3Identification and characterization of rice blast resistance gene Pid4 by a combination of transcriptomic profiling and genome analysis显示文摘Map-based cloning of plant disease resistance (R) genes is time-consuming. Here, we reported the isolation of blast R gene Pid4 using comparative transcriptomic profiling and genome-wide sequence analysis. Pid4 encodes a coiled-coil nucleotide-binding site leucine-rich repeat(CC-NBS-LRR) protein and is constitutively expressed at diverse developmental stages in the rice variety Digu. The Pid4 protein is localized in both the nucleus and cytoplasm. Introduction of Pid4 into susceptible rice cultivars confers race-specific resistance to leaf and neck blast. Amino acid sequence comparison and blast resistance spectrum tests showed that Pid4 is a novel R gene, different from the previously reported R genes located in the same gene cluster. A Pid4 Indel marker was developed to facilitate the identification of Pid4 in different rice varieties. 6The Classical and Regulatory Functions of C1q in Immunity and Autoimmunity显示文摘A classical function of C1q is to bind immune complexes and initiate complement activation producing membrane lytic complexes,opsonins and anaphylatoxins. This classical pathway of complement activation is also elicited when C1q binds some other ligands. Besides complement activation,C1q also regulates cell differentiation,adhesion,migration,activation and survival. C1q deficiency is associated with autoimmunity as well as increased susceptibility to infections. 8Heat shock protein 47 promotes tumor survival and therapy resistance by modulating AKT signaling via PHLPP1 in colorectal cancer显示文摘Objective:Heat shock protein 47(HSP47)is a collagen-specific molecular chaperone that facilitates collagen maturation.Its role in cancer remains largely unknown.In this study,we investigated the roles o f HSP47 in colorectal cancer(CRC)and therapy resistance.Methods:Expression o f HSP47 in CRC tissues was examined(1)in paired human CRC/adjacent normal tissues,using real time quantitative reverse transcription polymerase chain reaction(qRT-PCR),The Cancer Genome Atlas(TCGA)database,and 22 independent m icroarray databases(curated CRC).In vitro studies on several CRC cell lines(H C T 116,RKO and C C L228)with modulated HSP47 expression were conducted to assess cell viability and apoptosis(TU N EL assay and caspase-3/-7)during exposure to chemotherapy.AKT signaling and co-imm unoprecipitation studies were performed to examine HSP47 and PHLPP1 interaction.In vivo studies using tumor xenografts were conducted to assess the effects of HSP47 modulation on tum or growth and therapy response.Results:HSP47 was upregulated in CRC and was associated with poor prognosis in individuals with CRC.In vitro,HSP47 overexpression supported the survival of CRC cells,whereas its knockdown sensitized cells to 5-fluorouracil(5-FU).HSP47 promoted survival by inhibiting apoptosis,enhancing AKT phosphorylation,and decreasing expression of the AKT-specific phosphatase PHLPP1 when cells were exposed to chemotherapy.These effects were partly results of the interaction between HSP47 and PHLPP1,which decreased PHLPP1 stability and led to more persistent AKT activity.In vivoy HSP47 supported tumor growth despite 5-FU treatment.Conclusions:HSP47 supports the growth of CRC tumors and suppresses the efficacy of chemotherapy via modulation of AKT signaling.Yijye Chern Peter Zhang Hyelim Ju Isabella T.Tai 2020Cancer Biology & Medicine2020,17,2:3

11Efficient solvent extraction of antioxidant-rich extract from a tropical diatom,Chaetoceros calcitrans (Paulsen) Takano 1968显示文摘Objective: To compare the in vitro antioxidant capacity of a diatom, Chaetoceros calcitrans(C. calcitrans) extracted using six types of solvents.Methods: Each extract was evaluated in terms of extraction yield, total carotenoid,fucoxanthin content, total phenolic and antioxidant capacities(DPPH and ABTS +scavenging activity and iron chelating activity).Results: The methanol extract exhibited the highest yield [(22.71 ± 0.96) g/100 g dry weight(DW)], total carotenoid [(4.46 ± 0.36) mg/g DW], total phenolic [(2.49 ± 0.08) mg gallic acid equivalents/g DW] and second highest fucoxanthin content [(2.08 ± 0.03) mg fucoxanthin/g DW] as compared to other solvent extracts. Methanolic extract also exhibited significantly higher(P < 0.05) scavenging(DPPH, ABTS +) and iron chelating activities.Conclusions: Methanol was the recommended solvent for the production of antioxidant rich extract from C. calcitrans. Both carotenoids and phenolic acids were found to be positively correlated to the antioxidant capacities of C. calcitrans. 16Mesuaferrea stem bark extract induces apoptosis and inhibits metastasis in human colorectal carcinoma HCT 116 cells, through modulation of multiple cell signalling pathways显示文摘Considering the great potential of natural products as anticancer agents, the present study was designed to explore the molecular mechanisms responsible for anticancer activities of Mesua ferrea stem bark extract against human colorectal carcinoma. Based on MTT assay results, bioactive sub-fraction(SF-3) was selected for further studies using HCT 116 cells. Repeated column chromatography resulted in isolation of less active α-amyrin from SF-3, which was identified and characterized by GC-MS and HPLC methods. α-amyrin and betulinic acid contents of SF-3 were measured by HPLC methods. Fluorescent assays revealed characteristic apoptotic features, including cell shrinkage, nuclear condensation, and marked decrease in mitochondrial membrane potential in SF-3 treated cells. In addition, increased levels of caspases-9 and-3/7 levels were also observed in SF-3 treated cells. SF-3 showed promising antimetastatic properties in multiple in vitro assays. Multi-pathway analysis revealed significant down-regulation of WNT, HIF-1α, and EGFR with simultaneous up-regulation of p53, Myc/Max, and TGF-β signalling pathways in SF-3 treated cells. In addition, promising growth inhibitory effects were observed in SF-3 treated HCT 116 tumour spheroids, which give a hint about in vivo antitumor efficacy of SF-3 phytoconstituents.
