| 2 | ^40Ar/^39Ar and Rb-Sr Ages of the Tiegelongnan Porphyry Cu-(Au)Deposit in the Bangong Co-Nujiang Metallogenic Belt of Tibet,China:Implication for Generation of Super-Large Deposit显示文摘The Tiegelongnan deposit is a newly discovered super-large porphyry-epithermal Cu-(Au) deposit in the western part of the Bangong Co-Nujiang metallogenic belt, Tibet(China). Field geology and geochronology indicate that the porphyry mineralization was closely related to the Early Cretaceous intermediate-felsic intrusions(ca. 123–120 Ma). Various epithermal ore and gangue mineral types were discovered in the middle-shallow part of the orebody, indicating the presence of epithermal mineralization at Tiegelongnan. Potassic, propylitic, phyllic and advanced argillic alteration zones were identified. ^(40)Ar/^(39)Ar dating of hydrothermal biotite(potassic zone), sericite(phyllic zone), and alunite(advanced argillic zone) in/around the ore-bearing granodiorite porphyry yielded 121.1±0.6 Ma(1σ), 120.8±0.7 Ma(1σ) and 117.9±1.6 Ma(1σ), respectively. Five hydrothermal mineralization stages were identified, of which the Stage IV pyrite was Rb-Sr dated to be 117.5±1.8 Ma(2σ), representing the end of epithermal mineralization. Field geology and geochronology suggest that both the epithermal and porphyry mineralization belong to the same magmatic-hydrothermal system. | 4 | Antioxidant properties of magnesium lithospermate B contribute to the cardioprotection against myocardial ischemia/reperfusion injury in vivo and in vitro显示文摘OBJECTIVE: To determine the cardioprotective effect of magnesium lithospermate B (MLB) on myocardial ischemia/reperfusion (MI/R) injury and to investigate the antioxidant potential in vivo and in vitro. METHODS: MI/R injury was induced by the occlusion of left anterior descending coronary artery for 30 min followed by reperfusion for 3 h in rats. After reperfusion, hearts were harvested to assess infarct size, histopathological damages, the levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH) and malondialdehyde (MDA). Blood samples were col- lected to determine serum levels of creatine kinase-MB (CK-MB), cardiac troponin (cTnI) and lactate dehydrogenase (LDH). Furthermore, simulatedischemia/reperfusion (SI/R) injury in vitro was established by oxygen and glucose deprivation (OGD) for 2 h followed by 24-hour recovery period in cardiomyocytes. The activity of LDH in the cultured supernatant and the levels of intracellular reactive oxygen species (ROS), SOD and MDA in cardiomyocytes were also measured. Finally, cardiomyocytes apoptosis was determined with flow cytometry. RESULTS: MLB significantly limited infarct size, ameliorated histopathological damages and prevented leakage of CK-MB, cTnI and LDH. Additionally, SOD, CAT, GPx and GSH activities were notably increased by MLB, along with the MDA content decreased as compared with the model group in rats. In vitro study, MLB also decreased LDH activity in the cultured supernatant, increased SOD activity in cardiomyocytes, reduced intracellular ROS and MDA levels, and significantly suppressed cardiomyocytes apoptosis. CONCLUSION: MLB possessed remarkably cardioprotective effects on MI/R injury in vivo and in vitro. | 5 | Characterization of drug responses of mini patient-derived xenografts in mice for predicting cancer patient clinical therapeutic response显示文摘Background:Patient-derived organoids and xenografts(PDXs)have emerged as powerful models in functional diag-nostics with high predictive power for anticancer drug response.However,limitations such as engraftment failure and time-consuming for establishing and expanding PDX models followed by testing drug efficacy,and inability to subject to systemic drug administration for ex vivo organoid culture hinder realistic and fast decision-making in selecting the right therapeutics in the clinic.The present study aimed to develop an advanced PDX model,namely MiniPDX,for rapidly testing drug efficacy to strengthen its value in personalized cancer treatment.Methods:We developed a rapid in vivo drug sensitivity assay,OncoVee®MiniPDX,for screening clinically relevant regimens for cancer.In this model,patient-derived tumor cells were arrayed