1Overexpression of the M2 isoform of pyruvate kinase is an adverse prognostic factor for signet ring cell gastric cancer显示文摘AIM:To investigate M2 isoform of pyruvate kinase(PKM2) expression in gastric cancers and evaluate its potential as a prognostic biomarker and an anticancer target.METHODS:All tissue samples were derived from gastric cancer patients underwent curative gastrectomy as a primary treatment.Clinical and pathological information were obtained from the medical records.Gene expression microarray data from 60 cancer and 19 noncancer gastric tissues were analyzed to evaluate the expression level of PKM2 mRNA.Tissue microarrays were constructed from 368 gastric cancer patients.Immunohistochemistry was used to measure PKM2 expression and PKM2 positivity of cancer was determined by proportion of PKM2-positive tumor cells and staining intensity.Association between PKM2 expression and the clinicopathological factors was evaluated and the correlation between PKM2 and cancer prognosis was evaluated.RESULTS:PKM2 mRNA levels were increased more than 2-fold in primary gastric cancers compared to adjacent normal tissues from the same patients(log transformed expression level:7.6 ± 0.65 vs 6.3 ± 0.51,P < 0.001).Moreover,differentiated type cancers had significantly higher PKM2 mRNA compared to undifferentiated type cancers(log transformed expression level:7.8 ± 0.70 vs 6.7 ± 0.71,P < 0.001).PKM2 protein was mainly localized in the cytoplasm of primary cancer cells and detected in 144 of 368(39.1%) human gastric cancer cases.PKM2 expression was not related with stage(P = 0.811),but strongly correlated with gastric cancer differentiation(P < 0.001).Differentiated type cancers expressed more PKM2 protein than did the undifferentiated ones.Well differentiated adenocarcinoma showed 63.6% PKM2-positive cells;in contrast,signet-ring cell cancers showed only 17.7% PKM2-positive cells.Importantly,PKM2 expression was correlated with shorter overall survival(P < 0.05) independent of stage only in signet-ring cell cancers.CONCLUSION:PKM2 expression might be an adverse prognostic factor for signet-ring cell carcinomas.Its function and potential as a prognostic marker should be further verified in gastric cancer.Jae Yun Lim Sun Och Yoon So Young Seol Soon Won Hong Jong Won Kim Seung Ho Choi Jae Yong Cho 2012World Journal of Gastroenterology2012,18,30:19

3Diagnostic value for extrahepatic metastases of hepatocellular carcinoma in positron emission tomography/computed tomography scan显示文摘AIM: To evaluated the value of 18F-fluorodeoxyglucose (FDG) positron emission tomography (PET)/computed tomography (CT) scan in diagnosis of hepatocellular carcinoma (HCC) and extrahepatic metastases. METHODS: A total of 138 patients with HCC who had both conventional imaging modalities and 18F-FDG PET/CT scan done between November 2006 and March 2011 were enrolled. Diagnostic value of each imaging modality for detection of extrahepatic metastases was evaluated. Clinical factors and tumor characteristics including PET imaging were analyzed as indicative factors for metastases by univariate and multivariate methods. RESULTS: The accuracy of chest CT was significantly superior compared with the accuracy of PET imaging for detecting lung metastases. The detection rate of metastatic pulmonary nodule ≥ 1 cm was 12/13 (92.3%), when < 1 cm was 2/10 (20%) in PET imaging. The accuracy of PET imaging was significantly superior compared with the accuracy of bone scan for detecting bone metastases. In multivariate analy- sis, increased tumor size (≥ 5 cm) (P = 0.042) and increased average standardized uptake value (SUV) uptake (P = 0.028) were predictive factors for extrahepatic metastases. Isometabolic HCC in PET imaging was inversely correlated in multivariate analysis (P = 0.035). According to the receiver operating characteristic curve, the optimal cutoff of average SUV to predict extrahepatic metastases was 3.4. CONCLUSION: 18F-FDG PET/CT scan is invaluable for detection of lung metastases larger than 1 cm and bone metastases. 