| 2 | Common telomerase reverse transcriptase promoter mutations in hepatocellular carcinomas from different geographical locations显示文摘AIM: To determine the mutation status of human telomerase reverse transcriptase gene(TERT) promoter region in hepatocellular carcinoma(HCC) from different geographical regions.METHODS: We analyzed the genomic DNA sequences of 59 HCC samples comprising 15 cell lines and 44 primary tumors,collected from patients living in Asia,Europe and Africa.We amplified a 474 bp DNA fragment of the promoter region of TERT gene including the 1295228 and 1295250 sequence of chromosome 5 by using PCR.Amplicons were then sequenced by Sanger technique and the sequence data were analyzed with by using DNADynamo software in comparison with wild type TERT gene sequence as a reference.RESULTS: The TERT mutations were found highly frequent in HCC.Eight of the fifteen tested cell lines displayed C228 T mutation,and one had C250 T mutation with a mutation frequency up to 60%.All of the mutations were heterozygous and mutually exclusive.Ten out of forty-four tumors displayed C228 T mutation,and additional five tumors had C250 T mutation providing evidence for mutation frequency of 34% in primary tumors.Considering the geographic origins of HCC tumors tested,TERT promoter mutation frequencies were higher in African(53%),when compared to non-African(24%) tumors(P = 0.056).There was also a weak inverse correlation between TERT promoter mutations and murine double minute 2 single nucleotide polymorphism 309 TG polymorphism(P = 0.058).Mutation frequency was nearly two times higher in established HCC celllines(60%) compared to the primary tumors(34%).CONCLUSION: TERT promoter is one of most frequent mutational targets in liver cancer,and hepatocellular carcinogenesis is highly associated with the loss of telomere-dependent cellular senescence control. | Dilek Cevik Gokhan Yildiz Mehmet Ozturk | 2015 | World Journal of Gastroenterology2015，21，1： | 6 |
| 3 | Reflection and Transmission of Regular Waves from/Through Single and Double Perforated Thin Walls显示文摘In this paper, reflection and transmission coefficients of regular waves from/through perforated thin walls are investigated. Small scale laboratory tests have been performed in a wave flume firstly with single perforated thin Plexiglas plates of various porosities. The plate is placed perpendicular to the flume with the height from the flume bottom to the position above water surface. With this thin wall in the flume wave overtopping is prohibited and incident waves are able to transmit. The porosities of the walls are achieved by perforating the plates with circular holes.Model settings with double perforated walls parallel to each other forming so called chamber system, have been also examined. Several parameters have been used for correlating the laboratory tests' results. Experimental data are also compared with results from the numerical model by applying the multi-domain boundary element method(MDBEM)with linear wave theory. Wave energy dissipation due to the perforations of the thin wall has been represented by a simple yet effective porosity parameter in the model. | 4 | Redox active polymer metal chelates for use in flexible symmetrical supercapacitors:Cobalt-containing poly(acrylic acid)polymer electrolytes显示文摘The novel polymer metal chelate electrolytes(polychelates)were prepared by incorporation of cobalt sulfate(Co)into poly(acrylic acid)(PAA)host matrix.Quasi-solid state supercapacitor devices were fabricated using polychelates,PAA-Co X(X:3,5,7,and 10)where X represents the doping fraction(w/w)of Co in PAA.All polymer metal electrolytes were showed excellent bending-stretching properties,thermal stability and electrochemical durability with an optimum ionic conductivity of 3.15×10^(-4) S cm^(-1).Hierarchically porous activated carbon and nano-sized conductive carbon were used to form carbon composite symmetrical device electrodes.The electric double-layer capacitor(EDLC)and redox reactions of Co-incorporated polychelates at the interfaces of porous activated carbon provided an optimum specific capacitance of 341.33 F g^(-1) with a device of PAA-Co7,which is at least 15 times enhancement compared to the device of pristine PAA.The PAA-Co7 device also provided energy density of 21.25 Wh kg^(-1) at a power density of 117.69 W kg^(-1).A prolonged cyclic stability of the device exhibited superior capacitive performance after 10,000 charge-discharge cycles and the maintained 90%of its initial performance.In addition,the supercapacitor with a dimension of 1.5 cm×3 cm containing PAA-Co7 successfully operated the red-blue-green(RGB)LED light. | Emre Cevik Ayhan Bozkurt | 2021 | Journal of Energy Chemistry2021，30，4： | 3 |

