1Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. 3Hydrothermal synthesis of calcium sulfate whisker from flue gas desulfurization gypsum显示文摘Plenty of flue gas desulfurization(FGD) gypsum generated from coal-fired power plants for sulfur dioxide sequestration caused many environmental issues. Preparing calcium sulfate whisker(CSW) from FGD gypsum by hydrothermal synthesis is considered to be a promising approach to solve this troublesome problem and utilize calcium sulfate in a high-value-added way. The effects of particle size of FGD gypsum, slurry concentration,and additives on CSW were investigated in this work. The results indicated that fine particle size of FGD gypsum and moderately high slurry concentration were beneficial for crystal nucleation and growth. Three additives of magnesium chloride, citric acid, and sodium dodecyl benzene sulfonate(SDBS) were employed in this study. It was found that mean length and aspect ratio of CSW were both decreased by the usage of magnesium chloride,while a small quantity of citric acid or SDBS could improve the CSW morphology. When multi-additives of citric acid-SDBS were employed, the mean length and aspect ratio increased more than 20%. 5β-MnO_(2) with proton conversion mechanism in rechargeable zinc ion battery显示文摘Rechargeable aqueous zinc ion battery(RAZIB)is a promising energy storage system due to its high safety,and high capacity.Among them,manganese oxides with low cost and low toxicity have drawn much attention.However,the under-debate proton reaction mechanism and unsatisfactory electrochemical performance limit their applications.Nanorod b-MnO_(2) synthesized by hydrothermal method is used to investigate the reaction mechanism.As cathode materials for RAZIB,the Zn//b-MnO_(2) delivers 355 mA h g^(-1)(based on cathode mass)at0.1 A g^(-1),and retain 110 mA h g^(-1) after 1000 cycles at 0.2 A g^(-1).Different from conventional zinc ion insertion/extraction mechanism,the proton conversion and Mn ion dissolution/deposition mechanism of b-MnO_(2) is proposed by analyzing the evolution of phase,structure,morphology,and element of b-MnO_(2) electrode,the pH change of electrolyte and the determination of intermediate phase MnO OH.Zinc ion,as a kind of Lewis acid,also provides protons through the formation of ZHS in the proton reaction process.This study of reaction mechanism provides a new perspective for the development of Zn//MnO_(2) battery chemistry.Wenbao Liu Xiaoyu Zhang Yongfeng Huang Baozheng Jiang Ziwen Chang Chengjun Xu Feiyu Kang 2021Journal of Energy Chemistry2021,30,5:6
6Protective effect of oxysophoridine on cerebral ischemia/reperfusion injury in mice显示文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase. Protein and mRNA expression levels of N-methyl-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. 7High-Performance Aqueous Zinc-Ion Batteries Realized by MOF Materials显示文摘Rechargeable aqueous zinc-ion batteries(ZIB s) have been gaining increasing interest for large-scale energy storage applications due to their high safety,good rate capability,and low cost.However,the further development of ZIB s is impeded by two main challenges:Currently reported cathode materials usually suffer from rapid capacity fading or high toxicity,and meanwhile,unstable zinc stripping/plating on Zn anode seriously shortens the cycling life of ZIBs.In this paper,metal-organic framework(MOF) materials are proposed to simultaneously address these issues and realize high-performance ZIB s with Mn(BTC) MOF cathodes and ZIF-8-coated Zn(ZIF-8@Zn) anodes.Various MOF materials were synthesized,and Mn(BTC) MOF was found to exhibit the best Zn^2+-storage ability with a capacity of 112 mAh g^-1.Zn^2+ storage mechanism of the Mn(BTC) was carefully studied.Besides,ZIF-8@Zn anodes were prepared by coating ZIF-8 MOF material on Zn foils.Unique porous structure of the ZIF-8 coating guided uniform Zn stripping/plating on the surface of Zn anodes.As a result,the ZIF-8@Zn anodes exhibited stable Zn stripping/plating behaviors,with 8 times longer cycle life than bare Zn foils.Based on the above,high-performance aqueous ZIBs were constructed using the Mn(BTC) cathodes and the ZIF-8@Zn anodes,which displayed an excellent long-cycling stability without obvious capacity fading after 900 charge/discharge cycles.This work provides a new opportunity for high-performance energy storage system.Xuechao Pu Baozheng Jiang Xianli Wang Wenbao Liu Liubing Dong Feiyu Kang Chengjun Xu 2020Nano-Micro Letters2020,12,11:5