4Inhibition of pacemaker activity in interstitial cells of Cajal by LPS via NF-κB and MAP kinase显示文摘AIM:To investigate lipopolysaccharide(LPS) related signal transduction in interstitial cells of Cajal(ICCs) from mouse small intestine.METHODS:For this study,primary culture of ICCs was prepared from the small intestine of the mouse.LPS was treated to the cells prior to measurement of the membrane currents by using whole-cell patch clamp technique.Immunocytochemistry was used to examine the expression of the proteins in ICCs.RESULTS:LPS suppressed the pacemaker currents of ICCs and this could be blocked by AH6809,a prostaglandin E2-EP2 receptor antagonist or NG-Nitro-Larginine Methyl Ester,an inhibitor of nitric oxide(NO) synthase.Toll-like receptor 4,inducible NO synthase or cyclooxygenase-2 immunoreactivity by specific antibodies was detected on ICCs.Catalase(antioxidant agent) had no action on LPS-induced action in ICCs.LPS actions were blocked by nuclear factor kB(NF-kB) inhibitor,actinomycin D(a gene transcription inhibitor),PD 98059(a p42/44 mitogen-activated protein kinases inhibitor) or SB 203580 [a p38 mitogen-activated protein kinases(MAPK) inhibitor].SB 203580 also blocked the prostaglandin E2-induced action on pacemaker currents in ICCs but not NO.CONCLUSION:LPS inhibit the pacemaker currents in ICCs via prostaglandin E2-and NO-dependent mechanism through toll-like receptor 4 and suggest that MAPK and NF-kB are implicated in these actions.Dong Chuan Zuo Seok Choi Pawan Kumar Shahi Man Yoo Kim Chan Guk Park Young Dae Kim Jun Lee In Yeoup Chang Insuk So Jae Yeoul Jun 2013World Journal of Gastroenterology2013,19,8:9
5Anti-Proliferative Effects of Ginsenosides Extracted from Mountain Ginseng on Lung Cancer显示文摘Objective:To investigate the effect of three major ginsenosides from mountain ginseng as anticancer substance and explore the underlying mechanism involved in lung cancer.Methods:The inhibitory proliferation of lung cancer by major five ginsenosides(Rb1,Rb2,Rg1,Re,and Re) was examined using3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay.Calculated 50%inhibition(IC_(50)) values of five ginsenosides were determined and compared each other.Apoptosis by the treatment of single ginsenoside was performed by fluorescence-assisted cytometric spectroscopy.The alterations of apoptosis-related proteins were evaluated by Western blot analysis.Results:The abundance of ginsenosides in butanol extract of mountain ginseng(BX-MG) was revealed in the order of Rb1,Rg1,Re,Re and Rb2.Among them,Rb1 was the most effective to lung cancer cell,followed by Rb2 and Rg1 on the basis of relative IC_(50) values of IMR90 versus A549 cell.The alterations of apoptotic proteins were confirmed in lung cancer A549 cells according to the administration of Rb1,Rb2 and Rg1.The expression levels of caspase-3 and caspase-8 were increased upon the treatment of three ginsenosides,however,the levels of caspase-9 and anti-apoptotic protein Bax were not changed.Conclusion:Major ginsenosides such as Rb1,Rb2 and Rg1 comprising BX-MG induced apoptosis in lung cancer cells via extrinsic apoptotic pathway rather than intrinsic mitochondrial pathway.Dong Gyu Lee Sung-ll Jang Young-Rang Kim Kyeong Eun Yang So Jung Yoon Zee-Won Lee Hyun Joo An Ik-Soon Jang Jong-Soon Choi Hwa-Seung Yoo 2016Chinese Journal of Integrative Medicine2016,22,5:9
6Reliable high temperature, high humidity flexible thin film encapsulation using Al2O3/MgO nanolaminates for flexible OLEDs显示文摘Since most organic materials are very sensitive to moisture and oxygen, organic light emitting diodes (OLEDs) require an encapsulation layer to protect the active layer from these gases. Since light, flexible and portable OLEDs are being employed in more diverse climates and environmental conditions, the OLED encapsulation layer must retain robust mechanical properties and stability in high temperature/high humidity conditions. Al2O3 films have demonstrated excellent barrier performance, but they readily hydrolyze when exposed to prolonged harsh environments. In this study, we fabricated a thin film encapsulation (TFE) film that was resistant to hydrolysis, using Al2O3/MgO (AM) nanolaminates. MgO has superior resistance to harsh environments, and the aluminate phase generated by the chemical reaction of Al2O3 and MgO provided excellent barrier performance, even after storage in harsh conditions. A multi-barrier fabricated using the AM nanolaminate showed excellent barrier performance, close to the level required by OLEDs. It did not significantly deteriorate even after a bending test of 1,000 iterations at 0.63% strain. After 1,000 cycle of bending, the electrical properties of the passivated OLEDs were not significantly degraded at shelf-lifetime test where the fabricated device was stored for 50 days in a harsh environment of 60℃, 90% relative humidity. 