1Electrical storm: A clinical and electrophysiological overview显示文摘Electrical storm(ES) is a clinical condition characterized by three or more ventricular arrhythmia episodes leading to appropriate implantable cardioverterdefibrillator(ICD) therapies in a 24 h period. Mostly, arrhythmias responsible of ES are multiple morphologies of monomorphic ventricular tachycardia(VT), but polymorphic VT and ventricular fibrillation can also result in ES. Clinical presentation is very dramatic in most cases, strictly related to the cardiac disease that may worsen electrical and hemodynamic decompensation. Therefore ES management is challenging in the majority of cases and a high mortality is the rule both in the acute and in the long-term phases. Different underlying cardiomyopathies provide significant clues into the mechanism of ES, which can arise in the setting of structural arrhythmogenic cardiomyopathies or rarely in patients with inherited arrhythmic syndrome, impacting on pharmacological treatment, on ICD programming, and on the opportunity to apply strategies of catheter ablation. This latter has become a pivotal form of treatment due to its high efficacy in modifying the arrhythmogenic substrate and in achieving rhythm stability, aiming at reducing recurrences of ventricular arrhythmia and at improving overall survival. 2Pericyte-like differentiation of human adipose-derived mesenchymal stem cells:An in vitro study显示文摘BACKGROUND Adipose-derived mesenchymal stem cells(ASCs)are characterized by long-term self-renewal and a high proliferation rate.Under adequate conditions,they may differentiate into cells belonging to mesodermal,endodermal or ectodermal lineages.Pericytes support endothelial cells and play an important role in stabilizing the vessel wall at the microcirculation level.The loss of pericytes,as occurs in diabetic retinopathy,results in a breakdown of the blood-retina barrier(BRB)and infiltration of inflammatory cells.In this context,the use of pericytelike differentiated ASCs may represent a valuable therapeutic strategy for restoring BRB damage.AIM To test in vitro strategies to obtain pericyte-like differentiation of human ASCs(hASCs).METHODS Different culture conditions were tested:hASCs cultured in a basal medium supplemented with transforming growth factorβ1;and hASCs cultured in a specific pericyte medium(PM-hASCs).In a further sample,pericyte growth supplement was omitted from the PM.In addition,cultures of human retinal pericytes(hRPCs)were used for comparison.Pericyte-like differentiation of hASCs was tested by immunocytochemical staining and western blotting to evaluate the expression ofα-smooth muscle actin(α-SMA)and neural/glial antigen 2(NG2).Interactions between human retinal endothelial cells(hRECs)and different groups of hASCs were investigated in co-culture experiments.In these cases,the expression of typical junctional proteins such as vascular endothelial-Cadherin,zonula occludens-1 and Occludin were assessed in hRECs.In an in vitro model of the BRB,values of trans-endothelial electrical resistance were measured when hRECs were co-cultured with various groups of pretreated hASCs.The values observed were compared with co-cultures of hRECs and hRPCs as well as with cultures of hRECs alone.Three-dimensional co-cultures of hRECs and hRPCs or pericyte-like hASCs in Matrigel were designed to assess their reciprocal localization.RESULTS After 3-6 d of culture,α-SMA and NG2 immunocytochemistry showed that the closest pericyte-like phenotype was observed when hASCs were cultured in Pericyte Medium(PM-hASCs).In particular,α-SMA immunoreactivity,already visible at the basal level in pericytes and ASCs,was strongly increased only when transforming growth factor was added to the culture medium.NG2 expression,almost undetectable in most conditions,was substantially increased only in PMhASCs.Immunocytochemical results were confirmed by western blot analysis.The presence of pericyte growth supplement seems to increase NG2 expression rather thanα-SMA,in agreement with its role in maintaining pericytes in the proliferative state.In co-culture experiments,immunoreactivity of vascular endothelial-Cadherin,zonula occludens-1 and Occludin was considerably increased in hRECs when hRPCs or PM-hASCs were also present.Supporting results were found by trans-endothelial electrical resistance measurements,gathered at 3 and 6 d of co-culture.The highest resistance values were obtained when hRECs were co-cultured with hRPCs or PM-hASCs.The pericyte-like phenotype of PM-hASCs was also confirmed in three-dimensional co-cultures in Matrigel,where PM-hASCs and hRPCs similarly localized around the tubular formations made by hRECs.CONCLUSION PM-hASCs seem able to strengthen the intercellular junctions between hRECs,likely reinforcing the BRB;thus,hASC-based therapeutic approaches may be developed to restore the integrity of retinal microcirculation.Giuliana Mannino Florinda Gennuso Giovanni Giurdanella Federica Conti Filippo Drago Salvatore Salomone Debora Lo Furno Claudio Bucolo Rosario Giuffrida 2020World Journal of Stem Cells2020,12,10:3
