| 6 | Study protocol of the Asian XELIRI ProjecT(AXEPT):a multinational,randomized,non-inferiority,phase Ⅲ trial of second-line chemotherapy for metastatic colorectal cancer, comparing the eicacy and safety of XELIRI with or without bevacizumab versus FOLFIRI w显示文摘Background: Capecitabine and irinotecan combination therapy(XELIRI) has been examined at various dose levels to treat metastatic colorectal cancer(m CRC). Recently, in the Association of Medical Oncology of the German Cancer Society(AIO) 0604 trial, tri?weekly XELIRI plus bevacizumab, with reduced doses of irinotecan(200 mg/m^2 on day 1) and capecitabine(1600 mg/m^2 on days 1–14), repeated every 3 weeks, has shown favorable tolerability and eicacy which were comparable to those of capecitabine and oxaliplatin(XELOX) plus bevacizumab. The doses of capecit?abine and irinotecan in the AIO trial are considered optimal. In a phase I/II study, XELIRI plus bevacizumab(BIX) as second?line chemotherapy was well tolerated and had promising eicacy in Japanese patients.Methods: The Asian XELIRI Projec T(AXEPT) is an East Asian collaborative, open?labelled, randomized, phase Ⅲ clinical trial which was designed to demonstrate the non?inferiority of XELIRI with or without bevacizumab versus standard FOLFIRI(5?fluorouracil, leucovorin, and irinotecan combination) with or without bevacizumab as second?line chemo?therapy for patients with m CRC. Patients with 20 years of age or older, histologically conirmed m CRC, Eastern Coop?erative Oncology Group performance status 0–2, adequate organ function, and disease progression or intolerance of the irst?line regimen will be eligible. Patients will be randomized(1:1) to receive standard FOLFIRI with or with?out bevacizumab(5 mg/kg on day 1), repeated every 2 weeks(FOLIRI arm) or XELIRI with or without bevacizumab(7.5 mg/kg on day 1), repeated every 3 weeks(XELIRI arm). A total of 464 events were estimated as necessary to show non?inferiority with a power of 80% at a one?sided α of 0.025, requiring a target sample size of 600 patients. | 7 | Health behaviors of Korean adults with hepatitis B:Findings of the 2016 Korean National Health and Nutrition Examination Survey显示文摘AIM To assess the frequencies of five health-related behaviors(smoking, alcohol consumption, body weight, sleep dura-tion, and physical activity) in Korean adults with chronic hepatitis B.METHODS Data were obtained from the 2016 Korean National Health and Nutrition Examination Survey. In total, 5887 subjects(2568 males, 3319 females) over 19 years old were enrolled in this study. Interviews were performed to obtain information on demographic characteristics and medical conditions. A self-administered questionnaire and medical examination were used to assess the smoking history, alcohol use, physical activity, sleep duration, and body weight of the subjects. Chronic hepatitis B was diagnosed based on detection of hepatitis B surface antigen(HBs Ag). The subjects were categorized into HBs Ag positive and negative groups, and a complex sampling analysis was conducted to compare the health behaviors between these groups.RESULTS Among males, the current smoking rate in the HBs Ag positive group was higher than that in the negative group(45.5% vs 38.5%). In the positive group, the rates of monthly and high-risk alcohol use were 70.4% and 17.6% in males and 45.9% and 3.8% in females, respectively. The rate of alcohol use was similar between the two groups [P = 0.455(males) and P = 0.476(females)]. In the HBs Ag positive group, 32.3% and 49.9% of males and 26.5% and 49.6% of females were overweight and physically inactive, respectively. High-risk alcohol consumption and physical inactivity were significantly associated with self-perceived health status.CONCLUSION Our data demonstrate that a large proportion of Korean adults with chronic hepatitis B have poor health behaviors. | 9 | Cyanidin suppresses amyloid beta-induced neurotoxicity by inhibiting reactive oxygen speciesmediated DNA damage and apoptosis in PC12 cells显示文摘Amyloid beta(Aβ)-induced