7Tenofovir is a more suitable treatment than entecavir for chronic hepatitis B patients carrying naturally occurring rtM204I mutations显示文摘BACKGROUND Hepatitis B virus(HBV) DNA polymerase mutations usually occur to long term use of nucleos(t)ide analogues(NAs), but they can occur spontaneously in treatment-na?ve chronic hepatitis B(CHB) patients. The naturally occurring HBV DNA polymerase mutations might complicate antiviral therapy with NAs,leading to the generation of drug-resistant viral mutants and disease progression.The most common substitutions are known to be YMDD-motif mutations, but their prevalence and the influence on antiviral therapy is unclear.AIM To investigate prevalence of the naturally occurring rtM204I mutations in treatment-na?ve CHB genotype C2 patients and their influence on antiviral therapy.METHODS A total of 410 treatment-na?ve CHB patients infected with HBV genotype C2 strains were enrolled in this retrospective study. Among the 410 patients, 232 were treated with NAs for at least 12 mo. Significant fibrosis was defined as fibrosis-4 index > 3.25 or aspartate aminotransferase to platelet ratio index > 1.5.Complete viral response(CVR) during NAs was defined as undetectable serum HBV DNA(< 24 IU/m L). The rtM204I variants were analyzed by a newly developed locked nucleotide probe(LNA probe) based real-time PCR(LNA-RTPCR) method.RESULTS The LNA-RT-PCR could discriminate rtM204I mutant-type(17 patients, 4.2%)from rt M204 wild-type(386 patients, 95.8%) in 403 of 410 patients(98.3%sensitivity). Multivariate analysis showed that naturally occurring rtM204I variants were more frequently detected in patients with significant fibrosis [oddratio(OR) 3.397, 95% confidence-interval(CI) 1.119-10.319, P = 0.031]. Of 232 patients receiving NAs, multivariate analysis revealed that achievement of CVR was reversely associated with naturally occurring rtM204I variants prior to NAs treatment(OR 0.014, 95%CI 0.002-0.096, P < 0.001). Almost patients receiving tenofovir achieved CVR at 12 mo of tenofovir, irrespective of pre-existence of naturally occurring rtM204I mutations(CVR rates: patients with rtM204I, 100%;patients without rtM204I, 96.6%), whereas, pre-existence of naturally-occurring rtM204I-mutations prior to NAs significantly affects CVR rates in patients receiving entecavir(at 12 mo: Patients with rtM204I, 16.7%; patients without rtM204I, 95.6%, P < 0.001).CONCLUSION The newly developed LNA-RT-PCR method could detect naturally occurring rtM204I mutations with high-sensitivity. Theses mutations were more frequent in patients with liver fibrosis. 8Improvement of diabetes and hypertension after gastrectomy: A nationwide cohort study显示文摘AIM: To evaluate the effect of gastrectomy on diabetes mellitus(DM) and hypertension(HTN) in non-obese gastric cancer patients. METHODS: A total of 100000 patients, diagnosed with either type 2 DM or HTN, were randomly selected from the 2004 Korean National Health Insurance System claims. Among them, 360 diabetes and 351 hypertensive patients with gastric cancer who had been regularly treated without chemotherapy from January 2005 to December 2010 were selected. They were divided into three groups according to their treatment methods: total gastrectomy(TG), subtotal gastrectomy(STG) and endoscopic resection(ER). RESULTS: The drug discontinuation rate of antidiabetic and anti-hypertensive agents after gastric cancer treatment was 9.7% and 11.1% respectively. DM appeared to be improved more frequently(22.8%) and earlier(mean ± SE 28.6 ± 1.8 mo) in TG group than in the two other groups [improved in 9.5% of ER group(37.4 ± 1.1 mo) and 6.4% of STG group(47.0 ± 0.8 mo)]. The proportion of patients treated with multiple drugs decreased more notably in TG group compared to others(P = 0.001 in DM, and P = 0.035 in HTN). 