| 6 | Colorectal cancer: The epigenetic role of microbiome显示文摘Colorectal cancer(CRC)is the third most common cancer in men(746000 cases per year)and the second most common cancer in women globally(614000 cases per year).The incidence rate of CRC in developed countries(737000 cases per year)is higher than that in less developed countries(624000 cases per year).CRC can arise from genetic causes such as chromosomal instability and microsatellite instability.Several etiologic factors underlie CRC including age,diet,and lifestyle.Gut microbiota represent a proven cause of the disease,where they play pivotal roles in modulating and reshaping the host epigenome.Several active microbial metabolites have been found to drive carcinogenesis,invasion,and metastasis via modifying both the methylation landscape along with histone structure in intestinal cells.Gut microbiota,in response to diet,can exert both beneficial and harmful functions in humans,according to the intestinal balance of number and types of these bacteria.Although the intestinal microbial community is diverse among individuals,these microbes cumulatively produce 100-fold more proteins than the human genome itself,which calls for further studies to elaborate on the complicated interaction between these microorganisms and intestinal cells.Therefore,understanding the exact role that gut microbiota play in inducing CRC will help attain reliable strategies to precisely diagnose and treat this fatal disease. | Hussein Sabit Emre Cevik Huseyin Tombuloglu | 2019 | World Journal of Clinical Cases2019，7，22： | 3 |

| 8 | Investor sentiments and stock marketsduring the COVID-19 pandemic显示文摘This study examines the relationship between positive and negative investor sentiments and stock market returns and volatility in Group of 20 countries using variousmethods, including panel regression with fixed effects, panel quantile regressions, apanel vector autoregression (PVAR) model, and country-specific regressions. We proxyfor negative and positive investor sentiments using the Google Search Volume Indexfor terms related to the coronavirus disease (COVID-19) and COVID-19 vaccine, respectively. Using weekly data from March 2020 to May 2021, we document significantrelationships between positive and negative investor sentiments and stock marketreturns and volatility. Specifically, an increase in positive investor sentiment leads toan increase in stock returns while negative investor sentiment decreases stock returnsat lower quantiles. The effect of investor sentiment on volatility is consistent acrossthe distribution: negative sentiment increases volatility, whereas positive sentimentreduces volatility. These results are robust as they are corroborated by Granger causalitytests and a PVAR model. | 12 | Apoptotic cell death and its relationship to gastric carcinogenesis显示文摘AIM: To investigate the apoptotic process of cells with in the intestinal metaplasia areas co-localizing with chronic gastritis and gastric carcinomas and to analyze the involvement of proteins regulating apoptosis in the process of intestinal metaplasia related gastric carcinogenesis. METHODS: Forty-two gastric carcinoma and seventeen chronic gastritis cases were included in this study. All cases were examined for the existence of intestinal metaplasia. Ten cases randomly selected from each group were processed for TUNEL assay. TUNEL positive cells within the intestinal metaplasia areas, co-localizing either to gastric carcinoma or chronic gastritis, were counted and converted to apoptotic indices. In addition, p53, bcl-2 and bax expression patterns within these tissues were analyzed on the basis of immunohistochemistry. RESULTS: Twenty-eight of the cases were intestinal and 14 of the cases were diffuse type adenocarcinomas. 64% (27/42) of the gastric carcinoma cases had intestinal metaplasia. Intestinal metaplasia co-localized more with intestinal type carcinomas compared with diffuse type carcinomas [75% (21/28) vs 42% (6/14), respectively; P ≤0.05]. The mean apoptotic index in tumor cells was 0.70±0.08. The mean apoptotic index in intestinal metaplasias co-localizing to tumors was significantly higher than that of intestinal metaplasias co-localizing to chronic gastritis (0.70±0.03 vs 0.09±0.01, respectively; P≤0.05). p53 positivity was not observed in areas of intestinal metaplasia adjacent to tumors or chronic gastritis. Intestinal metaplasia areas adjacent to tumors showed lower cytoplasmic bcl-2 positivity compared to intestinal metaplasia areas adjacent to chronic gastritis [55.5% (15/27) vs 70.5% (12/17), respectively]. On the other hand, intestinal metaplasia areas adjacent to tumors showed significantly higher cytoplasmic bax positivity compared to intestinal metaplasia areas adjacent to chronic gastritis [44.4% (12/27) vs 11.7% (2/17), respectively; P ≤0.05]. CONCLUSION: Existence of apoptotic cells on the basis of TUNEL positivity is shown in intestinal metaplasias co-localizing to both diffuse and intestinal type gastric cancers in this study. Our results also suggested bax expression dependent induction of apoptosis especially in intestinal metaplasia areas adjacent to tumors. These findings strongly support the involvement of apoptotic mechanisms in the process of gastric carcinogenesis especially in the transition from intestinal metaplasia to gastric cancer.
