| 2 | Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold. | 4 | Icariside Ⅱ inhibits lipopolysaccharide-induced inflammation and amyloid production in rat astrocytes by regulating IKK/IκB/NF-κB/BACE1 signaling pathway显示文摘β-amyloid(Aβ)is one of the inducing factors of astrocytes activation and neuroinflammation,and it is also a crucial factor for the development of Alzheimer’s disease(AD).Icariside II(ICS II)is an active component isolated from a traditional Chinese herb Epimedium,which has shown to attnuate lipopolysaccharide(LPS)-induced neuroinflammation through regulation of NF-κB signaling pathway.In this study we investigated the effects of ICS II on LPS-induced astrocytes activation and Aβaccumulation.Primary rat astrocytes were pretreated with ICS II(5,10,and 20μM)or dexamethasone(DXMS,1μM)for 1 h,thereafter,treated with LPS for another 24 h.We found that ICS II pretreatment dose dependently mitigated the levels of tumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β),inducible nitric oxide synthase(iNOS),cyclooxygenase-2(COX-2)in the astrocytes.Moreover,ICS II not only exerted the inhibitory effect on LPS-induced IκB-αdegradation and NF-κB activation,but also decreased the levels of Aβ1–40,Aβ1–42,amyloid precursor protein(APP)and beta secretase 1(BACE1)in the astrocytes.Interestingly,molecular docking revealed that ICS II might directly bind to BACE1.It is concluded that ICS II has potential value as a new therapeutic agent to treat neuroinflammation-related diseases,such as AD. | Yong Zheng Yan Deng Jian-mei Gao Chun Lv Ling-hu Lang Jing-shan Shi Chang-yin Yu Qi-hai Gong | 2020 | Acta Pharmacologica Sinica2020，41，2： | 8 |

| 16 | Sex Differences in Diagnosis and Clinical Phenotypes of Chinese Children with Autism Spectrum Disorder显示文摘The aim of this study was to explore the differences between boys and girls in the diagnosis and clinical phenotypes of autism spectrum disorder(ASD) in China's mainland. Children diagnosed with ASD(n = 1064, 228 females) were retrospectively included in the analysis. All children were assessed using the Autism Diagnostic Interview-Revised(ADI-R) and Autism Diagnostic Observation Schedule(ADOS). The results showed that girls scored significantly higher in ADI-R socioemotional reciprocity than boys, and also scored lower in ADI-R and ADOS restricted and repetitive behaviors(RRBs). Meanwhile, the proportions of girls who satisfied the diagnostic cut-off scores in the ADI-R RRBs domain were lower than in boys(P \ 0.05). Our results indicated that girls with ASD show greater socio-emotional reciprocity than boys. Girls also tended to show fewer RRBs than boys, and the type of RRBs in girls differ from those in boys. | 17 | Isotope Geochronologic and Geochemical Constraints on the Magmatic Associations of the Collisional Orogenic Zone in the West Kunlun Orogen, China显示文摘The Jiajiwaxi pluton in the southern portion of the West Kunlun Range can be divided into two collision–related intrusive rock series, i.e., a gabbro–quartz diorite–granodiorite series that formed at 224±2.0 Ma and a monzonitic granite–syenogranite series that formed at 222±2.0 Ma. The systematic analysis of zircon U-Pb geochronology and bulk geochemistry is used to discuss the magmatic origin(material source and thermal source), tectonic setting, genesis and geotectonic implications of these rocks. The results of this analysis indicate that the parent magma of the first series, representing a transition from I-type to S-type granites, formed from thermally triggered partial melting of deep crustal components in an early island–arc–type igneous complex, similar to an I-type granite, during the continental collision orogenic stage. The parent magma of the second series, corresponding to an S-type granite, formed from the partial melting of forearc accretionary wedge sediments in a subduction zone in the late Palaeozoic–Triassic. During continued collision, the second series magma was emplaced into the first series pluton along a central fault zone in the original island arc region, forming an immiscible puncture-type complex. The deep tectonothermal events associated with the continent–continent collision during the orogenic cycle are constrained by the compositions and origins of the two series. | 19 | mTOR regulates TLR-induced c-fos and Th1 responses to HBV and HCV vaccines显示文摘Although IL-12 plays a critical role in priming Th1 and cytotoxic T lymphocyte(CTL) responses, Toll-like receptor(TLR) signaling only induces low amounts of IL-12 in dendritic cells and macrophages, implying the existence of stringent regulatory mechanisms. In this study, we sought to uncover the mechanisms underlying TLR-induced IL-12 expression and the Th1 response. By systemic screening, we identified a number of protein kinases involved in the regulation of TLRinduced IL-12 expression. In particular, PI3 K, ERK, and m TOR play critical roles in the TLR-induced Th1 response by regulating IL-12 and IL-10 production in innate immune cells. Moreover, we identified c-fos as a key molecule that mediates m TOR-regulated IL-12 and IL-10 expression in TLR signaling. Mechanistically, m TOR plays a crucial role in c-fos expression, thereby modulating NFκB binding to promoters of IL-12 and IL-10. By controlling the expression of a special innate gene program, m TOR can specifically regulate the TLR-induced T cell response in vivo. Furthermore, blockade of m TOR by rapamycin efficiently boosted TLR-induced antigen-specific T and B cell responses to HBV and HCV vaccines.
