In vivo theranostics with near-infraredemitting carbon dots—highly efficient photothermal therapy based on passive targeting after intravenous administration显示文摘Carbon dots that exhibit near-infrared fluorescence(NIR CDs)are considered emerging nanomaterials for advanced biomedical applications with low toxicity and superior photostability and targeting compared to currently used photoluminescence agents.Despite progress in the synthesis of NIR CDs,there remains a key obstacle to using them as an in vivo theranostic agent. This work demonstrates that the newly developed sulfur and nitrogen codoped NIR CDs are highly efficient in photothermal therapy(PTT)in mouse models(conversion efficiency of 59%)and can be readily visualized by photoluminescence and photoacoustic imaging. The real theranostic potential of NIR CDs is enhanced by their unique biodistribution and targeting.Contrary to all other nanomaterials that have been tested in biomedicine, they are excreted through the body’s renal filtration system. Moreover, after intravenous injection,NIR CDs are accumulated in tumor tissue via passive targeting, without any active species such as antibodies.Due to their accumulation in tumor tissue without the need for intratumor injection, high photothermal conversion, excellent optical and photoacoustic imaging performance, and renal excretion, the developed CDs are suitable for transfer to clinical biomedical practice. | Xin Bao Ye Yuan Jingqin Chen Bohan Zhang Di Li Ding Zhou Pengtao Jing Guiying Xu Yingli Wang Kateřina Holá Dezhen Shen Changfeng Wu Liang Song Chengbo Liu Radek Zbořil Songnan Qu | 2018 | Light(Science & Applications)2018,7,1: | 13
Altered chromatin recruitment by FOXA1 mutations promotes androgen independence and prostate cancer progression显示文摘Dear Editor,F0XA1,a forkhead(FKHD)family transcription factor,is highly expressed in the epithelium of endoderm-derived organs, including the prostate gland.1 Transgenic mouse studies have shown that F0XA1 expression is required for prostate epithelial cell differentiation and ductal morphogenesis during development and for the maintenance of this differentiated epithelial phenotype in the adult. Mechanistically, F0XA1 binds FKHD motifs in the DNA to open chromatin and in crease local accessibility. | Bohan Xu Bing Song Xiaodong Lu Jung Kim Ming Hu Jonathan C.Zhao Jindan Yu | 2019 | Cell Research2019,29,9: | 4
All in one theranostic nanoplatform enables efficient anti-tumor peptide I delivery for triple-modal imaging guided cancer therapy显示文摘Developing a reliable system to efficiently and safely deliver peptide drugs into tumor tissues still remains a great challenge since the instability of peptide drugs and low ability to traverse the cell membrane. Herein, we constructed a multifunctional nanoplatform based on porous europium/gadolinium (Eu/Gd)-doped NaLa(MoO4)2 nanoparticles (NLM NPs) to deliver antitumor peptide of B-cell lymphoma/leukemia-2-like protein 11 (BIM) for cancer therapy. The porous NLM NPs exhibited inherent photoluminescent, magnetic and X-ray absorbable properties, which enable them for triple-modal bioimaging, including fluorescence, magnetic resonance imaging (MRI) and computed tomography (CT). This triple-modal bioimaging can contribute to monitoring NLM NPs biodistribution and guiding therapy in vitro and in vivo. Furthermore, the NLM NPs showed negligible cytotoxicity in vitro and tissue toxicity in vivo. Importantly, NLM NPs could load the antitumor peptide of BIM and efficiently improve the resistance of peptide drugs to proteolysis. The BIM peptide was efficiently delivered into the tumor cells by NLM NPs, which can inhibit the growth and promote the apoptosis of cancer cells in vitro, significantly inhibit the tumor growth in vivo. Notably, NLM-BIM theranostic nanoplatform exhibits low systemic toxicity and fewer side effects in vivo. The NLM NPs can serve as a promising multifunctional peptide delivery nanoplatform for multi-modal bioimaging and cancer therapy. | Xiaoyan Qu Zhengqing Liu Bohan Ma Na Li Hongyang Zhao Tian Yang Yumeng Xue Xiaozhi Zhang Yongping Shao Ying Chang Jun Xu Bo Lei Yaping Du | 2019 | Nano Research2019,12,3: | 2
