| 1 | Integrative single-cell analysis of transcriptome,DNA methylome and chromatin accessibility in mouse oocytes显示文摘Oocyte growth is a key step in forming mature eggs that are ready to be fertilized.The states and modifications of chromatin represent critical sources of information for this process.However,the dynamics and interrelations of these chromatin characteristics remain elusive.In this study,we developed an improved scCOOL-seq technique (iscCOOL-seq),which is a multiomics,single-cell and single-base resolution method with high mapping rates,and explored the chromatin accessibility landscape and its relationship to DNA methylation in growing mouse oocytes.The most dramatic change in chromatin accessibility occurs during oocyte growth initiation,accompanied with prominent transcriptome alterations and an elevated variation in DNA methylation levels among individual oocytes.Unlike CpG islands (CGIs),partially methylated domains (PMDs)are associated with a low density of nucleosome-depleted regions (NDRs)during the whole maturation period.Surprisingly,highly expressed genes are usually associated with NDRs at their transcriptional end sites (TESs).In addition,genes with de novo methylated gene bodies during oocyte maturation are already open at their promoters before oocyte growth initiation.Furthermore,epigenetic and transcription factors that might be involved in oocyte maturation are identified.Our work paves the way for dissecting the complex, yet highly coordinated,epigenetic alterations during mouse oocyte growth and the establishment of totipotency. | Chan Gu Shanling Liu Qihong Wu Lin Zhang Fan Guo | 2019 | Cell Research2019,29,2: | 14 |
| 2 | A Model of Hydrothermal Dolomite Reservoir Facies in Precambrian Dolomite, Central Sichuan Basin, SW China and its Geochemical Characteristics显示文摘Hydrothermal mineral assemblages and related hydrothermally enhanced fracturing are common in the Precambrian Dengying Formation of Central Sichuan Basin. Petrographic and geochemical analyses of core samples show that the hydrothermal dolomite reservoirs of Dengying Formation consist of four main types of pores in the reservoir facies. These include: 1) hydrothermal dissolution vug(or pore), 2) intercrystalline pore, 3) residual inter-breccia vug(or pore), and 4) enlarged dissolved-fracture. There are three different fabrics dolomite in hydrothermal dolomite reservoirs, namely, saddle dolomite, fine-medium dolomite and micritic dolomite. Micritic dolomite is the original lithology of host rock. Saddle dolomite with curved or irregular crystal faces was directly crystallized from hydrothermal fluids(average temperature 192°C). Fine-medium dolomites are the products of recrystallization of micritic dolomite, resulting in abnormal geochemical characteristics, such as slight depletion of δ^(18)O, significant enrichment of Mn-Fe and ^(87)Sr/^(86)Sr, and positive Eu anomaly. A model for the distribution of various hydrothermal dolomite reservoir facies is proposed here, which incorporates three fundamental geological controls: 1) extensional tectonics and tectono-hydrothermal events(i.e., the Xingkai Taphrogenesis of Late Sinian-Early Cambrian, and Emei Taphrogenesis of Late Permian), 2) hydrothermal fluid storage in clastic rocks with large thickness(e.g., Nanhua System of Chengjiang Formation and part of Doushantuo Formation), and 3) confining bed for hydrothermal fluids(such as, the shale in Qiongzhusi Formation). The supply of hydrothermal fluid is critical. Large basement-rooted faults and associated grid-like fracture system may function as the channels for upward migration of hydrothermal fluid flow. | 3 | Gas sensing capabilities of TiO_(2) porous nanoceramics prepared through premature sintering显示文摘Pure and noble metal(Pt,Pd,and Au)doped TiO_(2)nanoceramics have been prepared from TiO_(2)nanoparticles through traditional pressing and sintering.For those samples sintered at 550℃,a typical premature sintering occurred,which led to the formation of a highly porous microstructure with a Brunauer-Emmett-Teller(BET)specific surface area of 23 m^(2)/g.At room temperature,only Pt-doped samples showed obvious response to hydrogen,with sensitivities as high as~500 for 1000 ppm H_(2)in N_(2);at 300℃,all samples showed obvious responses to CO,while the responses of noble metal doped samples were much higher than that of the undoped ones.The mechanism for the observed sensing capabilities has been discussed,in which the catalytic effect of Pt for hydrogen is believed responsible for the room-temperature hydrogen sensing capabilities,and the absence of glass frit as commonly used in commercial thick-film metal oxide gas sensors is related to the high sensitivities.It is proposed that much attention should be paid to metal oxide porous nanoceramics in developing gas sensors with high sensitivities and low working temperatures. | Yao XIONG Zilong TANG Yu WANG Yongming HU Haoshuang GU Yuanzhi LI Helen Lai Wah CHAN Wanping CHEN | 2015 | Journal of Advanced Ceramics2015,4,2: | 3 |
| 4 | Modified Zhibai Dihuang pill,a traditional Chinese medicine formula,on steroid withdrawal in systemic lupus erythematosus:A systematic review and meta-analysis显示文摘Background:Zhibai Dihuang pill(ZBDH),a Chinese herbal formula,has been widely used as an adjunctive therapy to help reduce the patient’s steroid dose and maintain low disease activity in systemic lupus erythematosus(SLE).Objective:This systematic review evaluates the therapeutic effect of modified ZBDH in reducing steroid use in patients with SLE.Search strategy:A systematic literature search was carried out using seven databases,including PubMed,Embase,Cochrane Central Register of Controlled Trials,Chinese Biomedical Literature Database,Chinese National Knowledge Infrastructure,Chinese VIP Information and Wanfang Database,from their inception to June 1 st,2019.The search terms included‘‘systemic lupus erythematosus,'‘‘Chinese medicine'and‘‘clinical trial,'and their synonyms.Subject headings matching the above terms were also used.Inclusion criteria:This meta-analysis included randomized controlled trials that evaluated the reduction of steroid dose in patients with SLE.Traditional Chinese medicine(TCM)formulas in experimental group should be prescribed based on ZBDH and used as adjunctive therapy and the comparator should contain steroids.Data extraction and analysis:Two authors independently conducted database search,study selection,data extraction and quality assessment.The extracted information contained study design,sample size,recruitment mode,diagnostic criteria,inclusion and exclusion criteria,participant characteristics,TCM patterns,TCM formulas and treatment outcomes.The primary outcome was the change of steroid dose.Secondary outcomes included SLE Disease Activity Index(SLEDAI),biomarkers of disease activity and clinical response rate.STATA 15.0 was used to analyze the pooled effects reported as weighted mean difference(WMD)or odds ratio,with a 95%confidence interval(CI).Results:In total,20 trials involving 1470 SLE patients were included.The pooled result showed that modified ZBDH taken in combination with standard care led to a larger reduction in steroid dose,compared to standard care alone(WMD:3.79;95%CI:2.58–5.01;P<0.001).Favorable outcomes were also seen in secondary outcome criteria,such as SLEDAI and complement 3.The modified ZBDH treatments were well tolerated without increasing adverse effects.Conclusion:The systematic review provided preliminary evidence supporting the use of ZBDH as a cotherapy to aid steroid dose reduction in patients with SLE.However,more rigorous studies should be conducted to validate these findings,and explore the mechanisms of ZBDH’s relevant bioactive constituents. | Liang Dai Ka Kei Chan Jian-chun Mao Yu Tianc Jun-hua Gu Jun Zhou Linda L.D.Zhong | 2020 | Journal of Integrative Medicine2020,18,6: | 3 |
