1Structure,Orientation and Finite Element Analysis of the Tail Club of Mamenchisaurus hochuanensis显示文摘The structure and orientation of the posterior extremity (tail club) of the caudal vertebrae of Mamenchisaurus hochuanensis Young and Chao, 1972 from the Upper Jurassic Shangshaximiao Formation has been analyzed to determine the tail club function using Finite Element Analysis. Of the four caudal vertebrae composing the tail club, the second largest (C'1') was probably the most proximal, and is fixed with the preceding sequence of the caudal vertebrae, whereas the smallest (C'4') is free and forms the termination of the tail club. Our analysis also suggests that the tail club is more efficient in lateral swinging rather than up-and-down motion, and that the best region for the tail club to impact is at the spine of the largest of the four caudals (C'2'), with a maximum load for impact at about 450 N. The tail club of Mamenchisaurus hochuanensis probably also had limitations as a defense weapon and was more possibly a sensory organ to improve nerve conduction velocity to enhance the capacity for sensory perception of its surroundings.XING Lida YE Yong SHU Chunkang PENG Guangzhao YOU Hailu 2009Acta Geologica Sinica(English Edition)2009,83,6:5

3AHR signaling pathway reshapes the metabolism of AML/MDS cells and potentially leads to cytarabine resistance显示文摘Emerging evidence suggests that aryl hydrocarbon receptor(AHR)promotes the initiation,invasion,progression,and metastasis of cancer cells.However,its effects in patients with myelodysplastic syndrome/acute myeloid leukemia(MDS/AML)remain undefined.In this study,we aimed to investigate the effects of AHR activation on malignant cells in patients with MDS/AML.We found that AHR was expressed aberrantly in patients with MDS/AML.Further studies demonstrated that inhibiting AHR decreased the mitochondrial dehydrogenase content and the mitochondrial membrane potential(MMP)in MDS/AML cells.Activating AHR with L-kynurenine(Kyn)increased AHR expression,which was accompanied by an increase in mitochondrial dehydrogenase content and MMP in MDS/AML cells.Moreover,the expression level of mitochondria-associated mitochondrial transcription factor A was increased after activating AHR with L-Kyn when compared with that in the control group but decreased after inhibiting the AHR signal.Activating AHR in MDS/AML cells enhanced the resistance to cytarabine.These findings indicated that activating the AHR signaling pathway reshaped the metabolism in MDS/AML cells,thus contributing to the resistance to cytarabine.Yan Jia Juan Guo Youshan Zhao Zheng Zhang Lei Shi Ying Fang Dong Wu Lingyun Wu Chunkang Chang 2021Acta Biochimica et Biophysica Sinica2021,53,4:1

5The expression of chemokine MCP-1 in colorectal carcinoma and its relationship to the infiltration of macrophage显示文摘Objective: To study the expression of MCP-1 in colorectal carcinoma and its relationship to the infiltration of the macrophage and to the biological behaviour of infiltration and metastasis of colorectal carcinoma. Methods: The expression of the MCP-1 mRNA was assessed in colorectal carcinoma collected freshly from surgical specimen by RT-PCR and the expres- sion of the MCP-1 protein was assessed in colorectal carcinoma collected from surgical specimen by immunohistochemistry. The tumor infiltrating cell and macrophage were also investigated by immunohistochemistry. Results: All the 12 specimens of colorectal carcinoma detected by RT-PCR expressed the MCP-1 mRNA; MCP-1 protein was detected in 90℅ (36/40) cases of the tumor; The expression of the MCP-1 protein in colorectal carcinoma correlated negatively with its state of metastasis and the Dukes’ stage. But a postive correlation was found between the expression of MCP-1 and the infiltrated macrophage. The stron- ger expression of MCP-1, the more number of the infiltrated macrophage. 10S100a9 deficiency accelerates MDS-associated tumor escape via PD-1/PD-L1 overexpression显示文摘In recent studies,the tolerable safety profile and positive bone marrow(BM)response suggest a beneficial use of anti-PD-1 agents in the treatment of Myelodysplastic Syndromes(MDS),but the underlying mechanism is still unknown.MDS is mainly characterized by ineffective hematopoiesis,which may contribute to inflammatory signaling or immune dysfunction.Our previous studies focused on inflammatory signaling,and the results showed that S100a9 expression was higher in low-risk MDS and lower in high-risk MDS.In this study,we combine the inflammatory signaling and immune dysfunction.SKM-1 cells and K562 cells co-cultured with S100a9 acquire apoptotic features.Moreover,we confirm the inhibitory effect of S100a9 on PD-1/PD-L1.Importantly,PD-1/PD-L1 blockade and S100a9 can both activate the PI3K/AKT/mTOR signaling pathway.The cytotoxicity is higher in lowerrisk MDS-lymphocytes than in high-risk MDS-lymphocytes,and S100a9 partially rescues the exhausted cytotoxicity in lymphocytes.Our study demonstrates that S100a9 may inhibit MDS-associated tumor escape via PD-1/PD-L1 blockade through PI3K/AKT/mTOR signaling pathway activation.Our findings indicate the possible mechanisms by which anti-PD-1 agents may contribute to the treatment of MDS.These insights may provide mutation-specific treatment as a supplementary therapy for MDS patients with high-risk mutations,such as TP53,N-RAS or other complex mutations.Roujia Wang Youshan Zhao Zijuan Li Juan Guo Sida Zhao Luxi Song Dong Wu Lan Wang Chunkang Chang 2023Acta Biochimica et Biophysica Sinica2023,55,2:0