Signaling pathways in obesity:mechanisms and therapeutic interventions显示文摘Obesity is a complex, chronic disease and global public health challenge.Characterized by excessive fat accumulation in the body, obesity sharply increases the risk of several diseases, such as type 2 diabetes, cardiovascular disease, and nonalcoholic fatty liver disease, and is linked to lower life expectancy.Although lifestyle intervention(diet and exercise)has remarkable effects on weight management, achieving long-term success at weight loss is extremely challenging, and the prevalence of obesity continues to rise worldwide.Over the past decades, the pathophysiology of obesity has been extensively investigated, and an increasing number of signal transduction pathways have been implicated in obesity, making it possible to fight obesity in a more effective and precise way.In this review,we summarize recent advances in the pathogenesis of obesity from both experimental and clinical studies, focusing on signaling pathways and their roles in the regulation of food intake, glucose homeostasis, adipogenesis, thermogenesis, and chronic inflammation. We also discuss the current antiobesity drugs,as well as weight loss compounds in clinical trials, that target these signals. The evolving knowledge of signaling transduction may shed light on the future direction of obesity research,as we move into a new era of precision medicine. | Xue Wen Bohan Zhang Beiyi Wu aitao Xiao Zehua Li Ruoyu Li Xuewen Xu Tao Li | 2022 | Signal Transduction and Targeted Therapy2022,7,9: | 2

New genetic and epigenetic insights into the chemokine system: the latest discoveries aiding progression toward precision medicine显示文摘Over the past thirty years, the importance of chemokines and their seven-transmembrane G protein-coupled receptors(GPCRs)has been increasingly recognized.Chemokine interactions with receptors trigger signaling pathway activity to form a network fundamental to diverse immune processes, including host homeostasis and responses to disease.Genetic and nongenetic regulation of both the expression and structure of chemokines and receptors conveys chemokine functional heterogeneity.Imbalances and defects in the system contribute to the pathogenesis of a variety of diseases, including cancer, immune and inflammatory diseases, and metabolic and neurological disorders, which render the system a focus of studies aiming to discover therapies and important biomarkers. The integrated view of chemokine biology underpinning divergence and plasticity has provided insights into immune dysfunction in disease states, including, among others, coronavirus disease 2019(COVID-19).In this review,by reporting the latest advances in chemokine biology and results fromanalyses of a plethora of sequencing-based datasets,we outline recent advances in the understanding of the genetic variations and nongenetic heterogeneity of chemokines and receptors and provide an updated view of their contribution to the pathophysiological network, focusing on chemokine-mediated inflammation and cancer.Clarification of the molecular basis of dynamic chemokine-receptor interactions will help advance the understanding of chemokine biology to achieve precision medicine application in the clinic. | Hanli Xu Shuye Lin Ziyun Zhou Duoduo Li Xiting Zhang Muhan Yu Ruoyi Zhao Yiheng Wang Junru Qian Xinyi Li Bohan Li Chuhan Wei Keqiang Chen Teizo Yoshimura Ji Ming Wang Jiaqiang Huang | 2023 | Cellular & Molecular Immunology2023,20,7: | 0
Structural engineering of BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(X=Cl,Br,I)heterostructures towards ultrastable and tunable photoluminescence显示文摘Construction of lead halide perovskite nanocrystals(LHP NCs)heterostructures is essential to obtain highly stable photoluminescence and expand their applications.Herein,a novel self-assembly strategy combining with a solvent-free thermal-assisted synthesis and a water-triggered reaction is developed to subsequently grow BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(X=Cl,Br,I)heterostructures at low nucleation temperature with high crystallinity. The as-obtained ternary BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(X=Cl,Br,I)heterostructures exhibit remarkably enhanced panchromatic emission and ultrastable luminescence ascribing to the low-defect growth based on lattice matching.Stable white light-emitting diodes(WLEDs)have been constructed with a high correlated color temperature(CCT)of 7225 K and luminous efficiency of 74.4 lm·W-1.Ln^(3+)-doped BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(Ln^(3+)=Eu^(3+),Tb^(3+),Dy^(3+),Sm^(3+),Yb^(3+)/Er^(3+))nanocomposites are further designed with excitation-dependent photoluminescence and thermochromic properties, making them excellent candidates for high-level anti-counterfeiting and encryption. This work offers a green and universal approach in assembling CsPbX_(3)(X=Cl,Br,I)on lattice-matched tungstate with adjustable panchromatic emission for versatile optical applications. | Peng Liu Yan Xu Bohan Li Yuchi Zhang Hongzhou Lian Jun Lin | 2024 | Nano Research2024,17,3: | 0
