2Injectable kartogenin and apocynin loaded micelle enhances the alleviation of intervertebral disc degeneration by adipose-derived stem cell显示文摘Cell transplantation has been proved the promising therapeutic effects on intervertebral disc degeneration(IVDD).However,the increased levels of reactive oxygen species(ROS)in the degenerated region will impede the efficiency of human adipose-derived stem cells(human ADSCs)transplantation therapy.It inhibits human ADSCs proliferation,and increases human ADSCs apoptosis.Herein,we firstly devised a novel amphiphilic copolymer PEG-PAPO,which could self-assemble into a nanosized micelle and load lipophilic kartogenin(KGN),as a single complex(PAKM).It was an injectable esterase-responsive micelle,and showed controlled release ability of KGN and apocynin(APO).Oxidative stimulation promoted the esterase activity in human ADSCs,which accelerate degradation of esterase-responsive micelle.Compared its monomer,the PAKM micelle possessed better bioactivities,which were attributed to their synergistic effect.It enhanced the viability,autophagic activation(P62,LC3 II),ECM-related transcription factor(SOX9),and ECM(Collagen II,Aggrecan)maintenance in human ADSCs.Furthermore,it is demonstrated that the injection of PAKM with human ADSCs yielded higher disc height and water content in rats.Therefore,PAKM micelles perform promoting cell survival and differentiation effects,and may be a potential therapeutic agent for IVDD.Chao Yu Dongdong Li Chenggui Wang Kaishun Xia Jingkai Wang Xiaopeng Zhou Liwei Ying Jiawei Shu Xianpeng Huang Haibin Xu Bin Han Qixin Chen Fangcai Li Jianbin Tang Chengzhen Liang Nigel Slater 2021Bioactive Materials2021,6,10:3
3Generation of transgenic wheat resistant to wheat yellow mosaic virus and identification of gene silence induced by virus infection显示文摘The plasmid containing the promoter Actl, the coat protein (cp) gene of wheat yellow mosaic virus (WYMV) and the selectable bar gene, was delivered via particle bombardment, directly into immature embryos of a wheat culti-vars. PCR and PCR-RFLP were employed to screen the existence of the cp gene in T0 and T1 generations. Seeds from the positive T1 plants were sowed in fields heavily contaminated with WYMV to detect their resistance. In field trial of virus infection, one of the transgenic wheat lines, P8-T2, exhibited highly disease-resistance. Western blot and RT-PCR analysis showed that the expression level of cp gene in the resistant transgenic line was reduced greatly compared to those susceptible to WYMV infection. 5Effects on the local symptoms of subgenomic RNAs expressions and their translational products of Tobacco necrosis virus A Chinese isolate显示文摘Based on a full-length infectious cDNA clone, gene modifications of Tobacco necrosis virus A Chinese isolate (TNV-AC) were made by site-directed mutagenesis or nucleotide deletions for in vitro transcrip-tion of mutant viral RNAs. Mechanical inoculations of Chenopodium amaranticolor with in vitro tran-scripts, containing a single nucleic acid substitution at the presumed transcriptional start sites for the two subgenomic (sg) RNAs, showed that the sgRNA1 and sgRNA2 of TNV-AC were initiated at G2184 or G2460, respectively. Mutagenesis of the translational initiation-codons for the open-reading frame (ORF) P8 or P6 encoded by sgRNA1 indicated that each of the two genes was essential for formation of local lesions on C. amaranticolor leaves, perhaps by blocking virus cell-to-cell movement, but were not necessary for viral RNA replication in the protoplast of tobacco cell BY-2. Results of prokaryotic expression showed that the ORF coding for coat protein on TNV-AC sgRNA2 was initiatively translated by the first AUG codon at nucleotides 2612―2614. Site-directed mutation of translational start codons, and deletion of the entire coding region, showed that the intact TNV-AC coat protein was dispensable for establishment of TNV-AC infection in C. amaranticolor, otherwise the numbers of local lesions and the viral RNA accumulation level were reduced, or the time to symptom appearance significantly delayed. These results suggested that the nucleotide sequence around the translational start codon coding for TNV-AC coat protein gene may play an important role in the local symptoms. 9Three-dimensional observation of the phase structure of high density polyethylene (HDPE)/poly(ethylene-co-butene) (PEB) blend by laser scanning confocal microscopy显示文摘In this paper,high density polyethylene (HDPE)/poly(ethylene-co-butene) (PEB) blend (50/50 wt%) was prepared through solution blending and then compression molding,and subsequently examined by laser scanning confocal microscopy (LSCM). The PEB used in this experiment was labeled with a small quantity of a fluorescein derivative to render fluorescence. The initial films showed uniform dye dis-tribution and no indication of phase separation within the resolution of optical microscopy. Sample films annealing at 140℃ followed by rapid cooling to room temperature showed obvious phase sepa-ration and bicontinuous structure. 10Functional analysis of beet necrotic yellow vein virus (BNYVV) RNA4 in fungal transmission显示文摘Full-length RNA4 of beet necrotic yellow vein virus (BNYVV) and its mutants, including a frame-shift and deletions within the coding region, were inserted into a transcription plasmid. Transcripts derived from these plasmids and RNAs extracted from a BNYVV isolate containing only RNAs 1, 2 and 3 were coinoculated to Tetragona expansa. Then these recombinant isolates were inoculated to sugar beet plants grown in a sand culture system containing Po-lymyxa betae that had been freed of virus particles to test efficiency of BNYVV transmission. The result showed that the efficiency of BNYVV transmission among the plants by Polymyxa betae was highly decreased by different deletions in the coding region of RNA4, but not affected by the frame-shift mutant with four nucleotides insertion. 11Effect of the RNA5 component on pathogenicity of beet necrotic yellow vein virus显示文摘The transcripts of beet necrotic yellow virus RNA5 were prepared from the cloned cDNA in a bacterio-phage T7 in vitro run-off transcription system. The RNA5 transcripts and RNAs extracted from two BNYVV mutants, Hu0 containing RNA1, 2 and Hu3 containing RNA1, 2, 3 respectively, were co-inoculated to Tetragonia expansa and sugar beet seedlings. The results indicated that the virus infections and accumulation in the hosts could be improved by presence of RNA5. Cooperating with RNA3, the RNA5 seems to cause more severe symptom and yield losses in sugar beet.