10Influenza A virus use of BinCARD1 to facilitate the binding of viral NP to importinα7 is counteracted by TBK1-p62 axis-mediated autophagy显示文摘As a major component of the viral ribonucleoprotein(vRNP)complex in influenza A virus(IAV),nucleoprotein(NP)interacts with isoforms of importinαfamily members,leading to the import of itself and vRNP complex into the nucleus,a process pivotal in the replication cycle of IAV.In this study,we found that BinCARD1,an isoform of Bcl10-interacting protein with CARD(BinCARD),was leveraged by IAV for efficient viral replication.BinCARD1 promoted the nuclear import of the vRNP complex and newly synthesized NP and thus enhanced vRNP complex activity.Moreover,we found that BinCARD1 interacted with NP to promote NP binding to importinα7,an adaptor in the host nuclear import pathway.However,we also found that BinCARD1 promoted RIG-I-mediated innate immune signaling by mediating Lys63-linked polyubiquitination of TRAF3,and that TBK1 appeared to degrade BinCARD1.We showed that BinCARD1 was polyubiquitinated at residue K103 through a Lys63 linkage,which was recognized by the TBK1-p62 axis for autophagic degradation.Overall,our data demonstrate that IAV leverages BinCARD1 as an important host factor that promotes viral replication,and two mechanisms in the host defense system are triggered—innate immune signaling and autophagic degradation—to mitigate the promoting effect of BinCARD1 on the life cycle of IAV.Xuyuan Wang Li Jiang Guangwen Wang Wenjun Shi Yuzhen Hu Bo Wang Xianying Zeng Guobin Tian Guohua Deng Jianzhong Shi Liling Liu Chengjun Li Hualan Chen 2022Cellular & Molecular Immunology2022,19,10:3
11Self-reducing bifunctional Ni-W/SBA-15 catalyst for cellulose hydrogenolysis to low carbon polyols显示文摘A series of self-reducing bifunctional Ni-W/SBA-15 catalysts were synthesized using biomass-based carbon source as the reducing agent without conventional further reduction step. The self-reducing catalysts were performed on the hydrogenolysis of cellulose to low carbon polyols. The effects of calcination temperature and metallic loading contents for cellulose hydrogenolysis reaction were investigated detailedly.The optimal calcination temperature was found to be 673 K by TG analysis. The active metal nanoparticles with a better dispersion were observed using SEM and element mapping technology. 12Influence of Surrounding Structures upon the Aerodynamic and Acoustic Performance of the Outdoor Unit of a Split Air-Conditioner显示文摘DC-inverter split air-conditioner is widely used in Chinese homes as a result of its high-efficiency and energy-saving. Recently, the researches on its outdoor unit have focused on the influence of surrounding structures upon the aerodynamic and acoustic performance, however they are only limited to the influence of a few parameters on the performance, and practical design of the unit requires more detailed parametric analysis. Three-dimensional computational fluid dynamics(CFD) and computational aerodynamic acoustics(CAA) simulation based on FLUENT solver is used to study the influence of surrounding structures upon the aforementioned properties of the unit. The flow rate and sound pressure level are predicted for different rotating speed, and agree well with the experimental results. The parametric influence of three main surrounding structures(i.e. the heat sink, the bell-mouth type shroud and the outlet grille) upon the aerodynamic performance of the unit is analyzed thoroughly. The results demonstrate that the tip vortex plays a major role in the flow fields near the blade tip and has a great effect on the flow field of the unit. The inlet ring's size and throat's depth of the bell-mouth type shroud, and the through-flow area and configuration of upwind and downwind sections of the outlet grille are the most important factors that affect the aerodynamic performance of the unit. Furthermore, two improved schemes against the existing prototype of the unit are developed, which both can significantly increase the flow rate more than 6 %(i.e. 100 m3·h^(-1)) at given rotating speeds. The inevitable increase of flow noise level when flow rate is increased and the advantage of keeping a lower rotating speed are also discussed. 13H7N9 virus infection triggers lethal cytokine storm by activating gasdermin E-mediated pyroptosis of lung alveolar epithelial cells显示文摘The H7N9 influenza virus emerged in China in 2013,causing more than 1560 human infections,39% of which were fatal.A ‘cytokine storm’ in the lungs of H7N9 patients has been linked to a poor prognosis and death;however,the underlying mechanism that triggers the cytokine storm is unknown.Here,we found that efficient replication of the H7N9 virus in mouse lungs activates gasdermin E(GSDME)-mediated pyroptosis in alveolar epithelial cells,and that the released cytosolic contents then trigger a cytokine storm.Knockout of Gsdme switched the manner of death of A549 and human primary alveolar epithelial cells from pyroptosis to apoptosis upon H7N9 virus infection,and Gsdme knockout mice survived H7N9 virus lethal infection.Our findings reveal that GSDME activation is a key and unique mechanism for the pulmonary cytokine storm and lethal outcome of H7N9 virus infection and thus opens a new door for the development of antivirals against the H7N9 virus.Xiaopeng Wan Jiqing Li Yupeng Wang Xiaofei Yu Xijun He Jianzhong Shi Guohua Deng Xianying Zeng Guobin Tian Yanbing Li Yongping Jiang Yuntao Guan Chengjun Li Feng Shao Hualan Chen 2022National Science Review2022,9,1:3
