2Comparative Analysis on Chemical Components and Sensory Quality of Aging Flue-Cured Tobacco from Four Main Tobacco Areas of China显示文摘Complicated changes are happening during the aging process of flue-cured tobacco (FCT) and understanding of the changing components is of great significance in improving the quality,shortening aging time,and enhancing production and economic efficiency in tobacco industry.The aging samples of FCT from four main producing areas of China,i.e.,Yunnan,Sichuan,Hubei,and Dongbei,were used to study the changing rule of the components such as alkali,acid,and carbohydrate as well as the aroma and their relationship with sensory quality;and based on the correlation among the components and the sensory quality index,multivariable models were established to predict the aging quality.The results showed that the sensory quality of FCT increased steadily during the aging time,and at the same time,the acidic components (total organic acids,volatile acids) increased gradually,while the alkaline substances (nicotine,volatile alkali),carbohydrate (total sugars,reducing sugar) and pH values showed a downward trend.Correlation analysis found that the sensory quality and pH values were negatively correlated (P<0.05),while the sensory quality with total organic acids and aroma were positively correlated.The optimal model for predicting the quality of FCT was y=56.75-0.63x12+50.09x2-13.27x22,(y:sensory quality;x1:pH;x2:total organic acids).The average predicating error was 1.04% with maximum predicating error of 2.89% and the accuracy rate of above 97%.SUN Jing-guo HE Jie-wang WU Feng-guang TU Shu-xing YAN Tie-jun SI Hui XIE Hao 2011Agricultural Sciences in China2011,10,8:20
3Safety of different acupuncture manipulations for posterior circulation ischemia with vertigo显示文摘Acupuncture at Fengchi(GB20) in the posterior neck improves vertigo.However,subarachnoid hemorrhage and spinal epidural hematoma have been reported to occur after acupuncture in the posterior neck.Therefore,in the present study,we assessed the safety of acupuncture at Fengchi.Laboratory tests and adverse event reports were used to evaluate the safety of different acupuncture manipulations for the treatment of posterior circulation ischemia with vertigo.A total of 136 patients were randomly assigned to four groups.Verum acupuncture was conducted with different needle insertion directions(contralateral paropia or prominentia laryngea) and different needle twisting frequencies(60 or 120 times/minute) at Fengchi and matching acupoints(for example,Zhongwan [CV12],Qihai [CV6],Zusanli [ST36],and Fenglong [ST40]).The patients received 14 treatments over 3–4 weeks.Routine blood analysis,hepatic and renal function tests,urine and feces tests and electrocardiography were performed before the first treatment session and after the final session.Adverse events were recorded after every session.Of the 136 patients,120 completed the study.There were no significant differences between pretreatment and posttreatment test results in any of the groups.Only five patients suffered from minor adverse events(needling pain,slight hematoma and transient chest tightness).No serious adverse events were found.Our results indicate that a 14-session course of needling at Fengchi is relatively safe for treating posterior circulation ischemia with vertigo.Yan Wen Chao Zhang Xiao-feng Zhao Shi-zhe Deng Si He Ling-hui Huang Guang Tian Zhi-hong Meng 2016Neural Regeneration Research2016,11,8:16
4Optimization of air quantity regulation in mine ventilation networks using the improved differential evolution algorithm and critical path method显示文摘In mine ventilation networks, the reasonable airflow distribution is very important for the production safety and economy. Three basic problems of the natural, full-controlled and semi-controlled splitting were reviewed in the paper. Aiming at the high difficulty semi-controlled splitting problem, the general nonlinear multi-objectives optimization mathematical model with constraints was established based on the theory of mine ventilation networks. A new algorithm, which combined the improved differential evaluation and the critical path method(CPM) based on the multivariable separate solution strategy,was put forward to search for the