within hollow fiber capsules,implanted subcutaneously into mice and cultured for 7 days.The cellular activity morphology and pharmacokinetics were systematically evaluated.MiniPDX performance(sensitivity,specificity,positive and negative predictive values)was examined using PDX as the reference.Drug responses were examined by tumor cell growth inhibition rate and tumor growth inhibition rate in PDX models and MiniPDX assays respectively.The results from MiniPDX were also used to evaluate its predictive power for clinical outcomes.Results:Morphological and histopathological features of tumor cells within the MiniPDX capsules matched those both in PDX models and in original tumors.Drug responses in the PDX tumor graft assays correlated well with those in the corresponding MiniPDX assays using 26 PDX models generated from patients,including 14 gastric cancer,10 lung cancer and 2 pancreatic cancer.The positive predictive value of MiniPDX was 92%,and the negative predictive value was 81%with a sensitivity of 80%and a specificity of 93%.Through expanding to clinical tumor samples,Min-iPDX assay showed potential of wide clinical application.Conclusions:Fast in vivo MiniPDX assay based on capsule implantation was developed-to assess drug responses of both PDX tumor grafts and clinical cancer specimens.The high correlation between drug responses of paired MiniPDX and PDX tumor graft assay,as well as translational data suggest that MiniPDX assay is an advanced tool for personalized cancer treatment. | Feifei Zhang Wenjie Wang Yuan Long Hui Liu Jijun Cheng Lin Guo Rongyu Li Chao Meng Shan Yu Qingchuan Zhao Shun Lu Lili Wang Haitao Wang Danyi Wen | 2018 | Cancer Communications2018,38,1: | 20 |

| 7 | Safety of different acupuncture manipulations for posterior circulation ischemia with vertigo显示文摘Acupuncture at Fengchi(GB20) in the posterior neck improves vertigo.However,subarachnoid hemorrhage and spinal epidural hematoma have been reported to occur after acupuncture in the posterior neck.Therefore,in the present study,we assessed the safety of acupuncture at Fengchi.Laboratory tests and adverse event reports were used to evaluate the safety of different acupuncture manipulations for the treatment of posterior circulation ischemia with vertigo.A total of 136 patients were randomly assigned to four groups.Verum acupuncture was conducted with different needle insertion directions(contralateral paropia or prominentia laryngea) and different needle twisting frequencies(60 or 120 times/minute) at Fengchi and matching acupoints(for example,Zhongwan [CV12],Qihai [CV6],Zusanli [ST36],and Fenglong [ST40]).The patients received 14 treatments over 3–4 weeks.Routine blood analysis,hepatic and renal function tests,urine and feces tests and electrocardiography were performed before the first treatment session and after the final session.Adverse events were recorded after every session.Of the 136 patients,120 completed the study.There were no significant differences between pretreatment and posttreatment test results in any of the groups.Only five patients suffered from minor adverse events(needling pain,slight hematoma and transient chest tightness).No serious adverse events were found.Our results indicate that a 14-session course of needling at Fengchi is relatively safe for treating posterior circulation ischemia with vertigo. | Yan Wen Chao Zhang Xiao-feng Zhao Shi-zhe Deng Si He Ling-hui Huang Guang Tian Zhi-hong Meng | 2016 | Neural Regeneration Research2016,11,8: | 16 |
| 8 | Therapeutic effects of different durations of acupuncture on rats with middle cerebral artery occlusion显示文摘Acupuncture is regarded as an effective therapy for cerebral ischemia. Different acupuncture manipulations and durations may result in different therapeutic effects. In the present study, the Neig uan(PC6) acupoint of rats with occluded middle cerebral arteries was needled at a fixed frequency(3 Hz) with different durations, i.e., 5, 60 and 180 seconds under a twisting-rotating acupuncture method. Results showed that different durations of acupuncture had different therapeutic effects, with 60 seconds yielding a better therapeutic effect than the other two groups. This