14Geochemical characteristics of hadal sediment in the northern Yap Trench显示文摘Two sediment cores were retrieved in the hadal zone of the Yap Trench,and their concentrations of six major elements Mg,Al,Ca,Ti,Mn and Fe and nine trace elements Sr,Ba,Pb,V,Cr,Co,Ni,Cu and Zn were determined in inductively coupled plasma atomic emission spectrometry(ICP-AES).According to the vertical distribution profiles of the 15 elements,the correlation of their concentrations,the ratios of Ni/Co,V/Cr,Fe/Al,and Ti/Al,and morphological characteristics of the sediment samples,the implications of the depositional environment and the sediment provenance were analyzed.The results show that the ratio of Ni/Co in all depths of the two sediment cores were below 5,and the ratio of V/Cr were lower than 2,indicating that the depositional environment of the hadal zone in the trench was oxidative and might have inflow of the Antarctic bottom oxygen-rich water.The sediment samples on the eastern side of the trench were siliceous mud mainly composed of diatoms,radiolarian,and sponge needles from surface to deep layer.The vertical profile of the elements,the concentration of TOC and the fossil record indicated that the sediment sample from station Dive113 was well mixed from surface to bottom layer.Based on the correlation of concentrations of the elements,the morphological characteristics of the sediment,and the ratios of Fe/Al and Ti/Al,we inferred that the sediment in the hadal zone of the trench had terrestrial,volcanic,biological,and authigenic sources.Major source of the sediment in the eastern side of the trench were terrestrial;whereas the sediment in the western side of the trench received more volcaniclastic input.HUANG Yuhuan SUN Chengjun YANG Guipeng YUE Xin’an JIANG Fenghua CAO Wei YIN Xiaofei GUO Chaonan NIU Jiaohong DING Haibing 2020Journal of Oceanology and Limnology2020,38,3:3
15Distribution characteristics of lipids in hadal sediment in the Yap Trench显示文摘In this study,cored sediment samples collected by the Jiaolong Submersible at 6779 m depth from the hadal zone of the Yap Trench in May 2016 were sliced in 1-cm interval from top to bottom,and lipids in each sediment layer were quantitatively and qualitatively analyzed.The vertical distribution profiles of the lipids in the sediment sample,their main existing forms,and their possible sources were investigated.The results show that the concentration of lipid in the surface sediment was the highest with the carbon number from 12 to 27,dominated by medium and short-chain lipids.The total concentration of fatty acids in surface sediment was much higher than those in the offshore and deep-sea areas,being up to 325.77μg/g due to the funnel effect caused by the“V”terrain of the trench.Fatty acid 18:0 was the most abundant lipids in the sediment sample.Abnormal high concentrations of fatty acid 18:1ω7 and alkanes indicated the existence of hydrothermal fluids in the study area.In addition,saturated fatty acids and monounsaturated fatty acids existed mainly in free form,and polyunsaturated fatty acids existed mainly in bound form.Most of the alkanes were in bound form,and their major source was autochthounous input.The carbon number of alcohols in the sediment sample ranged from 12 to 20,mainly existed in bound form.The source of fatty acids was mainly autochthonous input,and the neutral lipids had both marine and terrestrial origin.This is the first study of lipids in hadal sediment of the Yap Trench.The results will promote deeper understanding of organic carbon cycle in marine environment.YAN Yixin SUN Chengjun HUANG Yuhuan CAO Wei JIANG Fenghua YANG Guipeng DING Haibing 2020Journal of Oceanology and Limnology2020,38,3:2
16The study of the adductor muscle-shell interface structure in three Mollusc species显示文摘The adductor muscle scar(AMS) is the fixation point of adductor muscle to the shell. It is an important organicinorganic interface and stress distribution area. Despite recent advances, our understanding of the structure and composition of the AMS remain limited. Here, we report study on the AMS of three bivalves: Mytilus coruscus,Chlamys farreri and Ruditapes philippinarum. Results showed that there were significant differences among their AMS structures. Both M. coruscus and C. farreri were found to have a columnar layer above the nacreous platelet shell structure at the AMS and this layer was more organized in M. coruscus. There was no distinguishable twolayer structure in R. philippinarum. Atomic force microscopy(AFM) and Fourier transform infrared spectroscopy(FT-IR) results showed that the AMS was much smoother than the nacreous inner shell in all the three species and the AMS had minor different compositions from the nacreous shell layer. SDS-PAGE(sodium dodecyl-sulfate polyacrylamide gel electophoresis) study of the proteins isolated from the interface indicated that there was a 70 k Da protein which seemed to be specifically located to the highly organized columnar AMS structure in Mytilus coruscus.