3Photonic extreme learning machine by free-space optical propagation显示文摘Photonic brain-inspired platforms are emerging as novel analog computing devices,enabling fast and energyefficient operations for machine learning.These artificial neural networks generally require tailored optical elements,such as integrated photonic circuits,engineered diffractive layers,nanophotonic materials,or time-delay schemes,which are challenging to train or stabilize.Here,we present a neuromorphic photonic scheme,i.e.,the photonic extreme learning machine,which can be implemented simply by using an optical encoder and coherent wave propagation in free space.We realize the concept through spatial light modulation of a laser beam,with the far field acting as a feature mapping space.We experimentally demonstrate learning from data on various classification and regression tasks,achieving accuracies comparable with digital kernel machines and deep photonic networks.Our findings point out an optical machine learning device that is easy to train,energetically efficient,scalable,and fabrication-constraint free.The scheme can be generalized to a plethora of photonic systems,opening the route to real-time neuromorphic processing of optical data.DAVIDE PIERANGELI GIULIA MARCUCCI CLAUDIO CONTI 2021Photonics Research2021,9,8:2

14Localization and shock waves in curved manifolds显示文摘The investigation of the interplay between geometry and nonlinearity may open the road to the control of extreme waves. We study three-dimensional localization and dispersive shocks in a bent cigar shaped potential by the nonlinear Schro¨ dinger equation. At high bending and high nonlinearity, topological trapping is frustrated by the generation of curved wave-breaking. Four-dimensional parallel simulations confirm the theoretical model. 15Biosensing with free space whispering gallery mode microlasers显示文摘Highly accurate biosensors for few or single molecule detection play a central role in numerous key fields,such as healthcare and environmental monitoring.In the last decade,laser biosensors have been investigated as proofs of concept,and several technologies have been proposed.We here propose a demonstration of polymeric whispering gallery microlasers as biosensors for detecting small amounts of proteins,down to 400 pg.They have the advantage of working in free space without any need for waveguiding for input excitation or output signal detection.The photonic microsensors can be easily patterned on microscope slides and operate in air and solution.We estimate the limit of detection up to 148 nm/RIU for three different protein dispersions.In addition,the sensing ability of passive spherical resonators in the presence of dielectric nanoparticles that mimic proteins is described by massive ab initio numerical simulations.ANGELA CAPOCEFALO SILVIA GENTILINI LORENZO BAROLO PAOLA BAIOCCO CLAUDIO CONTI NEDA GHOFRANIHA 2023Photonics Research2023,11,5:0
16Electrical storm in systemic sclerosis: Inside the electroanatomic substrate显示文摘We report the case of a 63-year-old woman affected by a severe form of systemic scleroderma with pul-monary involvement(interstitial fibrosis diagnosed by biopsy and moderate pulmonary hypertension) and cardiac involvement(paroxysmal atrial fibrillation, right atrial flutter treated by catheter ablation, ventricular tachyarrhythmias, previous dual chamber implantable cardioverter defibrillator implant). Because of recurrent electrical storms refractory to iv antiarrhythmic drugs the patient was referred to our institution to undergo catheter ablation. During electrophysiological proce-dure a 3D shell of cardiac anatomy was created with intracardiac echocardiography pointing out a significant right ventricular dilatation with a complex aneurysmal lesion characterized by thin walls and irregular multiple trabeculae. A substrate-guided strategy of catheter ab-lation was accomplished leading to a complete electri-cal isolation of the aneurism and to the abolishment of all abnormal electrical activities. 