global optimal solution more efficiently. In each step of evolution,the feasible solutions of air quantity distribution are firstly produced by the improved differential evolution algorithm, and then the optimal solutions of regulator pressure drop are obtained by the CPM.Through finite steps iterations, the optimal solution can be given. In this new algorithm, the population of feasible solutions were sorted and grouped for enhancing the global search ability and the individuals in general group were randomly initialized for keeping diversity. Meanwhile, the individual neighborhood in the fine group which may be closely to the optimal solutions were searched locally and slightly for achieving a balance between global searching and local searching, thus improving the convergence rate. The computer program was developed based on this method. Finally, the two ventilation networks with single-fan and multi-fans were solved. The results show that this algorithm has advantages of high effectiveness, fast convergence, good robustness and flexibility. 5Recombinant adenovirus vector Ad-hIL-10 protects grafts from cold ischemia-reperfusion injury following orthotopic liver transplantation in rats显示文摘BACKGROUND:Interleukin 10(IL-10),a Th2 type cytokine,modulates inflammatory responses by inhibiting the production of proinflammatory cytokines.This study was designed to investigate the protective effects of adenovirus- mediated human IL-10(Ad-hIL-10)gene transfer on protecting grafts from cold ischemia-reperfusion injury following orthotopic liver transplantation in rats. METHODS:Adenoviruses encoding hIL-10 orβ-galactosidase (Ad-lacZ)were injected via the superior mesenteric vein into prospective donor animals.The donor liver was harvested 48 hours after transduction,and stored for 12 hours at 4℃ in lactated Ringer's solution prior to transplantation.The rats were divided into saline,Ad-lacZ,and Ad-hIL-10 groups.Liver function test,histopathological examination,reverse transcriptase-polymerase chain reaction(RT-PCR),and Western blotting were performed at 24 hours after transplantation in the three groups. RESULTS:Liver function(ALT and AST)was significantly improved,and the Suzuki score was significantly decreased in the Ad-hIL-10 group.The levels of hepatic TNF-α,MIP-2,ICAM-1 mRNA,and NF-κB protein in the Ad-hIL-10 group were significantly decreased.The expression of hIL-10 mRNA was detected by RT-PCR in Ad-hIL-10-treated grafts but not in controls treated with saline or Ad-lacZ. 6Spinal Cord Stimulation Frequency Influences the Hemodynamic Response in Patients with Disorders of Consciousness显示文摘Spinal cord stimulation(SCS) is a promising technique for treating disorders of consciousness(DOCs).However, differences in the spatio-temporal responsiveness of the brain under varied SCS parameters remain unclear. In this pilot study, functional near-infrared spectroscopy was used to measure the hemodynamic responses of 10 DOC patients to different SCS frequencies(5 Hz, 10 Hz, 50 Hz, 70 Hz, and 100 Hz). In the prefrontal cortex, a key area in consciousness circuits, we found significantly increased hemodynamic responses at 70 Hz and 100 Hz, and significantly different hemodynamic responses between 50 Hz and 70 Hz/100 Hz. In addition, the functional connectivity between prefrontal and occipital areas was significantly improved with SCS at 70 Hz. 7Enhanced Poly(ethylene terephthalate)Hydrolase Activity by Protein Engineering显示文摘Poly(ethylene terephthalate)hydrolase(PETase)from Ideonella sakaiensis exhibits a strong ability to degrade poly(ethylene terephthalate)(PET)at room temperature,and is thus regarded as a potential tool to solve the issue of polyester plastic pollution.Therefore,we explored the interaction between PETase and the substrate(a dimer of the PET monomer ethylene terephthalate,2PET),using a model of PETase and its substrate.In this study,we focused on six key residues around the substrate-binding groove in order to create novel high-efficiency PETase mutants through protein engineering.These PETase mutants were designed and tested.The enzymatic activities of the R61A,L88F,and I179F mutants,which were obtained with a rapid cell-free screening system,exhibited 1.4 fold,2.1 fold,and 2.5 fold increases,respectively,in comparison with wild-type PETase.The I179F mutant showed the highest activity,with the degradation rate of a PET film reaching 22.5 mg perμmol·L^-1 PETase per day.Thus,this study has created enhanced artificial PETase enzymes through the rational protein engineering of key hydrophobic sites,and has further illustrated the potential of biodegradable plastics.Yuan Ma Mingdong Yao Bingzhi Li Mingzhu Ding Bo He Si Chen Xiao Zhou Yingjin Yuan 2018Engineering2018,4,6:10