oxidative stress is a major pathologic hallmark of Alzheimer's disease. Cyanidin, a natural flavonoid compound, is neuroprotective against oxidative damage-mediated degeneration. However, its molecular mechanism remains unclear. Here, we investigated the effects of cyanidin pretreatment against Aβ-induced neurotoxicity in PC12 cells, and explored the underlying mechanisms. Cyanidin pretreatment significantly attenuated Aβ-induced cell mortality and morphological changes in PC12 cells. Mechanistically, cyanidin effectively blocked apoptosis induced by Aβ, by restoring the mitochondrial membrane potential via upregulation of Bcl-2 protein expression. Moreover, cyanidin markedly protected PC12 cells from Aβ-induced DNA damage by blocking reactive oxide species and superoxide accumulation. These results provide evidence that cyanidin suppresses Aβ-induced cytotoxicity, by preventing oxidative damage mediated by reactive oxide species, which in turn inhibits mitochondrial apoptosis. | 10 | Laser micromachining of CNT/Fe/Al_2O_3 nanocomposites显示文摘CNT/Fe/Al2O3 mixed powders were synthesized from Fe/Al2O3 nanopowders using thermal CVD for the homogeneous dispersion of carbon nanotubes CNTs. CNTs consisted of MWNT, and the diameter was approximately 20-30 nm. After sintering, CNTs were homogenously located throughout Al2O3 grain boundary and were buckled. A femto-second laser installed with special optical systems was used for micromachining of the nanocomposites. The relationship between material ablation rate and energy fluence was theoretically investigated and compared with experimental results from cross-sectional SEM analysis. | 11 | Zircon U–Pb geochronology and Hf isotope geochemistry of magmatic and metamorphic rocks from the Hida Belt, southwest Japan显示文摘Zircon U–Pb and Hf isotope data integrated in this study for magmatic and metamorphic rocks fromthe Hida Belt,southwest Japan,lead to a new understanding of the evolution of the Cordilleran arc system along the ancestral margins of present-day Northeast Asia.Ion microprobe data for magmatic zircon domains from eight mafic tointermediate orthogneisses in the Tateyama and Tsunogawa areas yielded weighted mean 206Pb/238U ages spanning the entire Permian period(302–254 Ma).Under cathodoluminescence,primary magmatic growth zones in the zircon crystals were observed to be partially or completely replaced by inward-penetrating,irregularly curved featureless or weakly zoned secondary domains that mostly yielded U–Pb ages of 250–240 Ma and relatively high Th/U ratios(>0.2).These secondary domains are considered to have been formed by solid-state recrystallization during thermal overprints associated with intrusions of Hida granitoids.Available whole-rock geochemical and Sr–Nd isotope data as well as zircon age spectra corroborate that the Hida Belt comprises the Paleozoic–Mesozoic Cordilleran arc system built upon the margin of the North China Craton,together with the YeongnamMassif in southern Korea.The arcmagmatismalong this systemwas commenced in the Carboniferous and culminated in the Permian–Triassic transition period.Highly positiveεHf(t)values(>+12)of late Carboniferous to early Permian detrital zircons in the Hida paragneisses indicate that there was significant input from the depleted asthenospheric mantle and/or its crustal derivatives in the early stage of arc magmatism.On the other hand,near-chondriticεHf(t)values(+5 to−2)of magmatic zircons from late Permian Hida orthogneisses suggest a lithospheric mantle origin.Hf isotopic differences between magmatic zircon cores and the secondary rims observed in some orthogneiss samples clearly indicate that the zircons were chemically open to fluids or melts during thermal overprints.Resumed highly positive zirconεHf(t)values(>+9)shared by Early Jurassic granitoids in the Hida Belt and Yeongnam Massif may reflect reworking of the Paleozoic arc crust. | Deung-Lyong Cho Tae-Ho Lee Yutaka Takahashi Takenori Kato Keewook Yi Shinae Lee Albert Chang-sik Cheong | 2021 | Geoscience Frontiers2021,12,4: | 2 |
