1Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 2Metformin inhibits pancreatic cancer metastasis caused by SMAD4 deficiency and consequent HNF4G upregulation显示文摘Pancreatic ductal adenocarcinoma(PDAC)has poor prognosis due to limited therapeutic options.This study examines the roles of genome-wide association study identified PDAC-associated genes as therapeutic targets.We have identified HNF4G gene whose silencing most effectively repressed PDAC cell invasiveness.HNF4G overexpression is induced by the deficiency of transcriptional factor and tumor suppressor SMAD4.Increased HNF4G are correlated with SMAD4 deficiency in PDAC tumor samples and associated with metastasis and poor survival time in xenograft animal model and in patients with PDAC(log-rank P=0.036;HR=1.60,95%CI=1.03–2.47).We have found that Metformin suppresses HNF4G activity via AMPK-mediated phosphorylation-coupled ubiquitination degradation and inhibits in vitro invasion and in vivo metastasis of PDAC cells with SMAD4 deficiency.Furthermore,Metformin treatment significantly improve clinical outcomes and survival in patients with SMAD4-deficient PDAC(log-rank P=0.022;HR=0.31,95%CI=0.14–0.68)but not in patients with SMAD4-normal PDAC.Pathway analysis shows that HNF4G may act in PDAC through the cell-cell junction pathway.These results indicate that SMAD4 deficiency-induced overexpression of HNF4G plays a critical oncogenic role in PDAC progression and metastasis but may form a druggable target for Metformin treatment.Chengcheng Wang Taiping Zhang Quan Liao Menghua Dai Junchao Guo Xinyu Yang Wen Tan Dongxin Lin Chen Wu Yupei Zhao 2021Protein & Cell2021,12,2:5
3Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 4Identification of powdery mildew resistance loci in wheat by integrating genome-wide association study(GWAS) and linkage mapping显示文摘Wheat powdery mildew(Blumeria graminis f.sp.tritici, Bgt) is a disease of increasing importance globally due to the adoption of high yielding varieties and modern sustainable farming technologies.Growing resistant cultivars is a preferred approach to managing this disease, and novel powdery mildew resistance genes are urgently needed for new cultivar development.A genome-wide association study was performed on a panel of 1292 wheat landraces and historical cultivars using 5011 single nucleotide polymorphism(SNP)markers.The association panel was evaluated for reactions to three Bgt inoculants, OKS(14)-B-3-1, OKS(14)-C-2-1, and Bgt15.Linkage disequilibrum(LD) analysis indicated that genome-wide LD decayed to 0.1 at 23 Mb, and population structure analysis revealed seven subgroups in the panel.Association analysis using a mixed linear model(MLM) identified three loci for powdery mildew resistance on chromosome 2 B, designated QPm.stars-2BL1,QPm.stars-2BL2, and QPm.stars-2BL3.To evaluate the efficacy of GWAS in gene discovery,QPm.stars-2BL2 was validated using F2 and F2:3 populations derived from PI420646 × OK1059060-126135-3.Linkage analysis delimited the powdery mildew resistance gene in PI 420646 to an interval where QPm.stars-2BL2 was located, lending credence to the GWAS results.QPm.stars-2BL1 and QPm.stars-2BL3, which were associated with four SNPs located at 457.7–461.7 Mb and two SNPs located at 696.6–715.9 Mb in the Chinese Spring reference IWGSC RefSeq v1.0, respectively, are likely novel loci for powdery mildew resistance and can be used in wheat breeding to improve powdery mildew resistance.Genqiao Li Xiangyang Xu Chengcheng Tan Brett F.Carver Guihua Bai Xuewen Wang J.Michael Bonman Yanqi Wu Robert Hunger Christina Cowger 2019The Crop Journal2019,7,3:2
5Effects of Ovarian Preservation Time on in vitro Maturation and in vitro Fertilization of Oocytes from Slaughtered Sheep显示文摘[Objectives] The aim was to study the effects of ovarian preservation time on in vitro fertilization of oocytes from slaughtered sheep. [Methods] The collected ovaries were randomly and evenly divided into four groups. They were preserved in physiological saline containing penicillin( 100 IU/ml) and streptomycin( 100 μg/ml) at 15-20 ℃ for 0( Control),6,12 and 18 h,respectively. Then,the oocytes were subjected to in vitro fertilization. [Results]The maturation rates,cleavage rates and blastocyst rates of the oocytes preserved for 6 and 12 h showed no significant differences compared with those of the oocytes preserved for0 h( 72. 03%,70. 87% vs. 73. 68%; 74. 12%,72. 60% vs. 74. 49%; 22. 22%,20. 75% vs. 23. 29%)( P > 0. 05). There were also no significant differences in maturation rate,cleavage rate or blastocyst rate between the oocytes preserved for 18 and 0 h( P > 0. 05). 7Heritability estimates for seed yield and its components in Cynodon dactylon var. dactylon (L.) Pers.