurements were also recorded. RESULTS: Liver biochemistries were increased in the majority of patients before death. Histology of liver bi- opsy specimens showed portal inflammation in 73.3%, centrilobular necrosis in 80%, lobular inflammation in 66.7%, hepatocellular apoptosis in 66.6% and cholan- gitis/cholangiolitis in 20% of patients. Mixed hepatitic/ cholestatic type of liver injury was observed in 6/15 (40%) patients and hepatitc in 9/15 (60%). Steatosis was ob- served in 11/15 (73.3%) patients affecting 5%-80% of liver parenchyma. Among the histological features, the presence of portal inflammation in liver biopsy was as- sociated with increased hospitalization in the ICU prior death (P = 0.026). 20Pathological,immunohistochemical and microbiologicalal analysis of lacrimal sac biopsies in patients with chronic dacrocystitis显示文摘AIM:To analyze cases of obstruction of the nasolacrimal duct which creates a fertile environment for secondary bacterial Infection and can result in dacryocystitis,which is a constant threat to cornea and orbital soft tissue and a potential source of endophthalmitis following intraocular surgery.The majority of obstructions of the lacrimal excretory outflow system are acquired ones occurring in adulthood and involving the distal parts of the system.Acquired obstruction may be primary/idiopathic or secondary to a wide variety of infectious,inflammatory,traumatic,mechanical,toxic or neoplastic causes mimicking idiopathic inflammation.These cases are treated by dacryocystorhinostomy(DCR).METHODS:The present study was conducted to determine the histopathologic,immunohistochemical and current microbiologic characteristics of lacrimal sac specimens in patients undergoing external dacryocystorhinostomy.RESULTS:Non-specific lacrimal sac pathology was present in all 33 cases and 81.8%of the cases showed moderate chronic inflammation with a chronic inflammatory score(CIS)ranging between 4 and 6,whereas 12.12%showed severe inflammatory changes with a CIS of 7.Mild degree of inflammation was seen in6.06%with a CIS of 3.The total prevalence of grampositive,gram-negative,and culture-negative samples were 59.4%,37.5%,and 3%respectively.CONCLUSION:Non-specific chronic inflammation withfibrosis is indeed the most commonly reported histopathological finding in lacrimal sac wall biopsy specimens.Rowayda Mahmoud Amin Faten Aly Hussein Hisham Farouk Idriss Nesrine Fathy Hanafy Dina Mohamed Abdallah 2013International Journal of Ophthalmology(English edition)2013,6,6:7