duration of treatment demonstrated rapid cerebral blood flow, encouraging recovery of neurological function, and small cerebral infarct volume. | 9 | Estrogenic activities of two synthetic pyrethroids and their metabolites显示文摘Synthetic pyrethroids(SPs) are among the most common pesticides in current use,and so far,several SPs have been assessed for their potential estrogenicities by various methods.Previous studies have shown that the estrogenicities partly come from their metabolites.Although considerable information is available with respect to the metabolism and environmental degradation of SPs,little is known about the estrogenicities of the metabolites.In this study,permethrin(PM) and β-cypermethrin(CP),as well as their metabolites(3-phenoxybenzoic alcohol(PBCOH),3-phenoxybenzaldehyde(PBCHO) and 3-phenoxybenzoic acid(PBCOOH) were evaluated for their estrogenic activities in the MCF-7 human breast carcinoma cell line.In the MCF-7 cell proliferation assay,PM and CP exhibited significant estrogenic activities at 10-7 mol/L,comparable to 17β-estradiol(E2 ) of 10-9 mol/L,with the relative proliferative effect ratios of 55.4% and 56.3%,respectively.The real-time quantitative polymerase chain reaction(qRT-PCR) results confirmed the estrogenicities of PM and CP with significant alteration of pS2 and ERα mRNA levels observed at 10-6 mol/L.For the three major metabolites,PBCOH and PBCOOH exhibited estrogenic activities in all assays,while no significant estrogenic responses was observed for PBCHO compared to the vehicle control.In particular,PBCOH had even slightly stronger estrogenic activity than its parent compounds,indicating that metabolism may be one of the reasons for the estrogenicities of the SPs.Given the widespread use of SPs,the toxicological effects of parent compounds and their metabolites should be taken into consideration in the risk assessment of SPs. | Meiqing Jin Ling Li Chao Xu Yuezhong Wen Meirong Zhao | 2010 | Journal of Environmental Sciences2010,22,2: | 13 |

| 12 | Reversal of multidrug resistance in gastric cancer cells by CDX2 downregulation显示文摘AIM: To explore the role of CDX2 in the multi-drug resistance (MDR) process of gastric cancer in vitro and in vivo . METHODS: A cisplatin-resistant gastric cancer cell line with stable downregulation of CDX2 was established. mRNA and protein expression levels of CDX2, survivin, cyclin D1, and c-Myc were detected by western blotting and semi-quantitative reverse-transcriptase polymerase chain reaction (RT-PCR). The influence of downregulation of CDX2 on MDR was assessed by measuring IC50 of SGC7901/DDP cells to cisplatin, doxorubicin, and 5-fluorouracil, rate of doxorubicin efflux, apoptosis, and cell cycle progression detected by flow cytometry. In addition, we determined the in vivo effects of CDX2 small interfering RNA (siRNA) on tumor size, and apoptotic cells in tumor tissues were detected by deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling and hematoxylin and eosin staining. RESULTS: CDX2 siRNA led to downregulation of endogenous CDX2 mRNA (0.31 ± 0.05 vs 1.10 ± 0.51, 0.31 ± 0.05 vs 1.05 ± 0.21, P = 0.003) and protein (0.12 ± 0.08 vs 0.51 ± 0.07, 0.12 ± 0.08 vs 0.55 ± 0.16, P = 2.57 × 10 -4) expression. It significantly promoted the sensitivity of SGC7901/DDP cells to cisplatin (0.12 ± 0.05 vs 0.33 ± 0.08, 0.12 ± 0.05 vs 0.39 ± 0.15, P = 0.001), doxorubicin (0.52 ± 0.13 vs 4.11 ± 1.25, 0.52 ± 0.13 vs 4.05 ± 1.44, P = 2.81 × 10-4), and 5-fluorouracil (0.82 ± 0.13 vs 2.81 ± 0.51, 0.82 ± 0.13 vs 3.28 ± 1.03, P = 1.71 × 10-4). Flow cytometry confirmed that the percentage of apoptotic cells increased after CDX2 downregulation (32.15% ± 2.15% vs 17.63% ± 3.16%, 32.15% ± 2.15% vs 19.3% ± 2.25%, P = 1.73 × 10-6). This notion was further supported by the observation that downregulation of CDX2 blocked entry into the S-phase of the cell cycle (31.53% ± 3.78% vs 65.05% ± 7.25%, 31.53% ± 3.78% vs 62.27% ± 5.02%, P = 7.55 × 10-7). Furthermore, downregulation of CDX2 significantly increased intracellular accumulation of doxorubicin (0.21 ± 0.06 vs 0.41 ± 0.11, 0.21 ± 0.06 vs 0.40 ± 0.08, P = 0.003). In molecular studies, semiquantitative RT-PCR and western blotting