Highly specific characterization and discrimination of monosodium urate crystals in gouty arthritis based on aggregation-induced emission luminogens显示文摘Existing technologies used to detect monosodium urate(MSU)crystals for gout diagnosis are not ideal due to their low sensitivity and complexity of operation. The purpose of this study was to explore whether aggregation-induced emission luminogens(AIEgens)can be used for highly specific imaging of MSU crystals to assist in the diagnosis of gout.First,we developed a series of luminogens(i.e.,tetraphenyl ethylene(TPE)-NH_(2),TPE-2NH_(2),TPE-4NH_(2),TPE-COOH,TPE-2COOH,TPE-4COOH,and TPE-Ketoalkyne),each of which was then evenly mixed with MSU crystals.Next, optimal fluorescence imaging of each of the luminogens was characterized by a confocal laser scanning microscope(CLSM).This approach was used for imaging standard samples of MSU,hydroxyapatite(HAP)crystals, and mixed samples with 1:1 mass ratio of MSU/HAP.We also imaged samples from mouse models of acute gouty arthritis,HAP deposition disease, and comorbidities of interest.Subsequently,CLSM imaging results were compared with those of compensated polarized light microscopy, and we assessed the biosafety of TPE-Ketoalkyne in the RAW264.7 cell line. Finally,CLSM time series and three-dimensional imaging were performed on MSU crystal samples from human gouty synovial fluid and tophi. As a promising candidate for MSU crystal labeling,TPE-Ketoalkyne was found to detect MSU crystals accurately and rapidly in standard samples, animal samples, and human samples, and could precisely distinguish gout from HAP deposition disease. This work demonstrates that TPE-Ketoalkyne is suitable for highly specific and timely imaging of MSU crystals in gouty arthritis and may facilitate future research on MSU crystal-related diseases. | Wenjuan Wang Guiquan Zhang Ziyi Chen Hanlin Xu Bohan Zhang Rong Hu Anjun Qin Yinghui Hua | 2023 | Bio-Design and Manufacturing2023,6,6: | 0
Low-temperature and flexible strategy to in-situ fabricate ZrSiO_(4)-based ceramic composites via doping and tuning solid-state reaction显示文摘Synthetic zircon(ZrSiO_(4))ceramics are typically fabricated at elevated temperatures(over 1500℃),which would lead to high manufacturing cost.Meanwhile, reports about preparing ZrSiO_(4)-based ceramic composites via controlling the solid-state reaction between zirconia(ZrO_(2))and silica(SiO_(2))are limited. In this work,we proposed a low-temperature strategy to flexibly design and fabricate ZrSiO_(4)-based ceramic composites via doping and tuning the solid-state reaction. Two ceramic composites and ZrSiO_(4) ceramics were in-situ prepared by reactive fast hot pressing(FHP)at approximately 1250℃ based on the proposed strategy,i.e.,a ZrSiO_(4)-SiO_(2) dual-phase composite with bicontinuous interpenetrating and hierarchical microstructures,a ZrSiO_(4)-ZrO_(2) dual-phase composite with a microstructure of ZrO_(2) submicron-and nano-particles embedded in a micron ZrSiO_(4) matrix, and ZrSiO_(4) ceramics with a small amount of residual ZrO_(2) nanoparticles. The results showed that the phase compositions, microstructure configurations, mechanical properties, and wear resistance of the materials can be flexibly regulated by the proposed strategy.Hence,ZrSiO_(4)-based ceramic composites with different properties can be easily fabricated based on different application scenarios. These findings would offer useful guidance for researchers to flexibly fabricate ZrSiO_(4)-based ceramic composites at low temperatures and tailor their microstructures and properties through doping and tuning the solid-state reaction. | Bohan Wang Le Fu Junjie Song Wenjun Yu Ying Deng Guofu Xu Jiwu Huang Wei Xia | 2023 | Journal of Advanced Ceramics2023,12,6: | 0
