1Prolonged high-fat-diet feeding promotes non-alcoholic fatty liver disease and alters gut microbiota in mice显示文摘BACKGROUND Non-alcoholic fatty liver disease (NAFLD) has become an epidemic largely due to the worldwide increase in obesity. While lifestyle modifications and pharmacotherapies have been used to alleviate NAFLD, successful treatment options are limited. One of the main barriers to finding safe and effective drugs for long-term use in NAFLD is the fast initiation and progression of disease in the available preclinical models. Therefore, we are in need of preclinical models that (1) mimic the human manifestation of NAFLD and (2) have a longer progression time to allow for the design of superior treatments. AIM To characterize a model of prolonged high-fat diet (HFD) feeding for investigation of the long-term progression of NAFLD. METHODS In this study, we utilized prolonged HFD feeding to examine NAFLD features in C57BL/6 male mice. We fed mice with a HFD (60% fat, 20% protein, and 20% carbohydrate) for 80 wk to promote obesity (Old-HFD group, n = 18). A low-fat diet (LFD)(14% fat, 32% protein, and 54% carbohydrate) was administered for the same duration to age-matched mice (Old-LFD group, n = 15). An additional group of mice was maintained on the LFD (Young-LFD, n = 20) for a shorter duration (6 wk) to distinguish between age-dependent and age-independent effects. Liver, colon, adipose tissue, and feces were collected for histological and molecular assessments.RESULTS Prolonged HFD feeding led to obesity and insulin resistance. Histological analysis in the liver of HFD mice demonstrated steatosis, cell injury, portal and lobular inflammation and fibrosis. In addition, molecular analysis for markers of endoplasmic reticulum stress established that the liver tissue of HFD mice have increased phosphorylated Jnk and CHOP. Lastly, we evaluated the gut microbial composition of Old-LFD and Old-HFD. We observed that prolonged HFD feeding in mice increased the relative abundance of the Firmicutes phylum. At the genus level, we observed a significant increase in the abundance of Adercreutzia, Coprococcus, Dorea, and Ruminococcus and decreased relative abundance of Turicibacter and Anaeroplasma in HFD mice. 2Do ankle braces provide similar effects on ankle biomechanical variables in subjects with and without chronic ankle instability during landing?显示文摘Purpose:The purpose of this study was to examine effects of a sport version of a semi-rigid ankle brace(ElementTM) and a soft ankle brace (ASO) on ankle biomechanics and ground reaction forces(GRFs) during a drop landing activity in subjects with chronic ankle instability(CAD compared to healthy subjects with no history of CAI. Methods:Ten healthy subjects and 10 subjects who had multiple ankle sprains participated in the study as the control and unstable subjects, respectively.The CAI subjects were age,body mass index and gender matched with the control subjects.The arch index and ankle functions of the subjects were measured in a subject screening session.During the biomechanical test session,participants performed five trials of drop landing from 0.6 m,wearing no brace(NB).Element? brace and ASO brace.Simultaneous recording of three-dimensional kinematic(240 Hz) and GRF(1200 Hz) data were performed. Results:The CAI subjects had lower ankle functional survey scores.The arch index and deformity results showed greater arch deformity of ElementTM against a static load than in NB and ASO due to greater initial arch position held by the brace.CAI participants had greater eversion velocity than healthy controls.The ASO brace reduced the first peak vertical GRF whereas ElementTM increased 2nd peak vertical GRF. ElementTMbrace reduced eversion range of motion(ROM) and peak eversion velocity compared to NB and ASO.In addition,ElementTM reduced dorsiflexion ROM and increased peak plantarflexion moment compared to NB and ASO. 6A tailored,electronic textile conformable suit for large-scale spatiotemporal