17Biophotonic rogue waves in red blood cell suspensions显示文摘Rogue waves are ubiquitous in nature,appearing in a variety of physical systems ranging from acoustics,microwave cavities,optical fibers,and resonators to plasmas,superfluids,and Bose–Einstein condensates.Unlike nonlinear solitary waves,rogue waves are extreme events that can occur even without nonlinearity by,for example,spontaneous synchronization of waves with different spatial frequencies in a linear system.Here,we report the observation of rogue-wave-like events in human red blood cell(RBC)suspensions under weak light illumination,characterized by an abnormal L-shaped probability distribution.Such biophotonic extreme events arise mostly due to the constructive interference of Mie-scattered waves from the suspended RBCs,whose biconcave shape and mutable orientation give rise to a time-dependent random phase modulation to an incident laser beam.We numerically simulate the beam propagation through the colloidal suspensions with added disorder in both spatial and temporal domains to mimic random scattering due to Brownian motion.In addition,at high power levels,nonlinear beam self-focusing is also observed,leading to a dual-exponential probability distribution associated with the formation of multiple soliton-like spots.Such rogue wave events should also exist in environments with cells of other species such as swimming bacteria,and understanding of their underlying physics may lead to unexpected biophotonic applications.YU-XUAN REN JOSHUA LAMSTEIN CHENSONG ZHANG CLAUDIO CONTI DEMETRIOS NCHRISTODOULIDES ZHIGANG CHEN 2023Photonics Research2023,11,11:0

19Phase-matching-free parametric oscillators based on two-dimensional semiconductors显示文摘Optical parametric oscillators are widely used as pulsed and continuous-wave tunable sources for innumerable applications,such as quantum technologies,imaging,and biophysics.A key drawback is material dispersion,which imposes a phase-matching condition that generally entails a complex design and setup,thus hindering tunability and miniaturization.Here we show that the burden of phase-matching is surprisingly absent in parametric microresonators utilizing mono-layer transition-metal dichalcogenides as quadratic nonlinear materials.By the exact solution of nonlinear Maxwell equations and first-principle calculations of the semiconductor nonlinear response,we devise a novel kind of phase-matching-free miniaturized parametric oscillator operating at conventional pump intensities.We find that different two-dimensional semiconductors yield degenerate and non-degenerate emission at various spectral regions due to doubly resonant mode excitation,which can be tuned by varying the incidence angle of the external pump laser.In addition,we show that high-frequency electrical modulation can be achieved by doping via electrical gating,which can be used to efficiently shift the threshold for parametric oscillation.Our results pave the way for the realization of novel ultra-fast tunable micron-sized sources of entangled photons—a key device underpinning any quantum protocol.Highly miniaturized optical parametric oscillators may also be employed in lab-on-chip technologies for biophysics,detection of environmental pollution and security.Alessandro Ciattoni Andrea Marini Carlo Rizza Claudio Conti 2018Light(Science & Applications)2018,7,1:0
20Non-locality and collective emission in disordered lasing resonators显示文摘Random lasing is observed in optically active resonators in the presence of disorder.As the optical cavities involved are open,the modes are coupled,and energy may pour from one state to another provided that they are spatially overlapping.Although the electromagnetic modes are spatially localized,our system may be actively switched to a collective state,presenting a novel form of non-locality revealed by a high degree of spectral correlation between the light emissions collected at distant positions.In a nutshell,light may be stored in a disordered nonlinear structure in different fashions that strongly differ in their spatial properties.This effect is experimentally demonstrated and theoretically explained in titania clusters embedded in a dye,and it provides clear evidence of a transition to a multimodal collective emission involving the entire spatial extent of the disordered system.Our results can be used to develop a novel type of miniaturized,actively controlled all-optical chip.Marco Leonetti Claudio Conti Cefe Lopez 2013Light(Science & Applications)2013,2,1:0
