1Spatiotemporal microRNA profile in peripheral nerve regeneration:miR-138 targets vimentin and inhibits Schwann cell migration and proliferation显示文摘While the peripheral nervous system has regenerative ability,restoration of sufficient function remains a challenge.Vimentin has been shown to be localized in axonal growth fronts and associated with nerve regeneration,including myelination,neuroplasticity,kinase signaling in nerve axoplasm,and cell migration;however,the mechanisms regulating its expression within Schwann cell(SC) remain unexplored.The aim of this study was to profile the spatial and temporal expression profile of micro RNA(mi RNA) in a regenerating rat sciatic nerve after transection,and explore the potential role of mi R-138-5 p targeting vimentin in SC proliferation and migration.A rat sciatic nerve transection model,utilizing a polyethylene nerve guide,was used to investigate mi RNA expression at 7,14,30,60,and 90 days during nerve regeneration.Relative levels of mi RNA expression were determined using microarray analysis and subsequently validated with quantitative real-time polymerase chain reaction.In vitro assays were conducted with cultured Schwann cells transfected with mi RNA mimics and assessed for migratory and proliferative potential.The top seven dysregulated mi RNAs reported in this study have been implicated in cell migration elsewhere,and GO and KEGG analyses predicted activities essential to wound healing.Transfection of one of these,mi RNA-138-5 p,into SCs reduced cell migration and proliferation.mi R-138-5 p has been shown to directly target vimentin in cancer cells,and the luciferase assay performed here in rat Schwann cells confirmed it.These results detail a role of mi R-138-5 p in rat peripheral nerve regeneration and expand on reports of it as an important regulator in the peripheral nervous system.Travis B.Sullivan Litchfield C.Robert Patrick A.Teebagy Shannon E.Morgan Evan W.Beatty Bryan J.Cicuto Peter K.Nowd Kimberly M.Rieger-Christ David J.Bryan 2018Neural Regeneration Research2018,13,7:6
2A tunable diode laser absorption spectrometer for formaldehyde atmospheric measurements validated by simulation chamber instrumentation显示文摘A tunable diode laser absorption spectrometer (TDLAS) for formaldehyde atmospheric measurements has been set up and validated through comparison experiments with a Fourier transform infrared spectrometer (FT-IR) in a simulation chamber.Formaldehyde was generated in situ in the chamber from reaction of ethene with ozone.Three HCHO ro-vibrational line intensities (at 2909.71,2912.09 and 2914.46 cm-1) possibly used by TDLAS were calibrated by FT-IR spectra simultaneously recorded in the 1600-3200 cm-1 domain during ethene ozonolysis,enabling the on-line deduction of the varying concentration for HCHO in formation.The experimental line intensities values inferred confirmed the calculated ones from the updated HITRAN database.In addition,the feasibility of stratospheric in situ HCHO measurements using the 2912.09 cm-1 line was demonstrated.The TDLAS performances were also assessed,leading to a 2σ detection limit of 88 ppt in volume mixing ratio with a response time of 60 sec at 30 Torr and 294 K for 112 m optical path.As part of this work,the room-temperature rate constant of this reaction and the HCHO formation yield were found to be in excellent agreement with the compiled literature data.V.Catoire F.Bernard Y.Mbarki A.Mellouki G.Eyglunent V.Daёle C.Robert 2012Journal of Environmental Sciences2012,24,1:4

4An integrated approach to site-specific management zone delineation显示文摘Dividing fields into a few relatively homogeneous management zones(MZs) is a practical and costeffective approach to precision agriculture. There are three basic approaches to MZ delineation using soil and/or landscape properties, yield information, and both sources of information. The objective of this study is to propose an integrated approach to delineating site-specific MZ using relative elevation, organic matter, slope, electrical conductivity, yield spatial trend map, and yield temporal stability map(ROSE-YSTTS) and evaluate it against two other approaches using only soil and landscape information(ROSE) or clustering multiple year yield maps(CMYYM). The study was carried out on two no-till corn-soybean rotation fields in eastern Illinois, USA. Two years of nitrogen(N) rate experiments were conducted in Field B to evaluate the delineated MZs for site-specific N management. It was found that in general the ROSE approach was least effective in accounting for crop yield variability(8.0%–9.8%), while the CMYYM approach was least effective in accounting for soil and landscape(8.9%–38.1%), and soil nutrient and pH variability(9.4%–14.5%). The integrated ROSE-YSTTS approach was reasonably effective in accounting for the three sources of variability(38.6%–48.9%, 16.1%–17.3% and 13.2%–18.7% for soil and landscape, nutrient and pH, and yield variability, respectively), being either the best or second best approach. It was also found that the ROSE-YSTTS approach was effective in defining zones with high,medium and low economically optimum N rates. It is concluded that the integrated ROSE-YSTTS approach combining soil, landscape and yield spatial-temporal variability information can overcome the weaknesses of approaches using only soil, landscape or yield information,and is more robust for MZ delineation. It also has the potential for site-specific N management for improved economic returns.