10Design and experimental study of a practical Osculating Inward Cone Waverider Inlet显示文摘A design method based on tip to tail streamline tracing and osculating inward cone methods is discussed for designing the integrated Osculating Inward Cone Waverider Inlet(OICWI). A practical geometrical constrained experimental model of OICWI is designed based on the validated design method. It has a total contraction ratio of 4.61 and inner contraction ratio is 2.0. Wind-tunnel tests have been conducted for the OICWI model at free stream Mach number(Ma_∞) of 4.0, 3.5 and 3.0 respectively. The experimental results show that the OICWI has high flow capture ratio and compression abilities. It can self-start at Ma_∞= 3.5 and 4.0 and its flow capture ratio is 0.73 at Ma_∞= 4.0, and Angle of Attack(AOA) 0°. The research results show that the OICWI has advantages of inward cone waverider and streamline tracing inlet. 11Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 12Recent progress on MOF-derived carbon materials for energy storage显示文摘Metal-organic frameworks(MOFs)are of quite a significance in the field of inorganic-organic hybrid crystals.Especially,MOFs have attracted increasing attention in recent years due to their large specific surface area,desirable electrical conductivity,controllable porosity,tunable geometric structure,and excellent thermal/chemical stability.Some recent studies have shown that carbon materials prepared by MOFs as precursors can retain the privileged structure of MOFs,such as large specific surface area and porous structure and,in contrast,realize in situ doping with heteroatoms(eg,N,S,P,and B).Moreover,by selecting appropriate MOF precursors,the composition and morphology of the carbon products can be easily adjusted.These remarkable structural advantages enable the great potential of MOF-derived carbon as high-performance energy materials,which to date have been applied in the fields of energy storage and conversion systems.In this review,we summarize the latest advances in MOF-derived carbon materials for energy storage applications.We first introduce the compositions,structures,and synthesis methods of MOF-derived carbon materials,and then discuss their applications and potentials in energy storage systems,including rechargeable lithium/sodium-ion batteries,lithium-sulfur batteries,supercapacitors,and so forth,in detail.Finally,we put forward our own perspectives on the future development of MOF-derived carbon materials.Jincan Ren Yalan Huang He Zhu Binghao Zhang Hekang Zhu Shenghui Shen Guoqiang Tan Feng Wu Hao He Si Lan Xinhui Xia Qi Liu 2020Carbon Energy2020,2,2:8
13Cloning and Characterization of a Salt Tolerance-Associated Gene Encoding Trehalose-6-Phosphate Synthase in Sweetpotato显示文摘Trehalose plays an important role in metabolic regulation and abiotic stress tolerance in a variety of organisms. In plants, its biosynthesis is catalyzed by two key enzymes: trehalose-6-phosphate synthase(TPS) and trehalose-6-phosphate phosphatase(TPP). In the present study, a TPS gene, named IbTPS, was first isolated from sweetpotato(Ipomoea batatas(L.) Lam.) cv. Lushu 3 by rapid amplification of cDNA ends(RACE). The open reading frame(ORF) contained 2 580 nucleotides encoding 859 amino acids with a molecular weight of 97.433 kDa and an isoelectric point(pI) of 5.7. The deduced amino acid sequence showed high identities with TPS of other plants. Real-time quantitative PCR analysis revealed that the expression level of IbTPS gene was significantly higher in stems of Lushu 3 than in its leaves and roots. Subcellular localization analysis in onion epidermal cells indicated that IbTPS gene was located in the nucleus. Transgenic tobacco(cv. Wisconsin 38) plants over-expressing IbTPS gene exhibited significantly higher salt tolerance compared with the control plant. Trehalose and proline content was found to be significantly more accumulated in transgenic tobacco plants than in the wild-type and several stress tolerance related genes were up-regulated. 