9Identification of TRPM7 channels in human intestinal interstitial cells of Cajal显示文摘AIM:To investigate the characteristics of slow elec-trical waves and the presence of transient receptor potential melastatin-type 7(TRPM7)in the human gas-trointestinal(GI)tract.METHODS:Conventional microelectrode techniques were used to record intracellular electrical responses from human GI smooth muscle tissue.Immunohisto-chemistry was used to identify TRPM7 channels in in-terstitial cells of Cajal(ICCs).RESULTS:The human GI tract generated slow electri-cal waves and had ICCs which functioned as pacemak-er cells.Flufenamic acid,a nonselective cation channel blocker,and 2-APB(2-aminoethoxydiphenyl borate)and La3+,TRPM7 channel blockers,inhibited the slowwaves.Also,TRPM7 channels were expressed in ICCs in human tissue.CONCLUSION:These results suggest that the human GI tract generates slow waves and that TRPM7 chan-nels expressed in the ICCs may be involved in the gen-eration of the slow waves.Byung Joo Kim Kyu Joo Park Hyung Woo Kim Seok Choi Jae Yeoul Jun In Youb Chang Ju-Hong Jeon Insuk So Seon Jeong Kim 2009World Journal of Gastroenterology2009,15,46:3

12Shape Estimation of Soft Manipulator Using Stretchable Sensor显示文摘The soft robot manipulator is attracting attention in the surgical fields with its intrinsic softness,lightness in its weight,and safety toward the human organ.However,it cannot be used widely because of its difficulty of control.To control a soft robot manipulator accurately,shape sensing is essential.This paper presents a method of estimating the shape of a soft robot manipulator by using a skin-type stretchable sensor composed of a multiwalled carbon nanotube(MWCNT)and silicone(p7670).The sensor can be easily fabricated and applied by simply attaching it to the surface of the soft manipulator.In its fabrication,MWCNT is sprayed on a teflon sheet,and liquid-state silicone is poured on it.After curing,we turn it over and cover it with another silicone layer.The sensor is fabricated with a sandwich structure to decrease the hysteresis of the sensor.After calibration and determining the relationship between the resistance of the sensor and the strain,three sensors are attached at 120°intervals.Using the obtained data,the curvature of the manipulator is calculated,and the entire shape is reconstructed.To validate its accuracy,the estimated shape is compared with the camera data.We experiment with three,six,and nine sensors attached,and the result of the error of shape estimation is compared.As a result,the minimum tip position error is approximately 8.9 mm,which corresponded to 4.45%of the total length of the manipulator when using nine sensors.Jinho So Uikyum Kim Yong Bum Kim Dong-Yeop Seok Sang Yul Yang Kihyeon Kim Jae Hyeong Park Seong Tak Hwang Young Jin Gong Hyouk Ryeol Choi 2021Cyborg and Bionic Systems2021,,1:2

15Foldable and washable textile-based OLEDs with a multi-functional near-room-temperature encapsulation layer for smart e-textiles显示文摘Wearable electronic devices are being developed because of their wide potential applications and user convenience.Among them,wearable organic light emitting diodes(OLEDs)play an important role in visualizing the data signal processed in wearable electronics to humans.In this study,textile-based OLEDs were fabricated and their practical utility was demonstrated.The textilebased OLEDs exhibited a stable operating lifetime under ambient conditions,enough mechanical durability to endure the deformation by the movement of humans,and washability for maintaining its optoelectronic properties even in water condition such as rain,sweat,or washing.In this study,the main technology used to realize this textile-based OLED was multi-functional nearroom-temperature encapsulation.The outstanding impermeability of TiO_(2)film deposited at near-room-temperature was demonstrated.The internal residual stress in the encapsulation layer was controlled,and the device was capped by highly crosslinked hydrophobic polymer film,providing a highly impermeable,mechanically flexible,and waterproof encapsulation.So Yeong Jeong Hye Rin Shim Yunha Na Ki Suk Kang Yongmin Jeon Seungyeop Choi Eun Gyo Jeong Yong Cheon Park Ha-Eun Cho Junwoo Lee Jeong Hyun Kwon Sung Gap Im Kyung Cheol Choi 2021npj Flexible Electronics2021,5,1:2