显示文摘Bermudagrass[Cynodon dactylon var.dactylon(L.)Pers.]is a major warm-season turf and forage grass worldwide.Seed yield is an important trait targeted for improvement in bermudagrass breeding programs because of the increased interest in seed-propagated cultivars.Understanding the nature of genetic variation for seed yield and its components in bermudagrass would aid the development of seed-propagated cultivars.The objective of this study was to estimate the genetic variation and narrow-sense heritability for seed yield and its two major components,inflorescences prolificacy and seed set percentage in bermudagrass.Twenty-five half-sib families and their respective clonal parents were evaluated at two Oklahoma locations,Perkins and Stillwater(Oklahoma,USA),over two years.Half-sib families were different for seed yield,inflorescences prolificacy and seed set percentage,indicating the expression of additive genes in controlling these traits.Family×location effects were observed for seed set percentage and seed yield.All three traits showed family×year interaction effects.There was a significant family×location×year interaction in inflorescences prolificacy and seed set percentage.Narrow-sense heritability estimates for seed yield was 0.18 based on variance component analysis among half-sib families and ranged from 0.26 to 0.68 based on parent-offspring regressions,indicating genetic complexity of seed yield.Heritability estimates were moderate(0.30−0.55)for inflorescences prolificacy and moderate to relatively high(0.41−0.78)for seed set percentage.The results indicate that sufficient magnitudes of additive genetic variation for seed set percentage and inflorescence prolificacy permit positive response to selection and conventional progeny-based genotypic evaluation is necessary for seed yield improvement.Citation:Tan C,Wu Y,Taliaferro CM,Bell GE,Martin DL,et al.2022.Heritability estimates for seed yield and its components in Cynodon dactylon var.Chengcheng Tan Yanqi Wu Charles M.Taliaferro Greg E.Bell Dennis L.Martin Mike W.Smith 2022Grass Research2022,2,1:0
8Next generation of neurological therapeutics:Native and bioengineered extracellular vesicles derived from stem cells显示文摘Extracellular vesicles(EVs)-based cell-free therapy,particularly stem cell-derived extracellular vesicles(SC-EVs),offers new insights into treating a series of neurological disorders and becomes a promising candidate for alternative stem cell regenerative therapy.Currently,SC-EVs are considered direct therapeutic agents by themselves and/or dynamic delivery systems as they have a similar regenerative capacity of stem cells to promote neurogenesis and can easily load many functional small molecules to recipient cells in the central nervous system.Meanwhile,as non-living entities,SC-EVs avoid the uncontrollability and manufacturability limitations of live stem cell products in vivo(e.g.,low survival rate,immune response,and tumorigenicity)and in vitro(e.g.,restricted sources,complex preparation processes,poor quality control,low storage,shipping instability,and ethical controversy)by strict quality control system.Moreover,SC-EVs can be engineered or designed to enhance further overall yield,increase bioactivity,improve targeting,and extend their half-life.Here,this review provides an overview on the biological properties of SC-EVs,and the current progress in the strategies of native or bioengineered SC-EVs for nerve injury repairing is presented.Then we further summarize the challenges of recent research and perspectives for successful clinical application to advance SC-EVs from bench to bedside in neurological diseases.Shilin Jin Zhongyue Lv Lin Kang Jiayi Wang Chengcheng Tan Liming Shen Liang Wang Jing Liu 2022Asian Journal of Pharmaceutical Sciences2022,17,6:0
9Co_2O_3-doping Effect on the Formation of 3CaO 3Al_2O_3 CaSO_4显示文摘The Co2O3-doping effect on the formation of 3CaO-3Al2O3-CaSO4 from CaCO3-Al2O3-CaSO4-2H2O mixtures was investigated by means of SO2 emission behavior, chemical analysis, X-ray diffraction, differential thermal analysis and scanning electron microscopy. The experimental results show that Co2O3 addition increases the reactivity of the CaCO3-Al2O3-CaSO4-2H2O system significantly, by reducing SO2 emissions in combustion and f-CaO contents in the clinkers, promoting the nucleation and growth of 3CaO-3Al2O3-CaSO4, and intensifying the formation of 3CaO-3Al2O3-CaSO4. 10Facile Preparation of Fe-N-C Oxygen Reduction Electrocatalysts from Metal Organic Frameworks for Zn-Air Battery显示文摘It is critical to study efficient,stable oxygen reduction reaction(ORR)electrocatalysts due to insufficient stability and expensive price of Pt/C catalysts for Zn-air batteries.Fe–N–C electrocatalysts was synthesized by a facile solvent-green method and the efficiency of Fe–N–C optimized was studied as potential ORR electrocatalysts under alkaline condition.Results indicated that it had excellent ORR activity with E_(1/2)of 0.93 V,which was competitive to that of Pt/C-JM under the same conditions.Moreover,the assembled Zn-air battery exhibited discharge potential and charge potential of 1.2 V,2.32 V at 5 mA cm^(−2)with high stability,respectively.Overall,all results illustrated that Fe–N–C is an excellent ORR electrocatalyst in the field of metal air battery.Additionally,this work opens a good way to synthesize highly efficient electrocatalysts from metal organic framework and to investigate ORR mechanism of efficient chemical energy to electricity conversion.Chengcheng Wang Dawei Luo Bingxue Hou Mortaza Gholizadeh Zanxiong Tan Xiaojie Han 2022Journal of Renewable Materials2022,10,5:0
