1Seismic performance of recycled concrete-filled square steel tube columns显示文摘An experimental study on the seismic performance of recycled concrete-filled square steel tube(RCFST) columns is carried out.Six specimens were designed and tested under constant axial compression and cyclic lateral loading.Two parameters, replacement percentage of recycled coarse aggregate(RCA) and axial compression level, were considered in the test.Based on the experimental data, the hysteretic loops, skeleton curves, ductility, energy dissipation capacity and stiffness degradation of RCFST columns were analyzed.The test results indicate that the failure modes of RCFST columns are the local buckling of the steel tube at the bottom of the columns, and the hysteretic loops are full and their shapes are similar to normal CFST columns.Furthermore, the ductility coefficient of all specimens are close to 3.0, and the equivalent viscous damping coefficient corresponding to the ultimate lateral load ranges from 0.323 to 0.360, which demonstrates that RCFST columns exhibit remarkable seismic performance.Chen Zongping Jing Chenggui Xu Jinjun Zhang Xianggang 2017Earthquake Engineering and Engineering Vibration2017,16,1:8
2Bounding surface plasticity model for stress-strain and grain-crushing behaviors of rockfill materials显示文摘Crushing of grains can greatly influence the strength,dilatancy,and stress-strain relationship of rockfill materials.The critical state line(CSL)in the void ratio versus mean effective stress plane was extended to the breakage critical state plane(BCSP).A state void-ratio-pressure index that incorporated the effect of grain crushing was proposed according to the BCSP.Rowe’s stress-dilatancy equation was modified by adding the breakage voidratio-pressure index,which was also incorporated into the formulations of the bounding stress ratio and plastic modulus.A BCSP-based bounding surface plasticity model was proposed to describe the state-dependent stressstrain behaviors and the evolution of grain crushing during shearing process of rockfill materials,and was shown to sufficiently capture the breakage phenomenon.Yang Xiao Zengchun Sun Armin WStuedlein Chenggui Wang Zhijun Wu Zhichao Zhang 2020Geoscience Frontiers2020,11,2:5

4Generation of transgenic wheat resistant to wheat yellow mosaic virus and identification of gene silence induced by virus infection显示文摘The plasmid containing the promoter Actl, the coat protein (cp) gene of wheat yellow mosaic virus (WYMV) and the selectable bar gene, was delivered via particle bombardment, directly into immature embryos of a wheat culti-vars. PCR and PCR-RFLP were employed to screen the existence of the cp gene in T0 and T1 generations. Seeds from the positive T1 plants were sowed in fields heavily contaminated with WYMV to detect their resistance. In field trial of virus infection, one of the transgenic wheat lines, P8-T2, exhibited highly disease-resistance. Western blot and RT-PCR analysis showed that the expression level of cp gene in the resistant transgenic line was reduced greatly compared to those susceptible to WYMV infection. 9Exploration on Separation of Periplaneta Americana Small Peptides with Tricine-SDS-PAGE System显示文摘[Objective] This study aimed to explore a Tricine-SDS-PAGE method for separation and purification of Periplaneta Americana small peptides. [Method] By comparing the separation effects of three types of gels with different ratios on P. Americana small peptides, an appropriate Tricine-SDS-PAGE method for separation of small peptides was established. [Result] Standard low molecular weight Marker was preliminarily separated to a certain extent with polyacrylamide gel electrophoresis, while the sample mixture was not separated due to the different levels of aggregation in separating gel, spacer gel and stacking gel. 14Three-dimensional observation of the phase structure of high density polyethylene (HDPE)/poly(ethylene-co-butene) (PEB) blend by laser scanning confocal microscopy显示文摘In this paper,high density polyethylene (HDPE)/poly(ethylene-co-butene) (PEB) blend (50/50 wt%) was prepared through solution blending and then compression molding,and subsequently examined by laser scanning confocal microscopy (LSCM). The PEB used in this experiment was labeled with a small quantity of a fluorescein derivative to render fluorescence. The initial films showed uniform dye dis-tribution and no indication of phase separation within the resolution of optical microscopy. Sample films annealing at 140℃ followed by rapid cooling to room temperature showed obvious phase sepa-ration and bicontinuous structure. 