revealed that CDX2 downregulation could inhibit expression of c-Myc, survivin and cyclin D1. The dolomite of the Changxing Formation is divided into four diagenetic systems: (1) penecontemporaneous stage syngenetic brine diagenetic system, (2) early diagenetic stage strata seal brine diagenetic system, (3) middle-late diagenetic stage mixed hot brine diagenetic system and (4) tectonic uplift stage mixed hydrothermal fluid diagenetic system. New understanding of the controlling factors and distribution of dolomite reservoir development is discussed. Reef shoal facies belts controlled regional reservoir distribution and the scale of development. Burial dolomitization of a strata seal brine diagenetic system is the foundation of reservoir development, mainly developing pore reservoir. Burial dolomitization of mixed hot brine diagenetic system expanded the reservoir distribution and improved the reservoir quality, mainly developing pore-vug reservoir. In another group (n = 64) experiencing significant levels of depression or anxiety, the L-allele frequency was also significantly higher. In healthy participants, L-carriers had reduced functional and anatomical connectivity between the amygdala and prefrontal cortex (PFC), which was correlated with anxiety or depression scores. Our findings demonstrated that in Chinese Han participants, in contrast to Caucasians, the L-allele confers vulnerability to anxiety or depression and weakens top-down emotional control between the PFC and amygdala. | 18 | Long-term hepatic consequences of chemotherapy-related HBV reactivation in lymphoma patients显示文摘AIM: To investigate the long-term consequences of chemotherapy-related HBV reactivation in patients with lymphoma.METHODS: This study was based on the database of published prospective study evaluating HBV reactivation in HBV lymphoma patients during chemotherapy.Deteriorated liver reserve (DLR) was defined as development of either one of the following conditions during follow-up: (1) newly onset parenchyma liver disease, splenomegaly or ascites without evidence of lymphoma involvement; (2) decrease of the ratio (albumin/globulin ratio) to less than 0.8 or increase of the ratio of INR of prothrombin time to larger than 1.2 without evidence of malnutrition or infection. Liver cirrhosis was diagnosed by imaging studies.RESULTS: A total of 49 patients were included. The median follow-up was 6.2 years (range, 3.9-8.1 years).There were 31 patients with and 18 patients without HBV reactivation. Although there was no difference of overall survival (OS) and chemotherapy response rate between the two groups, DLR developed more frequently in patients with HBV reactivation (48.4% vs 16.7%; P= 0.0342). Among the HBV reactivators, HBV genotype C was associated with a higher risk of developing DLR (P = 0.0768) and liver cirrhosis (P = 0.003). Four of five patients with sustained high titer of HBV DNA and two of three patients with multiple HBV reactivation developed DLR. Further, patients with a sustained high titer of HBV DNA had the shortest OS among the HBV reactivators (P= 0.0000). | 19 | Prognosis of hepatic cirrhosis patients with esophageal or gastric variceal hemorrhage: multivariate analysis显示文摘Objective: To study the effect of bacterial infection,use of antibiotics, active bleeding at endoscopy, andthe severity of liver disease as prognostic factors inhepatic cirrhotic patients during the first 5 days afterthe episode of esophageal or gastric variceal hemor-rhage.Methods: Seventy-six hepatic cirrhosis patients withesophageal or gastric variceal bleeding were enrolled.Bleeding was managed in a standardized protocol u-sing octreotide and vasopressin in sclerotherapy orband ligation for active bleeding at endoscopy. Thescreening protocol for bacterial infection consisted ofchest radiograph; blood, urine and ascitic fluid cul-tures; the severity of liver disease shown by Child-Pugh score.Results: Active bleeding was observed at endoscopyin 40 patients (53%). Failure to control bleedingWithin 5 days occurred in 36 patients (45%). Empir-ical antibiotic treatment was used in 53 patients(67%), whereas bacterial infections were documen-ted in 43 patients (57%).