physiological sensing in vivo显示文摘The rapid advancement of electronic devices and fabrication technologies has further promoted the field of wearables and smart textiles.However,most of the current efforts in textile electronics focus on a single modality and cover a small area.Here,we have developed a tailored,electronic textile conformable suit(E-TeCS)to perform large-scale,multimodal physiological(temperature,heart rate,and respiration)sensing in vivo.This platform can be customized for various forms,sizes and functions using standard,accessible and high-throughput textile manufacturing and garment patterning techniques.Similar to a compression shirt,the soft and stretchable nature of the tailored E-TeCS allows intimate contact between electronics and the skin with a pressure value of around~25 mmHg,allowing for physical comfort and improved precision of sensor readings on skin.The E-TeCS can detect skin temperature with an accuracy of 0.1℃ and a precision of 0.01℃,as well as heart rate and respiration with a precision of 0.0012 m/s^(2) through mechano-acoustic inertial sensing.The knit textile electronics can be stretched up to 30%under 1000 cycles of stretching without significant degradation in mechanical and electrical performance.Experimental and theoretical investigations are conducted for each sensor modality along with performing the robustness of sensor-interconnects,washability,and breathability of the suit.Collective results suggest that our E-TeCS can simultaneously and wirelessly monitor 30 skin temperature nodes across the human body over an area of 1500 cm2,during seismocardiac events and respiration,as well as physical activity through inertial dynamics.Irmandy Wicaksono Carson ITucker Tao Sun Cesar A.Guerrero Clare Liu Wesley M.Woo Eric J.Pence Canan Dagdeviren 2020npj Flexible Electronics2020,4,1:4
7Ron receptor-dependent gene regulation in a mouse model of endotoxin-induced acute liver failure显示文摘BACKGROUND:Prior experimentation has shown that loss of the tyrosine kinase(TK) signaling domain of the Ron receptor leads to marked hepatocyte protection in a model of lipopolysaccharideinduced acute liver failure(ALF) in D-galactosamine(GalN)sensitized mice.The aim of this study was to identify the role of Ron in the regulation of hepatic gene expression.METHODS:Microarray analyses were performed on liver RNA isolated sequentially from wild-type(WT) and TK-/mice during the progression of ALF.Gene array data were validated using Western and immunohistochemistry analyses as well as with ex vivo culture systems.RESULTS:At baseline,101 genes were differentially expressed between WT and TK-/-livers,which regulate processes involved in hypoxia,proliferation,apoptosis and metabolism.One hour after ALF induction,WT livers exhibited increased cytokine expression compared to TK-/-livers,and after 4 hours,an induction of suppressor of cytokine signaling(SOCS) genes as well as JAK-STAT pathway activation were prominent in TK-/livers compared to controls.CONCLUSION:Our studies suggest a novel hepato-protective mechanism in Ron TK-/-mice wherein increased and sustained SOCS production and JAK-STAT activation in the hepatocyte may inhibit the destructive proinflammatory milieu and promote survival factors which blunt hepatic death and the ensuing development of ALF.Rishikesh M Kulkarni Louis W Kutcher William D Stuart Daniel J Carson Mike A Leonis Susan E Waltz 2012Hepatobiliary & Pancreatic Diseases International2012,11,4:3