15A conductive supramolecular hydrogel creates ideal endogenous niches to promote spinal cord injury repair显示文摘The current effective method for treatment of spinal cord injury(SCI)is to reconstruct the biological microenvironment by filling the injured cavity area and increasing neuronal differentiation of neural stem cells(NSCs)to repair SCI.However,the method is characterized by several challenges including irregular wounds,and mechanical and electrical mismatch of the material-tissue interface.In the current study,a unique and facile agarose/gelatin/polypyrrole(Aga/Gel/PPy,AGP3)hydrogel with similar conductivity and modulus as the spinal cord was developed by altering the concentration of Aga and PPy.The gelation occurred through non-covalent interactions,and the physically crosslinked features made the AGP3 hydrogels injectable.In vitro cultures showed that AGP3 hydrogel exhibited excellent biocompatibility,and promoted differentiation of NSCs toward neurons whereas it inhibited over-proliferation of astrocytes.The in vivo implanted AGP3 hydrogel completely covered the tissue defects and reduced injured cavity areas.In vivo studies further showed that the AGP3 hydrogel provided a biocompatible microenvironment for promoting endogenous neurogenesis rather than glial fibrosis formation,resulting in significant functional recovery.RNA sequencing analysis further indicated that AGP3 hydrogel significantly modulated expression of neurogenesis-related genes through intracellular Ca2+signaling cascades.Overall,this supramolecular strategy produces AGP3 hydrogel that can be used as favorable biomaterials for SCI repair by filling the cavity and imitating the physiological properties of the spinal cord.Biao Yang Chengzhen Liang Di Chen Feng Cheng Yuang Zhang Shaoke Wang Jiawei Shu Xianpeng Huang Jingkai Wang Kaishun Xia Liwei Ying Kesi Shi Chenggui Wang Xuhua Wang Fangcai Li Qian Zhao Qixin Chen 2022Bioactive Materials2022,7,9:0
16A Comparative Study on Four Staining Methods for Antitumor Active Fraction of Periplaneta americana显示文摘[Objective] This study aimed to compare four staining methods for proteins of SDS-polyacrylamide gel electrophoresis(SDS-PAGE) and explore a suitable staining method for the antitumor active fraction of P.americana after polyacrylamide gel electrophoresis.[Method] BSA was used as the standard for the comparison of Coomassie brilliant blue staining method,potassium staining method,calcium staining method and silver staining method,on the basis,antitumor active fraction samples of P.americana were used for SDS-PAGE electrophoresis and staining.[Result] The results showed that silver staining method could be accurately,quickly and easily used for SDS-PAGE staining of the antitumor active fraction of P.americana.[Conclusion] This study laid the foundation for exploring the medicinal value of P.americana.Shaohui WU Chenggui ZHANG Xiumei WU Guangming LIU 2012Agricultural Biotechnology2012,1,1:0
17Caspase-1 and Gasdermin D Afford the Optimal Targets with Distinct Switching Strategies in NLRP1b Inflammasome-Induced Cell Death显示文摘Inflammasomes are essential complexes of innate immune system, which form the first line of host defense against pathogens. Mounting evidence accumulates that inflammasome signaling is highly correlated with coronavirus disease 2019 (COVID-19). However, there remains a significant gap in our understanding of the regulatory mechanism of inflammasome signaling. Combining mathematical modeling with experimental analysis of NLRP1b inflammasome signaling, we found that only the expression levels of caspase-1 and GSDMD have the potential to individually switch cell death modes. Reduction of caspase-1 or GSDMD switches cell death from pyroptosis to apoptosis. Caspase-1 and GSDMD present different thresholds and exert distinct pathway choices in switching death modes. Pyroptosis switches to apoptosis with an extremely low threshold level of caspase-1, but with a high threshold of GSDMD. Caspase-1-impaired cells employ ASC-caspase-8-dependent pathway for apoptosis, while GSDMD-impaired cells primarily utilize caspase-1-dependent pathway. Additionally, caspase-1 and GSDMD can severally ignite the cooccurrence of pyroptosis and apoptosis. Landscape topography unravels that the cooccurrence is dramatically different in caspase-1- and GSDMD-impaired cells. Besides pyroptosis state and apoptosis state, a potential new “coexisting” state in single cells is proposed when GSDMD acts as the driving force of the landscape. The “seesaw model” is therefore proposed, which can well describe the death states that are controlled by caspase-1 or GSDMD in single cells.