15Telomere-dependent and telomereindependent roles of RAP1 in regulating human stem cell homeostasis显示文摘RAP1 is a well-known telomere-binding protein, but its functions in human stem cells have remained unclea匚 Here we generated RAP1 -deficient human embryonic stem cells (hESCs) by using CRISPR/Cas9 technique and obtained RAP1-deficient human mesenchymal stem cells (hMSCs) and neural stem cells (hNSCs) via directed differentiation. In both hMSCs and hNSCs, RAP1 not only negatively regulated telomere length but also acted as a transcriptional regulator of RELN by tuning the methylation status of its gene promoter. 18Comparative genomic and transcriptomic analyses uncover the molecular basis of high nitrogen-use efficiency in the wheat cultivar Kenong 9204显示文摘Studying the regulatory mechanisms that drive nitrogen-use efficiency(NUE)in crops is important for sustainable agriculture and environmental protection.In this study,we generated a high-quality genome assembly for the high-NUE wheat cultivar Kenong 9204 and systematically analyzed genes related to nitrogen uptake and metabolism.By comparative analyses,we found that the high-affinity nitrate transporter gene family had expanded in Triticeae.Further studies showed that subsequent functional differentiation endowed the expanded family members with saline inducibility,providing a genetic basis for improving the adaptability of wheat to nitrogen deficiency in various habitats.To explore the genetic and molecular mechanisms of high NUE,we compared genomic and transcriptomic data from the high-NUE cultivar Kenong 9204(KN9204)and the low-NUE cultivar Jing 411 and quantified their nitrogen accumulation under high-and low-nitrogen conditions.Compared with Jing 411,KN9204 absorbed significantly more nitrogen at the reproductive stage after shooting and accumulated it in the shoots and seeds.Transcriptome data analysis revealed that nitrogen deficiency clearly suppressed the expression of genes related to cell division in the young spike of Jing 411,whereas this suppression of gene expression was much lower in KN9204.In addition,KN9204 maintained relatively high expression of NPF genes for a longer time than Jing 411 during seed maturity.Physiological and transcriptome data revealed that KN9204 was more tolerant of nitrogen deficiency than Jing 411,especially at the reproductive stage.The high NUE of KN9204 is an integrated effect controlled at different levels.Taken together,our data provide new insights into the molecular mechanisms of NUE and important gene resources for improving wheat cultivars with a higher NUE trait.Xiaoli Shi Fa Cui Xinyin Han Yilin He Long Zhao Na Zhang Hao Zhang Haidong Zhu Zhexin Liu Bin Ma Shusong Zheng Wei Zhang Jiajia Liu Xiaoli Fan Yaoqi Si Shuiquan Tian Jianqing Niu Huilan Wu Xuemei Liu Zhuo Chen Deyuan Meng Xiaoyan Wang Liqiang Song Lijing Sun Jie Han Hui Zhao Jun Ji Zhiguo Wang Xiaoyu He Ruilin Li Xuebin Chi Chengzhi Liang Beifang Niu Jun Xiao Junming Li Hong-Qing Ling 2022Molecular Plant2022,15,9:6

20Effect of Laser Power on Microstructure and Wear Resistance of WC_P/Ni Cermet Coating显示文摘Laser cladding nickel-based alloy coating (Ni60) and nickel-based composite coating doped with WC par-ticles by 35 %( WCP/Ni) were produced onthelow-carbon steel substrate by CO2continuous wavelaser with pow-er of 5 kWusing theinjected powder technique . The effect of laser power on microstructure and wear resistance oflaser cladding WCP/Ni cermet coating was investigated . The WC_P/Ni alloy coating with evenly distributed WCce-ramic phases and the better bond with the substrate alloy was obtained at a power of 2·2 kW. Diffusion solutionreaction happened between WC particles and the substrate alloy during laser cladding ,and led to the formation ofblock rich-tungsten carbide on the edges of the WC particles ,especially at higher power . The WC_P/Ni alloy coat-ing consists of the undissolved WC particles ,the block or dendritic rich-tungsten carbide ,the bar-like rich-chromi-umcarbide ,and dendrite solid solution and eutectic structure among the carbides .