8G-Quadruplex(G4) Motifs in the Maize(Zea mays L.) Genome Are Enriched at Specific Locations in Thousands of Genes Coupled to Energy Status,Hypoxia,Low Sugar,and Nutrient Deprivation显示文摘The G-quadruplex(G4) elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix.This G4 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA.The reversibility and structural diversity of G4 s make them highly versatile genetic structures,as demonstrated by their roles in various functions including telomere metabolism,genome maintenance,immunoglobulin gene diversification,transcription,and translation.Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci.To investigate their potential roles in a large-genome model plant species,we computationally identified 149,988 non-telomeric G4 motifs in maize(Zea mays L.,B73 AGPv2),29%of which were in non-repetitive genomic regions.G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand,one in the 5'UTR and the other at the 5'end of the first intron.Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro.The G4 motifs were prevalent in key regulatory genes associated with hypoxia(group VII ERFs),oxidative stress(DJ-1/GATase1),and energy status(AMPK/SnRK) pathways.They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis,sugar degradation,inositol metabolism,and base excision repair.Collectively,the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses.Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize,a model genetic system and major world crop species for feed,food,and fuel.Carson M.Andorf Mykhailo Kopylov Drena Dobbs Karen E.Koch M.Elizabeth Stroupe Carolyn J.Lawrence Hank W.Bass 2014Journal of Genetics and Genomics2014,41,12:2

10小肾肿瘤的冷冻消融治疗显示文摘目的:回顾分析小肾肿瘤冷冻消融治疗的临床资料并总结经验。方法:2004年8月~2010年11月行腹腔镜下(腹腔镜组,n=7)或经皮穿刺(经皮组,n=24)冷冻消融治疗小肾肿瘤患者30例(31个肿瘤)。31个肿瘤中7个位于肾上极,12个位于肾中极,12个位于肾下极。冷冻治疗前均行肿瘤穿刺活检病理检查,术后定期随访。结果:平均手术时间腹腔镜组(142.1±32.9)min明显长于经皮组(103.8±29.9)min,差异有统计学意义(P=0.009)。中位出血量腹腔镜组为55ml明显多于经皮组的5ml,差异有统计学意义(P<0.001)。病理结果示19个(61.3%)肾细胞癌,4个(12.9%)嗜酸细胞瘤,5个(16.1%)良性肿瘤,3个(9.7%)穿刺标本中未见肿瘤组织。随访3~60(19.2±16.1)个月,术前平均血肌酐浓度141.5μmol/L与术后3个月复查浓度176.8μmol/L差异无统计学意义意义(P=0.212)。1例(3.2%)患者术后出现肉眼血尿,1例(3.2%)肾细胞癌患者术后6个月复发。结论:冷冻消融是治疗小肾肿瘤安全有效的方法,适用于高危患者。顾晓 WONG Carson 2012临床泌尿外科杂志2012,27,11:2

14Levels and correlates of 24-hour movement behaviors among South Koreans:Results from the Korea National Health and Nutrition Examination Surveys,2014 and 2015显示文摘Purpose.This study aimed to examine the levels and correlates of 24-h movement behaviors(i.e.,physical activity,sedentary time,and sleep),and different patterns of these behaviors in a nationally representative sample of South Koreans aged 12 years and older.This study also aimed to examine the sociodemographic correlates of 24?h movement behaviors stratified by age groups.Methods.Self-reported,repeated cross-sectional data from 10,708 participants in the 2014 and 2015 Korea National Health and Nutrition Examination surveys were used.Key variables included moderate-to-vigorous intensity physical activity,muscular strengthening exercises,walking,active transportation,sedentary time,and sleep.Sociodemographic variables included age,sex,household income,area of residence,and education level.Descriptive statistics by sex and age as well as general linear models by age group were performed.Results.The proportions of individuals meeting the moderate-to-vigorous intensity physical activity,muscular strengthening exercises,and sleep guidelines were,respectively,21.6%,22.1%,and 32.5%in male youth;6.9%,4.5%,and 22.8%in female youth;55.5%,30.8%,and 54.0%in male adults;48.8%,14.4%,and 57.6%in female adults;44.0%,30.6%,and 45.5%in male older adults;and 29.5%,8.9%,and 37.3%in female older adults.The proportions of individuals showing the most ideal combinations of 24-h movement behaviors were only 3.2%in youth,0.4%in adults,and 0 in older adults.Universally,older age,female sex,or living in metro Seoul were associated with unfavorable patterns of 24-h movement behaviors across different age groups.However,the associations of income and education with movement behaviors were mixed across age groups.Conclusion.Overall,the proportion of South Koreans with a healthy 24-h movement behavior pattern is low.The sociodemographic correlates of different types of 24-h movement behaviors should be considered when designing targeted interventions for the promotion of healthy active living for South Koreans.Eun-Young,Lee Valerie Carson Justin Y.Jeon John C.Spence Mark S.Tremblay 2019Journal of Sport and Health Science2019,8,4:2
